The construction of advanced laboratories for precision instruments, such as electron microscopes, involves unique challenges that are influenced by the specific environmental conditions required for optimal functionality. These include mitigating interference from magnetic fields and vibrations, which are critical for maintaining the precision and accuracy of the instruments used. This study aims to offer enhanced project management strategies and detailed construction solutions that address the environmental and technical needs specific to electron microscopy labs, thereby facilitating effective lab operations and extending the lifecycle of high-end precision instruments. Case studies of existing laboratory constructions, onsite investigations, and comprehensive reviews of the technical and environmental requirements provide the basis for a best practice for constructing sophisticated electron microscopy labs. The approach integrates both pre-construction planning and post-construction adjustments to create optimal operational environments. The findings suggest that successful lab constructions are those that incorporate thorough onsite assessments, strategic location choices, and the use of advanced construction materials and techniques specifically designed to counteract environmental challenges like magnetic and vibration interferences. Actionable guidelines for both planning and executing the construction of electron microscope labs highlighted in this tutorial are intended as an important resource to troubleshoot or upgrade existing lab facilities and to consult in preparation of future lab construction projects. 展开更多

目的:研究热盐水灌胃导致大鼠萎缩性胃炎(CAG)胃粘膜组织细胞超微结构的变化,探讨长期热咸饮食与慢性萎缩性胃炎间的关系。方法:用热盐水灌胃(55℃,150g mL,2 5mL d)建立大鼠萎缩性胃炎动物模型。选取大鼠腺胃胃窦组织进行病理组织学检... 目的:研究热盐水灌胃导致大鼠萎缩性胃炎(CAG)胃粘膜组织细胞超微结构的变化,探讨长期热咸饮食与慢性萎缩性胃炎间的关系。方法:用热盐水灌胃(55℃,150g mL,2 5mL d)建立大鼠萎缩性胃炎动物模型。选取大鼠腺胃胃窦组织进行病理组织学检查,以扫描电镜观察胃粘膜上皮细胞超微结构的变化。结果:光镜下热盐水灌胃后第12周,大鼠胃粘膜出现腺体明显缩小,粘膜肌层的平滑肌呈束状增生插入粘膜固有层中。腺体上1 3至2 3腺上皮萎缩,腺管腔增宽,胃小凹颈部粘膜宽度变窄。扫描电镜见正常组大鼠胃粘膜被纵横交错的小沟分隔成许多胃小区,呈网状,胃小凹(胃腺开口)壁衬有圆形或椭圆形上皮细胞,体积基本一致,有短而稀的微绒毛。上皮细胞排列规则,被覆一薄层连续的粘液。热盐水组大鼠在12周时见胃粘膜细胞表面粗糙,粘膜表面糜烂、扁平;在32周时见胃粘膜细胞萎缩、腺腔直径增大、细胞表面破溃,并见纤维性渗出。结论:热盐水灌胃可引起胃粘膜上皮细胞损害,长期过咸及过热刺激可能会诱发萎缩性胃炎。 展开更多

Studies on mitochondrial macromolecular syntheses in various organs of aging animals labeled with 3H-precursors as revealed by electron microscopic radioautography

In order to study the aging changes of intramitochondrial macromolecular synthesis in various organs of aging animals during the development and aging, 10 groups of developing and aging mice, each consisting of 3 individuals, total 30, from fetal day 19 to postnatal newborn at day 1, 3, 9, 14 and adult at month 1, 2, 6, 12 to 24 were injected with 3H-thymidine a DNA precurson, another 10 groups consisting of 3 individuals, total 30, were injected with 3H-uridine a RNA precursor, and another 10 groups of 30 individuals were injected with 3H-leucine a protein precursor, total 90 individuals. Then, all the animals were sacrificed 1 hr after the injections and the liver tissues, the lung tissues, the kidney tissues, the testis and ovary tissues, the adrenal tissues were taken out, fixed and processed for electron microscopic radioautography. On electron microscopic radioautograms obtained from each animal, ten photographs in respective groups, numbers of mitochondria per cell profile area, numbers of labeled mitochondria per cell and the mitochondrial labeling index (LI) labeled with 3H-thymidine showing DNA synthesis, LI labeled with 3H-uridine showing RNA synthesis, and LI labeled with 3H-leucine showing protein synthesis, in various organs, were counted and the results in various organs in respective aging groups were compared, respectively. From the results, it was demonstrated that the numbers of mitochondria in hepatocytes as well as in 3 zones of the adrenal cortex, the zona glomerulosa, fasciculata and reticularis of mice at various ages increased from fetal day 19 to postnatal month 1 due to development and aging of animals. On the other hand, the numbers of labeled mitochondria and the labeling indices of intramitochondrial DNA syntheses incorporating 3H-thymidine in hepatocytes and 3 zones of cortical cells increased from fetal day 19 to postnatal month 1 and decreased to month 24. The numbers of labeled mitochondria and the labeling indices of intramitochondrial RNA syntheses incorporating 3H-uridine in hepatocytes and 3 zones of cortical cells increased from fetal day 19 to postnatal month 1 and decreased to month 24. The numbers of labeled mitochondria and the labeling indices of intramitochondrial RNA syntheses incorporating 3H-uridine in hepatocytes and 3 ones of cortical cells increased from fetal day 19 to postnatal month 1 and decreased to month 24. Moreover, some other organs such as the lung and the testis were also review and discussed. From the results, it was shown that the activity of intramitochnodrial DNA synthesis, RNA synthesis, and protein synthesis in hepatocytes, adrenal cortical cells, pulmonary cells and testicular cells in developing and aging mice as well as some other organs increased and decreased due to development and aging. 展开更多

This study shows that submicron/nanoparticles found in bacterial cells (S. aureus) incubated with polyurethane (a material commonly used for prostheses in odontostomatology) are a consequence of biodestruction. The presence of polyurethane nanoparticles into bacterial vesicles suggests that the internalization process occurs through endocytosis. TEM and FIB/SEM are a suitable set of correlated instruments and techniques for this multi facet investigation: polyurethane particles influence the properties of S. aureus from the morpho-functional standpoint that may have undesirable effects on the human body. S. aureus and C. albicans are symbiotic microorganisms;it was observed that C. albicans has a similar interaction with polyurethane and an increment of the biodestruction capacity is expected by its mutual work with S. aureus. 展开更多

In this paper we try to study the retentive peculiarity of soluble enriched uranium UO2F2 in subcellular level by electron microscopic autoradiography. The early dynamic accumulation of radioactivity in the body showed that enriched uranium UO2F2 was chiefly localized in kidney, especially accumulated in epicyte of kidney near- convoluted tubule. In liver cells, enriched uranium UO2F2 at first deposited in the nucleus and the cytoplasm, then accumulated in mitochondria selectively and lysosome as well. The electron microscopic autoradiographic study showed that the dynamic retention of radioactivity of enriched uranium UO2F2 in skeleton rose steadily throughout the exposure. Enriched uranium UO2F2 chiefly deposited in nucleus and mitochondria of the osteoblast as well as osteoclast. 展开更多

目的观察牙龈成纤维细胞表面形态和细胞核糖体S6蛋白激酶(P70s6k)的表达,评价镀氮化钛钕铁硼磁体的生物相容性。方法将托槽、镀氮化钛及未镀膜钕铁硼磁体加入DMEM中制备成浸提液,以10%浓度加入培养的大鼠牙龈成纤维细胞,通过扫描电镜观... 目的观察牙龈成纤维细胞表面形态和细胞核糖体S6蛋白激酶(P70s6k)的表达,评价镀氮化钛钕铁硼磁体的生物相容性。方法将托槽、镀氮化钛及未镀膜钕铁硼磁体加入DMEM中制备成浸提液,以10%浓度加入培养的大鼠牙龈成纤维细胞,通过扫描电镜观察细胞形态的改变,免疫印记分析P70s6k的表达。结果加入托槽、镀氮化钛磁体浸出液培养的细胞表面形态良好、细胞膜完整,高倍扫描显示细胞膜表面及边缘有较多、较长的微绒毛,未镀膜磁体浸出液培养的细胞表面形态可见不光滑、不完整,细胞表面及边缘微绒毛短小。免疫印记结果显示,P70s6k表达的平均灰度值托槽组(76.0±1.50)与镀氮化钛磁体组(75.5±2.65)无显著差异(P>0.05)。未镀膜磁体组(40.4±1.75)与托槽组差异有显著性(P<0.01)。结论镀氮化钛钕铁硼磁体具有良好的生物相容性。 展开更多

目的观察不同浓度毒鼠强中毒大鼠实质性器官的超微病理变化。方法采用灌胃方法使大鼠毒鼠强染毒,制作大鼠中毒模型,并以健康大鼠灌服生理盐水为对照,提取大鼠的脑皮质、脑干桥脑部、心、肝、脾、肾等实质性器官,进行超微病理观察。结果... 目的观察不同浓度毒鼠强中毒大鼠实质性器官的超微病理变化。方法采用灌胃方法使大鼠毒鼠强染毒,制作大鼠中毒模型,并以健康大鼠灌服生理盐水为对照,提取大鼠的脑皮质、脑干桥脑部、心、肝、脾、肾等实质性器官,进行超微病理观察。结果不同浓度的毒鼠强可对心肌细胞、脑神经细胞、神经纤维和肝细胞造成一定损伤,而对肾、脾组织细胞损伤不明显。脑皮质神经元细胞结构模糊,内质网轻度扩张,线粒体扩张水肿,且无明显的剂量-反应关系;脑干桥脑部可见多处弥散性软化灶,神经细胞核溶解,神经纤维细胞有坏死,血管内皮细胞肿胀,血管周围有间隙出现,脑干损伤有明显的剂量-反应关系;心肌细胞表现为不同程度的心肌细胞坏死、线粒体弥漫性崩解或肿胀、肌丝断裂溶解、肌溶灶普遍存在等变化,并呈现出明显的剂量-效应关系;肝细胞表现弥漫性肿胀,细胞模糊,肝窦狭小,内皮细胞肿胀,结构模糊不清,细胞质内线粒体明显肿胀,糖原含量减少,与中毒剂量和时间均呈明显相关性。结论脑、心、肝可能为其毒性作用的主要靶器官或靶组织。 展开更多

本研究旨在对水稻黄绿叶突变体ygl-9108进行表型分析和基因定位,为后续图位克隆该叶色相关基因奠定基础。以黄绿叶突变体ygl-9108为试材,利用突变体ygl-9108和‘五山丝苗’杂交的F2群体进行基因定位。结果表明,与野生型相比,突变体的株... 本研究旨在对水稻黄绿叶突变体ygl-9108进行表型分析和基因定位,为后续图位克隆该叶色相关基因奠定基础。以黄绿叶突变体ygl-9108为试材,利用突变体ygl-9108和‘五山丝苗’杂交的F2群体进行基因定位。结果表明,与野生型相比,突变体的株高、有效穗数、穗长、每穗粒数、结实率、粒宽、千粒重和单株谷重均显著下降,下降比例分别为21.5%、21.2%、14.6%、19.2%、11.0%、10.2%、12.9%和52.2%。透射电镜观察显示,突变体叶肉细胞的叶绿体数量大量减少,类囊体结构异常。遗传分析表明,突变体的表型由一个隐性核基因控制。利用图位克隆方法,ygl-9108被定位于水稻第11号染色体两InDel标记11Y39和11Y45之间,物理距离约为147 kb,该区间内未见有叶色相关基因的报道,表明ygl-9108是一个新的黄绿叶调控基因。本研究结果为ygl-9108的克隆和功能分析奠定了坚实的基础。 展开更多

The significance of electronic microscopc examination(EM) in the diagnosis of pulmonary neoplasms was evaIuated in 40 cases of Patients with different kinds of Pulmonary neoplasms.In 27 of the 40 cases,final diagnoses were made by light microscope(LM) examination,while in the remaining 13 cases,LM faded to reach definite diagnoses which were established with the help of EM.By analyzing our data,we conclude that in the following situations,EM helps meet in the diagnosis of pulmonary neoplasm:1.diagnosis of neuroendocrinal carcinomas of the lung;2.diagnosis of some rare pulmonary neoplasm;3.documentation of the histologic origins of the matastatic pulmonary neoplasms and 4.differentiation of malignant mesothelioma with pleural metastasis of Pulmonary adenocarcinoma. 展开更多

Objective:To estimate electroporation(EP) influence on malignant and normal cells.Methods: Two cell lines including human malignant melanoma(Me-43) and normal human gingival fibroblast(HCFs) were used.EP parameters were the following:230,1000,1 730,2 300 V/cm;30 μ s by 3 impulses for every case.The viability of cells after EP was estimated by MTT assay. The ullrastructural analysis was observed by transmission electron microscope(Zeiss EM 900). Results:In the current study we observed the intracellular effect following EP on Me-43 and HGF cells.At the conditions applied,we did not observe any significant damage of mitochondrial activity in both cell lines treated by EP.Conversely,we showed that EP in some conditions can stimulate cells to proliferation.Some changes induced by EP were only visible in electron microscopy.In fibroblast cells we observed significant changes in lower parameters of EP(230 and 1 000 V/cm).After applying higher electric field intensities(2 300 V/cm) we detected many vacuoles,myelin-like bodies and swallowed endoplasmic reticulum.In melanoma cells such strong pathological modifications after EP were not observed,in comparison with control cells. The ultrastructure of both treated cell lines was changed according to the applied parameters of EP.Conclusions:We can claim that EP conditions are cell line dependent.In terms of the intracellular morphology,human fibroblasts are more sensitive to electric field as compared with melanoma cells.Optimal conditions should be determined for each cell line.Summarizing our study,we can conclude that EP is not an invasive method for human normal and malignant cells. This technique can be safely applied in chemotherapy for delivering drugs into tumor cells. 展开更多

The study has been carried out using field emission microscope. Changes in electron work function of the iridium substrate which are produced by vapor of deposition of submonolayers of gold in ultra high vacuum have been measured by noting the changes in the slope of Fowler-Nordheim plots. The same procedure for studying the adsorption of copper on iridium?[1] was followed to study the adsorption of gold on iridium. By scanning and transmission electron microscopy, leukemlc celb were obwrved after CFU-Mix culture. Even though granulocytlc growth factor, erythropoietin and lymphocytlc growth factor were added at vitor, acute leukemlc celb still showed defects In differentiation and maturation. These were characterized by abnormal colony which consisted of smooth cells, bizarre shape, nuclear-cytoplasmic asynchrony In development, and appearance of nuclear bleb. However, chronic myelogenous leukemlc celb were more nature than the acute ones, manifesting in normal colony with finger- like projections and ruffled membrane. Macrophages and eosinophils could be observed. It b suggested that there b a difference In differentiation between acute and chronic leukemia cells. 展开更多
