The academic community has conducted a seri...As an important ecological tree species in northern China, Populus simonii plays a crucial role in maintaining ecological balance and promoting environmental sustainability. The academic community has conducted a series of in-depth studies on this species, covering key areas such as genomics, survival mechanisms, and genetic breeding. Through the analysis of the genomic structure and function of P. simonii, we have not only revealed the molecular basis for its adaptation to harsh environments but also identified key genes that promote its growth and resistance to pests and diseases. Furthermore, exploring the survival mechanisms of P. simonii has deepened our understanding of its stress resistance traits, including how it effectively copes with abiotic stresses such as drought, salinization, and heavy metal pollution. In genetic breeding, significant progress has been made through the application of modern biotechnology, improving the growth rate and wood quality of P. simonii and enhancing its environmental adaptability and disease resistance. These research findings have not only enriched our knowledge of the biological characteristics of P. simonii but also provided a solid scientific foundation for its application in ecological restoration, forestry production, and environmental management.展开更多

Toward using th...The role of plant eIF5A proteins in multiple biological processes, such as protein synthesis regulation, translation elongation, mRNA turnover, programmed cell death and stress tolerance is well known. Toward using these powerful proteins to increase stress tolerance in agricultural plants, in the present study, we cloned and characterized PsneIFSA2 and PsneIFSA4 from young poplar (P. simonii × P. nigra) leaves. The deduced amino acid sequences of PsneIF5A2 and PsneIF5A4 were 98 % similar to each other, and they are orthologs of eIF5A 1 in Arabidopsis. In a subcellular localization analysis, PsneIF5A2 and PsneIF5A4 proteins were localized in the nucleus and cytoplasm, qRT-PCR analysis showed that PsneIF5A2 and PsneIF5A4 were transcribed in poplar flowers, stem, leaves, and roots. In addition, they were also induced by abiotic stresses. Transgenic yeast expressing PsneIF5A2 and PsneIF5A4 had increased salt, heavy metal, osmotic, oxidative tolerance. Our results suggest that PsneIF5A2 and PsneIF5A4 are excellent candidates for genetic engineering to improve salt and heavy metal tolerance in agricultural plants.展开更多

Our results show that significant differences exist between plants of different provenances in morphological and physiological traits, except for leaf-stalk length and the number of leaf margins. The coefficient of variation for all traits ranges from 14.77% to 81.49%. The mean phenotypic coefficient of differentiation (VST) is 47.1%, which means that the variation within provenances is the major source for phenotypic variation in P. simonii. Given our cluster analysis of provenances based on an average linkage computing method, the five provenances ofP. simonii investigated could be divided into three groups. Our results provide a theoretical basis for genetic resource conservation and provenance selection of natural P. simonii in northern China.展开更多

[ Method] Plant expression vectors of AP1 genes were constructed and transformed into tobacco leaf disks with Agrobacterium-mediated method. Transgenic tobacco plants were identified by PCR. [ Result] AP1 genes were integrated into the genome of tobacco. Transgenic tobacco plants all presented an early flowering phenotype compared with wild-type tobacco. [ Conclusion] AP1 genes could promote early flowering in transgenic tobacco plants, which provided theoretical basis for molecular regulation of flowering in poplar.展开更多

The activities of two enzymes, lipoxygenase (LOX), a key enzyme ofjasmonate (JA) pathway, and phenylalanine ammonia lyase (PAL), a rate-limiting enzyme of phenyl- propanoid pathway, were measured in the leaves of one-year-old poplar (Populus simonii × P. pyramidalis 'Opera 8277') cuttings after Clostera anachoreta larvae attack. The results show that the increased activities of LOX and PAL were found not only in the leaves wounded by C. anachoreta larvae but also in their tipper systemic leaves, indicating that JA and phenylpropanoid pathways were activated, and the defense response was stimulated systemically. The increase in LOX and PAL activities in neighboring intact poplar cuttings sug- gested that there exists the interplant communication between poplar plants mediated by the herbivore-induced volatiles. Methyl jasmonate (MeJA) was also proved to be an airborne signal to induce defense response in P simonii × P pyramidalis 'Opera 8277' cuttings.展开更多

(PSM) and Populus simo...In order to investigate the effects of afforestation on soil microbial abundance, microbial biomass carbon and enzyme activity in sandy dunes, 20-year-old Pinus sylvestris var. mongolica Litv. (PSM) and Populus simonii Carri6re (PSC) mature forests were se- lected in Horqin Sandy Land, and mobile dunes was set as a control (CK). Results show that PSM and PSC plantations can im- prove soil physicochemical properties and significantly increase microbiological activity in mobile dunes. Soil microbial abun- dance, microbial biomass carbon and enzyme activity show an order of PS〉PSM〉CK. Total soil microbial abundance in PSM and PSC was respectively 50.16 and 72.48 times more than that in CK, and the differences were significant among PSM, PSC and CK Soil microbial biomass carbon in PSM and PSC was respectively 23.67 and 33.34 times more than that in CK, and the difference was insignificant between PSM and PSC. Soil enzyme activity, including dehydrogenase (DEH), peroxidase (PER), protease (PRO), urease (URE) and cellobiohydrolase (CEL) in PSM and PSC were respectively 19.00 and 27.54, 4.78 and 9.89, 4.05 and 8.67, 29.93 and 37.46, and 9.66 and 13.42 times of that in CK. R sylvestris and P. simonii can effectively improve soil physico- chemical and microbiological properties in sandy dunes and fix mobile dunes in Horqin Sandy Land. The Cmic:C ratio is an appli- cable indicator to estimate soil stability and soil water availability, and based on an overall consideration of plantation stability and sustainability, R sylvestris is better than R simonii in fixing mobile dunes in sandy land.展开更多

We examined organic P fractions...The aim of this study was to investigate the role of rhizosphere organic phosphorus (P) in soil P supply in semiarid forests and the effects of tree species on rihizosphere organic P. We examined organic P fractions in rhizosphere and bulk soils of mono-specific Simon poplar (Populus simonii) and Mongolian pine (Pinus sylvestris var. mongolica) plantations in a semiarid sandy soil of Horqin Sandy Land in Northeast China. Total organic P (TPo) accounted for 76% of total P across the two stands. The concentration of organic P (Po) fractions decreased in the order of NaOH-Po〉Res-Po〉HCI-Po〉NaHCO3-Po in both plantations. The concentration of NaHCO3-Po was 38% and 43% lower in rhizosphere soil than in bulk soil in Simon poplar and Mongolian pine plantations, respectively. In contrast, total P, TPo and NaOH-Po significantly accumulated in rhizosphere soil in Simon poplar plantations, but no change in Mongolian pine plantations. Soil recalcitrant organic P fractions were positively correlated with soil organic carbon. The results suggest that rhizosphere labile organic P was an important source of plant-available P in this semiarid region, but the dynamic of rhizosphere recalcitrant organic P fractions varied with tree species and was correlated to organic carbon dynamics.展开更多
