文摘Background:Non–small cell lung cancer(NSCLC)is a common malignant tumor with an increasing incidence.PIEZO-type mechanosensitive ion channel component 1(PIEZO1)is a mechanosensitive ion channel whose expression has been implicated in various cancers. However, its expression patterns, prognostic implications, and specific molecular mechanisms of action in NSCLC remain un-clear.This study aimed to characterize the expression profile of PIEZO1 in NSCLC in vitro and using bioinformatics analyses. Methods:To determine the expression profile of PIEZO1 in normal and cancerous human tissues and the mRNA expression of PIEZO1 in NSCLC,we analyzed data from The Cancer Genome Atlas.Various bioinformatics analyses were performed for correlation analyses, construction of survival curves, and identification of upstream mRNA targets and genes coexpressed with PIEZO1.Furthermore,the coexpressed genes were functionally annotated with Gene Ontology and subjected to pathway enrichment analyses.Additionally,we an-alyzed and compared PIEZO1 expression in normal and cancerous human tissue samples in vitro. Results:NSCLC tissue samples had lower PIEZO1 expression than adjacent tissues.PIEZO1 overexpression inhibited NSCLC progres-sion and cell migration and correlated with improved survival outcomes,as revealed by bioinformatics analyses.Additionally, analysis using the OncomiR database revealed that miR-942-5p upregulation contributed to the downregulation of PIEZO1 in NSCLC.Mechanistic in-vestigations suggested that PIEZO1 modulates NSCLC by regulating the mitogen-activated protein kinase pathway, particularly by targeting FLNA,RRAS,and MAP3K6.Bioinformatics analyses highlighted the potential of PIEZO1 as a therapeutic target and prognostic indicator of NSCLC.Conclusions:These novel findings reveal the expression profile of PIEZO1 in NSCLC and highlight the potential of PIEZO1 and miR-942-5p as diagnostic and therapeutic biomarkers of NSCLC and other cancer types.

文摘BACKGROUND The incidence of mixed neuroendocrine-non-neuroendocrine neoplasms(MiNEN)is low.To improve our understanding of this rare tumor type and optimally guide clinical treatment, associated risk factors, clinical manifestations, and prognosis must be explored.AIM To identify risk factors that influence the prognosis of patients with gastroenteropancreatic MiNEN(GEP-MiNEN).METHODS We retrospectively analyzed the clinical data of 46 patients who were diagnosed with GEP-MiNEN at the First Affiliated Hospital of Bengbu Medical College(Anhui,China)between January 2013 and December 2017.Risk factors influencing the prognosis of the patients were assessed using Kaplan-Meier curves and cox regression models. We compared the results with 55 randomly selected patients with gastroenteropancreatic GEP neuroendocrine tumors,47 with neuroendocrine carcinomas(NEC),and 58 with poorly differentiated adenocarcinoma.RESULTS Among the 46 patients with GEP-MiNEN,thirty-five had gastric tumors, nine had intestinal tumors(four in the small intestine and five in the colon and rectum),and two had pancreatic tumors. The median age of the patients was 66(41-84)years,and the male-to-female ratio was 2.83.Thirty-three(71.7%)patients had clinical stage III and IV cancers.Distant metastasis occurred in 14 patients,of which 13 had metastasis to the liver. The follow-up period was 11-72 mo,and the median overall survival was 30 mo.Ki-67 index≥50%,high proportion of NEC,lymph node involvement, distant metastasis, and higher clinical stage were independent risk factors affecting the prognosis of patients with GEP-MiNEN.The median overall survival was shorter for patients with NEC than for those with MiNEN(14 mo vs 30 mo,P=0.001),but did not significantly differ from those with poorly differentiated adenocarcinoma and MiNEN(30 mo vs 18 mo,P=0.453).CONCLUSION A poor prognosis is associated with rare, aggressive GEP-MiNEN.Ki-67 index, tumor composition, lymph node involvement, distant metastasis, and clinical stage are important factors for patient prognosis.

文摘E-jiao(Colla Corii Asini,CCA)has been widely used as a healthy food and Chinese medicine.Although authentic CCA is characterized by its typical sweet and neutral fragrance, its aroma components have been rarely investigated. This work investigated the aroma-active components and antioxidant activity of 19 CCAs from different geographical origins.CCA extracts obtained by simultaneous distillation and extraction were analyzed by gas chromatography-mass spectrometry(GC-MS),gas chromatography-olfactometry(GC-O)and sensory analysis. The antioxidant activity of CCAs was determined by ABTS and DPPH assays.A total of 65 volatile compounds were identified and quantified by GC-MS and 23 aroma-active compounds were identified by GC-O and aroma extract dilution analysis. The most powerful aroma-active compounds were identified based on the flavor dilution factor and their contents were compared among the 19 CCAs.Principal component analysis of the 23 aroma-active components showed 3 significant clusters.Canonical correlation analysis between antioxidant assays and the 23 aroma-active compounds indicates strong correlation(r=0.9776,p=0.0281).Analysis of aroma-active components shows potential for quality evaluation and discrimination of CCAs from different geographical origins.

文摘Background:Research on nutrition in early-life commonly focuses on the maturation of the intestine because the intestinal system is crucial for ensuring continued growth. To explore the importance of early nutrition regulation in animals, soy protein concentrate(SPC)was added to the early diet of broilers to investigate its effects on amino acid digestibility, intestinal development, especially intestinal microorganisms, and broiler metabolites.A total of 192 oneday-old Arbor Acres(AA)male broilers were randomly assigned to two experimental treatments with 8 replicates of 12 birds. The control group was fed a basal diet(control),and the treatment group was fed a basal diet supplemented with 12%SPC(SPC12)during the first 10 d(starter phase). From d 11 to 21(grower phase)and d 22 to 42(finisher phase),a basal diet was fed to both treatment groups. Results:SPC reduced the pH value and acid-binding capacity of the starter diet(P<0.05,d 10);SPC in the early diet enhanced the gizzard weight(P<0.05,d 10 and d 42)and the ileum weight(P<0.05,d 10)and decreased the weight and length of the jejunum(P<0.05,d 10)and the relative length of the duodenum and jejunum(P<0.05,d 10).At the same time,SPC enhanced villus height(P<0.05,d 10)and muscle thickness in the jejunum and ileum(P<0.05,d 10)and increased the number of goblet cells in the duodenum(P<0.05,d 10).Meanwhile,SPC increased the Chao1 index and the ACE index(P<0.05,d 10)and altered the composition of caecal microflora at d 10.SPC also increased the relative abundance of Alistipes,Anaerotruncus,Erysipelatoclostridium,Intestinimonas and Flavonifractor bacteria(P<0.05,d 10).At the same time, the concentrations of caecal butyric acid and total short-chain fatty acids(SCFAs)were also increased in the SPC12 group(P<0.05,d 10).Conclusions:In summary, the results showed that supplementing the starter diet of broilers with SPC has a significant effect on the early development of the intestine and the microflora.

文摘In this work,we employ the cache-enabled UAV to provide context information delivery to end devices that make timely and intelligent decisions.Different from the traditional network traffic, context information varies with time and brings in the ageconstrained requirement. The cached content items should be refreshed timely based on the age status to guarantee the freshness of user-received contents, which however consumes additional transmission resources. The traditional cache methods separate the caching and the transmitting, which are not suitable for the dynamic context information. We jointly design the cache replacing and content delivery based on both the user requests and the content dynamics to maximize the offloaded traffic from the ground network. The problem is formulated based on the Markov Decision Process(MDP).A sufficient condition of cache replacing is found in closed form, whereby a dynamic cache replacing and content delivery scheme is proposed based on the Deep Q-Network(DQN).Extensive simulations have been conducted.Compared with the conventional popularity-based and the modified Least Frequently Used(i.e.,LFU-dynamic)schemes,the UAV can offload around 30%traffic from the ground network by utilizing the proposed scheme in the urban scenario, according to the simulation results.

文摘As a representative technique in natural language processing(NLP),named entity recognition is used in many tasks, such as dialogue systems, machine translation and information extraction. In dialogue systems, there is a common case for named entity recognition, where a lot of entities are composed of numbers, and are segmented to be located in different places. For example,in multiple rounds of dialogue systems,a phone number is likely to be divided into several parts, because the phone number is usually long and is emphasized. In this paper, the entity consisting of numbers is named as number entity. The discontinuous positions of number entities result from many reasons. We find two reasons from real-world dialogue systems. The first reason is the repetitive confirmation of different components of a number entity, and the second reason is the interception of mood words. The extraction of number entities is quite useful in many tasks, such as user information completion and service requests correction. However, the existing entity extraction methods cannot extract entities consisting of discontinuous entity blocks. To address these problems,in this paper,we propose a comprehensive method for number entity recognition, which is capable of extracting number entities in multiple rounds of dialogues systems. We conduct extensive experiments on a real-world dataset, and the experimental results demonstrate the high performance of our method.

文摘We demonstrate the in situ growth of ultra-thin InA s nanowires with an epitaxial Al film by molecular-beam epitaxy. Our InAs nanowire diameter(~30 nm)is much thinner than before(~100 nm).The ultra-thin InAs nanowires are pure phase crystals for various different growth directions.Transmission electron microscopy confirms an atomically abrupt and uniform interface between the Al shell and the InAs wire.Quantum transport study on these devices resolves a hard induced superconducting gap and 2 e-periodic Coulomb blockade at zero magnetic field,a necessary step for future Majorana experiments.By reducing wire diameter, our work presents a promising route for reaching fewer sub-band regime in Major ana nanowire devices.
