Promethazine-Tetraphenyl Boron(III) Modified Carbon Paste Electrode for the Determination of Promethazine Hydrochloride

Promethazine modified carbon paste ion selective electrode has been prepared, based on the ion pair of promethazine hydrochloride Pm.Cl with sodium tetraphenyl boron(III) Pm-TPB dissolved in dioctyl phthalate DOP as a pasting liquid. The electrode showed the linear response with mean calibration graph slope of 58 mV/decade at 25℃;at a concentration range and a lower detection limit of 1 × 10-5 -6 × 10-2 M and 8 × 10-6 M promethazine hydrochloride, respectively. The change of pH of the tested solution within the range 4.2 -6.8 did not affect the electrode performance. The electrode showed a very good selectivity toward Pm+ cations with respect to a large number of organic and inorganic cations and compounds. The standard addition and potentiometric titration methods were applied for the determination of promethazine hydrochloride in pure solution and in pharmaceutical preparations with an?average recovery range of 97.3% -101.4% and a mean relative standard deviation (RSD) of 0.35% -1.83 % which were of comparable accuracy and precision to the previously reported works. 展开更多

Effect of Cinepazide Maleate Combined with Promethazine Hydrochloride in the Treatment of Emergency Vertigo

Objective:To explore the effect of cinepazide maleate combined with promethazine hydrochloride in the treatment of emergency vertigo.Methods:48 cases of emergency vertigo patients in our hospital from November 2017 to November 2019 were divided into experimental group(24 cases,treated with cinepazide maleate combined with promethazine hydrochloride)and control group (24 cases,treated with cinepazide maleate).The clinical efficacy,symptom relief time,adverse reactions and quality of life were compared.Results:The total effective rate of the experimental group(95.83%,23/24)was higher than that of the control group(75.00%),The remission time of nausea and vomiting(1.75±0.22)d,vertigo(3.54±0.63)d,deafiiess and tinnitus(3.47±0.58)d,night sweats(3.05±0.33)d in the experimental group were shorter than those in the control group,P<0.05;the incidence of adverse reactions in the experimental group(8.33%,2/24)was lower than that in the control group(33.33%),The scores of social function,material life attitude,physical health and psychological function in the experimental group were 59.14±7.23,54.05±8.04,53.58±0.86 and 60.11±8.44 respectively,P<0.05.Conclusion:In the process of clinical treatment of emergency vertigo patients,the combined application of cinepazide maleate and promethazine hydrochloride has definite curative effect,can relieve clinical symptoms in a short time,has less adverse reactions,and improves the quality of life of patients to a certain extent,which is worthy of promotion. 展开更多

Promethazine(PHZ)is used as a sedative in veterinary medicine,and its residue can threaten the health of human.The electrochemical detection of PHZ is suitable method for application in the field.However,the traditional electroanalysis is difficult to perform directly in meat samples due to matrix interference.This work integrates magnetic solid-phase extraction and differential pulse voltammetry for highly sensitive and selective determination of PHZ in beef and beef liver for the first time.CoFe_(2)O_(4)/graphene coated with C_(18)-functionalized mesoporous silica(MG@mSiO_(2)-C_(18))is synthesized as dispersed magnetic adsorbent to extract PHZ.Magnetic glassy carbon electrode modified with nitrogen-doped hollow carbon microspheres(HCM)attracts the MG@mSiO_(2)-C_(18)with PHZ,and directly detects the PHZ without elution procedure.MG@mSiO_(2)-C_(18)can separate PHZ to avoid the interference of impurities on following detection,and also concentrate PHZ on magnetic electrode.Additionally,the electrode modification with HCM can amplify the electrochemical signal of PHZ.Finally,the integrated PHZ determination method exhibits a wide linear range from 0.08μmol/L to 300μmol/L with a low limit of detection of 9.8 nmol/L.The beef sample analysis presents excellent recovery,demonstrating that this protocol is promising for the rapid and onsite detection of PHZ in real meat samples。 展开更多

Promethazine, a first generation antihistamine,has an antiarrhythmic effect on ischemia-reperfusion inducing arrhythmias^1 and experimental arrhythmias.^2 However, terfenadine as a second generation of antihistamine, has been reported to elicit hypotension, bradycardia, prolongation of the QTc interval and torsades de pointes (TdP) like ventricular arrhythmia.^3 This may be due to the blockage on rectifier postassium current (Ik) of terfenadine, resulting in the prolongation of the action potential duration (APD) and dispersion of the repolarization duration, which might provoke a specific form of polymorphic ventricular tachydysrhythmia, i.e. TdP.^4 In clinical practice,however, the class Ⅲ antiarrhythmic agents, which target on the lk and prolong the action potential duration and QTc interval, rarely lead to arrhythmias.Other actions must be considered to underlie the arrhythmogenic tendency of terfenadine besides its inhibition on Ik. Though both promethazine and terfenadine block the H1 receptor, there must be a different pharmacology profile between the two compounds on ion channels of cardiac myocytes.Whole-cell patch clamp technique was used to investigate the effects of these two antagonists of the H1 receptor on the main ion currents in cardiac electrical activities. 展开更多

The interactions of phenothiazine such as chlorpromazine hydrochloride(CPZ·HCl)and promethazine hydrochloride(PZ·HCl),with nucleic acids(NA)have been investigated.The CPZ-NA system can produce very intense resonance Rayleigh scattering(RRS),but the RRS intensity of the PZ-NA system is very weak.The energy relationship of the two reaction systems has been calculated by the AM1 and B3LYP of density functional methods for quantum chemistry,the binding site and interacting model of CPZ-NA and PZ-NA are studied.The results show that CPZ has strong binding ability with nucleic acids,but PZ hardly react with nucleic acids.It can be expected that RRS method may be a useful means for molecular recognition of nucleic acids and the molecular design of gene medicine in the future.Moreover,the method is used for the quan-titative determination of nucleic acids by employing chlorpromazine hydrochloride as reagent,which has very high sensitivity and good selectivity.Therefore,a sensitive,simple and rapid new method has been developed for the determination of nucleic acids. 展开更多

The present paper covers a simple, reliable and reproducible method, based on capillary zone electrophoresis(CZE) with amperometric detection(AD), for the separation and the determination of ephedrine hydrochloride, promethazine hydrochloride and codeine phosphate. Under the optimal conditions, the three analytes were base-line separated completely within 16 min. Good linear relationships between the peak heights and the concentrations of the three analytes were obtained with the correlation coefficients better than 0 9993. The method was directly applied to the determination of the active ingredients in pharmaceutical preparations and the assay results were satisfactory. 展开更多

Role of Histamine H1 Receptors in Vestibular Nucleus in Motion Sickness

Objectives To investigate the expression of histamine H1 receptors (H1R) in the vestibular nucleus of brainstem in rats and the role of H1R in motion sickness (MS). Methods A total of 24 healthy Sprague-Dawley rats were divided randomly into four groups (n=6 each) which determined if the animals would receive induction of MS or drug (promethazine) treatment: MS ( - )/Drug ( - ); MS(+)/Drug ( - ); MS ( - )/Drug ( + at 0.25 mg); and MS ( + )/ Drug(+). MS was induced by complex motion stimulation and the conditioned taste aversion was used as a behavioral indicator of MS. The volume of 0.15% sodium saccharin solution (SS) intake within 45 minutes after motion stimulation was measured. H1R in the vestibular nucleus was examined by immunofluorescence staining. The expression of H 1R protein in brainstem tissue at vestibular nucleus level was detected by western blot. Results The mean SS intake volume in the MS ( + )/Drug ( - ) group (8.8 ml) was significantly less than that of the MS ( - )/Drug ( - ) group (15.1 ml) (P 〈 0.01). The mean SS intake volume of the MS (-)/Drug (+) group (14.8 ml) was similar to that of the MS(-)/Drug(-) group. The mean SS intake volume (9.6 ml) of the MS(+)/Drug(+) group was more than that of the MS(+)/Drug(-)group (P〈0.01), but less than that of the MS(-)/Drug(-) group or MS(-)/Drug(+) group (P 〈 0.01). Immunofluorescence staining showed positive expression of H1R in the vestibular nucleus of brainstem and the expression was enhanced by motion stimulation. Western blot analysis showed that H1R protein expressed in the brainstem tissue at vestibular nucleus level and the expression also increased significantly after motion stimulation. The MS-induced increase of H1R was not affected significantly by promethazine. Conclusions H1Rs exist in the vestibular nucleus in rats and H 1R expression is up-regulated by motion stimulation, but not affected by promethazine. The findings indicate that the histaminergic system is involved in MS. Promethazine, as an H1R blocker, may play its anti-MS role by competing the binding site on H1Rs with histamine rather than inhibiting H1R expression. 展开更多
