为提高一株海洋来源的韩国普李斯特氏菌(Priestia koreensis FS-1)产淀粉酶的能力,对该株菌采用常压室温等离子体(atmospheric and room temperature plasma, ARTP)技术进行了诱变选育。结果显示:经过诱变选育后得到一株产淀粉酶活力较...为提高一株海洋来源的韩国普李斯特氏菌(Priestia koreensis FS-1)产淀粉酶的能力,对该株菌采用常压室温等离子体(atmospheric and room temperature plasma, ARTP)技术进行了诱变选育。结果显示:经过诱变选育后得到一株产淀粉酶活力较原始菌株提高了90%且稳定遗传的突变菌株P.koreensis FS-103。为解析该突变菌株酶活力增加的分子机制,对突变菌株P.koreensis FS-103进行了全基因组重测序及比对分析研究。相关分析显示:首先,P.koreensis FS-103的基因组中乙酰化修饰、抗氧化作用以及同义突变相关的基因发生了遗传变异;其次,对COG(clusters of orthologous groups)功能和变异基因进行富集分析,P.koreensis FS-103转录、翻译和翻译后修饰过程相关基因均上调。经过ARTP诱变以后的突变菌株P.koreensis FS-103淀粉酶活力提升的原因是以上功能基因发生上调效应。展开更多
