Background: It appeared that the conjunction inflammation and nerve damage (caused by surgery) generate the hyperalgesic component. But the probability of predicting hyperalgesia from the size of the surgical incision and/or the resulting inflammatory reaction is not well elucidated. This survey aims to study the influence of the size of the surgical incision and the resulting inflammatory reaction (interleukin 6 levels) in the occurrence of postoperative hyperalgesia in the population of Lubumbashi. Methods: The present study was descriptive cross-sectional. The data collection was prospective over 5 months, from February 1, 2024 to June 30, 2024. This study included any patient over the age of 18 who underwent surgery under general anesthesia. We used indirect signs to define hyperalgesia: higher (ENS > 6) and prolonged pain, postoperative overconsumption of morphine. Results: During our survey, we collected 48 operated patients who had severe postoperative pain, 16 of whom had hyperalgesia, i.e. a prevalence of hyperalgesia of 33.33%. The size of the incision most represented was between ≥20 and i.e. 62.50%. The type of surgery most affected by hyperalgesia was laparotomy. We observed an elevation of IL6 in 87.50% of patients. The largest elevation was 8.91 times the preoperative value and the smallest was 1.04 times. Pre- and postoperative IL6 levels were not associated with hyperalgesia (p = 0.265). Only the size of the surgical incision was associated with hyperalgesia (p = 0.04). Incision size values between [20 - 30] cm were those associated with hyperalgesia (p = 0.027). The model shows that making an incision greater than or equal to 20 cm increases the patient’s risk of developing hyperalgesia by more than 7.222 times and this is statistically significant (p = 0.004). Conclusion: According to this survey, the size of the surgical incision was associated with postoperative hyperalgesia and a size of more than 20 cm increases the patient’s risk of developing hyperalgesia by more than 7.222 times. 展开更多

Introduction: Rheumatoid arthritis (RA) is a chronic, erosive and deforming inflammatory rheumatic disease. In the era of biotherapies and the arrival of biosimilars in sub-Saharan Africa, the objective of this study was to describe plasma IL-6 variations in RA patients at Cité Verte District Hospital (Cameroon). Material and Methods: Descriptive and analytical cross-sectional study from December 1, 2021 to May 31, 2022. We included patients over 18 years old suffering from RA (ACR/EULAR 2010). Patients with an infection were not included. The data collected were age, sex, smoking status, family history, disease duration, disease activity by DAS28, CRP, rheumatoid factor, and plasma level of IL-6. Bone erosion was sought on radiography and ultrasound. Result: We included 31 patients, 25 of whom were women (80.6%). The mean age was 47.27 ± 17.97 years. Disease activity was predominantly moderate (32.3%) and severe (32.3%). Mean IL-6 level was 15.29 ± 2.36 pg/ml (extremes: 11.26 pg/ml and 20.15 pg/ml). IL-6 levels were higher in patients with a history of smoking. Similarly, IL-6 levels were higher in patients with mildly active RA in remission than in moderately and severely active RA. Mean IL-6 levels were significantly higher in patients with erosive RA (16.3 pg/ml VS 14.6 pg/ml). Conclusion: IL-6 levels were significantly elevated in men, weaned smokers and patients with bone erosions. 展开更多

AIM:To characterise expression of interleukin 6(IL-6),a potent proinflammatory cytokine,in the occurrence and development of inflammatory bowel disease(IBD) and investigate its effect on neuroimmunomodulation and immune homeostasis regulation.METHODS:In this study,rats with colitis induced by trinitrobenzene sulfonic acid(TNBS) were sacrificed on days 3,7,14,21 and 28 after induction.In the controls,the TNBS was just replaced by equivalent amount of phosphate buffered solution(PBS,0.01 mol/L).IL-6 mRNA expression in brain and colon tissues in each phase was evaluated by real-time reverse transcriptionpolymerase chain reaction,and cellular localisation and protein level of IL-6 was determined by immunohistochemistry.RESULTS:At day 7,mRNA expression of IL-6 was significantly higher in the colon and brain of IBD rats than that of the controls.The protein level was also significantly higher in colon,hypothalamus and cerebral cortex of IBD rats compared with the controls.So there are similar temporal trends in IL-6 mRNA expression and protein levels in all positions with a persistent increase to a peak at day 7,followed by a decline and gradual return to normal levels.CONCLUSION:These results revealed that changes in IL-6 expression in brain and colon tissues occur in different phases of IBD.Therefore,we propose that the nerve centre regulates and controls the occurrence and development of IBD via IL-6. 展开更多

Background: Recent studies indicate a strong functional relevance of the canonical inflammatory interleukin 6 signaling pathway in coronary artery disease (CAD). A genetic association of this signaling pathway with CAD has not been shown yet. We aimed to assess novel single nucleotide polymorphisms (SNPs) from genes of the Interleukin 6 signaling pathway. Results: To identify novel SNPs that are relevant for CAD, we employed a large-scale population-based case-control association study of 2199 cases and 1715 controls and assessed 73 SNPs from 12 genes out of the IL-6 signaling pathway. Results were adjusted to the CAD-related risk factors diabetes, hypertension, Body Mass Index, smoking and sex by logistic regression analysis. In a primary explorative study, we identified 5 SNPs that were significantly associated with CAD (MAPK1_rs6928, MAPK1_rs9340, MAPK1_ rs11913721, MAPK14_rs7757672, JAK1_rs310236). After adjustment to CAD-risk factors, MAPK1_ rs6928 showed the strongest association with CAD (P 0.0217, Odds Ratio 1.36, Confidence Interval 1.05 - 1.77). To reproduce this result, we performed a replication study employing independent patient and control panels. In this study we could not approve the association of rs6928 with CAD. Conclusion: In conclusion, we did not detect significant associations of SNPs from the IL-6 signaling pathway with CAD. Our investigation demonstrates the importance of independent replication studies to verify results from candidate-gene association studies in the quest to discover the underlying pathomechanism of CAD. 展开更多

Objective To investigate the effect of IL-6 on prostatic carcinoma cell lines, and differential effects on androgen-dependent and androgen-independent prostatic carcinoma cells. Methods The IL-6 producing capacities of LNCaP and PC-3 cells were determined, and effects of exogenous IL-6 and anti-IL - 6 antibodies on LNCaP and PC - 3 cells were examined. Results LNCaP produced a very small amount of IL-6, but PC-3 produced more, the concentraion of IL-6 being 190 pg/48 h per ml(1 × 106). The exogenous IL-6 inhibited LNCaP growth significantly,but had no obvious effect on PC -3 cells. Anti-IL-6 antibodies lowered PC-3 cells growth rate but had neutral effect on LNCaP. Conclusion PC-3 cells produces IL-6 massively in autocrine manner. IL-6 could be antagonized by anti-IL-6 antibodies,resulting in slowing PC-3 cells growth, and LNCaP cells growth could be inhibited by exogenous IL-6.7 refs,2 tabs. 展开更多

Methods: Expressions of IL-6 in bone marrow mononuclear cells (BMMNCs) were detected using double labeled immunofluorometry, and the IL-6 activities in the supernatants of co-culture system of myelomacell line KM3 and bone marrow stromal cells (BMSCs) were measured using B9 cell proliferation test. ResultS: IL6 molecules were expressed not only in cytoplasndc γ-globin positive cell but also in type I collagen or von Willebrand Factor(vwr)positive cells. After fixation, both myeloma cells and BMSCs hardly secreted IL-6. IncreasedIL-6 activities were found in the supernatants after co-culture of unfixed my8loma cell line KM3 and fixed BMSCsor of fixed KM3 cells and unfixed BMSCs. Furthermore, BMSCs from MM patients induced KM3 cells to secretsignificantly more IL-6 in the supernatants than in BMSCs did controls (P<0. 01). Objectives: The aim of this study was to assess the levels of Y-box binding protein 1 (YBX-1) and interleukin 6 (IL-6) in the sera of metastatic and non-metastatic breast cancer patients (BC), investigate their clinicopathological significance and to analyze their potential use as biomarkers of breast cancer metastasis. Methods: The study included ninety subjects sub-grouped equally into metastatic BC, non-metastatic BC and healthy volunteers. Serum YBX-1 and IL-6 were quantified using ELISA technique while CA 15-3 was quantified using IRMA kit. Clinical data were collected from patients’ records. Results: YBX-1 (p < 0.001), IL-6 (p < 0.001) and CA15-3 (p = 0.017, 0.001) were significantly elevated in metastatic and non-metastatic BC patients compared to healthy controls, however, only YBX-1 (p 0.001) showed a significant difference with cancer metastasis. Generally, YBX-1 and IL-6 were correlated with worse histological grade and late clinical stage in breast cancer patients and they were also associated with axillary lymph nodes involvement and positive vascular invasion in metastatic BC patients. Serum YBX-1 and IL-6 levels were positively correlated to each other (rs = 0.615, p < 0.001) and they showed high sensitivity and specificity compared to CA 15-3 (p < 0.001 and p = 0.004 for YBX-1 and IL-6 respectively) for predicting cancer metastasis. Conclusions: Serum YBX-1 and IL-6 are potential biomarkers of breast cancer patients with significant correlation with bad clinicopathological characteristics. Serum YBX-1 and IL-6 have superior sensitivity and specificity compared to CA15-3 and can serve as potential follow up and prognostic markers. 展开更多

In this study, serum concentrations of carboxyterminal propeptide of type Ⅰ collagen(PICP) and carboxyterminal cross-linked telopeptide of type Ⅰ collagen (ICTP), which represent the rates of synthesis and degradation of type Ⅰ collagen, were determined by radioimmunoassay in 56 patients with multiple myeloma (MM) and 22 healthy controls. It was discovered that serum concentrations of both PICP and ICTP were higher in MM than those in healthy controls (P<0. 01 ). With the disease progressing and the number of bone lesions increasing,serum concentration of ICTP elevated while serum concentration of PICP showed no significant change. Neither serum PICP nor ICTP concentration was related to M-component classes. Our results indicated that serum ICTP concentration was a good serological marker to reflect severity of bone lesions in MM and elevated serum PICP concentration might be due to compensatory increase in type Ⅰ collagen synthesis. BACKGROUND Accumulating evidence suggests that the inflammatory cytokine interleukin-6(IL-6)contributes to the pathophysiology of psychiatric disorders.However,there was no study concerning the relationship between IL-6 concentrations and clinical features in the chronic phase of early-onset schizophrenia(EOS).AIM To investigate the relationship between serum IL-6 concentration and the clinical features of EOS.METHODS We measured serum IL-6 Levels from 74 patients with chronic schizophrenia,including 33 with age at onset<21 years(EOS group)and 41 with onset≥21 years in[adult-onset schizophrenia(AOS)group],and from 41 healthy controls.Symptom severities were evaluated using the Positive and Negative Syndrome Scale(PANSS).RESULTS Serum IL-6 concentrations were higher in both EOS and AOS groups than healthy controls(F=22.32,P<0.01),but did not differ significantly between EOS and AOS groups(P>0.05)after controlling for age,body mass index,and other covariates.Negative symptom scores were higher in the EOS group than the AOS group(F=6.199,P=0.015).Serum IL-6 concentrations in the EOS group were negatively correlated with both total PANSS-negative symptom score(r=-0.389,P=0.032)and avolition/asociality subscore(r=-0.387,P=0.026).CONCLUSION Patients with EOS may have more severe negative symptoms than those with adult-onset schizophrenia during the chronic phase of the illness.IL-6 signaling may regulate negative symptoms and its avolition/asociality subsymptoms among the early-onset chronic schizophrenic patients. 展开更多

目的探讨老年肺部感染病人致病菌耐药情况及耐药程度对血清IL-6、IL-10、CRP水平的影响。方法选取2020年5月至2023年5月运城市中心医院收治的214例老年肺部感染病人,分析其致病菌耐药情况,并探讨致病菌种类、耐药程度对血清IL-6、IL-10... 目的探讨老年肺部感染病人致病菌耐药情况及耐药程度对血清IL-6、IL-10、CRP水平的影响。方法选取2020年5月至2023年5月运城市中心医院收治的214例老年肺部感染病人,分析其致病菌耐药情况,并探讨致病菌种类、耐药程度对血清IL-6、IL-10、CRP水平的影响,采用Spearman法分析血清IL-6、IL-10、CRP水平与不同致病菌耐药程度的相关性。结果214例老年肺部感染病人的痰标本共检出病原菌265株,革兰氏阴性菌、革兰氏阳性菌占比分别为76.23%(202/265)、23.77%(63/265)。其中,一般耐药47例(21.96%),多重耐药105例(49.07%),广泛耐药62例(28.97%)。革兰氏阴性菌感染病人与革兰氏阳性菌感染病人的耐药情况比较,差异无统计学意义(P>0.05);革兰氏阴性菌感染病人血清IL-6、IL-10、CRP水平均高于革兰氏阳性菌感染病人(P<0.05),且在同种类致病菌感染病人中,一般耐药病人IL-6、IL-10、CRP水平低于多重耐药病人,多重耐药病人低于广泛耐药病人(P<0.05)。Spearman秩相关分析结果显示,革兰氏阴性菌或革兰氏阳性菌感染病人血清IL-6、IL-10、CRP水平与致病菌耐药程度均呈正相关(P<0.05)。结论老年肺部感染致病菌以革兰氏阴性菌为主,且多重耐药菌占比较高。血清IL-6、IL-10、CRP水平与致病菌耐药程度存在相关性,临床可根据血清炎症因子水平初步判定致病菌耐药程度,合理使用抗生素。 展开更多

目的 观察真方白丸子加减治疗中风风痰入络证的疗效及对患者血清人脂蛋白相关磷脂酶A2(Lp-PLA2)、白细胞介素-6(IL-6)、同型半胱氨酸(Hcy)水平的影响。方法 参照随机数字表法将80例患者分为对照组与试验组各40例。对照组予以西医常规对... 目的 观察真方白丸子加减治疗中风风痰入络证的疗效及对患者血清人脂蛋白相关磷脂酶A2(Lp-PLA2)、白细胞介素-6(IL-6)、同型半胱氨酸(Hcy)水平的影响。方法 参照随机数字表法将80例患者分为对照组与试验组各40例。对照组予以西医常规对症治疗,试验组在此基础上给予真方白丸子加减治疗。治疗4周后观察对比两组患者的临床疗效、中医证候积分、神经功能[改良Rankin量表(MRS)评分和美国国立卫生研究院卒中量表(NIHSS)评分]、血清Lp-PLA2、IL-6与Hcy水平,统计治疗期间出现药物不良反应的情况。结果 治疗后,试验组的总有效率高于对照组(P<0.05),中医证候积分、MRS评分和NIHSS评分均明显降低且低于对照组(P<0.05)。与对照组比较,试验组治疗后血清Lp-PLA2、IL-6与Hcy水平均显著降低(P<0.05)。治疗期间实验组不良反应总发生率与对照组比较差异无统计学意义(P> 0.05)。结论 应用真方白丸子加减治疗中风风痰入络证具有良好的临床效果,在缓解患者临床症状、恢复其机体神经功能和改善血清Lp-PLA2、IL-6与Hcy水平方面均表现良好,且不良反应少。 展开更多

Objective: To determine the effect of flurbiprofen combined with prednisolone on interleukin-6 in elderly surgery patients. Methods: In this double-blind randomized controlled study, patients aged 65 to 80 who were undergoing spinal fusion surgery for disc herniation were administered flurbiprofen 100 mg (P group, flurbiprofen group), prednisolone 0.6 mg/kg (D group, prednisolone group), prednisolone 0.6 mg/kg plus flurbiprofen 100 mg (P + D group, flurbiprofen + prednisolone group) or normal saline (S group, saline group) 15 minutes before the induction of anesthesia. Plasma samples were collected before surgery (T0) and on day 1 (T1), day 2 (T2) and day 3 (T3) following surgery. At the same time, systemic inflammatory response syndrome (SIRS) was assessed by SIRS criteria. The levels of interleukin-6 (IL-6), tumor necrosis factor-α (TNF-α) and C-reactive protein (CRP) for collected samples were measured. Results: Other groups had significantly lower levels of IL-6, CRP and occurrence of SIRS than S group (p < 0.05). Compared to groups P and D, the levels of IL-6 and CRP in P + D group were significantly lower on T1 (p < 0.05). Peak levels of IL-6 in all groups were presented on T1 (p 0.05). The levels of CRP within three days were significantly different but did not show peak levels (p > 0.05). Conclusion: Compared to prednisolone or flurbiprofen, combining flurbiprofen with prednisolone in elderly surgery patients led to an increased suppression of IL-6. 展开更多

目的探讨血浆肝素结合蛋白(HBP)、白细胞介素-6(IL-6)和降钙素原(PCT)对脓毒症进展为脓毒症休克的早期预判价值。方法选取2022年1月—2023年12月在浙江大学医学院附属第二医院治疗的177例脓毒症患者为研究对象,依据入院6 h内是否发生休... 目的探讨血浆肝素结合蛋白(HBP)、白细胞介素-6(IL-6)和降钙素原(PCT)对脓毒症进展为脓毒症休克的早期预判价值。方法选取2022年1月—2023年12月在浙江大学医学院附属第二医院治疗的177例脓毒症患者为研究对象,依据入院6 h内是否发生休克将其分为休克组和未休克组。比较两组血浆HBP、IL-6和PCT差异,绘制受试者工作特征曲线分析各指标对脓毒症进展为脓毒症休克的预测价值,采用二分类Logistic回归分析探讨脓毒症进展为脓毒症休克的影响因素。结果休克组血浆HBP、IL-6、PCT高于未休克组,差异有统计学意义(P<0.05)。HBP、IL-6、PCT及三项联合检测预测脓毒症进展为脓毒症休克的敏感度、特异度分别为35.0%和89.7%、78.8%和60.8%、67.5%和53.6%、67.5%和68.0%。HBP、PCT、IL-6及三项联合检测早期预判脓毒症进展为脓毒症休克的曲线下面积分别为0.589、0.598、0.663和0.717。将脓毒症是否进展为脓毒症休克作为因变量,将HBP、PCT和IL-6早期预判脓毒症休克的最佳截断值作为临界值,进行二分类Logistic回归分析,结果显示HBP和IL-6为脓毒症进展为脓毒症休克的影响因素(P<0.05)。结论血浆HBP、PCT和IL-6对脓毒症进展为脓毒症休克有早期预判价值,三项联合检测预判价值更高,HBP和IL-6是脓毒症进展为脓毒症休克的影响因素。 展开更多

背景 急性肾损伤(AKI)是脓毒症常见并发症,机体免疫-炎症指标是预测脓毒症并发AKI患儿预后的常用指标,目前从微小RNA(miR)方面评估的研究较少,有待临床探究。目的 探究血清淀粉样蛋白A(SAA)、白介素6(IL-6)、肿瘤坏死因子α(TNF-α)及mi... 背景 急性肾损伤(AKI)是脓毒症常见并发症,机体免疫-炎症指标是预测脓毒症并发AKI患儿预后的常用指标,目前从微小RNA(miR)方面评估的研究较少,有待临床探究。目的 探究血清淀粉样蛋白A(SAA)、白介素6(IL-6)、肿瘤坏死因子α(TNF-α)及miR在脓毒症并发AKI患儿中的表达,并分析其对预后的评估价值。方法 选取2020年3月—2023年3月平顶山市第一人民医院收治的100例脓毒症并发AKI患儿为观察组,另选取同期80例单纯脓毒症患儿为对照组。收集患者一般资料,酶联免疫吸附试验(ELISA)检测血清SAA、IL-6、TNF-α水平,采用实时荧光定量PCR法检测miR-21-3p、miR-182-5p、miR-128-3p相对表达量。比较两组序贯性器官功能衰竭(SOFA)评分、急性生理与慢性健康(APACHEⅡ)评分。采用Pearson相关性检验分析血清SAA、IL-6、TNF-α及miR水平与SOFA、APACHEⅡ评分的相关性。绘制受试者工作特征(ROC)曲线探究血清SAA、IL-6、TNF-α及miR水平对脓毒症并发AKI患儿死亡的预测价值并计算ROC曲线下面积(AUC)。结果 观察组SOFA评分、APACHEⅡ评分、血清SAA、IL-6、TNF-α、miR-21-3p、miR-182-5p、miR-128-3p水平均高于对照组(P<0.05)。住院28 d后观察组74例患儿生存,26例患儿死亡。生存患儿血清SAA、IL-6、TNF-α、miR-21-3p、miR-182-5p、miR-128-3p均低于死亡患儿(P<0.05)。血清SAA、IL-6、TNF-α、miR-21-3p、miR-182-5p、miR-128-3p与SOFA、APACHEⅡ评分均呈正相关(P<0.05)。ROC曲线结果显示联合预测的AUC为0.926(95%CI=0.856~0.969,P<0.05)。结论 脓毒症并发AKI患儿血清SAA、IL-6、TNF-α、miR-21-3p、miR-182-5p、miR-128-3p异常高表达,临床检测各项指标水平对患儿预后评估有较高价值及预警作用。 展开更多

目的探究血清白细胞介素-6受体(IL-6R)和白细胞介素-22(IL-22)表达与溃疡性结肠炎严重程度及临床结局的关系。方法选取监利市人民医院2017年12月至2022年6月治疗的212例溃疡性结肠炎病人为研究对象,根据其病情严重程度分为轻症组与重症... 目的探究血清白细胞介素-6受体(IL-6R)和白细胞介素-22(IL-22)表达与溃疡性结肠炎严重程度及临床结局的关系。方法选取监利市人民医院2017年12月至2022年6月治疗的212例溃疡性结肠炎病人为研究对象,根据其病情严重程度分为轻症组与重症组,根据治疗2个月的预后情况分为预后良好组与预后不良组,采用酶联免疫吸附测定检测IL-6R、IL-22水平并收集病人临床资料,logistic多因素回归分析影响溃疡性结肠炎病人预后的相关因素;绘制受试者操作特征曲线(ROC曲线)预测IL-6R和IL-22对溃疡性结肠炎严重程度以及预后的临床诊断及预测价值。结果与轻症组[(11.10±1.21)ng/L、(50.24±6.35)ng/L]相比,重症组在入院时的血清IL-6R[(15.32±2.62)ng/L]、IL-22水平[(63.61±6.95)ng/L]显著增加(P<0.05),通过绘制ROC曲线发现IL-6R、IL-22以及两者联合诊断重症溃疡性结肠炎的曲线下面积(AUC)分别为0.85、0.85、0.92,并且二者联合诊断显著优于IL-6R、IL-22单独诊断(Z=3.58,3.42;P=0.003,0.006);通过分析病人一般临床资料发现,溃疡性结肠炎病人的预后与病人是否有肠外表现、C反应蛋白(CRP)、降钙素原(PCT)、红细胞沉降率(ESR)、粪便钙卫蛋白(FC)、粪便乳铁蛋白(FL)、IL-6R以及IL-22水平有关(P<0.05),与性别、年龄无关(P>0.05),logistic多因素回归分析发现,CRP、PCT、ESR、FC、FL、IL-6R以及IL-22均为影响溃疡性结肠炎病人预后的危险因素(P<0.05)。绘制ROC曲线发现IL-6R、IL-22以及两者联合预测溃疡性结肠炎病人预后的AUC分别为0.81、0.83、0.90,并且二者联合预测显著优于IL-6R、IL-22单独预测(Z=3.70,3.18;P<0.001,P=0.002)。结论溃疡性结肠炎病人血清IL-6R、IL-22水平随病人病情严重程度增加,均为影响溃疡性结肠炎病人预后的危险因素,并且对病人的病情严重程度及临床结局预测具有辅助诊断价值。 展开更多

目的 探讨膝关节假体周围感染(PJI)的MRI影像表现及其与血清α-防御素、白细胞介素-6、D-二聚体水平相关性。方法 选择2017年2月至2021年10月于本院行全膝关节置换术后的150例患者的临床资料,术后行MRI检查和血清α-防御素、白细胞介素-... 目的 探讨膝关节假体周围感染(PJI)的MRI影像表现及其与血清α-防御素、白细胞介素-6、D-二聚体水平相关性。方法 选择2017年2月至2021年10月于本院行全膝关节置换术后的150例患者的临床资料,术后行MRI检查和血清α-防御素、白细胞介素-6、D-二聚体水平检测。根据是否发生PJI将患者分为感染组(42例)和非感染组(108例)。比较两组患者的MRI表现和血清α-防御素、白细胞介素-6、D-二聚体水平差异,采用Spearman相关系数分析PJI的MRI表现与血清α-防御素、白细胞介素-6、D-二聚体水平的相关性。结果 感染组患者假体周围发生骨膜反应、存在假体关节囊积液以及假体周围肌内水肿所占比例显著高于非感染组(P<0.05),而感染组与非感染组之间假体周围骨质溶解和骨髓水肿所占比例差异均无统计学意义(P>0.05)。感染组患者血清α-防御素显著低于非感染组(P<0.05),而白细胞介素-6、D-二聚体水平显著高于非感染者(P<0.05)。Spearman相关系数分析显示,感染组患者MRI表现骨膜反应、假体关节囊积液、肌内水肿均与血清α-防御素水平呈负相关(P<0.05),而与血清白细胞介素-6、D-二聚体水平均呈正相关(P<0.05)。结论膝关节PJI的MRI影像表现具有一定特征性,其与血清α-防御素、白细胞介素-6、D-二聚体水平具有一定相关性,有助于为临床的定性诊断以及治疗决策提供更科学的影像学证据。 展开更多

目的探讨山柰酚(kaempferol,Kpf)调节白细胞介素-6(interleukin-6,IL-6)/信号转导与转录激活子3(signal transducer and activator of transcription 3,STAT3)信号通路对妊娠糖尿病(gestational dia-betes mellitus,GDM)大鼠炎症反应的... 目的探讨山柰酚(kaempferol,Kpf)调节白细胞介素-6(interleukin-6,IL-6)/信号转导与转录激活子3(signal transducer and activator of transcription 3,STAT3)信号通路对妊娠糖尿病(gestational dia-betes mellitus,GDM)大鼠炎症反应的影响。方法雌雄同笼与高脂高糖喂养联合腹腔注射链脲佐菌素(streptozotocin,STZ)构建GDM大鼠模型,将造模成功大鼠按照随机数字表达分为Model组、Kpf低、中、高剂量组、激活剂组(IL-6/STAT3通路激活剂rIL-6),每组12只,同批12只妊娠大鼠作为control组,各组腹腔注射相应药物,每天1次,直到妊娠19 d。罗氏血糖仪测量大鼠空腹血糖(fasting blood glucose,FBG);分析各组大鼠妊娠结局;ELISA法测定大鼠血清空腹胰岛素(fasting insulin,FINS)及胎盘中白细胞介素(interleukin,IL)-6、IL-1β、肿瘤坏死因子(tumor necrosis factor-α,TNF-α)水平;苏木精伊红(hematoxylin-eosin,HE)染色观察大鼠胎盘组织中病理损伤;TUNEL染色检测胎盘组织细胞凋亡;蛋白质印迹测定胎盘中IL-6/STAT3通路蛋白表达。结果与control组比较,Model组大鼠FBG、FINS、HOMA-IR、胎鼠质量、IL-6、IL-1β、TNF-α水平、胎盘组织IL-6、p-STAT3/STAT3水平及细胞凋亡率增加,活胎率、窝产子数下降(P<0.05);相较于Model组,Kpf低、中、高剂量组大鼠FBG、FINS、HOMA-IR、胎鼠质量、IL-6、IL-1β、TNF-α水平、胎盘组织IL-6、p-STAT3/STAT3水平及细胞凋亡率下降,活胎率、窝产子数增加(P<0.05);相较于Kpf高剂量组,激活剂组上述指标变化均显著逆转(P<0.05)。结论Kpf可能通过抑制IL-6/STAT3信号通路降低GDM大鼠胎盘炎症,减轻胰岛素抵抗。 展开更多
