BACKGROUND Extensive evidence has illustrated the promotive role of integrin binding sialoprotein(IBSP)in the progression of multiple cancers.However,little is known about the functions of IBSP in gastric cancer(GC)pr...BACKGROUND Extensive evidence has illustrated the promotive role of integrin binding sialoprotein(IBSP)in the progression of multiple cancers.However,little is known about the functions of IBSP in gastric cancer(GC)progression.AIM To investigate the mechanism underlying the regulatory effects of IBSP in GC progression,and the relationship between IBSP and cleavage and polyadenylation factor 6(CPSF6)in this process.METHODS The mRNA and protein expression of relevant genes were assessed through realtime quantitative polymerase chain reaction and Western blot,respectively.Cell viability was evaluated by Cell Counting Kit-8 assay.Cell invasion and migration were evaluated by Transwell assay.Pyroptosis was measured by flow cytometry.The binding between CPSF6 and IBSP was confirmed by luciferase reporter and RNA immunoprecipitation(RIP)assays.RESULTS IBSP exhibited higher expression in GC tissues and cell lines than in normal tissues and cell lines.IBSP knockdown suppressed cell proliferation,migration,and invasion but facilitated pyroptosis.In the exploration of the regulatory mechanism of IBSP,potential RNA binding proteins for IBSP were screened with catRAPID omics v2.0.The RNA-binding protein CPSF6 was selected due to its higher expression in stomach adenocarcinoma.Luciferase reporter and RIP assays revealed that CPSF6 binds to the 3’-untranslated region of IBSP and regulates its expression.Knockdown of CPSF6 inhibited cell proliferation,migration,and invasion but boosted pyroptosis.Through rescue assays,it was uncovered that the retarded GC progression mediated by CPSF6 knockdown was reversed by IBSP overexpression.CONCLUSION Our study highlighted the vital role of the CPSF6/IBSP axis in GC,suggesting that IBSP might be an effective biotarget for GC treatment.展开更多

METHODS The FD with liver depres...AIM To investigate gut microbial diversity and the interventional effect of Xiaoyaosan(XYS) in a rat model of functional dyspepsia(FD) with liver depression-spleen deficiency syndrome. METHODS The FD with liver depression-spleen deficiency syndrome rat model was established through classic chronic mild unpredictable stimulation every day. XYS group rats received XYS 1 h before the stimulation. The models were assessed by parameters including state ofthe rat, weight, sucrose test result and open-field test result. After 3 wk, the stools of rats were collected and genomic DNA was extracted. PCR products of the V4 region of 16 S rD NA were sequenced using a barcoded Illumina paired-end sequencing technique. The primary composition of the microbiome in the stool samples was determined and analyzed by cluster analysis.RESULTS Rat models were successfully established, per data from rat state, weight and open-field test. The microbiomes contained 20 phyla from all samples. Firmicutes, Bacteroidetes, Proteobacteria, Cyanobacteria and Tenericutes were the most abundant taxonomic groups. The relative abundance of Firmicutes, Proteobacteria and Cyanobacteria in the model group was higher than that in the normal group. On the contrary, the relative abundance of Bacteroidetes in the model group was lower than that in the normal group. Upon XYS treatment, the relative abundance of all dysregulated phyla was restored to levels similar to those observed in the normal group. Abundance clustering heat map of phyla corroborated the taxonomic distribution. CONCLUSION The microbiome relative abundance of FD rats with liver depression-spleen deficiency syndrome was significantly different from the normal cohort. XYS intervention may effectively adjust the gut dysbacteriosis in FD.展开更多
AIM To assess the validity and reliability of the European Organization for Research and Treatment of Cancer Quality of Life Questionnaire-Colorectal Cancer 29(EORTC QLQ-CR29) in Chinese patients with colorectal cance...AIM To assess the validity and reliability of the European Organization for Research and Treatment of Cancer Quality of Life Questionnaire-Colorectal Cancer 29(EORTC QLQ-CR29) in Chinese patients with colorectal cancer(CRC). METHODS From March 2014 to January 2015, 356 patients with CRC from four different hospitals in China were enrolled in the study, and all patients self-administered the EORTC QLQ-CR29 and the quality of life core questionnaire(EORTC QLQ-C30). Evaluation of the scores was based on the Karnofsky Performance Scale(KPS). The reliability and validity of the questionnaires were assessed by Cronbach's α coefficient, the Spearman correlation test and Wilcoxon rank sum test.RESULTS The EORTC QLQ-CR29 showed satisfactory reliability(α > 0.7), although the urinary frequency and blood and mucus in stool dimensions had only moderate reliability(α = 0.608). The multitrait scaling analyses showed good convergent(r > 0.4) and discriminant validity. Significant differences were obtained for each item in the different KPS subgroups(KPS ≤ 80; KPS > 80). body image and most single-item dimensions showed statistically significant differences in patients with a stoma compared with the rest of the patients. CONCLUSION The EORTC QLQ-CR29 exhibits high validity and reliability in Chinese patients with CRC, and can therefore be recommended as a valuable tool for the assessment of quality of life in these patients.展开更多
The novel severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)is the cause of a rapidly spreading illness,coronavirus disease 2019(COVID-19),affecting more than seventeen million people around the world.Diagnos...The novel severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)is the cause of a rapidly spreading illness,coronavirus disease 2019(COVID-19),affecting more than seventeen million people around the world.Diagnosis and treatment guidelines for clinicians caring for patients are needed.In the early stage,we have issued"A rapid advice guideline for the diagnosis and treatment of 2019 novel coronavirus(2019-nCoV)infected pneumonia(standard version)";now there are many direct evidences emerged and may change some of previous recommendations and it is ripe for develop an evidence-based guideline.We formed a working group of clinical experts and methodologists.The steering group members proposed 29 questions that are relevant to the management of COVID-19 covering the following areas:chemoprophylaxis,diagnosis,treatments,and discharge management.We searched the literature for direct evidence on the management of COVID-19,and assessed its certainty generated recommendations using the Grading of Recommendations,Assessment,Development and Evaluation(GRADE)approach.Recommendations were either strong or weak,or in the form of ungraded consensus-based statement.Finally,we issued 34 statements.Among them,6 were strong recommendations for,14 were weak recommendations for,3 were weak recommendations against and 11 were ungraded consensus-based statement.They covered topics of chemoprophylaxis(including agents and Traditional Chinese Medicine(TCM)agents),diagnosis(including clinical manifestations,reverse transcription-polymerase chain reaction(RT-PCR),respiratory tract specimens,IgM and IgG antibody tests,chest computed tomography,chest X-ray,and CT features of asymptomatic infections),treatments(including lopinavirritonavir,umifenovir,favipiravir,interferon,remdesivir,combination of antiviral drugs,hydroxychloroquine/chloroquine,interleukin-6 inhibitors,interleukin-1 inhibitors,glucocorticoid,qingfei paidu decoction,lianhua qingwen granules/capsules,convalescent plasma,lung transplantation,invasive or noninvasive ventilation,and extracorporeal membrane oxygenation(ECMO)),and discharge management(including discharge criteria and management plan in patients whose RT-PCR retesting shows SARS-CoV-2 positive after discharge).We also created two figures of these recommendations for the implementation purpose.We hope these recommendations can help support healthcare workers caring for COVID-19 patients.展开更多

METHODS: Immortalized human corneal epithelial cells (HCECs) were exposed to inactive #lsperg///us fumigatus (A. fumigatus) conidia at 0, 4, 8, 12, 16, and 24h respectively. The corneas of Wistar rats were exposed to active A. fumigatus at 0, 12, 24, 48h and the normal rat corneas were used for normal control. The mRNA level of S100B was evaluated by real time quantitative reverse transcription-polymerase chain reaction (qRT-PCR). Sl00B protein expression in cornea epithelium was detected by immunohistochemical/immunocytochemical staining (IHC/ICC). RESULTS: Histopathology revealed a significant inflammatory cell infiltration in fungal keratitis human and rat cornea. Corneal epithelial cells didn't express or rarely express S100B at baseline. A. fumigatus significantly induced S100B mRNA expression in cultured corneal epithelial cells in a time depended manner in vitro the mRNA began to rise significantly at 8h in vitro (P〈0.05) and continue to rise as time prolonged (P〈0.01). in vivo S100B mRNA level was low in the normal corneas. However, it was increased in keratitis corneas from 12h after infection (P〈0.05) and reached to a peak at 24h (P〈0.001). Immunochemistry revealed an obvious staining in fungal keratitis corneas as well as immortalized HCECs compared to the normal ones respectively, indicating an increased expression of SlOOB protein. CONCLUSION: S100B exists in corneal epithelial cells and is over-expressed under A. fumigatus stimulation. Sl00B may play an important role in the innate immune response of the corneal epithelium during A. fumigatus infection.展开更多
Background:Breast cancer(BC)is a common malignancy with highly female incidence.So far the function of notoginsenoside R1(NGR1),the extract from Panax notoginseng,has not been clearly elucidated in BC.Methods:Optimal ...Background:Breast cancer(BC)is a common malignancy with highly female incidence.So far the function of notoginsenoside R1(NGR1),the extract from Panax notoginseng,has not been clearly elucidated in BC.Methods:Optimal culture concentration and time of NGR1 were investigated by cell counting kit-8 assay.Cell proliferation ability was measured by colony formation assays.Transwell assay was used to detect the effect of NGR1 on cell migration and invasion.The apoptosis rate of cells between each group was measured by TUNEL assay.Results:NGR1 treatment has an inhibitory efect on proliferation,migration,invasion,and angiogenesis and a stimulating effect on cell cycle arrest and apoptosis of Michigan Cancer Foundation-7(MCF-7)cells.The 50%growth inhibitory concentration for MCF-7 cells at 24 h was 148.9 mmol/L.The proportions of MCF-7 cells arrested in the G0/G1 phase were 36.94+6.78%,45.06士5.60%,and 59.46+5.60%in the control group,75,and 150 mmol/L groups,respectively.Furthermore,we revealed that NGR1 treatment attenuates BC progression by targeted downregulating CCND2 and YBX3 genes.Additionally,YBX3 activates phosphatidylinositol 3-phosphate kinase(PI3K)/protein kinase B(Akt)signaling pathway by activating kirsten rat sarcoma viral oncogene,which is an activator of the PI3K/Akt signaling pathway.Conclusion:These results suggest that NGR1 can act as an efficacious drug candidate that targets the YBX3/PI3K/Akt axis in patients with BC.展开更多

Objective This study is aimed to investigate the differences in pharmaceutical quality and therapeutic efficacy of calcipotriol ointments between the original product and the generics.Methods We evaluated the pharmace...Objective This study is aimed to investigate the differences in pharmaceutical quality and therapeutic efficacy of calcipotriol ointments between the original product and the generics.Methods We evaluated the pharmaceutical quality of the original product and the generics including pH,content and related substance,and calcipotriol stability in the matrix.Intradermal retention of calcipotriol ointments in vitro was investigated using skin of Bama miniature pig.The pharmacodynamics study was performed in psoriasis guinea pig models.Results The matrix of original product is better than the generics,and influenced the drug diffusion,stability and in vitro retention,thus resulted in the difference in pharmacodynamics.The retention amount of calcipotriol in the original product was significantly higher than that of generic B,and approximately 4 folds higher than that of generic A.All of the three ointments improved pathological indexed in psoriasis guinea pigs,and there was a significant difference between the preparation group and the model group (P =0.001).Guinea pigs in the original group had the lowest interleukin-17 level (35.7 ± 1.13 μg/ml) and Baker Score (2.70 ± 0.97) compared with generic A (37.9 ±3.3 μg/ml,3.10 ± 0.22,respectively) and generic B (39 ± 4.6μg/ml,3.80 ± 0.57,respectively),but no statistical difference was found among the three groups (all P > 0.05).Conclusions Difference occurs in the internal quality between the original product and the generics.The original preparation owns suitable balance between matrix stability and transdermal ability so that showes superiority in pharmaceutical properties,intradermal retention and therapeutic effect in vitro and in vivo,which suggests that more attention should be paid to the formulation and process during the research and development of the generics.The original product is recommended with priority for treatment especially at emergency.展开更多

A. fumigatus significantly induced S100B mRNA expression in cultured corneal epithelial cells in a time depended manner in vitro the mRNA began to rise significantly at 8h in vitro (P〈0.05) and continue to rise as time prolonged (P〈0.01). in vivo S100B mRNA level was low in the normal corneas. However, it was increased in keratitis corneas from 12h after infection (P〈0.05) and reached to a peak at 24h (P〈0.001). Immunochemistry revealed an obvious staining in fungal keratitis corneas as well as immortalized HCECs compared to the normal ones respectively, indicating an increased expression of SlOOB protein. CONCLUSION: S100B exists in corneal epithelial cells and is over-expressed under A. fumigatus stimulation. Sl00B may play an important role in the innate immune response of the corneal epithelium during A. fumigatus infection.

文摘Background:Breast cancer(BC)is a common malignancy with highly female incidence.So far the function of notoginsenoside R1(NGR1),the extract from Panax notoginseng,has not been clearly elucidated in BC.Methods:Optimal culture concentration and time of NGR1 were investigated by cell counting kit-8 assay.Cell proliferation ability was measured by colony formation assays.Transwell assay was used to detect the effect of NGR1 on cell migration and invasion.The apoptosis rate of cells between each group was measured by TUNEL assay.Results:NGR1 treatment has an inhibitory efect on proliferation,migration,invasion,and angiogenesis and a stimulating effect on cell cycle arrest and apoptosis of Michigan Cancer Foundation-7(MCF-7)cells.The 50%growth inhibitory concentration for MCF-7 cells at 24 h was 148.9 mmol/L.The proportions of MCF-7 cells arrested in the G0/G1 phase were 36.94+6.78%,45.06士5.60%,and 59.46+5.60%in the control group,75,and 150 mmol/L groups,respectively.Furthermore,we revealed that NGR1 treatment attenuates BC progression by targeted downregulating CCND2 and YBX3 genes.Additionally,YBX3 activates phosphatidylinositol 3-phosphate kinase(PI3K)/protein kinase B(Akt)signaling pathway by activating kirsten rat sarcoma viral oncogene,which is an activator of the PI3K/Akt signaling pathway.Conclusion:These results suggest that NGR1 can act as an efficacious drug candidate that targets the YBX3/PI3K/Akt axis in patients with BC.

文摘Abstract In this paper, we get the H^jek-R^nyi-type inequalities for a pairwise NQD sequence, an L^T (r 〉 1) mixingale and a linear process, which have the concrete coefficients. In addition, we obtain the strong law of large numbers, strong growth rate and the integrability of supremum for the above sequences, which generalize and improve Corollary 2 for L^T(r 〉 1) mixingale of Hansen.

文摘Consider a discrete-time risk model with insurance and financial risks in a stochastic economic environment. Assume that the insurance and financial risks form a sequence of independent and identically distributed random vectors with a generic random vector following a wide type of dependence structure. An asymptotic formula for the finite-time ruin probability with subexponential insurance risks is derived. In doing so, the subexponentiality of the product of two dependent random variables is investigated simultaneously.

文摘In the paper,we establish some exponential inequalities for non-identically distributed negatively orthant dependent(NOD,for short)random variables.In addition,we also establish some exponential inequalities for the partial sum and the maximal partial sum of identically distributed NOD random variables.As an application,the Kolmogorov strong law of large numbers for identically distributed NOD random variables is obtained.Our results partially generalize or improve some known results.
文摘Objective This study is aimed to investigate the differences in pharmaceutical quality and therapeutic efficacy of calcipotriol ointments between the original product and the generics.Methods We evaluated the pharmaceutical quality of the original product and the generics including pH,content and related substance,and calcipotriol stability in the matrix.Intradermal retention of calcipotriol ointments in vitro was investigated using skin of Bama miniature pig.The pharmacodynamics study was performed in psoriasis guinea pig models.Results The matrix of original product is better than the generics,and influenced the drug diffusion,stability and in vitro retention,thus resulted in the difference in pharmacodynamics.The retention amount of calcipotriol in the original product was significantly higher than that of generic B,and approximately 4 folds higher than that of generic A.All of the three ointments improved pathological indexed in psoriasis guinea pigs,and there was a significant difference between the preparation group and the model group (P =0.001).Guinea pigs in the original group had the lowest interleukin-17 level (35.7 ± 1.13 μg/ml) and Baker Score (2.70 ± 0.97) compared with generic A (37.9 ±3.3 μg/ml,3.10 ± 0.22,respectively) and generic B (39 ± 4.6μg/ml,3.80 ± 0.57,respectively),but no statistical difference was found among the three groups (all P > 0.05).Conclusions Difference occurs in the internal quality between the original product and the generics.The original preparation owns suitable balance between matrix stability and transdermal ability so that showes superiority in pharmaceutical properties,intradermal retention and therapeutic effect in vitro and in vivo,which suggests that more attention should be paid to the formulation and process during the research and development of the generics.The original product is recommended with priority for treatment especially at emergency.
