文摘用精液保存液在4 °C下冷藏保存暗纹东方鲀(Fugu obscurus) 精液, 逐日测定保存液中以及扩散到生理盐水和淡水中的精子活动时间及其对成熟卵子受精率和孵化率的影响。结果, 精子活动时间随冷藏保存时间的延长而呈指数下降; 冷藏保存4 d 精液的受精率及保存3 d 精液的孵化率与原精液差别不明显。因此, 用保存6 d 内的精液生产上是可行的。
文摘为中华鲟淡水人工群体驯养与繁殖过程中的养殖水环境因子调控以及精子冷冻保存和建立精子库提供科学依据,进行了中华鲟子一代(Acipenser sinensis)精子在Na+、K+、Ca2+、Mg2+和葡萄糖溶液中的活力测定。结果表明:精子在纯水中的最高寿命(Life tmie,LT)和快速运动时间(Fast movement time,FT)分别为108.57s和39.76s。Na+浓度为4‰时精子LT和FT均达到最高(263.33s和45.33s)。K+浓度为0.01‰时精子LT最长(132.33s),最高FT(46.00s)则出现在K+浓度为0.008‰时。LT和FT在Ca2+浓度为4‰时均达到最高值(130.67s和53.67s)。Mg2+浓度为3‰时,LT达到最高(117.33s),FT最高值(27.20s)出现在Mg2+浓度为2‰时。LT在葡萄糖浓度为4%时达到最高值282.80s,FT最高值(55.88s)出现在浓度为3%时。表明,适量浓度的Na+、K+、Ca2+和Mg2+可以延长精子的LT和FT。LT和FT在葡萄糖的峰值均较其在Na+和K+溶液中高,表明精子能有效利用外源性物质延长其运动时间。与中华鲟野生亲本相比,子一代精子的LT和FT均有所下降,说明中华鲟子一代精子的质量较野生亲本低。
