To evaluate the effects of TCM treatment on gastric cancer patients, the authors performed a retrospective study to report the result of the integrated treatment of TCM with chemotherapy for stage IV non-surgical gastric cancer. METHODS: In this study, 182 patients with stage IV and non-surgical gastric cancer were retrospectively analyzed to evaluate the effects of TCM integrated with chemotherapy. Among the 182 cases, 88 cases received integrated therapy consisting of TCM and chemotherapy, while 94 cases received chemotherapy alone. The overall survival and Karnofsky performance status (KPS) score were measured as the main outcome. RESULTS: The median overall survival of the integrated therapy group and chemotherapy group were 16.9 and 10.5 months, respectively. The 1-, 3- and 5-year survival rates of integrated therapy group vs. chemotherapy group were 70% vs. 32%, 18% vs. 4%, and 11% vs. 0%, respectively. There was a significant difference between the two groups (x2 = 42.244, P 〈 0.001 ). After six-month treatment, KPS scores of the integrated therapy group and the chemotherapy group were 75.00 ± 14.78 and 60.64 ± 21.39, respectively (P 〈 0.001). The Cox regression analysis showed that TCM treatment is a protective factor for patients' overall survival. CONCLUSION: This study demonstrated that TCM integrated with chemotherapy may prolong overal survival and improve survival rate and life quality of patients with stage IV non-surgical gastric cancer展开更多

However, its potential mechanism has not yet been clearly addressed. This study aimed to predict targets and molecular mechanisms of ZYHT in treating GC by network pharmacology analysis and to explore the role of ZYHT in GC both in vitro and in vivo. Methods: Targets and molecular mechanisms of ZYHT were predicted via network pharmacology analysis. The effects of ZYHT on the expression of metastasis-associated targets were further validated by Western blot and quantitative real-time polymerase chain reaction. To explore the specific molecular mechanisms of the effects of ZYHT on migration and invasion, the runt-related transcription factor 3(RUNX3) gene was knocked out by clustered regularly interspaced short palindromic repeats/Cas9, and lentiviral vectors were transfected into SGC-7901 cells. Then lung metastasis model of GC in nude mice was established to explore the anti-metastasis effect of ZYHT. Western blot and immunohistochemistry were used to explore the impact of ZYHT on the expression of metastasis-related proteins with or without RUNX3 gene. Results: The network pharmacology analysis showed that ZYHT might inhibit focal adhesion, migration, invasion and metastasis of GC. ZYHT inhibited the proliferation, migration and invasion of GC cells in vitro via regulating the expression of metastasis-associated targets. Knocking out RUNX3 almost completely reversed the cell phenotypes(migration and invasion) and protein expression levels elicited by ZYHT.In vivo studies showed that ZYHT inhibited the metastasis of GC cells to the lung and prolonged the survival time of the nude mice. Knocking out RUNX3 partly reversed the metastasis of GC cells to the lung and the protein expression levels elicited by ZYHT.Conclusion: ZYHT can effectively inhibit the invasion and migration of GC in vitro and in vivo, and its molecular mechanism may relate to the upregulation of RUNX3 expression.展开更多

Based on the analysis of the pressure signal and the visual observation of the riser, it is shown that the additive of surfactant to the carrying liquid makes bubbles smaller in size but much larger in number in the upward intermittent flows. In addition, the additive of surfactant to a two-phase flow does not have a significant impact on the in-situ gas fraction, the pressure drop and the frequency of the liquid slug, but it reduces significantly the velocity of the liquid slug. When the superficial liquid velocity is set, an exponential relationship between the dimensionless velocity of the liquid slug and the Webber number can be obtained. These results might be used for estimating the characteristic parameters of the upward intermittent flow based upon the input operating conditions.展开更多

文摘Background:To systematically evaluate the efficacy and safety of traditional Chinese medicine in the treatment of refractory gastroesophageal reflux. Methods:PubMed,The Cochrane Library,Embase,Web of Science,China National Knowledge Infrastructure,Wanfang Database,China Science and Technology Journal Database and Chinese BioMedical Literature Database were searched for randomized controlled trials of traditional Chinese medicine in the treatment of refractory gastroesophageal reflux from database establishment time to December 2020.After two researchers independently screened the literature, extracted data and evaluated the bias risk included in the study,RevMan5.3 software was used for meta-analysis.Results:A total of 12 randomized controlled trials, were included, including 893 patients. The results of meta-analysis showed that the total effective rate of the treatment group(relative risk=1.28,95%confidence interval(CI)(1.19,1.38),P<0.00001),RGERDQ(refractory gastroesophageal reflux disease)score(mean difference(MD)=−3.35,95%CI(−4.13,−2.57),P<0.00001],acid reflux(acid in the stomach comes out of the mouth)[MD=−0.30,95%CI(−0.45,−0.15),P<0.00001],heartburn(the feeling that the heart is burned)(MD=−0.44,95%CI(−0.60,−0.29),P<0.00001,and retrosternal pain(MD=−0.27,95%CI(−0.44,−0.10),P<0.00001,belching(MD=−0.40,95%CI(−0.57,−0.24),P<0.00001],endoscopic mucosal score(MD=−0.62,95%CI(−0.78,−0.46),P<0.00001],the differences were statistically significant, and the effective rate of mucosal improvement under endoscopy was P=0.93>0.05,with no statistically significant difference. Conclusion:The current evidence shows that traditional Chinese medicine in the treatment of refractory gastroesophageal reflux disease is better than that of western medicine in the total effective rate, relieving acid reflux, heartburn, retrosternal pain and belching symptoms, but it is impossible to judge the improvement of mucosa under endoscope.Due to the limitations of the quality and quantity of included studies, more high-quality studies are needed to confirm the above conclusions.

文摘Additive manufacturing of aluminum alloys has received significant attention in the aerospace industry;however, achieving sufficient high strength, especially at elevated temperatures, remains challenging. Here,a crack-free and near-full dense Al-1Fe-0.6Cu-1.3Zr alloy was fabricated by the laser powder bed fusion(LPBF)technique. The Al-Fe-Cu-Zr alloy exhibits heterogeneous microstructures with two distinct zones. One is the so-called coarse-grain zones(CGZs)with an average grain size of 0.95μm,where(Al,Cu)Fe_(3) nanoparticles precipitate in the Al matrix and Fe and Cu cosegregate at the grain boundaries(GBs).The other is fine-grain zones(FGZs)with an average grain size of 0.45μm,where an Al 3 Zr nanoparti-cle precipitates in each of theα-Al grains(serves as the nuclei),and Fe-rich nanoprecipitates and Fe/Cu cosegregation appear at the GBs.As a result, the LPBF Al-Fe-Cu-Zr alloy, with these unique heteroge-neous structures, displays high strength at both room temperature and elevated temperatures,e.g.,with high yield strengths of 500 MPa at room temperature, and 163 MPa at 573 K,both are higher than those of additive manufactured Al-based alloys reported thus far.It is suggested that the high strength over a wide temperature range of the current LPBF Al alloy is mainly attributed to the combination of the precipitation strengthening mechanism and grain-boundary strengthening mechanism.

文摘OBJECTIVE: Traditional Chinese medicine (TCM) is regarded as an important treatment for gastric cancer patients, especially for those in advanced stage. CONCLUSION: This study demonstrated that TCM integrated with chemotherapy may prolong overal survival and improve survival rate and life quality of patients with stage IV non-surgical gastric cancer

文摘Objectives: Ziyin Huatan Recipe(ZYHT), a traditional Chinese medicine comprised of Lilii Bulbus, Pinelliae Rhizoma, and Hedyotis Diffusa, has shown promise in treating gastric cancer(GC). Knocking out RUNX3 partly reversed the metastasis of GC cells to the lung and the protein expression levels elicited by ZYHT.Conclusion: ZYHT can effectively inhibit the invasion and migration of GC in vitro and in vivo, and its molecular mechanism may relate to the upregulation of RUNX3 expression.

文摘The effect of the surfactant additive on the upward intermittent flows in a pipeline-riser system is studied experimentally, in a 3 m long horizontal pipe connected to a Perspex pipe of 2.0 m long and 25 mm in diameter, inclined to the horizontal plane by 7?, followed by the vertical PVC riser of 3.5 m high and 25 mm in diameter, operating at the atmospheric end pressure. These results might be used for estimating the characteristic parameters of the upward intermittent flow based upon the input operating conditions.

文摘Identification of cancer driver genes plays an important role in precision oncology research, which is helpful to understand cancer initiation and progression. However, most existing computational methods mainly used the protein–protein interaction(PPI)networks,or treated the directed gene regulatory networks(GRNs)as the undirected gene–gene association networks to identify the cancer driver genes, which will lose the unique structure regulatory information in the directed GRNs,and then affect the outcome of the cancer driver gene identification. Here, based on the multi-omics pan-cancer data(i.e.,gene expression, mutation, copy number variation, and DNA methylation),we propose a novel method(called DGMP)to identify cancer driver genes by jointing directed graph convolutional network(DGCN)and multilayer perceptron(MLP).DGMP learns the multi-omics features of genes as well as the topological structure features in GRN with the DGCN model and uses MLP to weigh more on gene features for mitigating the bias toward the graph topological features in the DGCN learning process. The results on three GRNs show that DGMP outperforms other existing state-of-the-art methods. The ablation experimental results on the Dawn Net network indicate that introducing MLP into DGCN can offset the performance degradation of DGCN,and jointing MLP and DGCN can effectively improve the performance of identifying cancer driver genes.DGMP can identify not only the highly mutated cancer driver genes but also the driver genes harboring other kinds of alterations(e.g.,differential expression and aberrant DNA methylation)or genes involved in GRNs with other cancer genes. The source code of DGMP can be freely downloaded from https://github.com/NWPU-903PR/DGMP.
