Effects of Root Growth of Deep and Shallow Rooting Rice Cultivars in Compacted Paddy Soils on Subsequent Rice Growth

Rice is often grown as multiple seasons in one year,alternating between flooded and upland systems.A major constraint,introduced from the flooded system,is a plough pan that may decrease rooting depth and productivity of follow-on upland rice.Roots penetrating the plough pan under flooded rice system can leave a legacy of weaker root growth pathways.Deeper rooting rice cultivars could have a bigger impact,but no direct evidence is available.To explore whether a deep rather than a shallow rooting rice cultivar grown in a flooded cropping cycle benefited deeper root growth of follow-on rice in an upland,reduced tillage cropping cycle,a simulated flooded paddy in greenhouse was planted with deep(Black Gora) and shallow(IR64) rooting cultivars and a plant-free control.Artificial plough pans were made in between the topsoil and subsoil to form different treatments with no plough pan(0.35 MPa),soft plough pan(1.03 MPa) and hard plough pan(1.70 MPa).After harvest of this ‘first season’ rice,the soil was drained and undisturbed to simulate zero-tillage upland and planted rice cultivar BRRI Dhan 28.The overall root length density(RLD),root surface area,the numbers of root tips and branching of BRRI Dhan 28 did not vary between plough pan and no plough pan treatments.Compared with the shallow rooting rice genotype,the deep rooting rice genotype as ‘first season’ crop produced 19% greater RLD,34% greater surface area and 29% more branching of BRRI Dhan 28 in the subsoil.In the topsoil,however,BRRI Dhan 28 had 28% greater RLD,35% greater surface area and 43% more branching for the shallow rather than deep rooting genotype planted in the ‘first season’.The results suggested that rice cultivar selection for a paddy cycle affects root growth of a follow-on rice crop grown under no-till,with benefits to subsoil access from deep rooting cultivars and topsoil proliferation for shallow rooting cultivars. 展开更多

Dalbergia sissoo Roxb. is one of the promising multipurpose tree species of South Asia. Most of the plantations of D. sissoo from seeds are facing severe threats due to the die-back disease, which ultimately causes death of this potential tree-species within a few months. Vegetative propagation could avoid the die-back disease. Thirty mother trees of different age-groups of D. sissoo were selected for evaluating the rooting behaviour of branch cuttings from D. sissoo as influenced by auxins (IAA or IBA at 100, 200, 500 mg·L^-1), ages of mother trees (10, 4 and 2 years old) and different environment conditions, i.e., different mediums (soil and sand) or light conditions (in shade and open condition). The results show that application of IAA and IBA induced more numbers of cuttings (collected from 10-year-old mother trees) to root compared to control. Branch cuttings of D. sissoo collected from 10-year-old mother trees and planted in soil bed in open conditions had 100.0% of cuttings to root in IAA (100 mg·L^-1) and IBA (200 mg·L^-1) treatments. Both rooting medium (Soil and sand) influenced significantly (p〈0.05) on rooting response of branch cuttings. Soil medium was found to achieve maximum no. of branch cuttings to root, compared to sand medium. 展开更多

Results show that IBA could significantly enhance rooting and root characteristics of cuttings and were best with IBA at 1500 mg/L. In the rooting process, the formation of adventitious roots was related to the consumption and accumulation of nutrients(soluble sugars and proteins) and the changes in endogenous hormones in phloem, leaf tips and leaf bases. The rooting of cuttings had a positive correlation with the consumption of soluble sugars during the period of callus formation and with the accumulation of soluble sugars during adventitious root formation and growth. Rooting was positively related to the breakdown of soluble proteins in the phloem when the callus formed, and had a positive correlation with its accumulation during adventitious root formation and growth. Leaf tips and leaf bases showed a reverse trend in changes of soluble protein. However, together with the phloem, leaf tips and leaf bases regulated and controlled the formation and development of adventitious roots. The main activities of soluble proteins exist in the leaf tips as this was the main source of soluble proteins. The relation between rooting and IAA(indole-3-acetic acid) content in phloem was positive and thus a high concentration of IAA could benefit the induction and formation of adventitious roots. However, rooting was negatively related with ABA(abscisic acid) and GA(gibberellic acid) and a high concentration of both could inhibit the induction and formation of adventitious roots. Rooting had a positive correlation with phloem IAA/ABA ratios, and higher ratios could improve rooting. Low concentrations of ZR(zeatin riboside) triggered the induction of adventitious roots, while higher concentrations promoted root growth.Endogenous hormones in leaf tips and bases had an impact on rooting. The activities of endogenous hormones mainly existed in leaf tips because they play a major role in the production and consumption of IAA and its ABA content increased during rooting. The ZR in leaf tips influenced the rooting of cuttings, especially in the callus formation and rooting stage. Leaf tips were the main source of GA. 展开更多

Picea crassifolia Kom, a perennial arbor spe- cies is recognized as one of the most adaptable plants found to date in Qilian Mountains. To explore the cutting reproduction technology of P. crassifolia and reveal its rooting mechanism, cuttings of P. crassifolia with different cutting types (softwood, hardwood and root), positions (top, upper, middle and bottom) and ages (7, 10, 15, 20, 25 year-old) were cultivated in a field experiment. One- year old softwood and hardwood cuttings were collected from 7-, 10-, 15-, 20-, and 25 year-old healthy ortets to analyze the changes from endogenous hormones and organic nutrients. Results indicate that the softwood cut- tings (0.5-1.0 cm in diameter) from upper branches of 15 year-old ortets shows better growth performance by improving rooting indexes, including a significant increasein rooting rate and a decrease in basal rot rate. Concomi- tantly, increasing rooting quantity and root length also increased. It is noteworthy that the high rooting rate of P. crassifolia cuttings due to its ability to accumulate high concentrations of indole-3-acetic acid (IAA) and total carbon (TC) rather than abscisic acid (ABA) and total nitrogen (TN). The rooting rate was mainly regulated by the IAA/ABA and TC/TN ratio. In summary, our results suggest that the softwood cuttings (0.5-1.0 cm in diameter) from upper branches of 15 year-old P. crassifolia can be considered as an effective strategy to improve cutting rooting rate, and the IAA/ABA and TC/TN ratio was one of the main factors limiting the cutting rooting rate of P. crassifolia. 展开更多

The rooting rate, the number of root and the length of root were studied after transplanting 20 d. The activity of peroxidase, nitrate reductase and fresh weight of roots were determined after transplanting 44 d. The results show that the optimum concentration range of La 3+ (1.0~3.0 μmol·L -1), Eu 3+ (2.0~3.0 μmol·L -1) in the rooted medium can increase the rooting rate and the fresh weight of roots, and promote the length of root and raise the activities of peroxidase and nitrate reductase significantly. La 3+ has more effect in improving the rooting rate, root length and the activities of peroxidase and less effect in promoting root fresh weight and the activities of nitrate reductase than Eu 3+. 展开更多

The effects of neodymium nitrate (Nd^3+) on the adventitious rooting of Dendrobium densiflorum shoot cuttings were studied. The addition of Nd^3+ (5 μmol/L) to culture medium significantly increased rooting frequency. Histological investigation showed that Nd^3+ did not change the process of root initiation. Nd^3+ did not influence total endogenous cytokinin levels, but significantly increased the level of endogenous indole-3-acetic acid (IAA) in the base of shoot cuttings. Compared to the control, the ratio of IAA/cytokinins was very high in the Nd^3+ treatment. These results suggested that the enhanced rooting frequency may be related to the increase in endogenous IAA level in Nd^3+ treatment. Analysis of enzyme activities showed that the enhanced accumulation of the endogenous IAA by Nd^3+ should not be attributed to inhibition of IAA decomposition by IAA oxidase or promotion of cytokinin decomposition by cytokinin oxidase. Besides, Nd^3+ increased the ratio of reduced glutathione (GSH) and oxidized glutathione (GSSG) in the process of adventitious rooting while the ratio of ascorbate (ASC) to dehydroascorbate (DHA) was not affected. 展开更多

Vegetative propagation is widely used as a means of genotype preservation in clone banks and seed orchards but despite this, the possibility of vegetative propagation using hormones is not clear for many indigenous tree species, including Balanites aegyptiaca. This research is on the vegetative propagation of B. aegyptiaca as influenced by rooting media and growth hormones.Juvenile stem cuttings were obtained from 4 months old seedlings. Nodal stem cuttings 4–5 cm long from 3–5 mm diameter stems were treated with indole-3-butyric acid or indole-3-acetic acid at 50, 100 and 150 mg/L and sown in three different rooting media: top soil, river sand, and sawdust. The results reveal a significant effect(p \ 0.05)of rooting media and growth hormones on B. aegyptiaca cuttings. B. aegyptiaca can be propagated vegitatively without the use of hormones. The control was recommended for vegetative propagation of B. aegyptiaca. 展开更多

The influence of low-vacuum helium cold plasma treatment on the rooting percentage,root growth and physiochemical properties of zoysiagrass stolon cuttings was studied.Zoysiagrass stolon cuttings were pre-treated with 0,100,200,300 and 400 W of cold plasma for 15 s.The cold plasma positively stimulated rooting and improved the root growth of the zoysiagrass stolon cuttings,and the 300 W treatment produced the best effect.The rooting percentage and root growth parameters,including the root length,total root surface area,total root volume,average root diameter,and root dry weight,significantly improved in response to the cold plasma treatment.In addition,the water uptake and relative conductivity of the stolon cuttings increased significantly in response to the cold plasma treatment.The results revealed that cold plasma-stimulated rooting and root growth appear to be a consequence of the improvement in permeability and water absorbing capacity of zoysiagrass stolon cuttings.The results of the present study will provide inspiration and support for the application of cold plasma in the vegetative propagation of plants. 展开更多

We investigated the influence of season on the rooting be- haviour of eight important bamboo species viz., Bambusa balcooa, B. bambos, B. nutans, B. tuMa, B. vulgaris, Dendrocalamus giganteus, D. hamiltonii and D. strictus. We collected 2-3 node culm cuttings in three growing seasons viz. spring (March), summer (June) and rainy (August) from superior candidate plus clumps (CPCs) centralized in the germplasm garden. Cuttings were placed horizontally in sand and were provided with intermittent misting at regular intervals. Bamboo species exhibited differential rhizogenesis behavior in different seasons. The study reveals significant variation in sprouting and rooting behaviour in different bamboo species. B. bambos had maximum rooting (78.89%), followed by B. vulgaris (74.44%). In general, the potential of different bamboo species for rooting was found to be in the order: B. bambos 〉 B. vulgaris 〉 B. balcooa 〉 D. hamiltonii 〉 B. tulda 〉 D. strictus 〉 D. giganteus 〉B. nutans. The maximum rooting was recorded in spring (56.67%), which was closely followed by summer (54.58%). In winter season, minimum rooting (36.67%) was observed. The interaction effect of species x season was also significant on sprouting and rooting pa- rameters. In B. balcooa, B. nutans, B. vulgaris, D. hamiltonii and D. strictus, cuttings collected in summer season showed maximum sprouting and rooting, whereas, B. bambos, B. tulda and D. giganteus had maximum rooting in spring. Deep rooting is an important trait in rice drought resistance.Genetic resources of deep-rooting varieties are valuable in breeding of water-saving and drought-resistant rice.In the present study,234BC2F7 backcross introgression lines were derived from a cross of Dongye 80(an accession of Dongxiang wild rice as the donor parent)and R974(an indica restorer line as the recurrent parent).A genetic linkage map containing 1977 bin markers was constructed by ddRADSeq for QTL analysis.Thirty-one QTLs for four root traits(the number of deep roots,the number of shallow roots,the total number of deep roots and the ratio of deep roots)were assessed on six rice chromosomes in two environments(2020 Shanghai and 2021 Hainan).Two of the QTLs,qDR5.1 and qTR5.2,were located on chromosome 5 in a 70-kb interval.They were detected in both environments.qDR5.1 explained 13.35%of the phenotypic variance in 2020 Shanghai and 12.01%of the phenotypic variance in 2021 Hainan.qTR5.2 accounted for 10.88%and 10.93%of the phenotypic variance,respectively.One QTL(qRDR2.2)for the ratio of deep roots was detected on chromosome 2 in a 210-kb interval and accounted for 6.72%of the phenotypic variance in 2020.The positive effects of these three QTLs were all from Dongxiang wild rice.Furthermore,nine and four putative candidate genes were identified in qRDR2.2 and qDR5.1/qTR5.2,respectively.These findings added to our knowledge of the genetic control of root traits in rice.In addition,this study will facilitate the future isolation of candidate genes of the deep-rooting trait and the utilization of Dongxiang wild rice in the improvement of rice drought resistance. 展开更多

Tree peony is a famous ornamental plant,while the low propagation rate is the main hurdles hindering the industry development.Till now,the highly efficient regeneration system for tree peony is not established.In this study,using Paeonia ostii’Fengdan’mature embryos,the effects of variations in inoculation method,initiating culture,adventitious shoot induction,rooting media,plant growth regulators(PGRs),and a nonconventional PGR(plant extracts)on regeneration from explants were evaluated.In embryo cultures,embryonic callus induction rate of 1/4 embryos was the highest among those of embryos with other three technical treatments(whole embryos,1/2 embryos,and pieces of embryos).The woody plant medium(WPM)containing 1.0 mg·L^(-1)6-BA,0.5 mg·L^(-1)GA3,30.0 g·L^(-1)sucrose,and 3.0 g·L^(-1)phytagel significantly improved shoot induction and multiplication.3.0 mg·L^(-1)plant extracts promoted hypocotyl germination,rooting,and root growth,in direct embryo culture,and a combination of 3.0 mg·L^(-1)plant extracts+2.0 mg·L^(-1)IBA+1.5 mg·L^(-1)IAA produced optimal rooting induction rate for multiple shoots in direct embryo culture and indirect somatic embryogenesis.For the three in vitro micropropagation methods,the highest shoot proliferation coefficient(5.4±0.2)was obtained with indirect somatic embryogenesis.Fortunately,the propagation ability of shoots remains high,even when culture propagation was continued for more than two years.Thus,a reliable system for plant regeneration from mature embryos derived from P.ostii’Fengdan’callus and two direct embryo culture systems have been established.The novel regeneration system could facilitate uniform seedling production. 展开更多

Commercial preparation (diluted 1:10 and 1:20) gave extensive rooting within five days after treatment. The presence of leaf with the stem cutting was necessary for optimal rooting as defoliation drastically reduced rooting even when the rooting time was extended from 5 to 10 days. The presence of leaf also promoted the levels of putrescine three days after the hormone treatment just at the time of root emergence. Cuttings that did not have leaf did not show an increase in putrescine levels and did not produce roots. Spermidine levels also increased in the cuttings that rooted but there was no change in the levels of spermine in the cuttings that rooted. It appears that putrescine and spermidine play a role in regulating rooting in bitter melon cuttings. The vegetative propagation conditions described show that it is a practically viable technique for small growers to produce large number of plantlets at the farm for selection and cultivation. 展开更多

Family;Sapotaceae) indigenous to the Sudano-sahelian zone of Africa has great economic and ecological potential and attributes. Commercial cultivation of the tree is however, hampered by the poor rooting success of its planting material with adverse consequences on field establishment and total fruit yield. A 3 × 2 factorial experiment arranged in a randomized complete block design was carried out in 2012 at Bole in the Interior Savanna agro-ecological zone. The objective was to assess the rooting success of Shea shoots by the air-layering technique using two media types (palm fibre and Sphagnum moss) and three IBA hormone concentrations (0, 5000 and 10,000 ppm) under contrasting climatic (wet and dry) conditions. Layered shoots which were sprayed with 10,000 ppm IBA and wrapped with Sphagnum moss gave significantly (P < 0.05) higher rooting success in terms of more roots per cutting (73.3%) and longer roots per cutting (9.0 cm) than palm fibre at both 5000 ppm (30.0%;3.7 cm) and 10,000 ppm (46.7%;7.9 cm) concentrations. Higher rooting success was significantly achieved in the wet season than in the dry season. Sphagnum moss treated with 10,000 ppm IBA facilitated the translocation of higher levels of sugar and total free phenol (TFP) to the base of the layered shoots which resulted in significantly (P < 0.05) higher number of roots and better roots protection from fungal infection than the other treatments. Rooting of layered shoots was affected to a greater extent by low temperatures (Rooting = 836 – 34.2 Temp (low);R2 = 82%, p < 0.001) than by high temperatures (Rooting = 5250 – 175.0 Temp (high);R2 = 64.5%, p < 0.009). Rooting of layered shoots was also significantly and negatively affected by the spread of the canopy of the selected tree such that closed canopy trees resulted in higher and better rooting that open canopy trees (Rooting = 113.87-23.697(Canopy spread);R2 = 89%, p = 0.002;n = 9). Furthermore, significant and positive correlations were found between IBA concentration level and simple sugars (r = 0.99;P < 0.0001;n = 9) and also between IBA concentration level and total free phenols (r = 0.98;P < 0.0002;n = 9). The study concluded that to achieve high rooting success in the propagation of Shea nut tree by the air-layering technique, a combination of Sphagnum moss treated with 10,000 ppm of IBA hormone should be used and the whole set up carried out in the wet season. 展开更多

The effect of stock-plant etiolation on adventitious rooting of single-node leafy cuttings (SNCs) made from coppice shoots from different age groups of donor plants were investigated for teak (Tectona grandis Linn. f.). When donor plants age were 1-, 2-, 3-, 4-, and 5-year-old, they were coppiced and maintained in the dark for etiolation, while a parallel set was kept in normal light in an open environment. After 20 days, coppice shoots were made into SNCs which were cultured under intermittent mist for rooting. Stock-plant etiolation significantly increased percent rooting, shoot and root length, and number of roots per SNC, but callusing at the base of SNCs decreased. Etiolated SNCs have shown 71.7% rooting, whereas for controls (non-etiolated), the response was 41.7%. The effect of aging on callusing, rooting and sprouting of etiolated and non-etiolated coppice shoots cuttings varied widely. Aging of donor plants decreased rooting and sprouting capability in SNCs and increased callusing at the base of etiolated cuttings. The results showed that stock-plant etiolation in 1-, 2-, 3-, 4-, and 5-year-old donor plants hastened rooting potential by rejuvenation of coppice shoots. 展开更多

The present study, conducted during 2016 and 2017 seasons, aimed to investigate the effect of IBA on rooting of Piper betle L. stem cuttings (softwood and semi-hardwood). The experiment was undertaken in misting house field 2 UPM using the sand media to determine the adventitious roots initiation and development using the histological method. The cuttings were treated with different IBA concentrations (0, 500, 1000, 1500 and 2000 mg/L). The nodes explants were used in the development of a protocol for in vitro propagation of P. betle L., with different concentrations of Clorox with different times of immersion (20% Clorox 10 minutes, 30% Clorox 10 minutes, 20% Clorox 20 minutes, and 30% 20 minutes). In multiplication of the plantlets, Murashige and Skoog (MS) medium with different concentrations of BAP (0, 0.5, 1.0, 2.0 mg/L) were used to investigate the rooting of the explants. The results indicated that the types of the cuttings were different in the rooting capacity and the length of the roots. Moreover, it was found that in comparison with the control treatment, by a rise in the concentrations of the IBA, there was a significant upsurge in the rooting percentage, the root diameter, and the number of the roots. The results indicated that the types of cutting with 1000, 1500 and 2000 mg/L IBA perform better in the root percentage (100%) in the semi hardwood cuttings. The best results, however, were 2000 mg/L IBA in the semi hardwood cuttings, with the number of the roots to be 35.05, and the fresh weight of the roots to be 3.94 g, the dry weight of the roots to be 0.33 g, the length of the roots to be 391.88 cm, the roots diameter to be 1.21 mm, the surface area of the roots to be 121.83 cm2, and the root volume to be 2.99 cm3. Nonetheless, the optimal concentration of Clorox with the time immersion was 20% with the 20-minute immersion time, which produced a shoot induction percentage of 30% dead explants and a mean number of 70.00 shoots per explant and the optimal concentration of benzylaminopurine (BAP) at 1.0 mg/L. It is of note that a shoot induction percentage of 22.29% and a mean number of 4.1% number of auxiliary bud per treatment. P. betle shoots in MS medium without PGR MS (0.0) yielded a good rooting. 展开更多

Clonal propagation of eucalypts has considerable importance due to the increasing demands for short rotation tree crops.Rooting quality is important as it governs the soil exploitation capacity of the plant and the anchorage of trees which are susceptible to wind damage.This study assesses the quality of adventitious rooting by coppice cuttings of commercially important Eucalyptus clones using multiple attribute ranking of the diff erences in parameters of root growth.The eff ects of diff erent concentrations of indole-3-butyric acid(IBA)on root development were observed.Cuttings were treated with 500,1000,4000,6000 mg L−1 IBA and tannic acid for 10 s,2 h and 24 h.Total length of root systems,number of roots,shoot to root ratios,number of root segments,extent of forking,rooting percentage,average root diameters,and number of root tips were measured.Grey relational analysis was used to create a comparability sequence to rank treatments.Reducing the concentration of auxin(IBA)and increasing the length of exposure produced better quality roots in Eucalyptus camaldulensis and interspecifi c hybrids(reciprocal hybrids of E.tereticornis and E.camaldulensis),while the opposite was observed with E.tereticornis clones.A root quality index was proposed,based on the Dickson quality index for the assessment of root system characteristics and considered total mass,shoot:root ratios,total length of root systems,and average root diameters.It has the advantage of implementation convenience.A positive correlation was obtained between grey relational analysis grades and root quality index.Rooting dynamics were studied by evaluating the total length of the root system at seven-day intervals and plotting daily current and medium increments using regression analysis.The curves showed the variation in growth rates among the diff erent clones,and their intersection gave the optimal time of permanence(time at which further growth is restricted)which varied considerably.The highest daily current increment was 35–40 days for all clones. 展开更多

Robinia pseudoacacia ‘Idaho' (Robinia x ambigua ‘Idahoensis', R. pseudoacacia x R. viscosa) modified by a mtLD gene went through five lines and had characteristics of drought tolerance. Three stages of their micropropagation had been studied by pre- vious investigators. The other two stages, in vitro shoot rooting and plantlet acclimatization, still remained unsolved in the laboratory. For this paper, we studied the later two stages based on the previous achievements. Results showed that the highest rooting rate of Idaho locust was 98.4% when the in vitro shoots, over 2.5 cm in height and 0.08 cm in diameter, were placed on a half strength MS basal medium with 0.4 mg.U1 IBA and 0.1 mg'U1 NAA as supplements and were solidified with 0.5% agar; the highest survival rate was 98.3% when the rooted plantlets were potted in vermiculite. All the stages for micropropagation of the Idaho locust, modified by the mtl-D gene, were assembled completely. The tissue culture plants grow well in the field. 展开更多
