Transport in water is the most common method for transporting live fish in China,however,transport is a strong stressor.Transport stress could lead to a reduced immune and antioxidant system function of tiger grouper,...Transport in water is the most common method for transporting live fish in China,however,transport is a strong stressor.Transport stress could lead to a reduced immune and antioxidant system function of tiger grouper,resulting in sickness and death.Besides,tiger grouper were continuously stressed during transport,which resulted in quality deterioration.It is necessary that find a way to relieve the stress of transportation of tiger grouper.Ascorbic acid is not only a good anti-stress agent,but it is also an effective immunostimulant.β-1,3-glucan is a feed additive that can enhance the immune response of fish.Therefore,this study evaluated the effects ofβ-1,3-glucan and ascorbic acid on the nutritional-immune response and antioxidant signaling pathways of live tiger grouper during simulated transport.Results indicated that addingβ-1,3-glucan and ascorbic acid in transport-water muted the increase of alanine aminotransferase(ALT)and aspartate aminotransferase(AST)activity.In addition,β-1,3-glucan and ascorbic acid activated Nrf2 and mediated TOR expression and then up-regulate related mRNA expression of antioxidant and immune enzymes.We concluded that the application ofβ-1,3-glucan and ascorbic acid inhibit the increase of metabolism enzymes and inflammatory factors and activate immune and antioxidant signaling pathways to relieve oxidant stress,immune response,and apoptosis.Reducing the loss of amino acids provided nutrients to relieve oxidative stress and immune response,which demonstrated immune-nutritional response in live tiger grouper during simulated transport.These results may provide a new solution for alleviating the decline of immune and antioxidant function of tiger grouper caused by transportation stress.展开更多

The cDNA of this gene is 1?644 nucleotides in length and the predicted polypeptide is 491 amino acids (aa)...The β-1,3-glucan recognition protein gene from Spodoptera exigua (SeβGRP) was cloned and characterized. The cDNA of this gene is 1?644 nucleotides in length and the predicted polypeptide is 491 amino acids (aa) in length, with a calculated molecular mass of 54.8 kDa. The first 22 aa encode a predicted secretion signal peptide. A BLAST search, multiple sequence alignment, and phylogenetic analysis of the aa sequence of SeβGRP revealed that this protein is most similar to the β-1,3-glucan recognition protein (βGRP) family of pattern recognition proteins. Using reverse-transcription polymerase chain reaction, we detected the presence of SeβGRP transcripts in the egg, larval, pupal, and adult stages of S. exigua. In addition, the SeβGRP transcript was expressed in all the tissues examined including the brain, hemocytes, fat body, intestine, and cuticle. There were no changes in SeβGRP mRNA levels in larvae infected with ultraviolet (UV)-killed Escherichia coli DH5α compared with the control larvae inoculated with the water; however, SeβGRP mRNA levels were markedly elevated 4–8 h after infection and slightly induced 12–24 h after infection in larvae injected with UV-killed Fusarium oxysporum. This may be because β-1,3-glucan is the main component of the cell wall of F. oxysporum, but not E. coli DH5α.展开更多

Many microalgae are now cultivated to produce bioactive substances. Odontella aurita is a marine diatom industrially cultured in outdoor open ponds and used for human nutrition. For the first time, we have systematically investigated the effects of culture conditions in cylindrical glass columns and fiat-plate photobioreactors, including nutrients (nitrogen, phosphorus, silicon, and sulfur), light intensity and light path, on O. aurita cell growth and biochemical composition (protein, carbohydrate, β-1,3-glucan, lipids, and ash). The optimal medium for photoautotrophic cultivation of O. aurita contained 17.65 mmol/L nitrogen, 1.09 mmol/L phosphorus, 0.42 mmol/L silicon, and 24.51 mmol/L sulfur, yielding a maximum biomass production of 6.1-6.8 g/L and 6.7-7.8 g/L under low and high light, respectively. Scale-up experiments were conducted with fiat-plate photobioreactors using different light-paths, indicating that a short light path was more suitable for biomass production of O. aurita. Analyses of biochemical composition showed that protein content decreased while carbohydrate (mainly composed of 15-1,3-glucan) increased remarkably to about 50% of dry weight during the entire culture period. The highest lipid content (19.7% of dry weight) was obtained under 0.11 mmol/L silicon and high light conditions at harvest time. Fatty acid profiles revealed that 80% were Cx4, C^6, and C20, while arachidonic acid and eicosapentaenoic acid (EPA) accounted for 1.6%-5.6% and 9%-20% of total fatty acids, respectively. High biomass production and characteristic biochemical composition profiles make O. aurita a promising microalga for the production ofbioactive components, such as EPA and D-1,3-glucan.展开更多
