ShCTR1 Interacts with ShRBOH1 to Positively Regulate Aerenchyma Formation in Saussurea inversa through ROS Mediation

Saussurea inversa is one of the Tibetan medicine with developed aerenchyma.In this study,we investigated the function of the ShCTR1 gene related to the formation of aerenchyma.The study of the occurrence and formation of aerenchyma in S.inversa has certain theoretical significance in revealing the special mechanism of alpine plants adapting to the environment.The results of yeast two-hybrid experiments showed that S.inversa ShCTR1 interacted with the ShRBOH1,which is a key producer of reactive oxygen species(ROS),and it was hypothesized that ShCTR1 regulation of aerenchyma formation in S.inversa mainly mediated by ROS.Aerenchyma induction and inhibition experiments were carried out on S.inversa seedlings,it was shown that ROS inducer(AT)and Ethene(ETH)induced the formation of aerenchyma in S.inversa well in roots,stems,and leaves,and Ethylene inhibitor(1-MCP)and ROS inhibitor DPI(diphenylene iodonium)inhibited the formation of aerenchyma of S.inversa in different degrees,which proved that ROS and ethylene(ET)were positively correlated with the formation of aerenchyma.Meanwhile,AT and ETH well induced the expression of the ShCTR1 gene,and 1-MCP and DPI inhibited the expression of the ShCTR1 gene to different degrees,which proved that ROS and ET were also positively correlated with the expression of the ShCTR1 gene.ShCTR1 was transferred into tobacco(Nicotiana tabacum L.),then repeat the above experiment.It was shown that positive rate and porosity were lowest and highest after DPI and AT treatments,respectively,it showed a positive correlation,which proved that the ShCTR1 gene was positively regulating the formation of aerenchyma.Overexpression of the ShCTR1 gene increased the content of ROS,consequently,the regulation of aerenchyma formation by the ShCTR1 gene is mainly mediated by ROS.It suggested that the formation of aerenchyma in S.inversa was related to hypoxia in the alpine environment,and ShCTR1 interacts with ShRBOH1 to positively regulate the formation of aerenchyma mainly mediated by ROS. 展开更多

Objective:To investigate the anti-inflammatory effects of the total flavonoids from Saussurea involucrata on lipopolysaccharides(LPS)-stimulated murine RAW264.7 macrophages and explore its underlying mechanism of action.Methods:Total flavonoids from Saussurea involucrata were extracted using chromatographic column method.Cell viability was determined by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium bromide assay.The production of nitric oxide was detected by Griess assay and the release of cytokines(IL-10 and TNF-α)and chemokines(MCP-1,MIP-1α,and CCL5/RANTES)was determined by ELISA to evaluate the anti-inflammatory activity of total flavonoids from Saussurea involucrata.Moreover,nuclear translocation of p65,c-Jun,and IRF3 was detected by immunofluorescence microscopy and Western blotting analysis was performed to determine the expression of related proteins.Results:Total flavonoids extracted from Saussurea involucrata were 751.5 mg/g and the content of rutin was 506.5 mg/g.The production of inflammatory mediators including nitric oxide,cytokines,and chemokines was effectively inhibited by total flavonoids from Saussurea involucrata.Meanwhile,total flavonoids also suppressed the nuclear translocation of p65,c-Jun,and IRF3 in LPS-stimulated RAW264.7 cells.The LPS-induced expression of iNOS and COX-2 was remarkably reduced by treatment with total flavonoids from Saussurea involucrata.Moreover,total flavonoids decreased the expression levels of p-IKKα/β,p-TBK1,p-p38,p-ERK,p-JNK,p-p65,p-c-Jun,and p-IRF3 in LPS-exposed RAW264.7 macrophages.Conclusions:Total flavonoids from Saussurea involucrata potentially inhibit the secretion of pro-inflammatory mediators,which may be related to inhibition of p65,c-Jun,and IRF3 signaling pathways in LPS-stimulated RAW264.7 cells. 展开更多

Objective:The antimicrobial activity of the ethanol extract of the Auklandia(Saussurea lappa)root plant was investigated to verify its medicinal use in the treatment of microbial infections.Methods:The antimicrobial activity of the ethanol extract was tested against clinical isolates ofsome multidrug-resistant bacteria using the agar well diffusion method.Commercial antibioticswere used as positive reference standards to determine the sensitivity of the clinical isolates.Results:The extracts showed significant inhibitory activity against clinical isolates of methicillinresistantStaphylococcus aureus,Pseudomonas aeruginosa,Escherichia coli,Klebsiella pneumonia,Extended Spectrum Beta-Lactemase,Acinetobacter baumannii.The minimum inhibitory concentration values obtained using the agar dilution test ranged from 2.0μg/μL-12.0μg/μL.In the contrary the water extract showed no activity at all against the tested isolates.Furthermore,theresults obtained by examining anti-resistant activity of the plant ethanolic extract showed thatat higher concentration of the plant extract(12μg)all tested bacteria isolates were inhibited with variable inhibition zones similar to those obtained when we applied lower extract concentrationusing the well diffusion assay.Conclusion:The results demonstrated that the crude ethanolicextract of the Auklandia(Saussurea lappa)root plant has a wide spectrum of activity suggestingthat it may be useful in the treatment of infections caused by the above clinical isolates(humanpathogens). 展开更多

The present study established a mouse model of Alzheimer's disease, and investigated the effects of treatment with flavone extract from artificially cultivated saussurea. A positive control group was treated with estradiol benzoate, and learning and memory ability were examined in the 8-arm radial maze. The learning and recognition ability of mice with Alzheimer's disease treated with flavone extract was significantly improved and the number of hippocampal neurons was significantly increased in the flavone-treated and positive control groups compared with the model group. The results indicate that flavone extract from artificially cultivated saussurea can improve learning and memory deficits in mice with Alzheimer's disease, exerting effects similar to those of estradiol benzoate. 展开更多

Cottonhead windhairdaisy (Saussurea laniceps Hand.-Mazz.) is one of the most famous and important medicinal herbs in China. Illegal collection from wild populations is increasingly threatening the present environment of S. laniceps. Estab- lishment of an efficient method for micropropagation is the best way to change its endangered situation. When mature seeds of S. laniceps were cultured on hormone-free MS medium, plantlets were formed from germinated seeds in 7–10 d. Then 0.5 cm × 0.5 cm leaf explants were transplanted to MS medium supplemented with 1-naphthalene-acetic acid (NAA)/2,4-D and benzyladenine (BA)/KT and callus was achieved 10 d after transfer. Shoot bud regeneration occurred from callus cultured on MS medium supple- mented with different growth regulators 20 d after culturing. The regeneration percentages varied with the different components of plant growth regulators. The percent regeneration from callus pretreated at low temperature of 5°C increased significantly compared with those incubated at 23/20°C directly. Optimal regeneration was observed with explants on media supplemented with 1.5 mg·L–1 BA plus 0.2 mg·L–1 NAA. In the presence of 0.2 mg·L–1 NAA in half-strength MS, 78% of the shoots formed roots. Plantlets from explants showed 63% survival after acclimatization. 展开更多

Saussurea medusa,Saussurea hypsipeta and Saussurea obvallata are typical alpine snowline plants growing in the Qinghai-Tibet plateau.They are characterized by a specialized morphology.S.medusa and S.hypsipeta have very dense trichomes on whole plant,whereas S.obvallata has transparent bracts covered inflorescences.The different forms reflect their adaptation to cold environments.To investigate the different mechanisms of adaptation of these species to cold temperatures,transcriptome sequencing was performed in three species of Saussurea DC.A total of 116394137237 and 113879 Unigenes were identified from S.medusa,S.hypsipeta and S.obvallata,respectively.Of these,55909(48.03%),65519(47.74%)and 51889(45.56%)Unigenes were matched in public databases.GO analysis identified that most of annotated Unigenes in the three species of plants were related to cellular,metabolic,and single−organism processes,and binding and catalytic activities.The differential expression of 37 genes related to environmental adaptation were discovered by pairwise comparisons.Of these,two candidate genes(Interaptin-like and CSLB3)related to trichome development were identified only in S.medusa and S.hypsipeta,which was consistent with their distinct morphology.Our data can provide a valuable resource for the further studies on the adaptive mechanisms of molecular and functional ecology in Saussurea DC. 展开更多

Objective:To investigate the effect and its underlying molecular mechanisms of essential oil from Saussurea costus in esophageal cancer cell line Eca109.Methods:The chemical composition of essential oil from Saussurea costus was investigated by gas chromatography-mass spectrometry(GC-MS).The anti-proliferative,anti-migrative,and apoptotic effects of essential oil from Saussurea costus against Eca109 cells were analyzed.Moreover,the expression of proteins associated with cell cycle,metastasis,and apoptosis was determined.Results:GC-MS analysis showed that essential oil from Saussurea costus was predominantly comprised of sesquiterpenes.Saussurea costus essential oil inhibited the viability of Eca109 cells in a dose-and time-dependent manner with IC_(50) values of(24.29±1.49),(19.16±2.27)and(6.97±0.86)μg/mL at 12,24,and 48 h,respectively.The expression levels of target proteins in the cell cycle(phase G_(1)/S),including cyclin D1,p21,and p53,were affected by Saussurea costus essential oil.The essential oil also downregulated the expression of metastasis-related proteins MMP-9 and MMP-2.Moreover,it induced apoptosis of Eca109 cells through the mitochondrial pathway,as well as inhibition of STAT3 phosphorylation.Conclusions:The essential oil from Saussurea costus exhibited anti-proliferative,anti-migrative,and apoptotic effects on Eca109 cells,and could be further explored as a potential anti-esophageal cancer agent. 展开更多

季节性雪被对植物的生长繁殖具有深刻影响。为阐明不同积雪梯度下钝苞雪莲各器官生物量、养分含量、繁殖分配及功能属性的变化规律,在青藏高原东缘红原县,通过人工堆积的方法开展积雪梯度(CK、S0、S1、S2、S3)控制实验,测量了不同积雪... 季节性雪被对植物的生长繁殖具有深刻影响。为阐明不同积雪梯度下钝苞雪莲各器官生物量、养分含量、繁殖分配及功能属性的变化规律,在青藏高原东缘红原县,通过人工堆积的方法开展积雪梯度(CK、S0、S1、S2、S3)控制实验,测量了不同积雪梯度下钝苞雪莲茎、叶、花苞的养分含量及繁殖分配等特征。结果表明:1)积雪变化没有显著改变钝苞雪莲的繁殖分配,但去除积雪(S0)显著降低了营养器官生物量。2)不同积雪处理下,个体大小与繁殖器官生物量均呈正线性关系(P<0.01)。3)茎部磷(P)含量随积雪量的增加显著升高(P<0.05);叶部P含量随积雪量的增加先上升后下降,即:S2>S1>CK>S3>S0,且存在显著差异(P<0.05);各处理下植物的不同器官在养分上主要受氮(N)元素的限制。4)去除积雪(S0)降低了茎干重、株高、茎高、茎分配和单株花苞量,过度积雪(S3)则降低了株高、茎高和花苞长度,中度积雪(S1、S2)则分别降低了花苞长度与单株叶片数。由此可见,不同积雪量并未显著改变钝苞雪莲的繁殖分配,但却改变了其不同器官的功能属性与养分含量,使磷元素成为植物响应积雪变化较为敏感的指标。 展开更多

长毛风毛菊提取物治疗肾性水肿的化学成分和作用机制研究

目的阐明藏药长毛风毛菊的药效及化学成分,探讨其有效成分、关键靶点和相关通路,明确其治疗肾性水肿的作用机制。方法将SD大鼠分为空白组、阿霉素诱导肾性水肿模型组,长毛风毛菊给药低剂量组、高剂量组。测定血清生化指标,探讨各组总蛋... 目的阐明藏药长毛风毛菊的药效及化学成分,探讨其有效成分、关键靶点和相关通路,明确其治疗肾性水肿的作用机制。方法将SD大鼠分为空白组、阿霉素诱导肾性水肿模型组,长毛风毛菊给药低剂量组、高剂量组。测定血清生化指标,探讨各组总蛋白(total protein,TP)、尿素氮(blood u-rea nitrogen,BUN)、血肌酐(serum creatinine,SCr)、甘油三酯(triglyceride,TG)、总胆固醇(serum total cholestero,TC)、低密度脂蛋白(low density lipoprotein,LDL-C)、高密度脂蛋白(high density lipopro-tein,HDL-C)指标的情况。采用UPLC-Q-TOF-MS/MS技术对藏药长毛风毛菊的化学成分进行分析;在SwissTargetPrediction、GeneCards数据库分别获取长毛风毛菊化学成分靶点和肾性水肿靶点,取交集获得治疗肾性水肿作用靶点;通过STRING数据库进行蛋白互作分析;利用Metascape平台进行gene ontology(GO)功能和京都基因和基因组百科全书(kyoto encyclopedia of genes and genomes,KEGG)通路分析;将药物、靶点等导入到Cytoscape3.7.2构建“药物-成分-靶点-疾病-通路”网络图。筛选出排名靠前的6个基因和12个有效成分,通过分子对接技术验证网络药理预测的准确性,揭示长毛风毛菊治疗肾性水肿的作用机制。结果大鼠血清中TP、HDL-C指标上调,BUN、SCr、TC、TG和LDL-C指标均下调。从藏药长毛风毛菊提取物中共鉴定了71个化合物,包括7个苯丙素类、16个绿原酸类和39个黄酮类等成分。通过预测得出75个长毛风毛菊治疗肾性水肿靶点,GO富集共涉及生物过程、细胞组分、分子功能3个方面,KEGG富集结果显示PI3K/AKT、AGE-RAGE、MAPK等是显著通路。分子对接结果显示,有效成分与关键靶点结合能Vina评分均小于0,高效验证了网络药理预测的准确性。结论该研究基于药效学实验,结合UPLC-Q-TOF-MS/MS鉴定、网络药理学分析和分子对接技术,表明长毛风毛菊能够治疗肾性水肿,可能通过调控PI3K/AKT、AGE-RAGE、MAPK信号通路中TP53、PI3K、AKT等相关靶点来发挥作用。 展开更多
