Protection of melatonin against damage of sperm mitochondrial function induced by reactive oxygen species

Aim: To study the mitochondrial function damage of sperm induced by reactive oxygen species (ROS) and the protection of melatonin (MLT) against the damage. Methods: Normal function spermatozoa were selected from semen samples by Percoll gradient centrifugation technique. The ROS generated by the hypoxan-thine xanthine oxidase system was incubated with the normal spermatozoa in the presence or absence of MLT (6 mmol/L) for 30 and 60 minutes. After incubation, the activity of succinate dehydroge-nase (SDH) in the mitochondria of spermatozoa was assessed by histochemical method and spermatozoa were labeled with specific fluorescent probe of Rhodamine 123 to measure the mitochondrial membrane potential (MMP) by flow cytometry. Results: After the normal spermatozoa were incubated with ROS, The sperm MMP was significantly decreased and the SDH activity of almost decreased to zero. MLT reduced the mitochondrial damage induced by ROS. Conclusion: ROS damage the mitochondrial function of sperm by affecting sperm MMP and SDH activity of. MLT protects sperm mitochondria from the damage induced by ROS through its effective antioxidative potential. 展开更多

Objective: To determine the concentration of malondialdehyde (MDA), product of lipid peroxidation, in seminal plasma and evalu ate its significance. Methods: Ninety-three cases of infertile pa tients were divided into the obstructive azoospermic (12 cases), the non-obstructive azoospermic (15 cases), the oligozoospermic (21 cases), the asthenozoospermic (19 cases), the oligoastheno zoospermic (16 cases) and the oligoasthenoteratozoospermic (10) groups. Eighteen fertile males served as the controls. MDA con centration was assessed by high-performance liquid chromatogra phy (HPLC). Results: With the exception of the obstructive azoospermic group, the MDA concentration in the seminal plasma was significantly different between the control group and the infer tile groups (P<0.01) as well as between the different infertile groups. Conclusion: Determination of MDA concentration in the seminal plasma is helpful to the diagnosis of male infertility induced by overproduction of reactive oxygen species. 展开更多

Recent advances in OCR show that end-to-end(E2E)training pipelines including detection and identification can achieve the best results.However,many existing methods usually focus on case insensitive English characters.In this paper,we apply an E2E approach,the multiplex multilingual mask TextSpotter,which performs script recognition at the word level and uses different recognition headers to process different scripts while maintaining uniform loss,thus optimizing script recognition and multiple recognition headers simultaneously.Experiments show that this method is superior to the single-head model with similar number of parameters in endto-end identification tasks. 展开更多

为了提高微型月季种质资源的利用率,从12个数量性状和8个质量性状对国外引进的微型月季主栽种质资源,进行了表型性状遗传多样性研究。结果表明,微型月季表型性状的种间变异系数为花直径的变异系数(20.61%)最大,侧叶宽变异系数(1.68%)最... 为了提高微型月季种质资源的利用率,从12个数量性状和8个质量性状对国外引进的微型月季主栽种质资源,进行了表型性状遗传多样性研究。结果表明,微型月季表型性状的种间变异系数为花直径的变异系数(20.61%)最大,侧叶宽变异系数(1.68%)最小;种内变异系数为柄叶数变异系数(15.14%)最大,花瓣长变异系数(4.95%)最小;数量性状的Shannon-weaver多样性信息指数H'在0.852~2.777之间,平均值为1.960。基于20个表型性状数据,将种质资源分为花型、叶型、叶刺、花色等5个不同类型。数量性状的前7个主成分累计方差贡献率达到91.75%,反映的信息与聚类分析和相关分析结果基本一致。花色、花型等经济性状表型性状指标的影响较大,结合相关性分析和主成分分析,20个数量性状可简化为花型、叶型、花色、叶刺等5个主要性状。以上结果为微型月季的资源评价和品种选育提供了重要的物质基础和理论依据。 展开更多

目的探讨中央气道原发涎腺型肿瘤(SGT)的影像表现及预后特点,以提高对该病的认识。方法回顾性分析2009年10月至2017年11月本院经病理证实的25例中央气道原发SGT的临床及影像资料,并随访统计患者生存率。依据病理结果分为腺样囊性癌(ACC... 目的探讨中央气道原发涎腺型肿瘤(SGT)的影像表现及预后特点,以提高对该病的认识。方法回顾性分析2009年10月至2017年11月本院经病理证实的25例中央气道原发SGT的临床及影像资料,并随访统计患者生存率。依据病理结果分为腺样囊性癌(ACC)14例(ACC组),黏液表皮样癌(MEC)10例(MEC组),上皮肌上皮癌(EMC)1例。患者术前均行CT检查及MPR后处理,其中CT增强扫描20例;PET-CT检查8例;MR检查1例。观察并记录肿瘤CT征象。采用独立样本t检验比较ACC组与MEC组患者年龄的差异。采用非参数检验比较两组病灶最大径的差异。采用Fisher精确概率法比较两组患者性别、吸烟史、TNM分期及CT征象的差异。采用Kaplan-Meier法估计总生存率(OS),采用log-rank方法比较两组患者OS的差异。结果ACC组患者发病年龄大于MEC组,差异有统计学意义(t=3.154,P<0.05)。ACC多位于气管或主支气管(13/14),MEC多位于叶或段支气管(6/10);ACC形态以分叶状肿块或管壁弥漫性增厚为主(13/14),MEC多表现为表面光滑的类椭圆形肿块(7/10);CT增强扫描ACC以轻中度强化为主(9/10),MEC多显著强化(8/10);MEC伴气道阻塞征象(9/10)较ACC(4/14)常见;ACC组与MEC组以上CT征象的差异均有统计学意义(P值均<0.05)。1例EMC表现为气管腔内类圆形结节。8例PET/CT检查中6例SUVmax≥2.2。25例患者中,1例失随访,其余患者随访时间2个月~9年(平均45个月),OS为87.5%;ACC组与MEC组患者OS的差异无统计学意义(χ^2=0,P=1.000)。年龄≤60岁、临床TNM分期为Ⅰ~Ⅱ期、行手术切除的患者OS高于年龄>60岁、临床TNM分期为Ⅲ~Ⅳ期、非手术切除的患者(χ^2值分别为13.799、13.799、13.171,P值均<0.05)。结论中央气道原发SGT浸润性弱,患者年龄偏低且预后良好,ACC与MEC的病灶部位、形状、强化程度及是否伴气道阻塞征象有差异。 展开更多

黄秋葵(Abelmoschus esculentus)组织中富含影响其DNA提取质量的多糖、多酚。为探究这一特性,本研究以黄秋葵幼叶为材料,探索了一种新的基因组DNA提取方法——硅胶膜离心柱法,利用硅胶膜离心柱与自配的溶液提取了高质量的DNA。此法先用... 黄秋葵(Abelmoschus esculentus)组织中富含影响其DNA提取质量的多糖、多酚。为探究这一特性,本研究以黄秋葵幼叶为材料,探索了一种新的基因组DNA提取方法——硅胶膜离心柱法,利用硅胶膜离心柱与自配的溶液提取了高质量的DNA。此法先用不含十六烷基三甲基溴化铵(cetyl-trimethyl-ammonium bromide, CTAB)的缓冲液去除一部分次生代谢物;然后用3%CTAB缓冲液分离DNA;再用氯仿/异戊醇(24:1)抽提,之后用硅胶膜离心柱纯化DNA。运用琼脂糖凝胶电泳、ISSR-PCR扩增和限制性酶切等方法检测DNA的质量,并与传统CTAB法、十二烷基硫酸钠(sodium dodecyl sulfate, SDS)法和高盐低pH法进行比较分析。结果表明,本法提取的DNA完整性好,纯度高;能被Eco RⅠ完全消化;ISSR扩增得到的条带最清晰、多态性好,明显优于其它3种传统方法。此外,本方法也成功用于石斛(Dendrobium)、葡萄(Vitis spp.)、无花果(Ficus carica)、芦荟(Aloe vera)等植物基因组DNA的提取,因此具有广泛的适用性,可为富含多糖植物基因组DNA的提取提供参考。 展开更多
