Ovalbumin(OVA)is the major allergenic protein that can induce T helper 2(Th2)-allergic reactions,for which current treatment options are inadequate.In this study,we developed a polymerized hypoallergenic OVA product via laccase/caffeic acid(Lac/CA)-catalyzed crosslinking in conjunction with galactomannan(Man).The formation of high molecular weight crosslinked polymers and the Ig G-binding were analyzed by sodium dodecyl sulfate-polyacrylamide gel electrophoresis(SDS-PAGE)and Western blotting.The study indicated that Lac/CA-catalyzed crosslinking plus Man conjugation substantially altered secondary and tertiary structures of OVA along with the variation in surface hydrophobicity.Gastrointestinal digestion stability assay indicated that crosslinked OVA exhibited less resistance in simulated gastric fluid(SGF)and simulated intestinal fluid(SIF).Mouse model study indicated that Lac-Man/OVA ameliorated eosinophilic airway inflammatory response and efficiently downregulated the expression of Th2-related cytokines(interleukin(IL)-4,IL-5,and IL-13),and upregulated IFN-γand IL-10 expression.Stimulation of bone marrow-derived dendritic cells with Lac-Man/OVA suppressed the expression of phenotypic maturation markers(CD80 and CD86)and MHC class II molecules,and suppressed the expression levels of proinflammatory cytokines.The knowledge obtained in the present study offers an effective way to acquire a hypoallergenic OVA product that can have a therapeutic effect in alleviating OVA-induced allergic asthma. 展开更多

Echinochloa crus-galli, one of the world's most serious weeds, influences seriously the yield and quality of cereal crop plant.It is well known that E.crus-galli grain is dormant, but its dormant type, as well as its dormancy release and germination in relation to galactomannan-hydrolysing enzyme activity were poorly understood.The cooperation of endo-β-mannanase(EC 3.2.1.78), β-mannosidase(EC 3.2.1.25) and α-galactosidase(EC 3.2.1.22) can hydrolyze the cell walls rich in mannan-based polymers.In the present paper, the mature grains are used as experimental materials, we investigated the water uptake of grains, the effect of removing partial endosperm, after-ripening, stratification and phytohormone on grain germination, and the change in endo-β-mannanase, β-mannosidase and α-galactosidase activities of grains during after-ripening and germination.The results showed that the freshly-collected grains were water-permeable and had only phase I and II of water uptake, while the grains after-ripened for 150 d had an obvious phase III of water uptake.In alternating photoperiod, the germination of grains freshly-collected was zero at 10–35°C, and that of half grains was 11% at 20°C only.The grain germination was notably promoted by after-ripening and stratification, but not by gibberellic acid and 6-benzyladenine at 0.0001–1 mmol L–1.β-Mannosidase activity increased during 0 to 300 d of after-ripening and then decreased.The activity of endo-β-mannanase and α-galactosidase of grains decreased with after-ripening.During grain germination, endo-β-mannanase and β-mannosidase activities obviously increased, while α-galactosidase activity decreased.Our data showed that E.crus-galli grain was a deep physiological dormant, the dormancy release by after-ripening was related to an increasing β-mannosidase activity, and its germination was closely associated with an increasing endo-β-mannanase and β-mannosidase activity; which have provided new knowledge to decrease the harm of E.crus-galli on production of cereal crop plant. 展开更多

Objective: To evaluate the antidiabetic effect of galactomannans extracted from Adenanthera pavonina's L.seeds(GAP) in streptozotocin(STZ) induced diabetic mice.Methods:The preliminary galactomannan yield from Adenanthera pavonina L.plant and extraction products composition were evaluated.Various chemical characterization methods like thin layer chromatography, Fourier transform infrared spectroscopy, ~1H and ^(13)C nuclear magnetic resonance, and molecular weight by gel permeation chromatography have been employed to characterize the extracted galactomannan.The mice were divided in four groups: Normal control, diabetic control, GAP(1% and 2%) treated and standard drug treated groups.Diabetic mice received treatment daily for 30 d.Diabetes was induced by STZ at a single dose of 120 mg/kg.Body weight, water and food intake, fasting blood glucose, total cholesterol and triglycerides were measured.Histopathological analysis of pancreas and liver were performed to evaluate STZ-induced tissue injuries.Results: The isolated and extracted galactomannan from Adenanthera pavonina was confirmed by various chemical characterization methods.GAP exhibited a 1.46:1 mannose: galactose ratio, and high molar weight.Both GAP enriched food decreased glycaemia, total cholesterol and triacylglycerol.GAP didn't interfere on food intakes or body weight, although it increased water intake.Furthermore, the relative liver weight indicated toxic galactomannan effects on the histopathological changes of the pancreas in STZ induced diabetes.Conclusions: It is concluded that GAP is a natural product that contains potent galactomannan and is useful in preventing and treating diabetes. 展开更多

Discovery of Unusual Highly Branched Galactomannan from Seeds of <i>Desmanthus illinoensis</i>

A galactomannan was isolated from seeds of a leguminous plant, Desmanthus illinoensis, which is grown in Okinawa, Japan. D-Galactose (molar ratio, 1.0) and D-mannose (0.82) were identified via High-performance Anion Exchange Chromatography Coupled with a Pulse Amperometric Detector. The molecular mass and specific rotation were estimated to be 1000 kDa and +53.8&deg;, respectively. The infrared spectrum indicated that the galactomannan was involved in both α- and β-linkages, and two types of α-linkages were detected at 814 and 830 cm-1. The 1H- and 13C-NMR spectra indicated that the majority of the β-D-mannan main chain was substituted with mono α-D-galactose or α-D-galacto-disaccharide-side chains. Methylation analysis was used to identify 2,3,4,6-tetra-O-methyl-D-galactose (molar ratio, 3.3), 2,3,4-tri-O-methyl- D-galactose (1.0) and 2,3-di-O-methyl-D-mannose (3.1). Specifically, unique 2,3,4-tri-O-methyl D-galactose residue was identified from mass spectrum. The results suggested that the galactomannan was 1,4-linked-β-D-mannan substituted with α-D-galactose or 1,6-linked-α-D-galacto-disaccharide side chains at C-6 on the main chain. The galactomannan isolated from D. illinoensis was an unusual highly branched polysaccharide, and its chemical structure was proposed. This work is the first to report on the galactomannan involving 1,6-linked α-D-galacto-disaccharide side chains in addition to α-D-galactose mono side chains. 展开更多

The galactomannan from Antrodia cinnamomea(AC)is characterized as one of the important bioactive components that exhibits potential immunostimulatory propriety.The biological function of its corresponding oligosaccharide fragments has not been revealed yet.In this study,we reported the first chemical synthesis of the series of oligosaccharide fragments related to AC galactomannan via the convergent glycosylation strategy.The preliminary immunological evaluation of these synthesized AC oligosaccharides disclosed that the backbone tetrasaccharide 1d showed the best immunomodulatory ability on enhancing proliferation,phagocytosis and cytokines secretion of Raw264.7 macrophages in vitro,indicating its immense potential as an immunostimulant candidate. 展开更多

Background Critically ill chronic obstructive pulmonary disease (COPD) patients admitted to an intensive care unit (ICU) due to respiratory failure are at particularly high risk of Aspergillus infection.The serum galactomannan index (GMI) has proven to be one of the prognostic criteria for invasive pulmonary aspergillosis (IPA) in classical immunocompromised patients.However,the prognostic value of serum GMI in critically ill COPD patients needs evaluation.The purpose of this study is to investigate the prognostic value of serum GMI in patients with severe COPD.Methods In this single-center prospective cohort study,serum samples for GMI assay were collected twice a week from the first day of ICU admission to the day of the patients' discharge or death.Patients were divided into two groups according to their clinical outcome on the 28th day of their ICU admission.Univariate analysis and survival analysis were tested in these two groups.Results One hundred and fifty-three critically ill COPD patients were included and were divided into survival group (106 cases) and non-survival group (47 cases) according to their outcome.Univariate analysis showed that the highest GMI level during the first week after admission (GMI-high 1st week) was statistically different between the two groups.Independent prognostic factors for poor outcome in severe COPD patients were:GMI-high 1st week >0.5 (RR:4.04,95% CI:2.17-7.51) combined with accumulative dosage of corticosteroids >216 mg before the RICU admission (RR:2.25,95% CI:1.11-4.56) and clearance of creatinine (Ccr) <64.31 ml/min (RR:2.48,95% CI:1.22-5.07).Conclusions The positive GMI-high 1st week (>0.5) combined with an accumulative dosage of corticosteroids >216 mg before the ICU admission and a low Ccr may predicate a poor outcome of critically ill COPD patients. 展开更多

目的 探究二代基因测序技术(NGS)联合半乳甘露聚糖(GM)检测对重症肺炎合并真菌感染的诊断价值。方法 选取2020年3月—2023年3月大庆龙南医院急诊重症监护室收治的重症肺炎患者148例。收集支气管肺泡灌洗液行病原微生物培养及肺泡灌洗液... 目的 探究二代基因测序技术(NGS)联合半乳甘露聚糖(GM)检测对重症肺炎合并真菌感染的诊断价值。方法 选取2020年3月—2023年3月大庆龙南医院急诊重症监护室收治的重症肺炎患者148例。收集支气管肺泡灌洗液行病原微生物培养及肺泡灌洗液NGS检测,同期行血清GM检测。以病原微生物培养为金标准,分析血清GM及肺泡灌洗液NGS诊断重症肺炎合并真菌感染的敏感性、特异性,评估血清GM检测、肺泡灌洗液NGS检测单独及联合诊断重症肺炎合并真菌感染结果与病原微生物培养结果的一致性。结果 148例重症肺炎中合并真菌感染30例,其中肺孢子菌10例、白色念珠菌8例、曲霉菌7例、根霉菌1例、肺孢子菌感染合并白色念珠菌3例、肺孢子菌感染合并鲍曼不动杆菌1例。血清GM检测结果显示,阳性28例,阴性120例,与病原学培养诊断真菌感染的Kappa值为0.700。肺泡灌洗液NGS诊断结果显示,阳性34例,阴性114例,与病原学培养诊断真菌感染的Kappa值为0.841。血清GM联合肺泡灌洗液NGS诊断结果显示,阳性30例,阴性118例,病原学培养诊断真菌感染的Kappa值为0.916。血清GM、肺泡灌洗液NGS单独及联合诊断重症肺炎合并真菌感染的敏感性分别为73.3%(95%CI:0.538,0.870)、93.3%(95%CI:0.765,0.988)、93.3%(95%CI:0.765,0.988),特异性分别为94.9%(95%CI:0.888,0.979)、94.9%(95%CI:0.889,0.979)、98.3%(95%CI:0.934,0.997),曲线下面积分别为0.841、0.941和0.958。结论 血清GM联合肺泡灌洗液NGS诊断重症肺炎合并真菌感染,在肺孢子菌、白色念珠菌及曲霉菌等常见菌种中的诊断效能良好。 展开更多

肺泡灌洗液半乳甘露聚糖试验鉴别诊断非粒缺患者肺部细菌感染与真菌感染的临床价值

目的分析肺泡灌洗液(BALF)半乳甘露聚糖(GM)检测试验鉴别诊断非粒缺患者肺部细菌感染和真菌感染的临床价值。方法回顾性分析2021年1—12月抚州市第一人民医院收治的70例非粒缺肺部感染患者的临床资料,根据痰培养结果分为真菌组(n=26)与... 目的分析肺泡灌洗液(BALF)半乳甘露聚糖(GM)检测试验鉴别诊断非粒缺患者肺部细菌感染和真菌感染的临床价值。方法回顾性分析2021年1—12月抚州市第一人民医院收治的70例非粒缺肺部感染患者的临床资料,根据痰培养结果分为真菌组(n=26)与细菌组(n=44)。比较两组血清3-β-D葡聚糖(BG)、血清GM、BALF BG、BALF GM水平;采用多因素Logistic回归分析非粒缺患者肺部真菌感染的影响因素;绘制ROC曲线分析血清BG、血清GM、BALF BG、BALF GM水平鉴别诊断非粒缺患者肺部细菌感染和真菌感染的价值。结果真菌组血清BG、血清GM、BALF BG、BALF GM水平均高于细菌组,差异有统计学意义(P<0.05)。多因素Logistic回归分析结果显示,血清BG、血清GM、BALF BG、BALF GM高水平是非粒缺患者肺部真菌感染的独立危险因素(P<0.05)。ROC曲线分析结果显示,BALF BG、BALF GM水平鉴别诊断非粒缺患者肺部细菌感染和真菌感染的AUC均较大,分别为0.968、0.985,BALF GM对细菌感染和真菌感染鉴别诊断的AUC最大,且灵敏度为92.31%,特异度为100.00%,截断值为1.34μg/L。结论BALF GM试验鉴别诊断非粒缺肺部细菌感染和真菌感染的灵敏度、特异度均较高,可为临床诊断和制订治疗方案提供参考依据。 展开更多

含GM肠内营养对重症脑卒中患者营养状态及胃肠功能的影响

目的:探索肺泡灌洗液的GM、IL-8水平在侵袭性肺曲霉病的临床诊断价值和意义。方法:选取2021年01月~2023年04月在洛阳市中心医院住院的52例肺曲霉菌病患者为研究对象(包括确诊组10例,临床诊断组42例)。选取同期住院的93例肺部细菌性感染... 目的:探索肺泡灌洗液的GM、IL-8水平在侵袭性肺曲霉病的临床诊断价值和意义。方法:选取2021年01月~2023年04月在洛阳市中心医院住院的52例肺曲霉菌病患者为研究对象(包括确诊组10例,临床诊断组42例)。选取同期住院的93例肺部细菌性感染患者为对照组。所有住院患者均于治疗前完成支气管检查,并完成血液GM、IL-8水平和肺泡灌洗液GM、IL-8水平测定,并进行比较和诊断效能分析。结果:血清GM、肺泡灌洗液GM、血清IL-8、肺泡灌洗液IL-8水平四项指标在IPA组和非IPA组对比,U值分别为3448.500、1320.000、3922.000、3796.500,P值均小于0.001,差异有统计学意义。血清GM水平在0.365时,曲线下面积为0.713(P<0.001)(95%置信区间0.616~0.810),诊断效能最高,灵敏度59.6%,特异度81.7%;肺泡灌洗液GM水平在1.0时,曲线下面积为0.728(P<0.001)(95%置信区间0.643~0.813),诊断效能最高,灵敏度38.5%,特异度96.8%;血清IL-8水平在55.0时,曲线下面积为0.811(P<0.001)(95%置信区间0.728~0.893),诊断效能最高,灵敏度73.1%,特异度84.9%;肺泡灌洗液IL-8水平在933.5时,曲线下面积为0.785(P<0.001)(95%置信区间0.705~0.865),诊断效能最高,灵敏度50%,特异度97.8%。血清GM、肺泡灌洗液GM、血清IL-8、肺泡灌洗液IL-8水平四项指标联合诊断,灵敏度90.4%,特异度71.0%,PPV63.5%,NPV93%,约登指数0.614。结论:肺泡灌洗液GM和IL-8在IPA的诊断中具有一定的临床诊断价值,两者联合有望成为新的诊断方式。 展开更多

分别在25和35℃下模拟皂荚种子发芽过程,对比研究不同发芽时间下皂荚胚芽子叶和内胚乳内α-D-半乳糖苷酶的酶活及皂荚半乳甘露聚糖的分子质量、单糖含量和流变特性等理化性质。结果表明:25℃时皂荚内胚乳内α-D-半乳糖苷酶的酶活在3.45~... 分别在25和35℃下模拟皂荚种子发芽过程,对比研究不同发芽时间下皂荚胚芽子叶和内胚乳内α-D-半乳糖苷酶的酶活及皂荚半乳甘露聚糖的分子质量、单糖含量和流变特性等理化性质。结果表明:25℃时皂荚内胚乳内α-D-半乳糖苷酶的酶活在3.45~5.78 U/g上下波动(发芽第2~8天)后迅速增加至86.12 U/g(发芽第14天),35℃时皂荚内胚乳内α-D-半乳糖苷酶的酶活在3.31~9.24 U/g之间上下波动。皂荚胚芽子叶内α-D-半乳糖苷酶的酶活大多高于内胚乳内α-D-半乳糖苷酶的酶活。种子发芽前期,较高发芽温度(35℃)更有利于半乳甘露聚糖的降解。随着发芽时间的延长,皂荚半乳甘露聚糖的分子质量和表观黏度逐渐降低,25℃下溶液在第12天从假塑性流体转变为牛顿流体;皂荚半乳甘露聚糖中甘露糖和半乳糖的含量逐渐降低,甘露糖与半乳糖质量比值(M/G值)逐渐增加,35℃发芽第14天M/G值达到最大值为3.31,有望成为价格昂贵的刺槐豆胶半乳甘露聚糖的替代产品。 展开更多

通过水提醇沉法制备沙苑子精多糖(记为SACP),再通过乙醇分级沉淀法分离出纯度较高的主要多糖组分(SACP-25)。采用比色法测定各组分多糖的理化性质,再应用尺寸排阻色谱-多角度光散射联用仪和离子色谱分别测定多糖的分子质量分布、溶液构... 通过水提醇沉法制备沙苑子精多糖(记为SACP),再通过乙醇分级沉淀法分离出纯度较高的主要多糖组分(SACP-25)。采用比色法测定各组分多糖的理化性质,再应用尺寸排阻色谱-多角度光散射联用仪和离子色谱分别测定多糖的分子质量分布、溶液构象和单糖组成,结果表明:SACP-25是高纯度的均一多糖组分,总糖含量为87.40%,主要由半乳糖(42.92%)和甘露糖(57.08%)组成,在0.1 mol/L NaNO_(3)溶液中呈无规则线团链。甲基化分析与核磁共振结果表明,SACP-25的主链由β-1,4-糖苷键连接的吡喃型甘露糖组成,末端吡喃型半乳糖通过α-1,6-糖苷键形成侧链。本研究发现沙苑子可以作为半乳甘露聚糖这一水溶性膳食纤维较好的天然来源,为多糖结构有序性理论提供数据支撑,同时也为沙苑子多糖在功能性食品和医药领域的开发和应用提供一定理论依据。 展开更多

以植物源半乳甘露聚糖(GM)为原料,1-苯基-2-丙炔-1-酮(PPK)为醚化试剂,通过绿色高效的羟基-炔点击化学改性技术,成功地在室温条件下制备得到兼具优异力学性能、疏水性能和紫外屏蔽性能的半乳甘露聚糖苯丙基烯酮醚(GMPPK)薄膜。结果表明... 以植物源半乳甘露聚糖(GM)为原料,1-苯基-2-丙炔-1-酮(PPK)为醚化试剂,通过绿色高效的羟基-炔点击化学改性技术,成功地在室温条件下制备得到兼具优异力学性能、疏水性能和紫外屏蔽性能的半乳甘露聚糖苯丙基烯酮醚(GMPPK)薄膜。结果表明:羟基-炔点击化学可以实现GM表面羟基的高效醚化。与未改性的GM薄膜相比,由于PPK分子的引入,GMPPK薄膜表现新的紫外屏蔽性(GMPPK2在275 nm处的屏蔽率高达98.86%)、疏水性(水接触角从74.9°增加至96.5°)和耐水溶胀性。此外,随—OH∶PPK的反应摩尔比增加,GMPPK2薄膜的力学性能进一步增加,其拉伸应力为44.7 MPa,韧性为1.7 MJ/m3,杨氏模量为25.3 MPa,分别是未改性GM薄膜的2.2倍、1.9倍和2.4倍。研究结果可为开发高性能半乳甘露聚糖包装薄膜材料的应用研究奠定理论基础。 展开更多
