背景内脏高敏感的发生与肠系膜淋巴结树突状细胞(mesentericlymph node dendritic cells,MLNDC)的异常活化相关;蛋白质二硫键异构酶A3(protein disulfideisomerase A3,PDIA3)可以调节信号转导与转录激活因子3(signal transduction and a... 背景内脏高敏感的发生与肠系膜淋巴结树突状细胞(mesentericlymph node dendritic cells,MLNDC)的异常活化相关;蛋白质二硫键异构酶A3(protein disulfideisomerase A3,PDIA3)可以调节信号转导与转录激活因子3(signal transduction and activator of transcription 3,STAT3)通路的活性.我们假设,在MLNDC中,PDIA3可以抑制STAT3通路,激活MLNDC,并最终引起内脏高敏感.目的初步探讨PDIA3/STAT3在内脏高敏感小鼠肠道树突状细胞异常免疫活化中的可能作用方法40只C57BL/6小鼠随机分成对照组(20只)和模型组(20只),采用束缚应激建立小鼠肠易激综合征内脏高敏感模型,用腹部撤回反射评估小鼠肠道敏感性,磁珠分选技术分离两组小鼠MLNDC,流式检测分选纯度以及比较两组MLNDC表面主要组织相容性复合体Ⅱ(major histocompatibility complex II,MHC-II)类分子的表达情况,蛋白印记技术和免疫荧光技术比较两组MLNDC中PDIA3和磷酸化-信号转导与转录激活因子3(phosphorylated signal transduction andactivator of transcription 3,P-STAT3)的表达及分布情况,混合淋巴细胞反应(mixed lymphocyte reaction,MLR)比较两组DC对初始T淋巴细胞的激活能力.结果模型组小鼠结肠扩张容量阈值明显低于对照组[(0.53mL±0.07 mL)vs(0.85 mL±0.12 mL),t=-10.845,P=0.000];流式检测磁珠分选所得的小鼠树突状细胞特异性抗原CD11c阳性MLNDC为86.61%±7.02%;模型组MLNDC表面MHC-Ⅱ类分子表达显著高于对照组[(73.08%±2.64%)vs(53.20%±4.50%),t=-7.617,P=0.000];与对照组相比,模型组小鼠MLNDC表达PDIA3明显升高[(0.77±0.04)vs(0.42±0.03),t=11.086,P=0.000],同时P-STAT3的表达明显下降[(0.73±0.03)vs(1.24±0.18),t=-4.737,P=0.038)],MLR中刺激初始T淋巴细胞增殖的能力明显增强[(34.24%±2.95%)vs(17.22%±3.47%),t=6.472,P=0.003].结论内脏高敏感小鼠MLNDC的异常免疫活化或许和PDIA3/STAT3有关. 展开更多

Traditional Chinese medicine for prevention and treatment of hepatocarcinoma: From bench to bedside 被引量:22

Traditional Chinese medicine(TCM) has played a positive role in the management of hepatocarcinoma. Traditional Chinese medicine(TCM) has played a positive role in the management of hepatocarcinoma. Hepatocarcinoma patients may present Qi-stagnation, damp-heat, blood stasis, Qi-deficiency, Yin-deficiency and other TCM syndromes(Zheng). Modern treatments such as surgery, transarterial chemoembolization(TACE) and high intensity focus ultrasound treatment would influence the manifestation of TCM syndromes. Herbs with traditional efficacy of tonifying Qi, blood and Yin, soothing liver-Qi stagnation, clearing heat and detoxifying and dissolving stasis, have been demonstrated to be potent to prevent hepatocarcinogenesis. TCM has been widely used in all aspects of integrative therapy in hepatocarcinoma, including surgical resection, liver transplantation, TACE, local ablative therapies and even as monotherapy for middle-advanced stage hepatocarcinoma. Clinical practices have confirmed that TCM is effective to alleviate clinical symptoms, improve quality of life and immune function, prevent recurrence and metastasis, delay tumor progression, and prolong survival time in hepatocarcinoma patients. The effective mechanism of TCM against hepatocarcinoma is related to inducing apoptosis, autophagy, anoikis and cell senescence, arresting cell cycle, regulating immune function, inhibiting metastasis and angiogenesis, reversing drug resistance and enhancing effects of chemotherapy. Along with the progress of research in this field, TCM will contribute more to the prevention and treatment of hepatocarcinoma. 展开更多

AIM To study the role of microbial metabolites in the modulation of biochemical and physiological processes in irritable bowel syndrome(IBS).METHODS In the current study, using a metabolomic approach, we analyzed the key metabolites differentially excreted in the feces of control mice and mice with IBS, with or without Clostridium butyricum(C. butyricum) treatment. C57 BL/6 mice were divided into control, IBS, and IBS + C. butyricum groups. In the IBS and IBS + C. butyricum groups, the mice were subjected to water avoidance stress(WAS) for 1 h/d for ten days. Gas chromatography/mass spectrometry(GC-MS) together with multivariate analysis was employed to compare the fecal samples between groups. RESULTS WAS exposure established an appropriate model of IBS in mice, with symptoms of visceral hyperalgesia and diarrhea. The differences in the metabolite profiles between the control group and IBS group significantly changed with the progression of IBS(days 0, 5, 10, and 17). A total of 14 differentially excreted metabolites were identified between the control and IBS groups, and phenylethylamine was a major metabolite induced by stress. In addition, phenylalanine metabolism was found to be the most relevant metabolic pathway. Between the IBS group and IBS + C. butyricum group, 10 differentially excreted metabolites were identified. Among these, pantothenate and coenzyme A(Co A) biosynthesis metabolites, as well as steroid hormone biosynthesis metabolites were identified as significantly relevant metabolic pathways.CONCLUSION The metabolic profile of IBS mice is significantly altered compared to control mice. Supplementation with C. butyricum to IBS mice may provide a considerable benefit by modulating host metabolism. 展开更多

Clinically significant endoscopic findings in patients of dyspepsia with no warning symptoms:A cross-sectional study

BACKGROUND Dyspepsia is one of the commonest clinical disorder.However,controversy remains over the role of endoscopy in patients with dyspepsia.No studies have evaluated the diagnostic value of endoscopy in patients with no warning symptoms according to the Rome IV criteria.AIM To study the diagnostic value of endoscopy in dyspeptic patients with no warning symptoms.METHODS This cross-sectional study included dyspeptic patients with no warning symptoms who met the inclusion and exclusion criteria at The First Affiliated Hospital,Zhejiang Chinese Medical University from April 2018 to February 2019.The clinical data were collected using questionnaires,including dyspeptic information,warning symptoms,other diseases,family history and basic demographic data.Based on dyspeptic symptoms,patients can be divided into epigastric pain syndrome,postprandial distress syndrome or overlapping subtypes.RESULTS A total of 1016 cases were enrolled,304(29.9%)had clinically significant findings that were detectable by endoscopy.The endoscopy findings included esophageal lesions in 180(17.7%)cases,peptic ulcers in 115(11.3%)cases and malignancy in 9(0.89%)patients.Multivariate logistic regression analysis showed that males[odds ratio(OR)=1.758,P<0.001],body mass index>25(OR=1.660;P=0.005),epigastric pain(OR=1.423;P=0.019)and Helicobacter pylori infection(OR=1.949;P<0.001)were independently associated with risk factors for the presence of clinically significant findings on endoscopy.CONCLUSION Chinese patients with dyspepsia with no warning symptoms should undergo endoscopy,particularly males,patients with body mass index>25,epigastric pain or Helicobacter pylori infection. 展开更多

BACKGROUND The incidence of infection with Mycobacterium abscessus(M.abscessus)has increased in recent years.This increase is partly associated with invasive cosmetic procedures.CASE SUMMARY The purpose of this case summary is to increase clinicians'awareness of M.abscessus infection and reduce mycobacterial infection caused by cosmetic procedures.We report the case of a 45-year-old woman who received acetyl hexapeptide-8(argireline)injections in the forehead and temples,and erythema,nodules,and abscesses appeared at the injection sites after one week.The pus specimens were examined by microbiological culture and confirmed to be positive for M.abscessus.Clarithromycin 500 mg twice daily and moxifloxacin 400 mg once daily were administered for 5 mo and the lesions gradually subsided.CONCLUSION We report here for the first time a case of infection with M.abscessus after argireline injection.This condition is easily misdiagnosed as a common bacterial infection.Microbiological examinations are helpful for diagnosis and standardized cosmetic procedures can prevent infection with M.abscessus. 展开更多

Constructing controllable thermal conduction networks is the key to efficiently improve thermal conductivities of polymer composites.In this work,graphite oxide(GO)and functionalized carbon nanotubes(f-CNTs)are combined to prepare“Line-Plane”-like hetero-structured thermally conductive GO@f-CNTs fillers,which are then performed to construct controllable 3D GO@f-CNTs thermal conduction networks via selfsacrificing template method based on oxalic acid.Subsequently,thermally conductive GO@f-CNTs/polydimethylsiloxane(PDMS)composites are fabricated via casting method.When the size of oxalic acid is 0.24 mm and the volume fraction of GO@f-CNTs is 60 vol%,GO@f-CNTs/PDMS composites present the optimal thermal conductivity coefficient(λ,4.00 W·m^(-1)·K^(-1)),about 20 times that of theλof neat PDMS(0.20 W·m^(-1)·K^(-1)),also much higher than theλ(2.44 W·m^(-1)·K^(-1))of GO/f-CNTs/PDMS composites with the same amount of randomly dispersed fillers.Meanwhile,the obtained GO@f-CNTs/PDMS composites have excellent thermal stability,whoseλdeviation is only about 3%after 500 thermal cycles(20-200℃). 展开更多

Herein, functionalized graphene oxide(GO) was prepared by the covalent functionalization with amino acids(lysine, glycine, glutamic acid and tyrosine) in this study. Zeta potential results demonstrated that covalent functionalization of GO with amino acids was favourable for their homogeneous dispersion in water and organic solvents. Based on the higher absolute value of zeta potential and the better dipersion stability of GO-lysine, the PVDF/GO-lysine hybrid membranes were then prepared via the phase inversion induced by immersion precipitation technique. SEM images showed a better pore diameter and porosity distribution on the PVDF/GO-lysine membrane surface. The zeta potential absolute value of the PVDF/GO-lysine membrane surface was higher than that of the virgin PVDF membrane. Furthermore, the PVDF/GO-lysine membranes surface exhibited good hydrophilicity. The water flux of PVDF/GO-lysine membranes can reach two times of that of the virgin PVDF membrane. And the BSA adsorbed amount on PVDF/GO-lysine surface was decreased to 0.82 mg/cm^2 for PVDF/GO-lysine-8% membrane. Filtration experiment results indicated that the fouling resistance was significantly improved for the PVDF/GO-lysine membranes. As a result, lysine functionalized GO will provide a promising method to fabricate graphene oxide based hybrid membranes with effective antifouling property and hydrophilicity. 展开更多

Background: Teng-Long-Bu-Zhong-Tang(TLBZT) is a traditional Chinese herbal medicine used to treat colorectal cancer(CRC). In the present study, we observed the anti-cancer effects of TLBZT on human RKO CRC. Materials and Methods: Mice were subcutaneously transplanted with RKO cells, divided into control, 5-Fu-administered, TLBZT-administered, and TLBZT and 5-Fu combination-administered groups, and treated with 5-Fluorouracil(5-Fu) and/or TLBZT. Apoptosis was detected by Td T-mediated d UTP nick-end labeling assay. The activity of caspase-3,-8, and-9 was detected using specific commercial kits. Cell senescence was assessed using senescence β-galactosidase staining. Protein expression was evaluated by immunohistochemistry. Results: TLBZT inhibited RKO CRC tumor growth, enhanced the anti-cancer effects of 5-Fu, induced apoptosis, and activated caspase-3,-8, and-9. TLBZT induced cell senescence accompanied by the downregulation of cyclin E1 and cyclin-dependent kinase 2 expressions. TLBZT also inhibited angiogenesis and the expression of hypoxia-inducible factor 1 subunit alpha and vascular endothelial growth factor-A. Conclusions: TLBZT inhibited RKO CRC tumor growth and enhanced the anti-cancer effects of 5-Fu, and it could be associated with apoptosis and cell senescence induction, and angiogenesis inhibition. 展开更多
