实验利用线粒体COI基因和Cytb基因的片段序列分析比较了黑锦鲤(Cyprinus carpio Karasugoi Koi)、三色锦鲤(C.carpio Taisho Sanshoku Koi)、红白锦鲤(C.carpio Kohaku Koi)、黄金锦鲤(C.carpio Kigoi Koi)、全白锦鲤(C.carpio Platinum... 实验利用线粒体COI基因和Cytb基因的片段序列分析比较了黑锦鲤(Cyprinus carpio Karasugoi Koi)、三色锦鲤(C.carpio Taisho Sanshoku Koi)、红白锦鲤(C.carpio Kohaku Koi)、黄金锦鲤(C.carpio Kigoi Koi)、全白锦鲤(C.carpio Platinum Ogon Koi)和全红锦鲤(C.carpio Higoi Koi)6个锦鲤(Cyprinus carpio Koi)养殖品系的遗传多样性和系统进化关系。结果显示:基因序列分析得到的单倍型数分别为7、6。6个品系均具有较高的单倍型多样性指数和较低的核苷酸多样性指数。群体间遗传分化系数Fst、遗传距离和AMOVA等结果显示品种间变异高于品种内变异,黑锦鲤和黄金锦鲤间、红白锦鲤和三色锦鲤间不存在显著遗传差异,其余品系间差异显著。根据COI和Cytb基因单倍型构建的品系间NJ系统进化树得到相同结果,黑锦鲤和全红锦鲤遗传关系较近,聚为一支。三色锦鲤、红白锦鲤和黄金锦鲤遗传关系较近,聚为一支。全白锦鲤与其他品系的亲缘关系均较远,单独形成一个分支。 展开更多

The common carp (Cyprinus carpio) has a large variety of strains. The more popular are the koi (Japanese ornamental carp), which are still bred today to generate creative colors and patterns, giving rise to multiple phenotypes. Since koi are in great demand, there is a challenge to determine the genetics defining their quality. Two methods: 1) direct sequencing of five candidate gene regions, i.e., mitochondrial (cytochrome b, 12S gene and the D-loop) and nuclear (red sensitive opsin and Rag-1) loci, to detect single nucleotide polymorphisms (SNP)s and 2) random amplification of polymorphic DNA (RAPD), were used to differentiate among four koi strains (Kohaku, Sanke, Ghost and Ohgon) and the common carp. Novel SNPs, distinguishing between koi and the common carp, were revealed in cytochrome b, the D-loop and in the red sensitive opsin;one was a missense mutation in cytochrome b at position 15860, in which threonine in the common carp became alanine in all koi strains examined. The Kohaku strain was found to have two alleles in the mitochondrial fragments, forming two different haplotypes (subpo-pulations). These novel SNPs distinguished between koi strains and the common carp, and the RAPD method enabled further differentiation among the four koi strains. 展开更多

Fish Production, Water Quality and Bacteriological Parameters of Koi Carp Ponds Under Live-food and Manure Based Management Regimes

To test the effectiveness of introducing live zooplankton against direct manuring in ornamental fish ponds upon their survival and production, larvae of koi carp, Cyprinus carpio L., were cultured for 11 weeks in earthen ponds maintained according to four management regimes: (1) live zooplankton fed to carp larvae (LF); (2) direct fertilization with poultry manure (PM); (3) direct fertilization with cowdung (CD); and (4) a control treatment (C). There were three replicates for each treatment. The growth of heterotrophic bacteria and pathogenic microorganisms like Aeromonas sp. and Pseudomonas sp. were also examined in response to pond management. Values of dissolved oxygen were significantly higher (P〈0.05) in the water of LF ponds, compared to other treatments, while the PM and CD treatments recorded were significantly higher (P〈0.05) values of PO4 - P, NH4 - N, NO3 - N, NO2 - N, specific conductivity, alkalinity, and BOD, compared to the LF and C treatments. The percentages of organic carbon and total nitrogen in the bottom sediments were higher in the PM and CD treatments compared to LF (P〈0.05). Average counts of heterotrophic bacteria in the water of PM and CD ponds were significantly higher than other treatments (P〈0.05). The development of Aeromonas sp. and Pseudomonas sp. were significantly higher (P〈0.05) in the PM and CD treatments. Weight gain of koi carp stocked in LF was significantly higher (P〈0.05) than that of fish in the other treatments. There was a significant difference in the survival rate of koi carp among the treatments ranging from 67.21% in C to 90.11% in LF. The results suggest that raising koi carp larvae in ponds and feeding them exogenously with zooplankton would support high rates of survival and production through maintenance of better water quality and greater abundance of zooplankton in the system. Significantly lower abundance of Aeromonas sp. and Pseudomonas sp. in the LF treatment considerably lowered any possibility of occurrence of bacterial disease. 展开更多

[ Method] Primers and corresponding TaqMan probe were designed based on the conserved sequence of Koi herpes virus (KHV) pol-ymerase gene (Sph) to establish a rapid and effective fluorescence quantitative PCR method for Koi herpes virus detection. The cell cultures were detected by using the established fluorescence quantitative PCR assay, and the results were com- pared with that of conventional PCR. [ Result] The sensitivity of fluorescence quantitative PCR was higher than that of conventional PCR. The minimum copy num- ber that could be detected was 1.6 - 102 copies/p.1. The established method was adopted for sample detection, and a reliable diagnostic result could be obtained within 4 h. [Conclusion] The established method is rapid, sensitive, specific and repeatable, which is conducive to the rapid detection of Koi herpes virus. Bacteria evaluated were Aeromonas hydrophila, Pseudomonas aeruginosa and Edwardseilla tarda. A. hydrophila was resistant to ampicilin, cephalothin, sulphatriad, cefpodoxime and cefoxitin. Highest sensitivity was recorded to Colistin methane sulphonate and Ceftriaxone. High resistance was observed for P. aeruginosa in Kanamysin, Cephalothin, Sulphatriad, Colistin methane sulphonate, Tobramysin, Nitro Furantoin, Cefpodoxime, Cefoxitin and Aztreonam. Highest sensitivity recorded with Moxifloxacin. High resistance was observed in Co-trimoxamole and higher sensitivity performed for E. tarda by Tetracyclin, Cephotaxime, Ciprofloxacin, Norfloxacin, Ofloxacin, Moxifloxacin, Gatifloxacin and Levofloxacin. Out of that highest sensitivity recorded with Moxifloxacin High sensitivity was recorded for Moxifloxacin, Ciprofloxacin, Gatifloxacin, Levofloxacin. By means of antibiotic sensitivity test highest resistance was recorded to P. aeruginosa (MAR index-0.3) and least resistant was recorded to E, tarda (MAR index- 0.033). 展开更多

The aim of the present study was to develop a formulated feed for improving skin pigmentation and growth performance of koi carp juveniles. Two experiments were conducted for determination of an optimal dietary astaxanthin and FM (fish meal) ration for koi carp juveniles. In the first experiment, three isonitrogenous and isoenergetic dietary treatments were designed with three different levels of astaxanthin consisted of 60, 80 and 100 mg/kg and a commercial feed served as a control. For the second experiment, four isonitrogenous and isoenergetic dietary treatments were formulated with graded inclusions of dietary FM replaced from 0% to 60% at 20% increments by SM (soybean meal), PBM (poultry by-product meal), and a CD (control diet). Obtained results demonstrated that skin pigmentation of koi carp juveniles fed the diet containing 80 mg/kg astaxanthin, 36.02% protein, 7.78% lipid, 4.20 Kcal/g GE (gross energy) were more better than those at the diet with lower estaxanthin content and commercial diet. Moreover, the highest growth and feed utilization of fish were observed at this diet with WG (weight gain), SGR (specific growth rate) and FCR (feed conversion ratio) were 121.80%, 0.95 (%/day) and 1.6, respectively. 展开更多

A poor environment increases fish’s susceptibility to myxosporean infection that can cause the death of larval fish,especially for koi fish(Cyprinus carpio).This study aimed to determine the effect of probiotics,local anti-parasitic drugs(kutuklin),and the chemical compound diflubenzuron treatments on the koi immune response.This study used PCR with specific primer 18S SSU rDNA and DNA sequencing to detect Myxobulus phylogenetic.The treatments were divided into 5 groups:Treatment(A)(healthy koi without treatment),(B)(infected koi without treatment),(C)(infected koi with 0.55 mL/30 L probiotics),(D)(infected koi with 1μL/g of feed kutuklin),and(E)(infected koi with 0.02 mg/5 L dimilin).Myxospore has observed with Scanning Electron Microscopy(SEM)and 4′,6-diamidino-2-phenylindole(DAPI)fluorescence staining.The histological analysis using semi-quantitative scoring methods,and flow cytometry was conducted to analyse the immune response of Cluster of differentiation 4(CD4^(+)),Cluster of differentiation 8(CD8^(+)),Tumor Necrosis Factor-alpha(TNF-α),Interferon gamma(IFN-γ)cells in the gills.Results show that the histological analysis indicated edema,hyperplasia,lamella fusion,congestion,and hypertrophy lesions in infected koi.Treatment with probiotics shows the lowest damage(30.6%).The immune responses of CD4^(+)and CD8^(+)cells to dimilin treatment were 10.54%and 16.86%,respectively.The largest TNF-αand IFN-γresponse were for the kutuklin treatment(29.26%)and probiotics treatment(8.23%). 展开更多

基于融合非局部操作的YOLOv5s高密度锦鲤鱼苗检测方法

针对现有方法在高密度锦鲤鱼苗目标检测任务中适用性差的问题,提出一种基于非局部操作的YOLOv5s(MS-Non-local BIFPN coordinate attention YOLOv5s,NBC-YOLOv5s)目标检测算法。首先,在YOLOv5s的主干网络中,添加多尺度非局部操作算子(mu... 针对现有方法在高密度锦鲤鱼苗目标检测任务中适用性差的问题,提出一种基于非局部操作的YOLOv5s(MS-Non-local BIFPN coordinate attention YOLOv5s,NBC-YOLOv5s)目标检测算法。首先,在YOLOv5s的主干网络中,添加多尺度非局部操作算子(multi scale non-local,MS-Non-local),增强模型对高密度锦鲤鱼苗的特征提取能力;其次,在颈部网络使用双向加权特征金字塔结构(bi-directional feature pyramid network,BIFPN)提升模型特征融合效率;最后,在网络的特征融合处,引入坐标注意力机制(coordinate attention,CA),增加模型对图片关键信息的关注度。为验证本文算法的有效性,结合真实渔场环境建立锦鲤鱼苗数据集。实验结果表明,NBC-YOLOv5s的精确率、召回率、平均精度均值(mAP)分别为88.5%、89.7%、93.7%,与YOLOv5s相比,改进后网络较原模型分别提升0.6、9.0、4.4百分点。为验证MS-Non-local对YOLOv5s的性能提升效果,本文对比了卷积注意力(convolutional block attention module,CBAM)、通道注意力(squeeze and excitation,SE)、双层路由注意力(bi-level routing attention,BRA)3种机制。结果表明,MS-Non-local的mAP相较于CBAM、SE、BRA分别提升了2.6、2.1、0.9百分点。并且通过模型拆解,分析了本文方法对不同密度锦鲤鱼苗图像的检测有效性,结果显示,该算法可实现真实场景下对高密度锦鲤鱼苗的检测,能够为筛选高品质锦鲤提供有效技术支撑。 展开更多

锦鲤色彩鲜艳,游姿飘逸,观赏价值高,被称作“会游泳的艺术品”。鱼类体色主要是由皮肤或鳞片上的色素细胞的种类和分布决定的。本文对黑色、白色、黄色和红色4种不同体色锦鲤的鳞片和皮肤的色素细胞组成、分布及形态结构等进行了比较观... 锦鲤色彩鲜艳,游姿飘逸,观赏价值高,被称作“会游泳的艺术品”。鱼类体色主要是由皮肤或鳞片上的色素细胞的种类和分布决定的。本文对黑色、白色、黄色和红色4种不同体色锦鲤的鳞片和皮肤的色素细胞组成、分布及形态结构等进行了比较观察,结果显示,鳞片和皮肤的色素细胞组成一致,4种锦鲤均存在着丰富的虹彩细胞,在黑色锦鲤中还观察到了黑色素细胞,黄色锦鲤和红色锦鲤还分布着黄/红色素细胞,在白色锦鲤中只观察到虹彩细胞。利用透射显微镜观察了4种体色锦鲤的虹彩细胞的超微结构,虹彩细胞主要有3种不同形态:梭形、短棒状和空泡状;采用实时荧光定量PCR检测了8个虹彩细胞发育相关基因在4种不同体色锦鲤皮肤中的表达情况,结果显示,8个虹彩细胞发育相关基因在不同体色锦鲤皮肤中均有表达,但表达存在差异,其中alk、pka、sox10、pax3、foxd3和pnp4a基因在红色和黄色锦鲤中相对表达量较高,ltk和alx4a基因在白色锦鲤中相对表达量最高。为进一步证实pnp4a基因在虹彩细胞中的作用,构建了pnp4a过表达重组质粒,并将其显微注射至AB斑马鱼受精卵中,过表达斑马鱼中虹彩细胞明显多于对照组,进一步证实了pnp4a在鱼类体色形成尤其是在虹彩细胞发生方面发挥着重要作用。 展开更多

试验旨在研究饲料中添加柴胡及其提取物对锦鲤幼鱼生长、生理生化、肝脏抗氧化及抗菌能力的影响。选取初始体重(21.0±0.5)g的锦鲤幼鱼360尾,随机分为3组,每组3个重复,每个重复40尾锦鲤幼鱼。K组、C组和T组锦鲤幼鱼分别饲喂基础饲... 试验旨在研究饲料中添加柴胡及其提取物对锦鲤幼鱼生长、生理生化、肝脏抗氧化及抗菌能力的影响。选取初始体重(21.0±0.5)g的锦鲤幼鱼360尾,随机分为3组,每组3个重复,每个重复40尾锦鲤幼鱼。K组、C组和T组锦鲤幼鱼分别饲喂基础饲料、基础饲料+5‰柴胡生粉和基础饲料+5‰柴胡提取物。试验期14 d。结果显示,锦鲤幼鱼血清中,14 d时,C组的溶菌酶(LZM)和C组、T组的碱性磷酸酶(AKP)活性显著高于0 d,并显著高于K组(P<0.05)。锦鲤幼鱼肝脏中,14 d时C组的LZM、酸性磷酸酶(ACP)和T组的AKP活性显著高于0 d(P<0.05)。14 d时,C组的超氧化物歧化酶(SOD)活性及总抗氧化能力(T-AOC)显著高于0 d(P<0.05),丙二醛(MDA)含量低于0、7 d。攻毒试验中,24 h内K组锦鲤幼鱼的死亡率为41.7%;C组死亡率为18.3%,保护率为56%;T组死亡率为21.7%,保护率为48%。C组锦鲤幼鱼血清中48 h的白细胞介素-13(IL-13)含量显著高于24、96 h(P<0.05);C组、T组48 h的免疫球蛋白M(IgM)含量显著高于24 h(P<0.05)。肝脏中,C组锦鲤幼鱼48 h的白细胞介素-4(IL-4)、IL-13和IgM含量显著高于24 h,IL-13含量显著高于K组(P<0.05)。研究表明,饲料中添加柴胡及其提取物投喂14 d可以有效提高锦鲤幼鱼的生理生化指标和抗维氏气单胞菌能力,但两者的作用时间和作用效果具有差异性,柴胡的作用效果更为突出。 展开更多

本试验旨在研究不同脂肪源饲料对锦鲤生长性能、肌肉脂肪酸组成、血清生化指标、肠道消化酶活性、肠道菌群组成及脂质代谢影响。选取平均体重为(27.65±0.37)g的锦鲤450尾,随机分成5组,每组3个重复,每个重复30尾。各组分别饲喂在基... 本试验旨在研究不同脂肪源饲料对锦鲤生长性能、肌肉脂肪酸组成、血清生化指标、肠道消化酶活性、肠道菌群组成及脂质代谢影响。选取平均体重为(27.65±0.37)g的锦鲤450尾,随机分成5组,每组3个重复,每个重复30尾。各组分别饲喂在基础饲料中分别添加鱼油(FO组)、豆油(SO组)、黑水虻油(BO组)、黄粉虫油(TO组)、蝇蛆油(MO组)作为单一脂肪源配制成的等氮等脂试验饲料。试验期8周。结果表明:1)各组之间存活率、增重率、饲料系数、肥满度、肝体比及脏体比均无显著差异(P>0.05)。2)MO组的肌肉饱和脂肪酸和单不饱和脂肪酸含量显著高于其他各组(P<0.05),FO组的肌肉多不饱和脂肪酸、二十碳五烯酸和二十二碳六烯酸含量显著高于其他各组(P<0.05)。3)FO和BO组的前肠蛋白酶活性显著高于SO和TO组(P<0.05),FO组的中肠蛋白酶活性显著高于SO、TO和MO组(P<0.05)。BO组的前肠脂肪酶活性显著高于其他各组(P<0.05),FO组的中肠脂肪酶活性显著高于TO和MO组(P<0.05),MO组的后肠脂肪酶活性显著高于FO、SO和TO组(P<0.05)。BO组的前肠淀粉酶活性显著高于其他各组(P<0.05),BO组的中肠淀粉酶活性显著高于SO和FO组(P<0.05)。4)TO组的血清白蛋白含量显著高于FO和BO组(P<0.05),FO、SO和MO组的血清甘油三酯含量显著高于BO和TO组(P<0.05),TO组的血清总胆固醇和低密度脂蛋白含量显著高于其他各组(P<0.05),TO和MO组的血清溶菌酶含量显著高于其他各组(P<0.05)。5)SO和BO组的Shannon指数、Chao1指数和Ace指数显著高于其他各组(P<0.05),Simpson指数显著低于其他各组(P<0.05)。FO、BO和MO组肠道菌群组成和丰富度相似。6)FO组的肝脏脂肪合成酶(FAS)和脂蛋白酯酶(LPL)活性显著高于其他各组(P<0.05)。FO和MO组的肝脏FAS mRNA相对表达量显著高于其他各组(P<0.05),MO组的肝脏LPL mRNA相对表达量显著高于其他各组(P<0.05)。由此可见,饲料中添加虫油(黑水虻油、黄粉虫油和蝇蛆油)不会影响鱼体生长,并且添加黑水虻油、蝇蛆油可以一定程度上提高肠道消化酶活性,且与鱼油的肠道菌群组成和丰富度相似,能够保证鱼体正常的脂质水平,并且提高机体脂质代谢酶活性。 展开更多

为建立一种简便、快捷的现地检测锦鲤疱疹病毒(KHV)的方法,本研究结合重组酶介导扩增(RAA)和侧向流试纸条(LFD)技术,根据RAA引物设计原则,以KHV Sph基因保守区为靶位点,设计5对引物并筛选出5号引物作为最优引物,在其下游引物、探针的5&#... 为建立一种简便、快捷的现地检测锦鲤疱疹病毒(KHV)的方法,本研究结合重组酶介导扩增(RAA)和侧向流试纸条(LFD)技术,根据RAA引物设计原则,以KHV Sph基因保守区为靶位点,设计5对引物并筛选出5号引物作为最优引物,在其下游引物、探针的5'端分别标记Biotin、FAM荧光基因。采用方阵法对RAA反应时间和温度等条件优化,结果显示,该检测方法在34℃~40℃恒温反应10 min即可实现对KHV目的基因片段的有效扩增,并且扩增产物结果可直接观察,由此初步建立了KHV的RAA-LFD检测方法。特异性试验结果显示,建立的RAA-LFD检测方法与鲤浮肿病毒(CEV)、鲫造血器官坏死病毒(CHNV)、流行性造血器官坏死病毒(EHNV)、蛙虹彩病毒(BIV)等常见水生动物病原核酸均无交叉反应。敏感性试验结果显示,该方法对重组质粒标准品p EASY-KHV的检测限为50拷贝/μL,与行业标准常规PCR的敏感性相当。重复性试验结果显示,该方法对5份p EASY-KHV的检测结果均一致;对室温放置7 d的p EASY-KHV检测结果也均一致。采用该方法和常规PCR对60份临床样品检测,结果显示二者的阴性、阳性符合率以及总符合率均为100%。本研究首次建立了用于检测KHV的RAA-LFD方法,该方法特异性强、敏感性高、重复性好、操作方便,可用于KHV的临床样品检测,为KHV现地检测提供了一种简便、快捷的技术手段。 展开更多

运气信念对在线抽奖促销参与意愿的影响:概率与价值视角

中奖概率和奖品价值是影响个体参与抽奖意愿的两个关键维度。基于此,设计出“小概率/大价值”和“大概率/小价值”两种抽奖组合,并设计了三个心理学实验,以揭示在线抽奖概率/价值维度、运气信念和不中次数对个体参与抽奖意愿的影响。研... 中奖概率和奖品价值是影响个体参与抽奖意愿的两个关键维度。基于此,设计出“小概率/大价值”和“大概率/小价值”两种抽奖组合,并设计了三个心理学实验,以揭示在线抽奖概率/价值维度、运气信念和不中次数对个体参与抽奖意愿的影响。研究结果显示:(1)相比大概率/小价值组合,人们普遍更偏爱小概率/大价值组合,从而验证了在线抽奖决策过程存在小概率高估的事实,也阐明了人们热衷参与小概率/大价值在线抽奖活动的基本原理。(2)运气信念对人们的在线抽奖选择具有一定调节作用,运气信念低的个体更倾向于选择大概率/小价值的在线抽奖活动,运气信念高的个体则更喜欢小概率/大价值的锦鲤类抽奖活动。(3)对多次不中仍继续参与在线抽奖的现象进行研究,发现运气信念与奖品价值/概率在初次参与抽奖时存在交互效应,但多次参与抽奖不中后这种交互效应消失,此时即使高信念水平的个体也不再热衷于大价值/小概率的抽奖。这一结论为企业更好地开展抽奖促销活动、消费者更理性的抽奖决策以及政府对抽奖销售更有效的监管均提供了重要启示。 展开更多

为建立一种快速、灵敏并适用于临床检测Ⅲ型鲤疱疹病毒(CyHV-3,KHV)的方法,实验根据KHV SphI-5基因的保守序列片段设计引物及探针,采用重组酶聚合酶扩增技术结合侧流层析试纸条(RPA-LFD)检测KHV。重组酶聚合酶扩增技术(RPA)具恒温扩增... 为建立一种快速、灵敏并适用于临床检测Ⅲ型鲤疱疹病毒(CyHV-3,KHV)的方法,实验根据KHV SphI-5基因的保守序列片段设计引物及探针,采用重组酶聚合酶扩增技术结合侧流层析试纸条(RPA-LFD)检测KHV。重组酶聚合酶扩增技术(RPA)具恒温扩增及高灵敏度特点,简化了设备要求的同时又能做到高效检测病毒,再结合侧向流动试纸条(LFD)将RPA结果快速地可视化,提高了该疾病的检测效率。结果表明,在38℃的最适反应温度下,采用RPA-AGE技术仅需10min便可检测出病原的目标片段,结合LFD方法仅需5min便可将RPA结果通过试纸条可视化呈现。研究研发的KHV RPA-LFD检测方法简单、快捷,可为实验条件有限的养殖场快速诊断需求提供技术支撑。 展开更多
