文摘White Hypsizygus marmoreus is a popular edible mushroom. Its mycelium is easy to be contaminated by Penicillium, which leads to a decrease in its quality and yield.Penicillium could compete for limited space and nutrients through rapid growth and produce a variety of harmful gases, such as benzene, aldehydes, phenols, etc.,to inhibit the growth of H.marmoreus mycelium.A series of changes occurred in H.marmoreus proteome after contamination when detected by the label-free tandem mass spectrometry(MS/MS)technique.Some proteins with up-regulated expression worked together to participate in some processes, such as the non-toxic transformation of harmful gases, glutathione metabolism, histone modification, nucleotide excision repair, clearing misfolded proteins, and synthesizing glutamine, which were mainly used in response to biological stress. The proteins with down-regulated expression are mainly related to the processes of ribosome function, protein processing, spliceosome, carbon metabolism, glycolysis, and gluconeogenesis. The reduction in the function of these proteins affected the production of the cell components, which might be an adjustment to adapt to growth retardation. This study further enhanced the understanding of the biological stress response and the growth restriction adaptation mechanisms in edible fungi. It also provided a theoretical basis for protein function exploration and edible mushroom food safety research.

文摘BACKGROUND Colorectal cancer is currently the third most common malignant tumor and the second leading cause of cancer-related death worldwide.Neoadjuvant chemoradiotherapy(nCRT)is standard for locally advanced rectal cancer(LARC).Except for pathological examination after resection,it is not known exactly whether LARC patients have achieved pathological complete response(pCR)before surgery. To date, there are no clear clinical indicators that can predict the efficacy of nCRT and patient outcomes.AIM To investigate the indicators that can predict pCR and long-term outcomes following nCRT in patients with LARC.METHODS Clinical data of 128 LARC patients admitted to our hospital between September 2013 and November 2022 were retrospectively analyzed.Patients were categorized into pCR and non-pCR groups.Univariate analysis(using the χ^(2) test or Fisher’s exact test)and logistic multivariate regression analysis were used to study clinical predictors affecting pCR.The 5-year disease-free survival(DFS)and overall survival(OS)rates were calculated using Kaplan-Meier analysis, and differences in survival curves were assessed with the log-rank test.RESULTS Univariate analysis showed that pretreatment carcinoembryonic antigen(CEA)level,lymphocyte-monocyte ratio(LMR),time interval between neoadjuvant therapy completion and total mesorectal excision, and tumor size were correlated with pCR.Multivariate results showed that CEA≤5 ng/mL(P=0.039),LMR>2.73(P=0.023),and time interval>10 wk(P=0.039)were independent predictors for pCR.Survival analysis demonstrated that patients in the pCR group had significantly higher 5-year DFS rates(94.7%vs 59.7%,P=0.002)and 5-year OS rates(95.8%vs 80.1%,P=0.019)compared to the non-pCR group.Tumor deposits(TDs)were significantly correlated with shorter DFS(P=0.002)and OS(P<0.001).CONCLUSION Pretreatment CEA,LMR,and time interval contribute to predicting nCRT efficacy in LARC patients.Achieving pCR demonstrates longer DFS and OS.TDs correlate with poor prognosis.

文摘Inflammation is a multifaceted cellular and molecular response triggered by injury, infection,or various pathological conditions.Serving as a protective defense mechanism, the inflammatory response involves clinical signs like redness, swelling, pain, and increased body temperature.Immune cells, notably neutrophils and macrophages, play key roles in orchestrating this response. The delicate balance between proinflammatory and anti-inflammatory mediators, including cytokines and chemokines, regulates the inflammatory cascade. While acute inflammation is crucial for tissue repair, chronic inflammation may indicate an imbalance, contributing to conditions like autoimmune diseases.Understanding these mechanisms is vital for developing therapeutic strategies and managing chronic diseases.

文摘Thermo-poro-mechanical responses along sliding zone/surface have been extensively studied. However,it has not been recognized that the potential contribution of other crucial engineering geological interfaces beyond the slip surface to progressive failure. Here,we aim to investigate the subsurface multiphysics of reservoir landslides under two extreme hydrologic conditions(i.e.wet and dry),particularly within sliding masses.Based on ultra-weak fiber Bragg grating(UWFBG)technology,we employ specialpurpose fiber optic sensing cables that can be implanted into boreholes as“nerves of the Earth”to collect data on soil temperature, water content, pore water pressure, and strain. The Xinpu landslide in the middle reach of the Three Gorges Reservoir Area in China was selected as a case study to establish a paradigm for in situ thermo-hydro-poro-mechanical monitoring. These UWFBG-based sensing cables were vertically buried in a 31 m-deep borehole at the foot of the landslide, with a resolution of 1 m except for the pressure sensor. We reported field measurements covering the period 2021 and 2022 and produced the spatiotemporal profiles throughout the borehole. Results show that wet years are more likely to motivate landslide motions than dry years. The annual thermally active layer of the landslide has a critical depth of roughly 9 m and might move downward in warmer years. The dynamic groundwater table is located at depths of 9e15 m,where the peaked strain undergoes a periodical response of leap and withdrawal to annual hydrometeorological cycles. These interface behaviors may support the interpretation of the contribution of reservoir regulation to slope stability, allowing us to correlate them to local damage events and potential global destabilization. This paper also offers a natural framework for interpreting thermo-hydro-poro-mechanical signatures from creeping reservoir bank slopes, which may form the basis for a landslide monitoring and early warning system.

文摘Underground hydrogen storage(UHS)and compressed air energy storage(CAES)are two viable largescale energy storage technologies for mitigating the intermittency of wind and solar power. Therefore,it is meaningful to compare the properties of hydrogen and air with typical thermodynamic storage processes. This study employs a multi-physical coupling model to compare the operations of CAES and UHS,integrating gas thermodynamics within caverns, thermal conduction, and mechanical deformation around rock caverns.Gas thermodynamic responses are validated using additional simulations and the field test data.Temperature and pressure variations of air and hydrogen within rock caverns exhibit similarities under both adiabatic and diabatic simulation modes.Hydrogen reaches higher temperature and pressure following gas charging stage compared to air,and the ideal gas assumption may lead to overestimation of gas temperature and pressure.Unlike steel lining of CAES,the sealing layer(fibre-reinforced plastic FRP)in UHS is prone to deformation but can effectively mitigates stress in the sealing layer. In CAES,the first principal stress on the surface of the sealing layer and concrete lining is tensile stress, whereas UHS exhibits compressive stress in the same areas. Our present research can provide references for the selection of energy storage methods.

文摘In order to better understand the specific substituent effects on the electrochemical oxidation process of β-O-4 bond, a series of methoxyphenyl type β-O-4 dimer model compounds with different localized methoxyl groups, including 2-(2-methoxyphenoxy)-1-phenylethanone, 2-(2-methoxyphenoxy)-1-phenylethanol, 2-(2-methoxyphenoxy)-1-(4-methoxyphenyl)ethanone, 2-(2-methoxyphenoxy)-1-(4-methoxyphenyl)ethanol, 2-(2,6-dimethoxyphenoxy)-1-(4-methoxyphenyl)ethanone, 2-(2,6-dimethoxyphenoxy)-1-(4-methoxyphenyl)ethanol have been selected and their electrochemical properties have been studied experimentally by cyclic voltammetry, and FT-IR spectroelectrochemistry. Combining with electrolysis products distribution analysis and density functional theory calculations, oxidation mechanisms of all six model dimers have been explored. In particular, a total effect from substituents of both para-methoxy(on the aryl ring closing to Cα) and Cα-OH on the oxidation mechanisms has been clearly observed, showing a significant selectivity on the Cα-Cβbond cleavage induced by electrochemical oxidations.

文摘Mg-Gd-Y-Zr alloy,as a typical magnesium rare-earth(Mg-RE)alloy,is gaining popularity in the advanced equipment manufacturing fields owing to its noticeable age-hardening properties and high specific strength. However,it is extremely challenging to prepare wrought components with large dimensions and complex shapes because of the poor room-temperature processability of Mg-Gd-Y-Zr alloy.Herein,we report a wire-arc directed energy deposited(DED)Mg-10.45Gd-2.27Y-0.52Zr(wt.%,GW102K)alloy with high RE content presenting a prominent combination of strength and ductility, realized by tailored nanoprecipitates through an optimized heat treatment procedure.Specifically, the solution-treated sample exhibits excellent ductility with an elongation(EL)of(14.6±0.1)%,while the aging-treated sample at 200°C for 58 h achieves an ultra-high ultimate tensile strength(UTS)of(371±1.5)MPa.Besides,the aging-treated sample at 250°C for 16 h attains a good strength-ductility synergy with a UTS of(316±2.1)MPa and a EL of(8.5±0.1)%.Particularly,the evolution mechanisms of precipitation response induced by various aging parameters and deformation behavior caused by nanoprecipitates type were also systematically revealed. The excellent ductility resulted from coordinating localized strains facilitated by active slip activity. And the ultra-high strength should be ascribed to the dense nano-β'hampering dislocation motion.Additionally, the shearable nano-β1 contributed to the good strength-ductility synergy. This work thus offers insightful understanding into the nanoprecipitates manipulation and performance tailoring for the wire-arc DED preparation of large-sized Mg-Gd-Y-Zr components with complex geometries.

文摘This study aims to investigate mechanical properties and failure mechanisms of layered rock with rough joint surfaces under direct shear loading.Cubic layered samples with dimensions of 100 mm×100 mm×100 mm were casted using rock-like materials, with anisotropic angle(α)and joint roughness coefficient(JRC)ranging from 15°to 75°and 2-20,respectively.The direct shear tests were conducted under the application of initial normal stress(σ_(n)) ranging from 1-4 MPa.The test results indicate significant differences in mechanical properties, acoustic emission(AE)responses, maximum principal strain fields, and ultimate failure modes of layered samples under different test conditions. The peak stress increases with the increasingαand achieves a maximum value atα=60°or 75°.As σ_(n) increases, the peak stress shows an increasing trend, with correlation coefficients R² ranging from 0.918 to 0.995 for the linear least squares fitting. As JRC increases from 2-4 to 18-20,the cohesion increases by 86.32%whenα=15°,while the cohesion decreases by 27.93%whenα=75°.The differences in roughness characteristics of shear failure surface induced byαresult in anisotropic post-peak AE responses, which is characterized by active AE signals whenαis small and quiet AE signals for a largeα.For a given JRC=6-8 andσ_(n)=1 MPa,asαincreases, the accumulative AE counts increase by 224.31%(αincreased from 15°to 60°),and then decrease by 14.68%(αincreased from 60°to 75°).The shear failure surface is formed along the weak interlayer whenα=15°and penetrates the layered matrix whenα=60°.Whenα=15°,as σ_(n) increases, the adjacent weak interlayer induces a change in the direction of tensile cracks propagation, resulting in a stepped pattern of cracks distribution. The increase in JRC intensifies roughness characteristics of shear failure surface for a smallα,however,it is not pronounced for a largeα.The findings will contribute to a better understanding of the mechanical responses and failure mechanisms of the layered rocks subjected to shear loads.

文摘This study is the result of long-term efforts of the authors’team to assess ground response of gob-side entry by roof cutting(GSERC)with hard main roof, aiming at scientific control for GSERC deformation.A comprehensive field measurement program was conducted to determine entry deformation, roof fracture zone, and anchor bolt(cable)loading. The results indicate that GSERC deformation presents asymmetric characteristics. The maximum convergence near roof cutting side is 458 mm during the primary use process and 1120 mm during the secondary reuse process. The entry deformation is closely associated with the primary development stage, primary use stage, and secondary reuse stage. The key block movement of roof cutting structure,a complex stress environment, and a mismatch in the supporting design scheme are the failure mechanism of GSERC.A controlling ideology for mining states, including regional and stage divisions, was proposed. Both dynamic and permanent support schemes have been implemented in the field.Engineering practice results indicate that the new support scheme can efficiently ensure long-term entry safety and could be a reliable approach for other engineering practices.

文摘The predominant presence of weak interlayers primarily composed of mudstone renders them highly susceptible to a reduction in bearing capacity due to the water-rock weakening effect, significantly impacting the safety of open-pit mining operations. This study focuses on the weak mudstone layers within open-pit mine slopes. The mineral composition of mudstone and the microstructure evolution characteristics before and after water wetting were analyzed by X-ray diffraction(XRD)and scanning electron microscope(SEM).The meso-structure and parameter variation characteristics of mudstone interior space after water-rock interaction were quantified by computed tomography scanning test, and the damage variable characterization method was proposed.Additionally, according to the uniaxial compression test, the degradation characteristics of the macroscopic mechanical behavior of mudstone under different water wetting time were explored, and the elastic modulus and strength attenuation model of mudstone based on mesoscopic damage were established. Finally, building upon the macro-meso structural response characteristics of mudstone,an exploration of the failure characteristics and deterioration mechanism under the influence of water-rock interactions was undertaken. The results show that the water-rock interaction makes the internal defects of mudstone gradually develop and form a fracture network structure, which eventually leads to the deterioration of its macroscopic mechanical properties. The porosity, fractal dimension and damage characteristics of mudstone show an exponential trend with the increase of water wetting time. Moreover, the deterioration mechanism of mudstone after water wetting are postulated to encompass factors such as the hydrophilicity of mineral molecular structures, hydration stress and expansion effects on clay particles,as well as the spatial distribution of microcracks and the phenomenon of fracture adsorption. The outcomes of this research endeavor aim to provide certain reference value for further understanding the water-rock interaction and stability control of mudstone slope.

文摘On December 18,2023,an M_(s)6.2 earthquake occurred in Jishishan,Gansu Province,China. This earthquake happened in the eastern region of the Qilian Orogenic Belt, which is situated at the forefront of the NE margin of the Tibetan Plateau(i.e.,Qinghai-Tibet Plateau),encompassing a rhombic-shaped area that intersects the Qilian-Qaidam Basin,Alxa Block,Ordos Block, and South China Block. In this study,we analyzed the deep tectonic pattern of the Jishishan earthquake by incorporating data on the crustal thickness, velocity structure, global navigation satellite system(GNSS)strain field, and anisotropy. We discovered that the location of the earthquake was related to changes in the crustal structure. The results showed that the Jishishan M_(s)6.2 earthquake occurred in a unique position, with rapid changes in the crustal thickness,Vp/Vs,phase velocity, and S-wave velocity. The epicenter of the earthquake was situated at the transition zone between high and low velocities and was in proximity to a low-velocity region.Additionally, the source area is flanked by two high-velocity anomalies from the east and west. The principal compressive strain orientation near the Lajishan Fault is primarily in the NNE and NE directions, which align with the principal compressive stress direction in this region. In some areas of the Lajishan Fault, the principal compressive strain orientations show the NNW direction, consistent with the direction of the upper crustal fast-wave polarization from local earthquakes and the phase velocity azimuthal anisotropy. These features underscore the relationship between the occurrence of the Jishishan M_(s)6.2 earthquake and the deep inhomogeneous structure and deep tectonic characteristics. The NE margin of the Tibetan Plateau was thickened by crustal extension in the process of northeastward expansion, and the middle and lower crustal materials underwent structural deformation and may have been filled with salt-containing fluids during the extension process. The presence of this weak layer makes it easier for strong earthquakes to occur through the release of overlying rigid crustal stresses. However,it is unlikely that an earthquake of comparable or larger magnitude would occur in the short term(e.g.,in one year)at the Jishishan east margin fault.

文摘The effects of deformation temperature on the transformation-induced plasticity(TRIP)-aided 304L,twinning-induced plasti-city(TWIP)-assisted 316L,and highly alloyed stable 904L austenitic stainless steels were compared for the first time to tune the mechan-ical properties, strengthening mechanisms, and strength-ductility synergy. For this purpose, the scanning electron microscopy(SEM),electron backscattered diffraction(EBSD),X-ray diffraction(XRD),tensile testing,work-hardening analysis, and thermodynamics calcu-lations were used. The induced plasticity effects led to a high temperature-dependency of work-hardening behavior in the 304L and 316L stainless steels. As the deformation temperature increased, the metastable 304L stainless steel showed the sequence of TRIP,TWIP,and weakening of the induced plasticity mechanism;while the disappearance of the TWIP effect in the 316L stainless steel was also observed. However, the solid-solution strengthening in the 904L superaustenitic stainless steel maintained the tensile properties over a wide temper-ature range, surpassing the performance of 304L and 316L stainless steels. In this regard, the dependency of the total elongation on the de-formation temperature was less pronounced for the 904L alloy due to the absence of additional plasticity mechanisms. These results re-vealed the importance of solid-solution strengthening and the associated high friction stress for superior mechanical behavior over a wide temperature range.

文摘The 2D limit equilibrium method is widely used for slope stability analysis. However, with the advancement of dump engineering, composite slopes often exhibit significant 3D mechanical effects.Consequently,it is of significant importance to develop an effective 3D stability calculation method for composite slopes to enhance the design and stability control of open-pit slope engineering.Using the composite slope formed by the mining stope and inner dump in Baiyinhua No.1 and No.2 open-pit coal mine as a case study, this research investigates the failure mode of composite slopes and establishes spatial shape equations for the sliding mass.By integrating the shear resistance and sliding force of each row of microstrip columns onto the bottom surface of the strip corresponding to the main sliding surface,a novel 2D equivalent physical and mechanical parameters analysis method for the strips on the main sliding surface of 3D sliding masses is proposed.Subsequently,a comprehensive 3D stability calculation method for composite slopes is developed, and the quantitative relationship between the coordinated development distance and its 3D stability coefficients is examined. The analysis reveals that the failure mode of the composite slope is characterized by cutting-bedding sliding, with the arc serving as the side interface and the weak layer as the bottom interface, while the destabilization mechanism primarily involves shear failure. The spatial form equation of the sliding mass comprises an ellipsoid and weak plane equation. The analysis revealed that when the coordinated development distance is 1500 m,the error rate between the 3D stability calculation result and the 2D stability calculation result of the composite slope is less than 8%,thereby verifying the proposed analytical method of equivalent physical and mechanical parameters and the 3D stability calculation method for composite slopes.Furthermore,the3D stability coefficient of the composite slope exhibits an exponential correlation with the coordinated development distance, with the coefficient gradually decreasing as the coordinated development distance increases. These findings provide a theoretical guideline for designing similar slope shape parameters and conducting stability analysis.

文摘Through combined applications of the transfer-matrix method and asymptotic expansion technique,we formulate a theory to predict the three-dimensional response of micropolar plates. No ad hoc assumptions regarding through-thickness assumptions of the field variables are made, and the governing equations are two-dimensional,with the displacements and microrotations of the mid-plane as the unknowns.Once the deformation of the mid-plane is solved,a three-dimensional micropolar elastic field within the plate is generated, which is exact up to the second order except in the boundary region close to the plate edge.As an illustrative example, the bending of a clamped infinitely long plate caused by a uniformly distributed transverse force is analyzed and discussed in detail.

文摘Background:According to clinical practice guidelines, transarterial chemoembolization(TACE)is the standard treatment modality for patients with intermediate-stage hepatocellular carcinoma(HCC).Early prediction of treatment response can help patients choose a reasonable treatment plan. This study aimed to investigate the value of the radiomic-clinical model in predicting the efficacy of the first TACE treatment for HCC to prolong patient survival. Methods:A total of 164 patients with HCC who underwent the first TACE from January 2017 to September 2021 were analyzed. The tumor response was assessed by modified response evaluation criteria in solid tumors(mRECIST),and the response of the first TACE to each session and its correlation with overall survival were evaluated. The radiomic signatures associated with the treatment response were identified by the least absolute shrinkage and selection operator(LASSO),and four machine learning models were built with different types of regions of interest(ROIs)(tumor and corresponding tissues)and the model with the best performance was selected. The predictive performance was assessed with receiver operating characteristic(ROC)curves and calibration curves. Results:Of all the models, the random forest(RF)model with peritumor(+10 mm)radiomic signatures had the best performance[area under ROC curve(AUC)=0.964 in the training cohort,AUC=0.949 in the validation cohort].The RF model was used to calculate the radiomic score(Rad-score),and the optimal cutoff value(0.34)was calculated according to the Youden’s index.Patients were then divided into a high-risk group(Rad-score>0.34)and a low-risk group(Rad-score≤0.34),and a nomogram model was successfully established to predict treatment response. The predicted treatment response also allowed for significant discrimination of Kaplan-Meier curves.Multivariate Cox regression identified six independent prognostic factors for overall survival, including male[hazard ratio(HR)=0.500,95%confidence interval(CI):0.260–0.962,P=0.038],alpha-fetoprotein(HR=1.003,95%CI:1.002–1.004,P<0.001),alanine aminotransferase(HR=1.003,95%CI:1.001–1.005,P=0.025),performance status(HR=2.400,95%CI:1.200–4.800,P=0.013),the number of TACE sessions(HR=0.870,95%CI:0.780–0.970,P=0.012)and Rad-score(HR=3.480,95%CI:1.416–8.552,P=0.007).Conclusions:The radiomic signatures and clinical factors can be well-used to predict the response of HCC patients to the first TACE and may help identify the patients most likely to benefit from TACE.
