BACKGROUND Pain in the back or pelvis or fear of back pain may affect the timing or cocontraction of the core muscles. In both static and dynamic movements, the Sahrmann core stability test provides an assessment of core muscle activation and a person's ability to stabilize the lumbopelvic complex.Preparatory cues and images can be used to increase the activation of these muscles. To attain optimal movement patterns,it will be necessary to determine what cueing will give the most effective results for core stability.AIM To investigate the effects of external and internal cues on core muscle activation during the Sahrmann five-level core stability test.METHODS Total 68 participants(21.83±3.47 years)were randomly allocated to an external(n=35)or internal cue group(n=33).Participants performed the Sahrmann fivelevel core stability test without a cue as baseline and the five-level stability exercises with an internal or external cue.External cue group received a pressure biofeedback unit(PBU),and the internal cue group received an audio cue.A Delsys Trigno^(TM)surface electromyography unit was used for muscle activation from the rectus abdominis, external oblique, and transverse abdominis/internal oblique muscles.RESULTS Linear mixed effects model analysis showed that cueing had a significant effect on core muscle activation(P=0.001);however,there was no significant difference between cue types(internal or external)(P=0.130).CONCLUSION Both external and internal cueing have significant effects on core muscle activation during the Sahrmann five-level core stability test and the PBU does not create higher muscle activation than internal cueing. 展开更多

In this paper, the practicality and feasibility of Active Force Control (AFC) integrated with Fuzzy Logic(AFCAFL) applied to a two link planar arm actuated by a pair of Pneumatic Artificial Muscle (PAM) is investigated. The study emphasizes on the application and control of PAM actuators which may be considered as the new generation of actuators comprising fluidic muscle that has high-tension force, high power to weight ratio and high strength in spite of its drawbacks in the form of high nonlinearity behaviour, high hysteresis and time varying parameters. Fuzzy Logic (FL) is used as a technique to estimate the best value of the inertia matrix of robot arm essential for the AFC mechanism that is complemented with a conventional Propor- tional-Integral-Derivative (PID) control at the outermost loop. A simulation study was first performed followed by an experi- mental investigation for validation. The experimental study was based on the independent joint tracking control and coordinated motion control of the arm in Cartesian or task space. In the former, the PAM actuated arm is commanded to track the prescribed trajectories due to harmonic excitations at the joints for a given frequency, whereas for the latter, two sets of trajectories with different loadings were considered. A practical rig utilizing a Hardware-In-The-Loop Simulation (HILS) configuration was developed and a number of experiments were carried out. The results of the experiment and the simulation works were in good agreement, which verified the effectiveness and robustness of the proposed AFCAFL scheme actuated by PAM. 展开更多

The expression levels of these two proteins closely correlate with the extracellular mechanical microenvironment. We investigated how the expression of α-SMA and tenascin-C was altered in the periodontal ligament (PDL) under orthodontic loading to indirectly reveal the intrinsic mechanical microenvironment in the PDL. In this study, we demonstrated the synergistic effects of transforming growth factor-β1 (TGF-β1) and mechanical tensile or compressive stress on myofibroblast differentiation from human periodontal ligament cells (hPDLCs). The hPDLCs under higher tensile or compressive stress significantly increased their levels of α-SMA and tenascin-C compared with those under lower tensile or compressive stress. A similar trend was observed in the tension and compression areas of the PDL under continuous light or heavy orthodontic load in rats. During the time-course analysis of expression, we observed that an increase in α-SMA levels was matched by an increase in tenascin-C levels in the PDL under orthodontic load in vivo. The time-dependent variation of α-SMA and tenascin-C expression in the PDL may indicate the time-dependent variation of intrinsic stress under constant extrinsic loading. 展开更多

The functional formula diet AHP, containing polysaccharides from Auricularia auricular, polyphenolic compounds from Hawthorn (Crataegus) and Pueraria radix, has been recently developed as a dietary intervention against dyslipidemia in our previous study. In the present study, its antioxidant activities and protective effects against proliferation of vascular smooth muscle cells (VSMCs) were investigated. AHP possessed the potent radical-scavenging effects against hydroxyl and superoxide radicals, and also the inhibitory effects against peroxidation of low density lipoprotein induced by Cu2+ in vitro. The protective effects of AHP against proliferation of VSMCs were evaluated through the methodology of serum pharmacology. The serum containing AHP significantly inhibited the proliferation of VSMCs induced by oxidized low density lipoprotein in a time- and dose-dependent manner, and also promoted the nitric oxide production of VSMCs. Our study indicated that this functional formula diet would be a potent alternative as a functional diet to prevent atherosclerosis at early stage. 展开更多

Serum testosterone does not correlate with androgen tissue activity, and it is critical to optimize tools to evaluate such activity in males. Ultrasound measurement of bulbocavernosus muscle (BCM) was used to assess the relationship between the number of CAG repeats (CAGn) in the androgen receptor (AR) and the BCM size; the changes in the number of CAGn over age were also evaluated. Transperineal ultrasound measurement of the BCM was also performed. AR CAGn were determined by high performance liquid chromatography, and morning hormone levels were determined using immunoassays. Forty-eight men had CAG repeat analysis. Twenty-five were 〈30 years of age, mean 23.7 years (s.d, = 3.24) and 23 were 〉45 years of age, mean 53years (s.d. = 5.58). The median CAGn was 21 (13-29). BCM area was greater when the number of CAGn were 〈18 as compared to the number of CAGn 〉24 (P= 0.04). There was a linear correlation between the number of CAGn and the BCM area R^2= 16% (P= 0.01). In the 45 to 65-years-old group, a much stronger negative correlation (R^2 = 29%, P= 0.01) was noticed. In the 19 to 29-years-old group, no such correlation was found (R2 = 4%, P = 0.36). In older men, the number of CAGn increased with age (R^2 = 32%, P= 0.01). The number of CAGn in the AR correlates with the area of the BCM. Ultrasound assessment of the BCM is an effective surrogate to evaluate end-organ activity of androgens. The number of CAGn may increase with age. 展开更多

Background:Forefoot strike(FFS) and rearfoot strike(RFS) runners differ in their kinematics, force loading rates, and joint loading patterns, but the timing of their muscle activation is less clear. Methods:Forty recreational and highly trained runners ran at four speeds barefoot and shod on a motorized treadmill. "Barefoot" runners wore thin,five-toed socks and shod runners wore neutral running shoes.Subjects were instructed to run comfortably at each speed with no instructions about foot strike patterns. Results:Eleven runners landed with an FFS when barefoot and shod and eleven runners landed with an RFS when barefoot and shod. The 18remaining runners shifted from an FFS when barefoot to an RFS when shod(shifters).Shod shifters ran with a lower stride frequency and greater stride length than all other runners. All FFS runners landed with more plantarflexed ankles and more vertical lower legs at the beginning of stance compared to RFS runners.FFS runners activated their plantarflexor muscles 1 1%earlier and 10%longer than RFS runners. Conclusion:This earlier and longer relative activation of the plantarrlexors likely enhances the capacity for the passive structures of the foot and ankle to store elastic energy, and may also enhance the performance of the active muscle by increasing the storage of elastic strain energy in the cross-bridaes and activated thin. 展开更多

Mammography is carried out in a special position, i.e. “an image is taken while the breast is compressed, stretched and kept in a fixed position”. The purpose of this study was to demonstrate quantitatively and qualitatively the physical and psychological burden due to positioning and breast compression during mammography. Muscle activity of each part of the body during positioning for the standard imaging method, to obtain craniocaudal (CC) view images, was measured in 15 adult females using surface EMG. The associated pain was analyzed using visual analogue scale (VAS) scores. During positioning for the CC view, muscle activity was highest in the biceps (24.44 iEMG/s) followed by the trapezius (17.78 iEMG/s) on the imaging side. Muscle activity of the biceps and the sternocleidomastoid on the imaging side showed significant differences compared with pre-imaging activity (biceps P < 0.031, sternocleidomastoid P < 0.005). The pain during mammography was rated as moderate to severe pain (VAS = 3.3 - 6.7) for CC views. As a result, the activities of not only the muscles directly involved in mammography positioning but also those indirectly involved were high as compared with the normal state. Measurement of muscle activity during mammography is expected to be used effectively, such as in the care to reduce pain for the subjects. 展开更多

Striated muscle tissue contains fibers with high oxidative capacity (heart muscle), higher oxidative capacity (type I and IIA fibers of skeletal muscle) and low oxidative capacity (type IIB/X fibers of skeletal muscle). Muscle fibers with higher oxidative capacity contain large mitochondria tightly packed with cristae as well as small forms of mitochondria containing relatively few cristae. The intensive development of the mitochondrial apparatus in the post-activity period reflects the adaptive processes, which is intended to supply the increased energy requirements of muscle fibers with higher oxidative capacity. Muscle fibers with low oxidative capacity contain significantly less mitochondria than fibers with higher capacity. It is typical to type IIB fibers that after intensive muscle activity there are damaged myofibrils in a relatively small area, some myofibrils are twisted and lose the connection with the neighboring structures. It is still not fully known how skeletal muscles with different oxidative capacity respond to an increased functional activity and what differences exist in these fibers between oxidative capacity and function of myofibrils. The aim of the present short review was to compare structural-functional changes in mitochondrial and myofibrillar compartments of heart and skeletal muscle fibers with different oxidative capacity and the effect of increased functional activity on the interaction of these compartments. 展开更多

To investigate the relationship between intracellular free Ca^2+ concentration ([Ca^2+ ]i ) and calcium-activated chloride (Clca) channels of pulmonary artery smooth muscle cells (PASMCs) in rats under acute and chronic hypoxic conditions, acute hypoxia-induced contraction was observed in rat pulmonary artery by using routine blood vascular perfusion in vitro. The fluorescence Ca^2+ indicator Fura-2/AM was used to observe [Ca^2+ ]i of rat PASMCs under normal and chronic hypoxic condition. The effect of Clca channels on PASMCs proliferation was assessed by MTT assay. The Clca channel blockers niflumic acid (NFA) and indaryloxyacetic acid (IAA-94) exerted inhibitory effects on acute hypoxia-evoked contractions in the pulmonary artery. Under chronic hypoxic condition, [Ca^2+ ]i was increased. Under normoxic condition, [Ca^2+ If was (123.634-18.98) nmol/ L, and in hypoxic condition, [Ca^2+]i wag (281. 754-16.48) nmol/L (P〈0. 01). Under normoxic condition, [Ca^2+ ]i showed no significant change and no effect on Clca channels was observed (P〉 0. 05). Chronic hypoxia increased [Ca^2+ ]i which opened Clca channels. The NFA and IAA-94 blocked the channels and decreased [Ca^2+ ]i from (281.75± 16.48) nmot/L to (117.66 ±15.36) nmol/L (P〈0.01). MTT assay showed that under chronic hypoxic condition NFA and IAA-94 decreased the value of absorbency (A value) from 0. 459±0. 058 to 0. 224±0. 025 (P〈0. 01). Hypoxia increased [Ca^2+ ]i which opened Cl~ channels and had a positive-feedback in [Ca^2+ ]i. This may play an important role in hypoxic pulmonary hypertension. Under chronic hypoxic condition, Clca channel may play a part in the regulation of proliferation of PASMCs. 展开更多

In this study, we investigated the effects of a combination of Ginkgo biloba extracts (GBE) and phosphodiesterase type 5 (PDE-5) inhibitors on the muscular tone of the corpus cavernosum and potassium channel activity of corporal smooth muscle cells. Strips of corpus cavernosum from male New Zealand white rabbits were mounted in organ baths for isometric tension studies. After contraction with 1 × 10^-5 mol I^-1 norepinephrine, GBE (0.01-1 mg ml^-1) and mirodenafil (0.01-100 nmol I^-1) were added together into the organ bath. In electrophysiological studies, whole-cell currents were recorded by the conventional patch-clamp technique in cultured smooth muscle cells of the human corpus cavernosum. The corpus cavemosum was relaxed in response to GBE in a dose-dependent manner (from 0.64%±8.35% at 0.01 mg ml^-1 to 52.28%±11.42% at 1 mg ml^-1). After pre-treatment with 0.03 mg ml^-1 of GBE, the relaxant effects of mirodenafil were increased at all concentrations, After tetraethylammonium (TEA) (1 mmol I^-1) administration, the increased effects were inhibited (P〈0.01). Extracellular administration of GBE increased the whole-cell K^+ outward currents in a dose-dependent fashion. The increase of the outward current was inhibited by I mmol 1-1 TEA. These results suggest that GBE could increase the relaxant potency of mirodenafil even at a minimally effective dose. The K+ flow through potassium channels might be one of the mechanisms involved in this synergistic relaxation. 展开更多

Cultured porcine endothelial cells (EC) produce and secrete plasminogenactivators (PA). If the serum free media incubated by vascular smooth muscle cells(SMC-CM) were mixed with the same media incubated by endothelial cells (EC-CM),the PA activities of the latter decreased significantly. Cocultivation of EC with SMC alsoresulted in a significant decrease (70.7%) of PA activities produced by EC. Sodiumdodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE) analysis of SMC-CMfollowed by reverse fibrin autography demonstrated that the PA inhibitor had a molecularweight of 49000-62000. In this study we also investigated the effect of a Chinese herbalmedicine-Radix Salviae Miltiorrhizae (RSM) on the inhibitory activity of SMC. Sports compression garments are common-selected sportswear to improve athletic performance and reduce sports injury. Since they consist of elastic textile that exerts compression and pressure onto the muscle, muscle activation is of interest to sports compression garment manufactures who seek to enhance sports performance through gradient compression garment design. However, there are few scientific investigations to explore this effect of muscle activation to evaluate the performance of sports compression garments during sport activity. In this paper, a systemic experimental protocol was presented to improve the quality of evaluation test under a standardized running test. Then muscle activation with and without wearing sport compression garments to highlight the changes caused by compression garment was investigated. The electromyography( EMG) signal was collected and processed by the proposed method to represent the muscle activation combined with gait analysis of running. As a result, the average muscle activation of gait cycle for the major muscles such as rectus femoris( RF) and gastrocnemius medialis( GM) revealed that they needed to exert more muscle force without wearing garment at certain periods for mid-stand and pre-swing phase within gait cycle. These lower muscle activation levels prolonged activity duration, reduced fatigue, and presented low risk for sport injury.Such an evaluation study therefore would scientifically prove the effect of compression garment and provide information on muscle activation and the ergonomic efficacy of gradient compression garment design. 展开更多

Most women like wearing high-heeled shoes for the benefit of sensuous attractiveness and self-esteem while musculoskeletal problems and gait patterns changes are often associated. The present study aimed to identify changes during the gait stance and swing phases in some lower limb muscles. In addition, abdominal muscle was included due to its importance in dynamic trunk stability, and lack of studies on the subject. Here, we found that the use of high-heeled shoes elicited not only the increasing of the electrical activity from the muscles involved in the gait cycle, but also altered the temporal sequence of their recruitment. As practical applications, these changes may be strategies to maintain stability and minimize risks of falling, but they are often associated to diseases. Women that use high-heeled shoes for prolonged time must apply specific muscle exercises to minimize its long-term effects. 展开更多

The aim of this short review is to describe the role of myosin isoforms during the adaptation of skeletal muscle to prolonged physical activity (for example endurance exercise) and to show the coordination between changes in muscle oxidative capacity and myofibrillar apparatus in slow-twitch and fast-twitch muscles. Adaptational changes in myosin isoforms during long lasting muscle activity (decrease of MyHC IIb isoforms relative content and increase of that MyHC IIa and decrease of MyLC 1 fast isoforms in fast-twitch muscles) are in good coordination with changes of muscle oxidative capacity. These changes show that during regular endurance exercise fast-twitch muscle fibers (type IIA) are also recruited and create the potential source of increase in endurance capacity during the process of adaptation to the prolonged physical activity. 展开更多

In this experiment, expression of tissue-type plasminogen activator (t-PA) in smooth muscle cells (SMCs) was measured at different intervals after the arterial injury. In the normal iliac arteries, only low levels of t-PA activity were estimated. t-PA activity in extracts of the iliac arteries increased significantlyat the 4th day after the injury, equivalent to the process that SMCs migrated from the media to the intima, and the t-PA activity was then decreased approximately to the normal level at the 7th day. Coexistent to the above data, results from in situ hybridization showed that the expression of t-PA mRNA in the intima as well as media increased also significantly at the 4th day after the arterial injury, and at the 7th day, tPA mRNA was detected only in those SMCs locating closely adjacent to the internal elastic lamina. These results suggest that t-PA might play an important role in SMC migration following endothelial injury, andantagonism of t-PA expression and/or activity within the vessel wall might be helpful in intervening the de velopment of restenosis following angioplasty. 展开更多

A water-soluble polysaccharide from abalone muscle(AMPP)was isolated. The contents of carbohydrate, protein, uronic acid, and sulfate in AMPP were 83.5%,0.5%,2.7%,and 2.6%,respectively.High-performance liquid chromatography analysis indicated that AMPP was homogeneous and had an average molecular weight of approximately 3.2 kDa.The main monosaccharides of AMPP were glucose(Glc)and mannose with a molar ratio of 99.7:0.3.The structural characteristics of AMPP were elucidated through methylation analysis,Fourier transform infrared spectroscopy, and nuclear magnetic resonance spectroscopy. The linkages of AMPP consisted of terminal,1,4-linked,1,6-linked,and 1,4,6-linked Glcp with a molar ratio of 3.1:7.2:1.0:2.5.In one repeat unit of the proposed AMPP structure, the backbone chain was composed of eight 1→4 glycosidic bonds and one 1→6 glycosidic bond, with three branch chains linked by 1→6 glycosidic bond.In addition,AMPP was found to possess potent immunostimulatory activity via rising phagocytosis of RAW264.7 cells and promoting secretion of TNF-α. 展开更多

The study examined the inhibitory effect of Atractylodes macrocephala (AM) on the uterine contraction during premature delivery and explored its electrophysiological mechanism by studying the effects of AM on the Ca^2+-activated K^+ currents of pregnant human myometrial smooth muscle cells with or without the treatment with intedeukin-6. Single cells were acutely isolated from pregnant human myometrial smooth muscles. Whole-cell Ca^2+-activated K^+ currents were recorded by using an Axopatchl-D amplifier. The cells were divided into three groups: group A in which AM was added into perfusate, group B, in which interleukin-6 was added into perfusate) and group C in which AM was added into perfusate after addition of interleukin-6. IL-6 10 ng/mL inhibited BKca by 36.9%±13.7% as compared with control (P〈0.01). AM at 2 mg/mL raised BKca by 36.7%±22.6% or 45.2%±13.7% with or without the treatment of IL-6, respectively (P〈0.01). It is concluded that AM was able to enhance the BKca of pregnant human myometrial smooth muscle cells treated or untreated with interleukin-6 and its effect on the BKca IL-treated cells was stronger that its effect on BKca of untreated cells. Our results suggested that AM can help to maintain the membrane potentials and the resting status of pregnant human myometrial smooth muscle cells. 展开更多

Objective:To investigate the effects of calcium-activated chloride (ClCa) channels on proliferation of pulmonary artery smooth muscle cells(PASMCs) in rats under chronic hypoxic condition. Methods:The cultured PASMCs were placed under normoxic and chronic hypoxic conditions:The cells were observed by light and electron microscope; The cell cycles were observed by flow-cytometry; Immunocytochemistry staining was used to detect the expressions of PCNA, c-fos and c-jun of PASMCs; Cytoplasmic free Ca^2+ concentration ([Ca^2+]i) in PASMCs was investigated by fluorescent quantitation using fluorospectrophotometer. Results:The PASMCs were contractile phenotype under normoxic conditions. Observation by transmission electron microscope: In kytoplasm of contractile phenotype cells, myofilament bundles were abundant and the content of cell organs such as Golgi's bodies were rare. The PASMCs were synthetic phenotype under chronic hypoxic condition. There were increased free ribosomes, dilated rough endoplasmic reticulums, highly developed Golgi complexes, decreased or disappeared thick filaments and dense body in kytoplasm of synthetic phenotype cells. After NFA and IAA-94, the situations were reversed The number of S +G2M PASMCs were significantly increased in chronic hypoxic condition; The NFA and IAA-94 were shown to significantly decrease them from (28.6±1.0)% to (16.0±1.6)% and the number of G0G1 PASMCs significantly increased from (71.4± 1.9)% to (83.9 ± 1.6)% (P〈 0.01). In chronic hypoxic conditions, the expression of proliferating cell nucleus antigen was significantly increased; The NFA and IAA-94 were shown to significantly decrease it from (81 ± 6)% to (27 ± 7)%(P 〈 0.01). The expression of c-fos and c-jun were significantly increased in'chronic hypoxic conditions; The NFA and IAA-94 were shown to significantly decrease them from 0.15 ±0.02, 0.32 ± 0.05 to 0.05 ± 0.01, 0.12 ± 0.05, respectively (P〈 0.01); Under chronic hypoxic conditions, [Ca^2+]i was increased; The NFA and IAA-94 decreased it from (281.8±16,5)nmol/L to (117.7 ± 15.4)nmol/L(P 〈 0.01). Conclusion:Hypoxia initiated the change of PASMCs from contractile to synthetic phenotype and increased proliferation of PASMCs. NFA and IAA-94 depressed cell proliferation by blocking ClCa channels in hypoxic condition. These may play an important role in proliferation of PASMCs under chronic hypoxic conditions. 展开更多
