We used to find the MTHFR gene polymorphisms are related with blood lead levels among pregnant women. Pregnancy-stimulated bone turnover may be associated with MTHFR gene polymorphisms too. In this article, we aimed to determine the relationship between MTHFR gene polymorphisms and bone turnover rates among the pregnant women. The participants including pregnant and non-pregnant women were selected and recruited during their routine prenatal or physical examination from July to October in 2012. A total of 1000 participants, including 250 pregnant women in the first, second, and third trimesters and 250 non-pregnant women, were enrolled in the study. Finally, after excluding 27 participants unable to provide blood samples, 973 eligible participants (i.e., 234, 249, and 248 pregnant women in the first, second, and third trimesters, respectively, and 242 non-pregnant women) were included in the research. The MTHFR gene 1298CC homozygote carriers were more susceptible to yield higher plasma homocysteine levels than the 1298AA/AC carriers, with standardized coefficients of 0.086 (P〈0.05) and 0.104 (P〈0.01) of all the participants and the pregnant women, respectively. The MTHFR gene 1793AA homozygote carriers more likely showed higher plasma osteocalcin levels (standardized β=0.091, P〈0.01) than the 1793GG/GA carriers among all the subjects. Plasma homocysteine levels were positively correlated with blood lead levels among the participants and the pregnant women with standardized coefficients of 0.320 (P〈0.01) and 0.179 (P〈0.01), respectively. Plasma osteocalcin levels were positively associated with blood lead levels among pregnant and non-pregnant women with standardized coefficients of 0.084 (P〈0.05) and 0.125 (P〈0.01), respectively. In conclusion, homocysteine and osteocalcin contents in plasma are associated with the MTHFR gene A1298C polymorphism and blood lead levels among pregnant women. The MTHFR gene A1298C polymorphism-related homocysteine is a possible risk factor for increased blood lead levels among Chinese women.展开更多

Methods: This case-control study conducted in Srinagar, Kashmir (North India) between 2013 and 2015 was designed to evaluate risk induced due to MTH-FR 677C>T gene polymorphisms to contribute in susceptibility for migraine in Kashmir population (North India). Using the polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) method, we tested the genotype distribution of 100 migraine patients in comparison with 120 healthy migraine-free controls from the same geographical region. Results: The genotypic frequencies of the patients and controls were not significantly associated (p > 0.05). Higher distribution of TT mutant genotype was found in controls as against the cases (5% versus 1%) but association was not significant (p > 0.05). Per copy frequency of T allele (Val) was found to be 0.14 in cases versus 0.19 in controls (p 0.05). Similar scenario was observed when migraine without aura was compared with controls where variant genotype (16% cases versus 39.0% controls: p > 0.05) as well as allele frequency was found to be less in cases (cases 0.15 versus 0.19 controls: p > 0.05). Conclusions: We conclude that MTHFR gene C677T polymorphism has no role in predisposition to the migraine in our population and cannot serve as a predictive factor for the risk of migraine.展开更多

The pathogenesis of migraine is known to be related to genetic risk factors such as the presence of common polymorphisms C677T (rs1801133) and A1298C (rs1801131) in the MTHFR gene. The aim of the study was to examine the role of these two MTHFR polymorphisms as risk factors for pediatric migraine and examine the gender-specific differences. Methods: 56 patients (M = 26;F = 30) with migraine were randomly selected from Tartu University Hospital Children’s Clinic. 29 patients had migraine with aura (M = 13, F = 16) and 27 migraine without aura (M = 13, F = 14). The average age of the patients was 12.7 yrs (SD = 3.3). The control group consisted of 168 healthy people (M = 78, F = 90), ages 22 - 35 yrs (average 29.2 ± 3.4). Results: Among patients with aura, girls had higher frequency of A1298C and C677T mutations than boys, but not with statistical significance (minor allele frequency 0.406 and 0.231, p = 0.16;minor allele frequency 0.313 and 0.269, p = 0.72;respectively). In the migraine without aura group, girls had higher frequency in C677T (minor allele frequency 0.357 vs 0.231, p = 0.31) than boys. Comparing the migraine group with controls, higher frequencies of minor allele of A1298C and C677T in female patients were found (p = 0.63 and p = 0.32 respectively). In contrast, the C677T minor allele occurred in male controls with higher frequency (p = 0.34). Conclusion: Our results indicate a possible contribution of common MTHFR gene polymorphisms to migraine risk in children. 91% of children with migraine carried the variant alleles. Among girls, MTHFR polymorphisms occurred more frequently than among boys, but these differences were not statistically significant. We found that girls with migraine (with and without aura) showed an increased prevalence of C677T polymorphisms.展开更多
