Effect of basic fibroblast growth factor(bFGF) on the treatment of exposure of the orbital implants 被引量:13

Methods: We review... Objective: To evaluate the efficacy and the indication of basic fibroblast growth factor (bFGF) in the treatment of exposure of orbital implants. Design: Retrospective and observational case series. Methods: We reviewed 41 patients (41 eyes) suffering exposure of orbital implants from Jan. 2000 to June 2006. The study group patients with mild exposure received com-bined treatment with bFGF and antibiotic drops, and while the control group patients with mild exposure were treated with anti-biotic drops only. The study group patients with moderate and severe exposure received combined treatment with bFGF and antibiotic drops, and after 2 months they were subjected to amniotic membrane transplantation, while the control group patients with moderate and severe exposure underwent amniotic membrane transplantation after using antibiotic drops. Observation of the growth of conjunctival epithelium and comparison of the healing rate of the two groups. Results: The healing rates of the mild, moderate and severe exposure study group were 100% and 92.3%. The healing rates of the mild, moderate and severe exposure control group were 55.6% and 66.7% respectively. The difference of the healing rates of the mild exposure study group and the control group was significant (P=0.033). And the difference of the healing rates of the moderate and severe exposure study group and the control group was not significant (P=0.167). Conclusion: bFGF may promote obviously the healing of orbital implant exposure, particularly it can be the first choice for the treatment of mild degree exposure. For the moderate and severe cases, it can be administered before surgical repair to enhance neovascularization and will tend to increase the success rate of surgical repair. 展开更多

Purpose: The usefulness of dissolving microneedles (DMs) for local skin therapy by basic fibroblast growth factor (bFGF) was studied in rats. Methods: We prepared four kinds of bFGF-loaded DMs, approximately 500 μm length and 300 μm diameter at the bottom. Long-term stability and dissolution studies were performed by HPLC method. Pharmacokinetic and pharmacological evaluations were performed after administration of bFGF loaded DMs to rats. Results: The bFGF contents were 2.15 ± 0.07, 1.07 ± 0.04, 0.56 ± 0.07 and 0.12 ± 0.03 μg. The 100.2 ± 3.4%, 100.2 ± 3.3%, 99.3 ± 1.4% and 100.4 ± 3.0% of bFGF were recovered after 1, 3 and 6 months and 1 year incubation at 40°C. The bFGF was released from DMs within 5 min. In a pharmacokinetic study using 2.0 and 1.0 μg bFGF-loaded DMs, no systemic exposure of bFGF was detected. The initial bFGF concentrations in the rat skin tissue after administration of bFGF-loaded DMs to the hair-removed rat abdominal skin were 510.2 ± 20.1 ng/g wet weight for 2 μg bFGF DMs and 264.2 ± 56.5 ng/g wet weight for 1 μg DMs, declining slowly thereafter to 226.3 ± 33.5 and 105.1 ± 27.4 ng/g wet weight at 6 hr after administration. Good dose-dependency was observed. Pharmacological evaluation of bFGF-loaded DMs of 2.0, 1.0, 0.5, and 0.1 μg, in the wound healing rat model, all used DMs, but 0.1 μg DMs, showed good healing effects. Considered collectively, these results suggest the usefulness of bFGF-loaded DMs for local therapy of skin wound disease. 展开更多

Methods.Myocardial infarction was induced in 24 New Zealand rabbits by ligating the root of left anterior descending coronary artery.Group Ⅰ(n=8) served as control, group Ⅱ(n=8) as a blank microcapsule group, group Ⅲ(n=8, each microcapsule contains 1μg bFGF) as micrpcapsule group.In group Ⅱ and Ⅲ, 5 blank microcapsules or bFGF slow-release microcapsules were implanted into myocardium underneath the epicardium between the left anterior descending coronary artery and left circumflex artery.Infarct size was evaluated by infarcted weight/left ventricle weight ratio and angiogenesis was evaluated by immunohistochemical examinations 5 weeks later. [WT5”BX] Results.As compared with group Ⅰ and Ⅱ, rabbits treated with bFGF slow-release microcapsules showed higher microvessel counts (group Ⅰ3775±450, group Ⅱ3837±498,vs.group Ⅲ 13550±481,P<0001) and less infarcted weight /left ventricle weight (group Ⅰ168%±04%,group Ⅱ167%±05%,vs.group Ⅲ 70%±02%,P<0001). Conclusions.Subepicardial administration of bFGF slow-release microcapsule in the infarcted rabbit model results in effective angiogenesis and reduction in infarct size. 展开更多

Methods Ten cases of normal ovarian tissues and 75 cases of ovarian epithelial neoplasm tissues were detected by immunohistochemical methods: S P for bFGF, FGFR 1,double immunohistochemistry Lab SA for Ki 67 antigen and bFGF. Results The expression level of bFGF, FGFR 1in ovarian epithelium and ovarian epithelial neoplasm showed a step wise increase in the following order:normal <benign <borderline <malignant; The expression level and intensity of bFGF and FGFR 1 were increased with the decrease of differentiation degree and increase of clinical stage in ovarian carcinoma; There was no statistical difference between the expression of bFGF, FGFR 1 in serous cystadenocarcinoma and that of mucinous cystadenocarcinoma; The expression of bFGF was correlated with that of FGFR 1 in neoplastic tissues; There were positive expression rates of bFGF and Ki 67 antigen in ovarian epithelial neoplasm. Objective: To explore the expression of vascular endothelial growth factor(VEGF) and basic fibroblast growth factor(bFGF) in non-small cell lung cancer(NSCLC) and theIR clinical significance. Methods: The expression of VEGF and bFGF was examined at the protein levels by immunohistochemical staining in 96 NSCLC patients, and in 36 of which at the mRNA levels by reverse transcription-PCR analysis. Results: VEGF mRNAs were expressed predominately as its secretory forms (VEGF121 and VEGF165) in NSCLC tissues. The positive ratios of VEGF121 and VEGF165 were 69.5%(25 of 36) and 41.7%(15 of 36) respectively. The positive ratio of bFGF was 52.8(19 of 36) in the same tumor specimens. The positive ratios of VEGF and bFGF at protein levels were 55.55%(20 of 36) and 58.33%(21 of 36) respectively. A significant positive correlation was observed between VEGF and bFGF expression in NSCLC tissues(P=0.002). No significant interrelationship between VEGF, bFGF expression and clinical data(age, sex, histological subtype differentiation, P-stage, metastasis and survival) was found. Conclusions: VEGF and bFGF may play an important role in angiogenesis and act in a synergistic manner in NSCLC. 展开更多

【目的】探讨自拟补肺健脾益肾方治疗慢性阻塞性肺疾病(COPD)稳定期患者的临床疗效及对血清转化生长因子β1(TGF-β1)、碱性成纤维细胞生长因子(bFGF)、超氧化物歧化酶(SOD)水平的影响。【方法】将86例COPD稳定期肺脾肾虚证患者随机分... 【目的】探讨自拟补肺健脾益肾方治疗慢性阻塞性肺疾病(COPD)稳定期患者的临床疗效及对血清转化生长因子β1(TGF-β1)、碱性成纤维细胞生长因子(bFGF)、超氧化物歧化酶(SOD)水平的影响。【方法】将86例COPD稳定期肺脾肾虚证患者随机分为对照组和观察组,每组各43例。对照组给予布地奈德吸入剂吸入治疗,并予以常规戒烟、预防呼吸道感染和呼吸功能锻炼。观察组在对照组的基础上联合自拟补肺健脾益肾方治疗,疗程为3个月。观察2组患者治疗前后临床症状评分、慢性阻塞性肺疾病评估测量表(CAT)评分、肺功能指标以及血清TGF-β1、bFGF、SOD水平的变化情况,评价2组患者的临床疗效,并采用改良呼吸困难量表(m MRC)评估患者治疗后的呼吸困难改善情况。【结果】(1)治疗3个月后,观察组的总有效率为95.35%(41/43),对照组为81.40%(35/43),组间比较,观察组的疗效明显优于对照组(P<0.05)。(2)治疗后,2组患者的中医临床症状评分(包括主症评分和次症评分)、CAT评分和急性加重次数均较治疗前降低(P<0.05),且观察组的降低作用均明显优于对照组(P<0.05或P<0.01)。(3)治疗后,2组患者的1秒用力呼气容积(FEV1)、用力肺活量(FVC)及FEV1/FVC等肺功能指标均较治疗前改善(P<0.05),且观察组的改善作用均明显优于对照组(P<0.05或P<0.01)。(4)治疗3个月后,观察组患者的呼吸困难改善程度明显优于对照组(P<0.05)。(5)治疗后,2组患者血清SOD水平均较治疗前升高(P<0.05),血清TGF-β1、b FGF水平均较治疗前降低(P<0.05),且观察组对血清SOD水平的升高作用及对血清TGF-β1、bFGF水平的降低作用均明显优于对照组(P<0.01)。【结论】自拟补肺健脾益肾方对COPD稳定期患者疗效显著,可有效改善患者呼吸困难等临床症状与肺功能,降低急性加重发作次数,提高患者生活质量,其机制可能与清除自由基、降低气道炎症反应及纤维化有关。 展开更多

目的:探讨原发性急性闭角型青光眼(APACG)患者的房水中趋化因子受体2(CXCR2)、碱性成纤维细胞生长因子(bFGF)水平与小梁切除术后预后的关系。方法:收集2020-06/2022-01期间本院收治的80例80眼行小梁切除术的APACG患者纳入病例组,依据术... 目的:探讨原发性急性闭角型青光眼(APACG)患者的房水中趋化因子受体2(CXCR2)、碱性成纤维细胞生长因子(bFGF)水平与小梁切除术后预后的关系。方法:收集2020-06/2022-01期间本院收治的80例80眼行小梁切除术的APACG患者纳入病例组,依据术后疗效分为成功组60例60眼和失败组20例20眼;收集同期本院行白内障超声乳化术且眼压正常的白内障患者86例86眼纳入对照组。采用酶联免疫吸附法检测房水中CXCR2、bFGF水平;采用ROC曲线分析房水中CXCR2、bFGF水平预测APACG患者小梁切除术失败的价值;APACG患者小梁切除术失败的影响因素采用多因素Logistic回归分析。结果:病例组房水中CXCR2、bFGF水平显著高于对照组(P<0.05)。失败组房水中CXCR2、bFGF水平及术后浅前房发生患者比例显著高于成功组(P<0.05)。房水中CXCR2、bFGF水平单独及联合预测APACG患者小梁切除术后失败的AUC分别为0.885、0.883、0.953。CXCR2、bFGF是APACG患者小梁切除术后失败的危险因素(P<0.05)。结论:APACG患者房水中CXCR2、bFGF水平显著升高,且二者均是小梁切除术后失败的危险因素。 展开更多

Hepatocellular carcinoma (HCC) is one of the most common cancers worldwide. Basic fibroblast growth factor (bFGF), which is highly expressed in developing tissues and malignant cells, regulates cell growth, differentiation, and migration. Its expression is essential for the progression and metastasis of HCC. This study aims to investigate the effects of bFGF on the expression of angiogenin, another growth factor, which plays an important role in tumor angiogenesis, and on cell proliferation in H7402 human hepatoma cells. The bFGF sense cDNA or antisense cDNA was stably transfected into H7402 cells. Genomic DNA PCR analysis demonstrated that human bFGF sense cDNA or antisense cDNA was inserted into the genome. Furthermore, the expression of bFGF and angiogenin was examined by RT-PCR and Western blot assays. MTT and colony formation assays were employed to determine cell proliferation. Stable bFGF over-expressing and under-expressing transfectants were successfully established. Expression of angiogenin was decreased in the over-expressing bFGF cells (sense transfectants) and was increased in the under-expressing bFGF cells (antisense transfectants). Cell proliferation increased in the bFGF sense transfectants and decreased in the bFGF antisense transfectants. These results demonstrated that the endogenous bFGF may not only negatively regulate the angiogenin expression but also contribute to the overall cell proliferation in H7402 human hepatoma cells. This study may be helpful in finding a potential therapeutic approach to HCC. 展开更多

目的 :了解以纤维蛋白胶 (Fibrinsealant,FS )为载体的注射型骨修复材料异位诱导成骨的作用 ,为其临床的应用提供实验依据。方法 :实验分组为 :实验组b (FS +bFGF+bBMP)、对照组b1(FS +bBMP)、对照组b2 (bBMP)、实验组r (FS +bFGF +rhBM... 目的 :了解以纤维蛋白胶 (Fibrinsealant,FS )为载体的注射型骨修复材料异位诱导成骨的作用 ,为其临床的应用提供实验依据。方法 :实验分组为 :实验组b (FS +bFGF+bBMP)、对照组b1(FS +bBMP)、对照组b2 (bBMP)、实验组r (FS +bFGF +rhBMP 2 )、对照组r1(FS +rhBMP)、对照组r2 (rhBMP)、对照组FS及空白对照组。将各组材料注射或植入小鼠肌袋内 ,采用放射学、形态学、碱性磷酸酶 (ALP)检测等方法对其成骨效应进行研究。结果 :在以bBMP为成骨因子的实验区中 ,实验组b具有高效的骨诱导活性 ,其成骨量显著高于对照组b1、对照组b2、对照组FS及空白对照组(P <0 .0 1) ;在以rhBMP 2为成骨因子的实验区中 ,实验组r同样具有高效的骨诱导活性 ,其成骨量也显著高于对照组r1、对照组r2、对照组FS及空白对照组 (P <0 .0 1)。结论 :以FS为载体复合BMP和bFGF的注射型骨修复材料具有高效的骨诱导活性 ,bFGF可明显增强BMP的骨诱导活性。 展开更多

利用大鼠坐骨神经钳夹损伤模型,观察了碱性成纤维细胞生长因子(bFGF)对大鼠脊髓前角运动神经元的保护作用。SD成年大鼠40只,分成实验组和对照组,钳夹损伤大鼠坐骨神经。在实验组损伤侧受损的神经周围放入浸有bFGF的明... 利用大鼠坐骨神经钳夹损伤模型,观察了碱性成纤维细胞生长因子(bFGF)对大鼠脊髓前角运动神经元的保护作用。SD成年大鼠40只,分成实验组和对照组,钳夹损伤大鼠坐骨神经。在实验组损伤侧受损的神经周围放入浸有bFGF的明胶海绵,对照组则用生理盐水浸泡海绵置于受损神经处。以大鼠右侧坐骨神经为正常自身空白对照。术后隔日一次向损伤侧腓肠肌肌注bFGF,对照组注射等量生理盐水。术后2、3、4、5周分别对每只大鼠进行心脏灌注固定,切取脊髓腰4~6节段,恒冷箱冷冻切片,Nisl染色,观察脊髓前角运动元的形态,计算其数目,将损伤侧与正常侧比较,将bFGF治疗组与非治疗组作比较,统计学处理。结果:术后3、4、5周治疗组的损伤侧脊髓前角运动神经元存活率均大于对照组,且统计差异有显著性意义(P<0.05)。表明bFGF对坐骨神经钳夹损伤所导致的脊髓前角运动神经元胞体死亡有保护作用,也许可能成为运动神经元新的神经营养因子。 展开更多

背景与目的:人端粒酶逆转录酶(human telomerase reverse transcriptase,hTERT)是端粒酶激活的决定因子,与肿瘤的形成和恶性程度有关,其表达受多种因素的调控。本研究检测胶质瘤中hTERT、maspin和碱性成纤维细胞生长因子(basic fibrobla... 背景与目的:人端粒酶逆转录酶(human telomerase reverse transcriptase,hTERT)是端粒酶激活的决定因子,与肿瘤的形成和恶性程度有关,其表达受多种因素的调控。本研究检测胶质瘤中hTERT、maspin和碱性成纤维细胞生长因子(basic fibroblast growth factor,bFGF)蛋白的表达,并分析它们的相关性及其与临床病理因素之间的关系。方法:应用原位杂交法、免疫组织化学SP法分别检测128例胶质瘤和8例正常脑组织中hTERT及maspin、bFGF蛋白的表达;检测结果的半定量评定参照Gatalica的H-score(H=I×P)系统;两组间率的比较用χ2检验;不同级别胶质瘤中hTERT、maspin和bFGF的表达采用Spearman等级相关分析;hTERT与maspin和bFGF表达之间的关系采用直线相关分析。结果:(1)胶质瘤中hTERT、maspin、bFGF阳性率分别为51.6%(66/128)、46.9%(60/128)、62.5%(80/128)。8例正常脑组织均不表达hTERT、bFGF,7例表达maspin。Ⅱ、Ⅲ、Ⅳ级胶质瘤中,hTERT阳性率分别为32.6%(14/43)、54.5%(30/55)、73.3%(22/30);maspin阳性率分别为58.1%(25/43)、49.1%(27/55)、26.7%(8/30);bFGF阳性率分别为39.5%(17/43)、72.7%(40/55)、76.7%(23/30)。hTERT、maspin和bFGF在正常脑组织和不同级别胶质瘤中的表达差异均有显著性(P﹤0.05),hTERT和bFGF表达与病理分级呈正相关(ρ=0.515,P﹤0.01;ρ=0.611,P﹤0.01),maspin表达与病理分级呈负相关(ρ=-0.425,P﹤0.05)。(2)hTERT表达与maspin蛋白表达呈负相关(r=-0.658,P<0.01),与bFGF蛋白表达呈正相关(r=0.627,P<0.01);maspin和bFGF蛋白表达呈负相关(r=-0.501,P<0.01)。(3)hTERT表达与年龄、性别、肿瘤直径和细胞密度无关(P>0.05),与核分裂相、血管密度、坏死显著相关(P<0.05)。结论:hTERT、maspin和bFGF三者间的表达有相关性,均与胶质瘤恶性程度有关。 展开更多

背景与目的:血管内皮生长因子(vascularendothelialgrowthfactor,VEGF)和碱性成纤维细胞生长因子(basicfibroblastgrowthfactor,bFGF)都能促进血管内皮细胞分裂和诱导血管形成,是肿瘤的生长、浸润、和转移过程中非常重要的物质。但它们... 背景与目的:血管内皮生长因子(vascularendothelialgrowthfactor,VEGF)和碱性成纤维细胞生长因子(basicfibroblastgrowthfactor,bFGF)都能促进血管内皮细胞分裂和诱导血管形成,是肿瘤的生长、浸润、和转移过程中非常重要的物质。但它们在单发性和多发性浅表膀胱移行细胞癌中表达的差异则未见报道,本研究主要是探讨VEGF和bFGF在浅表膀胱移行细胞癌中的表达及意义。方法:采用免疫组化法对60例浅表膀胱移行细胞癌组织及10例正常膀胱组织进行VEGF和bFGF的检测,观察单发性和多发性浅表膀胱移行细胞癌组织中VEGF和bFGF表达的关系。结果:多发性浅表膀胱移行细胞癌的VEGF阳性率为55.6%、bFGF阳性率为50.0%,以及VEGF和bFGF共同表达阳性率为50.0%,均明显高于单发者的水平,而高水平表达的多发性肿瘤患者的术后复发率61.1%也明显高于单发组的复发率(单发组的复发率16.7%)。结论:VEGF和bFGF表达水平的高低与浅表膀胱移行细胞癌的生物学行为有关。 展开更多
