Effect of Fouling in Daya Bay Scallop chlamys nobilis (reeve) Cultivation:Food Competition and Nutrient Release 被引量:3

Fouling organisms may reduce growth and survival rate of the cultured species. Fouler are often filter feeders, so they are potential competitors for food resource with the cultured species. The present study was conducted to measure the impact of fouling on food uptake and nutrient release in April, 2006 in Daya Bay near Guangzhou, China. Results showed that fouling organisms had significant effect on food uptake and nutrient release. The chlorophyll a uptake rate of fouled scallops was 7.53 / Lh ±1.416 / Lh in April, significantly higher than those of cleaned scallops, Le., 4.23 / Lh±2.744 / Lh. The consumption of total particulate matter by fouled scallops in April was 5.52 / Lh± 0.818 / Lh; the corresponding results for cleaned scallops are 2.49 / Lh ±0.614 / Lh Fouling increased ammonia release significantly. The ammonia release rate of fouled scallops was 33.81/Lh ±7.699 / Lh in April, while cleaned scallops released 2.46 / Lh ±0.511 / Lh ammonia. Phosphate uptake of fouled scallops was 2.01± 0.699 / Lh and cleaned scallops released phosphate 6.01 / Lh + 0.876 / Lh in April. There was not significant difference in nitrate consumption between fouled and cleaned scallops. According to the phytoplankton classification of input and output water samples, fouled scallops filtered more phytoplankton species than cleaned scallops. Therefore, this study showed that fouling contributed much to phytoplankton depletion and concentration increase of ammonia in water. 展开更多

Correlation and path coefficient analysis for phenotypic traits of noble scallop Chlamys nobilis 被引量:2

In April of 2006, a base population of the noble scallop Chlamys nobilis was established by collecting parental breeders from the stocks in Wushi, Zhanjiang. In December of 2006, 200 individuals were randomly sampled from the base population and subjected for correlation and path coefficient analysis. It was found that there were statistically significant phenotypic correlations among the traits (P 〈 0.01). Total weight was significantly and positively correlated with the shell length (r = 0.934 3), shell height (r = 0.895 9), shell width (r = 0.899 1 ), muscle weight (r = 0.882 0) and shell weight (r = 0.937 9), respectively. Shell length, shell width, muscle weight, shell height and shell weight had positive and direct effects on the total weight, with values of 0.397 1, 0.321 9, 0.172 1, 0.089 6 and 0.066 9, respectively. Shell length, shell width and muscle weight had higher direct effects on the total weight than shell height and shell weight. A combined evaluation of correlation, direct effects and indirect effects showed that direct selection for shell length, shell width, muscle weight, shell height and shell weight would be effective to improving the total weight. It was concluded that these traits could be regarded as the selection criteria in breeding programs of the species. 展开更多

The present study was conducted to measure the impact of fouling on food uptake and nutrient release in April and June, 2006 in Daya Bay near Guangzhou, China. Results showed that fouling organisms had significant effect on food uptake and nutrient release. The chlorophyll a uptake rate of fouled scallops was 7.53 Lh-1 ± 1.416 Lh-1 and 11.94 Lh-1± 2.497 Lh-1 in April and June, respectively, significantly higher than those of cleaned scallops, i.e., 4.23 Lh-1 ± 2.744 Lh-1 and 2.57 Lh-1 ± 1.832 Lh-1 respectively. The consumption of total particulate matter by fouled scallops in April and June was 5.52 Lh-1 ± 0.818 Lh-1 and 3.07 Lh-1 ± 0.971 Lh-1, respectively; the corresponding results for cleaned scallops are 2.49 Lh-1 ± 0.614 Lh-1 and 2.37 ± 1.214 Lh-1, respectively. Fouling in- creased ammonia release significantly. The ammonia release rate of fouled scallops was 33.81 Lh-1±7.699 Lh-1 and 76.39 Lh-1 ± 9.251 Lh-1 in April and June, while cleaned scallops released 2.46 Lh-1 ± 0.511 Lh-1 and 7.23 Lh-1 ± 1.026 Lh-1 ammonia, respectively. Phos- phate release of fouled scallops was 22.72 Lh-1 ± 9.978 Lh-1 in June and cleaned scallops released phosphate 6.01 Lh-1 ± 0.876 Lh-1 in April. Therefore, fouling contributed much to food reduction and concentration increase of ammonia and phosphate in water. 展开更多

The processes of scallop meats can produce a large amount of by-products like scallop skirts,which are rich in nutrients. This work aimed to obtain the information about antioxidant peptides derived from scallop skirts of Mimachlamys nobilis. The hydro-lysates obtained by trypsin exhibited the highest antioxidant activity( 75.6%,79.5% and 76.7% using DPPH,hydroxyl and superoxide anion radical scavenging activity assays,respectively). The antioxidant peptides( MNAP) were separated and purified using UF membranes,GFC processes and RP-HPLC,and the antioxidant activity of MNAP was identified to be 85.7% using DPPH radical scavenging activity assay. The peptides( MNAP) were composed of two peptides with their molecular weights2 855.6 Da and 3 007.2 Da using MALDI-TOF-MS,respectively. The results suggested that the scallop skirts of Mimachlamys nobilis could be explored as a source of antioxidant peptides. 展开更多

采用单一自由度独立比较的方法,研究了两个栉-华杂交组合HCH(栉孔扇贝长岛群体×华贵栉孔扇贝)和HJH(栉孔扇贝胶南群体×华贵栉孔扇贝)以及栉孔扇贝种内交配组合ZZ的胚胎发生及幼虫生长发育过程。结果表明,栉孔扇贝和华贵栉孔... 采用单一自由度独立比较的方法,研究了两个栉-华杂交组合HCH(栉孔扇贝长岛群体×华贵栉孔扇贝)和HJH(栉孔扇贝胶南群体×华贵栉孔扇贝)以及栉孔扇贝种内交配组合ZZ的胚胎发生及幼虫生长发育过程。结果表明,栉孔扇贝和华贵栉孔扇贝的种间杂交F1在生长和发育的两个环节上是不一致的,在涉及器官及其功能形成的发育环节上,杂交表现负效应;而在涉及形态变化和增大的生长环节上,杂交表现正效应,存在杂种优势现象。两个杂交组合中,HJH综合效果较好,授精22h平均孵化率达到95.8%,与ZZ基本接近;30日龄平均变态率达61.5%,明显好于组合HCH的42.0%;8日龄幼虫平均大小达143.4μm×123.2μm,表现出更强的生长优势。这些结果为全面评价栉孔扇贝与华贵栉孔扇贝杂交体系应用价值和选配优良杂交组合提供了理论依据。 展开更多

Bivalve live in aquatic environment and the water temperature can affect their immunity directly.In this research,the scallop Chlamys farreri was injected with 10^4 or 10^7 CFU mL^-1 Vibrio anguillarum and cultured at 11℃,17℃,23℃,and 28℃,respectively.For the control scallop,only phosphate-buffered saline(PBS)was injected.Then total haemocytes and granulocytes were measured by ELISA using monoclonal antibodies.In the meantime,expressions of six immunity-related genes,including lipopolysaccharide andβ-1,3-glucan binding protein(CfLGBP),C-type lectin(CfLec-2),Toll-like receptor(Cf TLR),Lysozyme(CfLYZ),superoxide dismutase(SOD),and phenoloxidase(CfPO)in haemocytes were measured using quantitative real-time PCR.The results showed that total haemocytes counts in 10^4 CFU mL^-1 injection groups showed no differences compared to the control group at all temperatures.However,they varied significantly in 10^7 CFU mL^-1 injection groups at 3 h at 11℃,6–12 h at 17℃,3–48 h at 23℃,and 12–48 h at 28℃.Granulocytes counts in 10^4 CFU mL^-1 injection groups showed no variance compared to the control group at all temperatures,except for 12 h at 23℃,and 24–36 h at 28℃.They were significantly decreased in 10^7 CFU mL^-1 injection groups during 6–48 h at 11℃,12–48 h at 17℃,3–48 h at 23℃,and 3–72 h at 28℃.The expression levels of six immunity-related genes in haemocytes of 10^7 CFU mL^-1 injection groups were significantly higher than those of control group and 10^4 CFU mL^-1 injection groups at all temperatures.The results indicated that infected with high concentration of vibrios,haemocyte counts,granulocyte counts and the expressions of immunity-related genes in scallop C.farreri were significantly affected by environmental temperature. 展开更多

Scallop is a popular sea food and an important aquaculture shellfish.Identification of genes and genetic variants relating to scallop growth could benefit high-yielding scallop breeding.Myostatin(MSTN) is a conservative regulator of muscle growth,and has become one of the most important target genes for genetic improvement of the production of farmed animals.In this study,four single nucleotide polymorphisms(SNPs) were identified in the 5' flanking region of MSTN gene(Cf MSTN) in Zhikong scallop(Chlamys farreri).The association of these SNPs with scallop growth traits,including shell length,shell height,body weight and striated muscle weight was analyzed.The SNP g-1162G>T was found to associate with shell length,shell height,and striated muscle weight.The TT type scallops showed significantly higher trait values than those of GT type,and the GG type individuals exhibited median values.On the contrary,significantly more Cf MSTN transcripts were detected in the striated muscle of GT type scallops than in those of TT and GG type ones.Our results suggested that Cf MSTN might regulate the scallop muscle growth negatively,and SNP g-1162G>T can be used as a candidate marker for the selective breeding of high-yielding scallop. 展开更多

Sequence variation of the first internal transcribed spacer of ribosomal DNA ( ITS - 1 ) was examined and its application to the study of genetic variation was explored in four populations of farter' s scallop Chlamys farreri. ITS - 1 fragments, with a length of about 300 bp,of 78 individuals collected from Dalian, Qingdao, Yantai in China and Korea respectively were amplified via PCR, cloned and sequenced. Intra-genomic variation was examined by sequencing several clones of single individuals. Alignment and polymorphism analysis detected 44 haplotypes and 50 polymorphic sites which consist of 30 substitutions and 20 indels, indicating a high level of polymorphisms. Sequence analysis also showed a very low level of intra-individual variation. All these features validated the feasibility of application of ITS - 1 fragment to population analysis. Polymorphism analysis showed that the Korea sample has the richest genetic variation, followed by Yantai and Qingdao samples. AMOVA (analysis of molecular variance) showed that the majority (96.26%) of genetic variation was distributed within populations and 3.74% resulted from among populations, but with P 〈 0.05 ( = 0.042), indicating that the populations in this study have significant divergence. This output was basically concordant with the result arising from RAPD data and different from that from mitochondrial 16S rDNA sequence data. Wild populations of Chlamys farreri derived from Japan and China are used as parents in a cross breeding experiment conducted in Zhangzi Island of Liaoning Province in the early May of 2000. Four different first filial generations of the crossbreeding, namely JC, CJ, CC and JJ, which represent different cross combinations of the parents, are differentiated from each other in some observed quantitative traits including survival rate, hatchery rate and attachment rate, etc. By ANOVA analysis, significant differences in growth rate are found among the four populations of the offspring after one year of culturing in Shandong Province. JJ population shows the fastest growth rate and has shell height of 46.1±0.71(SD)mm in average, whereas the other three populations, CC, CJ and JC, has the shell height of 43.1±0.42mm, 41.5±0.48mm and 37.2 ±0.31mm, respectively. 以栉孔扇贝内脏团为原料,对其中抗氧化肽的制备、纯化及性质进行了研究。首先采用硫酸铵沉淀、DEAE-Sephadex A-50阴离子交换层析的方法,进行了扇贝提取蛋白的制备,其次采用海洋蛋白酶YS-80水解、SephadexG-25凝胶层析的方法,进行扇贝... 以栉孔扇贝内脏团为原料,对其中抗氧化肽的制备、纯化及性质进行了研究。首先采用硫酸铵沉淀、DEAE-Sephadex A-50阴离子交换层析的方法,进行了扇贝提取蛋白的制备,其次采用海洋蛋白酶YS-80水解、SephadexG-25凝胶层析的方法,进行扇贝抗氧化肽的制备,获得多肽粗品(海洋肽,PCF),并进一步采用CM Sepharose阳离子交换层析和反相高效液相色谱的方法,对扇贝抗氧化肽进行了纯化,结果得到组分PCF-3A,反相高效液相显示为单一峰。采用茚三酮反应、Sephadex G-15凝胶层析和AccQ.Tag氨基酸分析柱,进行了PCF-3A的理化性质研究。结果表明,茚三酮反应呈阳性;分子量约为794D;由7种氨基酸组成,分别为天冬氨酸、甘氨酸、苏氨酸、丙氨酸、缬氨酸、赖氨酸和脯氨酸。采用邻苯三酚自氧化体系和Fenton反应体系对PCF-3A的抗氧化活性进行了研究,发现该肽可有效清除羟自由基和超氧阴离子,半抑制浓度(IC50)分别为0.39mg/ml和2.85mg/ml,具有明显的抗氧化、抗衰老作用。 展开更多

采用组织化学、免疫组化和生物化学方法对栉孔扇贝血淋巴中的一氧化氮和一氧化氮合酶(EC1.14.13.39;NOS)进行了研究。结果表明,大部分血细胞呈NOS组化阳性,少量呈强阳性或弱阳性。神经型一氧化氮合酶(nNOS)和内皮型一氧化氮合酶(eNOS)... 采用组织化学、免疫组化和生物化学方法对栉孔扇贝血淋巴中的一氧化氮和一氧化氮合酶(EC1.14.13.39;NOS)进行了研究。结果表明,大部分血细胞呈NOS组化阳性,少量呈强阳性或弱阳性。神经型一氧化氮合酶(nNOS)和内皮型一氧化氮合酶(eNOS)在大部分血细胞呈阳性,且多在其细胞质局部集中分布;诱生型一氧化氮合酶(iNOS)在大部分血细胞也呈阳性,但多较均匀分布。分光光度测定结果表明,血清和血细胞中均存在NOS和NO,结构型一氧化氮合酶(cNOS,nNOS+eNOS)活力分别为2.87U和2.61U;iNOS活力分别为0.32U和0.45U;NO的含量分别为33.64和41.67μmol/g蛋白。病毒感染后,血清和血细胞中总一氧化氮合酶(tNOS)活力实验组均高于对照组,3h时差异显著,9h和24h时差异极显著;血清和血细胞中iNOS活力在3h、9h和24h时,实验组均高于对照组,且均差异极显著;血清和血细胞中NO含量在3h、9h和24h时,实验组也均高于对照组,且均差异极显著。栉孔扇贝血淋巴中存在的NO NOS体系可在抵抗病原生物感染方面发挥重要的作用。 展开更多

利用RAPD技术对栉孔扇贝(Chlamysfarreri)的四个自然群体(大连、烟台、青岛、韩国)的遗传多样性进行了分析。12个随机引物在四个群体中共检测到123个位点,其中多态位点109个,多态位点比例为88.62%;四个群体中均未检测到各群体特有的扩... 利用RAPD技术对栉孔扇贝(Chlamysfarreri)的四个自然群体(大连、烟台、青岛、韩国)的遗传多样性进行了分析。12个随机引物在四个群体中共检测到123个位点,其中多态位点109个,多态位点比例为88.62%;四个群体中均未检测到各群体特有的扩增带。韩国、烟台、大连、青岛四个群体的遗传多样性指数(Shannon指数)分别为0.2557、0.2557、0.2344和0.2249,以韩国和烟台群体遗传多样性较高,其次为大连群体,青岛群体最低,但它们之间无明显差异。不同群体的遗传多样性指数的平均数Hpop为0.2427,整个栉孔扇贝自然群体的遗传多样性指数Hsp为0.2503。遗传多样性剖分显示,绝大多数(97%)的遗传多样性是由群体内不同个体间的差异造成的,只有很小部分(3%)的遗传多样性与群体间的遗传分化有关。而采用基于单倍型(等位基因)之间的进化距离的AMOVA分析得到的这两个数据则分别为99%和1%,Φ值为0.010,且P=0.05,表明群体间遗传分化明显程度位于临界值。 展开更多

闭壳肌是华贵栉孔扇贝Chlamys nobilis(Reeve)的产品形式,它的大小成为选择育种的一个重要目标性状,由于其不易测性给选育工作带来困难。采用多元回归和通径分析来了解各经济性状对闭壳肌重的影响程度,从而找出与闭壳肌重的关联度最大... 闭壳肌是华贵栉孔扇贝Chlamys nobilis(Reeve)的产品形式,它的大小成为选择育种的一个重要目标性状,由于其不易测性给选育工作带来困难。采用多元回归和通径分析来了解各经济性状对闭壳肌重的影响程度,从而找出与闭壳肌重的关联度最大的性状作为选育目标性状是本研究的目的。随机挑取同一养殖群体中的152只1龄华贵栉孔扇贝,对其壳长、壳高、壳宽、铰合线长、活体重、软体部重和闭壳肌重等7个性状进行测定并计算性状间的相关系数。以闭壳肌重为依变量,其它性状为自变量进行多元回归和通径分析,计算出对应的通径系数和决定系数。结果表明,7个性状间的相关系数均达到极显著水平(P<0.01);通径分析表明软体部重对闭壳肌重的直接影响最大(0.712),其次是壳宽(0.394),其它经济性状对闭壳肌重的影响是通过软体部重、壳宽来间接实现的;决定系数分析表明其结果与通径分析呈现一致的变化趋势。经多元回归分析,逐步剔除偏回归系数不显著的壳高和铰合线长,建立了壳长、壳宽、活体重和软体部重对闭壳肌重的回归方程:Y=-4.403+0.032X1+0.243X2-0.035X3+0.228X4,R2=0.856。本研究为华贵栉孔扇贝的选择育种提供理论依据。 展开更多
