血清ESM-1、E-cad对ACI-LAA溶栓后HT的预测价值

目的 探讨血清内皮细胞特异性分子-1 (ESM-1)、E-钙黏连蛋白(E-cad)对大动脉粥样硬化型脑梗死(ACI-LAA)溶栓后出血转化(HT)的预测价值。方法 选取2020年1月至2022年12月新疆医科大学第一附属医院收治的110例ACI-LAA患者,根据是否发生HT... 目的 探讨血清内皮细胞特异性分子-1 (ESM-1)、E-钙黏连蛋白(E-cad)对大动脉粥样硬化型脑梗死(ACI-LAA)溶栓后出血转化(HT)的预测价值。方法 选取2020年1月至2022年12月新疆医科大学第一附属医院收治的110例ACI-LAA患者,根据是否发生HT分为HT组和non-HT组。对比两组基础资料及血清ESM-1、E-cad水平。采用ROC曲线分析ESM-1、E-cad预测ACI-LAA患者发生HT的价值。结果 HT组患者NIHSS评分高于non-HT组,梗死面积大于non-HT组(P <0.05),HT组患者血清ESM-1、E-cad水平也高于non-HT组(P<0.05)。Logistic回归分析显示,梗死面积大、NIHSS评分高、血清ESM-1和E-cad水平升高是ACI-LAA患者发生HT的危险因素(P<0.05)。ESM-1联合E-cad预测ACI-LAA患者发生HT的曲线下面积为0.859,预测灵敏度为84.6%,特异度为72.6%。结论 血清ESM-1和E-cad水平与ACI-LAA患者HT密切相关,可作为早期预测发生HT的参考指标。 展开更多

miR-200c/ZEB1/E-cadherin轴在胆管癌转移侵袭中的作用及对胆管癌预后评估的价值

目的:探讨微小RNA-200c(miR-200c)/锌指转录因子(ZEB1)/E-钙黏蛋白(E-cadherin)在胆管癌(HCCA)转移、侵袭中的作用及对HCCA预后评估的临床价值。方法:选取2020年08月至2022年12月间我院收治的胆管癌患者36例为研究对象,同时选取胆管良... 目的:探讨微小RNA-200c(miR-200c)/锌指转录因子(ZEB1)/E-钙黏蛋白(E-cadherin)在胆管癌(HCCA)转移、侵袭中的作用及对HCCA预后评估的临床价值。方法:选取2020年08月至2022年12月间我院收治的胆管癌患者36例为研究对象,同时选取胆管良性疾病患者(胆总管结石或胆管良性狭窄)20例作为对照。采用原位杂交、实时荧光定量PCR检测miR-200c在HCCA患者组织及血清中的表达;生存曲线分析miR-200c表达与总生存期的相关性;Transwell实验检测细胞的迁移及侵袭能力;免疫印迹检测ZEB1及E-cadherin蛋白的表达。体内研究构建裸鼠成瘤实验,采用免疫组化检测ZEB1及E-cadherin蛋白的表达;Tunel技术检测组织中的凋亡情况。受试者工作特征(ROC)曲线评估生物标志物对胆管癌预后的诊断效能。结果:miR-200c低表达于HCCA组织及血清中;生存曲线分析显示,miR-200c表达与患者的总生存期呈正相关性,TNM分期与患者的总生存期呈负相关性;miR-200c mimics抑制了胆管癌细胞的迁移和侵袭能力,并降低ZEB1表达而诱导E-cadherin表达;转染ZEB1逆转miR-200c的作用效应,促进胆管癌细胞的迁移和侵袭特性,抑制胆管癌细胞的凋亡。CA199、CEA和miR-200c联合检测诊断胆管癌预后的ROC曲线下面积为0.892,灵敏度0.8611,特异度0.8000。结论:miR-200c/ZEB1/Ecadherin轴参与胆管癌的转移侵袭;CA199、CEA和miR-200c联合检测对胆管癌预后的诊断效能良好,对临床胆管癌的治疗策略具有重要指导价值。 展开更多

目的探讨血清E-钙粘连蛋白(E-cadherin)、血管内皮生长因子(VEGF)、细胞角蛋白19片段抗原21-1(CYFRA21-1)水平在甲状腺癌患者手术前后的变化及其对术后复发转移的预测价值。方法选取118例行甲状腺癌根治术的分化型甲状腺癌(DTC)患者为... 目的探讨血清E-钙粘连蛋白(E-cadherin)、血管内皮生长因子(VEGF)、细胞角蛋白19片段抗原21-1(CYFRA21-1)水平在甲状腺癌患者手术前后的变化及其对术后复发转移的预测价值。方法选取118例行甲状腺癌根治术的分化型甲状腺癌(DTC)患者为研究组,118例健康体检者为对照组;根据术后6个月复发转移情况,将DTC患者分为复发转移组33例与未复发转移组85例。比较对照组与研究组(术前)、研究组手术前后、复发转移组与未复发转移组血清E-cadherin、VEGF、CYFRA21-1水平,采用ROC曲线分析上述各指标对术后复发转移的预测价值。结果研究组血清E-cadherin水平显著低于对照组,血清VEGF、CYFRA21-1水平显著高于对照组(P<0.05);研究组术后2周血清E-cadherin水平显著高于术前,血清VEGF、CYFRA21-1水平显著低于术前(P<0.05);未复发转移组术后2周血清E-cadherin水平显著高于术后复发转移组,血清VEGF、CYFRA21-1水平显著低于术后复发转移组(P<0.05)。术后2周上述各指标联合检测预测DTC患者术后复发转移的AUC为0.862,敏感度为90.91%,特异度为75.29%。结论DTC患者术后血清中E-cadherin表达上调,VEGF、CYFRA21-1表达下调,各指标联合检测对复发转移具有较高的预测效能,可作为临床评估DTC患者术后复发转移情况的辅助指标。 展开更多

The autotetraploid Carassius auratus(4nRR,4n=200,RRRR)is derived from whole-genome duplication of Carassius auratus red var.(RCC,2n=100,RR).In the current study,we demonstrated that chromatophores and pigment changes directly caused the coloration and variation of 4nRR skin(red in RCC,brownish-yellow in4nRR).To further explore the molecular mechanisms underlying coloration formation and variation in 4nRR,we performed transcriptome profiling and molecular functional verification in RCC and 4nRR.Results revealed that scarb1,associated with carotenoid metabolism,underwent significant down-regulation in 4nRR.Efficient editing of this candidate pigment gene provided clear evidence of its significant role in RCC coloration.Subsequently,we identified four divergent scarb1 homeologs in 4nRR:two original scarb1 homeologs from RCC and two duplicated ones.Notably,three of these homeologs possessed two highly conserved alleles,exhibiting biased and allelespecific expression in the skin.Remarkably,after precise editing of both the original and duplicated scarb1homeologs and/or alleles,4nRR individuals,whether singly or multiply mutated,displayed a transition from brownishyellow skin to a cyan-gray phenotype.Concurrently,the proportional areas of the cyan-gray regions displayed a gene-dose correlation.These findings illustrate the subfunctionalization of duplicated scarb1,with all scarb1genes synergistically and equally contributing to the pigmentation of 4nRR.This is the first report concerning the functional differentiation of duplicated homeologs in an autopolyploidfish,substantiallyenrichingour understanding of coloration formation and change within this group of organisms. 展开更多

Space metallurgy is an interdisciplinary field that combines planetary space science and metallurgical engineering.It involves systematic and theoretical engineering technology for utilizing planetary resources in situ.However,space metallurgy on the Moon is challenging because the lunar surface has experienced space weathering due to the lack of atmosphere and magnetic field,making the mi-crostructure of lunar soil differ from that of minerals on the Earth.In this study,scanning electron microscopy and transmission electron microscopy analyses were performed on Chang’e-5 powder lunar soil samples.The microstructural characteristics of the lunar soil may drastically change its metallurgical performance.The main special structure of lunar soil minerals include the nanophase iron formed by the impact of micrometeorites,the amorphous layer caused by solar wind injection,and radiation tracks modified by high-energy particle rays inside mineral crystals.The nanophase iron presents a wide distribution,which may have a great impact on the electromagnetic prop-erties of lunar soil.Hydrogen ions injected by solar wind may promote the hydrogen reduction process.The widely distributed amorph-ous layer and impact glass can promote the melting and diffusion process of lunar soil.Therefore,although high-energy events on the lun-ar surface transform the lunar soil,they also increase the chemical activity of the lunar soil.This is a property that earth samples and tradi-tional simulated lunar soil lack.The application of space metallurgy requires comprehensive consideration of the unique physical and chemical properties of lunar soil. 展开更多

Quantifying freeze-melt dynamics of lakes on the Tibetan Plateau using Sentinel-1 synthetic aperture radar imagery

The ice phenology of alpine lakes on the Tibetan Plateau(TP)is a rapid and direct responder to climate changes,and the variations in lake ice exhibit high temporal frequency characteristics.MODIS and passive microwave data are widely used to monitor lake ice changes with high temporal resolution.However,the low spatial resolutions make it difficult to effectively quantify the freeze-melt dynamics of lakes.This work used Sentinel-1 synthetic aperture radar(SAR)data to derive high-resolution ice maps(about 6 days),then with the aid of Sentinel-2 optical images to quantify freeze-melt processes in three typical lakes on the TP(e.g.Selin Co,Ayakekumu Lake,and Nam Co).The results showed that three lakes had an average annual ice period of 125-157 days and a complete ice cover period of 72-115 days,from 2018 to 2022.They exhibit different ice phenology patterns.Nam Co is characterized by repeated episodes of freezing,melting,and refreezing,resulting in a prolonged freeze-up period.Meanwhile,the break-up period of Nam Co lasts for a longer duration(about 19 days),and the break-up exhibits a smooth process.Similarly,Ayakekumu Lake showed more significant inter-annual fluctuations in the freeze-up period,with deviations of up to 28 days observed among different years.Compared to the other two lakes,Selin Co experienced a relatively short freeze-up and break-up period.In short,Sentinel-1 SAR data can effectively monitor the weekly and seasonal variations in lake ice on the TP.Particularly,this data facilitates quantification of the freeze-melt dynamics. 展开更多

目的:探究不同卵巢浆液性肿瘤中E-钙黏蛋白(E-cadherin)、乳腺癌易感基因1(BRCA1)、P53的表达变化情况。方法:选取2017年12月—2022年2月赣州市妇幼保健院的50例卵巢浆液性癌患者为A组,同时期的40例卵巢交界性浆液性肿瘤患者为B组,30例... 目的:探究不同卵巢浆液性肿瘤中E-钙黏蛋白(E-cadherin)、乳腺癌易感基因1(BRCA1)、P53的表达变化情况。方法:选取2017年12月—2022年2月赣州市妇幼保健院的50例卵巢浆液性癌患者为A组,同时期的40例卵巢交界性浆液性肿瘤患者为B组,30例卵巢良性浆液性囊腺瘤患者为C组。检测及比较三组的组织E-cadherin、BRCA1、P53表达情况,并比较A组中不同年龄、淋巴结转移情况、FIGO分期及不同病理分级者的检测结果。结果:A组的E-cadherin及BRCA1阳性率显著均低于B组及C组,B组则显著均低于C组(P<0.05);A组P53阳性率显著高于B组及C组,B组则显著高于C组(P<0.05)。A组中低级别浆液性癌者E-cadherin和BRCA1阳性率均高于高级别浆液性癌者,低级别浆液性癌者P53阳性率显著低于高级别浆液性癌者,差异均有统计学意义(P<0.05)。A组中不同年龄及FIGO分期者的组织E-cadherin、BRCA1及P53阳性率比较,差异均无统计学意义(P>0.05)。A组有淋巴结转移者E-cadherin和BRCA1阳性率均低于无淋巴结转移者,有淋巴结转移者P53阳性率高于无淋巴结转移者,差异均有统计学意义(P<0.05)。结论:卵巢浆液性癌患者中E-cadherin及BRCA1呈低表达,而P53呈高表达,且不同临床病理情况者存在差异,因此在卵巢浆液性癌的早期筛查检测中有一定的参考价值。 展开更多

目的分析探讨稽留流产绒毛组织中胰岛素样生长因子-1(IGF-1)、E-钙黏附蛋白(E-cadherin)表达情况、微血管密度(MVD)与药物治疗结局的相关性。方法收集2019年9月至2021年8月在巴中市中心医院妇产科收治的进行药物治疗的稽留流产患者95例... 目的分析探讨稽留流产绒毛组织中胰岛素样生长因子-1(IGF-1)、E-钙黏附蛋白(E-cadherin)表达情况、微血管密度(MVD)与药物治疗结局的相关性。方法收集2019年9月至2021年8月在巴中市中心医院妇产科收治的进行药物治疗的稽留流产患者95例为研究组、正常早孕患者108例为对照组;所有患者均采用米非司酮配伍米索前列醇的统一药物方案进行治疗,采用免疫组织化学法检测绒毛组织中IGF-1、E-cadherin和MVD的表达,并分析药物治疗发生不完全流产的影响因素。结果研究组绒毛组织中IGF-1、E-cadherin的阳性表达率和MVD相对表达量均低于对照组(χ^(2)=4.67,P=0.03;χ^(2)=18.43,P<0.01;t=46.71,P<0.01);两组患者接受药物治疗后,不完全流产的发生率为18.72%;研究组不完全流产发生率(28.42%)高于对照组(10.19%),差异有统计学意义(χ^(2)=11.05,P<0.01);多因素Logistic回归分析结果显示,痛经(OR=2.24,95%CI:1.36~3.71,P<0.01)是药物治疗后发生不完全流产的独立危险因素;绒毛组织中E-cadherin的表达(OR=0.69,95%CI:0.55~0.70,P<0.01)和MVD的表达(OR=0.72,95%CI:0.68~0.78,P<0.01)均为患者接受药物治疗后发生不完全流产的独立保护因素。结论稽留流产患者绒毛组织中IGF-1、E-cadherin表达和MVD含量较低;接受药物治疗后不完全流产患者绒毛组织中E-cadherin、MVD相对较低。 展开更多

目的观察血小板内皮细胞黏附分子-1(PECAM-1)、E-选择素和血小板反应蛋白-1(TSP-1)对妊娠期高血压疾病(HDCP)患者发生妊娠结局不良的诊断效能。方法选取2020年1月至2022年1月在该院住院治疗的95例HDCP患者作为HDCP组,另选取同期在该院... 目的观察血小板内皮细胞黏附分子-1(PECAM-1)、E-选择素和血小板反应蛋白-1(TSP-1)对妊娠期高血压疾病(HDCP)患者发生妊娠结局不良的诊断效能。方法选取2020年1月至2022年1月在该院住院治疗的95例HDCP患者作为HDCP组,另选取同期在该院孕期保健检查的48例孕妇作为正常妊娠组。比较两组及不同严重程度HDCP患者血清PECAM-1、E-选择素和TSP-1水平,分析妊娠结局不良的影响因素,以及HDCP患者血清PECAM-1、E-选择素和TSP-1水平对妊娠结局不良的诊断效能。结果HDCP组血清PECAM-1水平明显低于正常妊娠组,其水平随着HCDP患者严重程度升高而降低,而HDCP组血清E-选择素和TSP-1水平均明显高于正常妊娠组,并且随着HDCP严重程度升高而升高,差异均有统计学意义(P<0.05)。妊娠结局不良组发病孕周、分娩孕周和血清PECAM-1水平均明显低于妊娠结局良好组,差异均有统计学意义(P<0.05);妊娠结局不良组血清E-选择素和TSP-1水平均明显高于妊娠结局良好组,差异均有统计学意义(P<0.05);妊娠结局不良组年龄、初产妇、孕前体质量指数、心率、收缩压和舒张压与妊娠结局良好组比较,差异均无统计学意义(P>0.05)。多因素Logistic回归分析结果显示,血清PECAM-1水平降低,血清E-选择素和TSP-1水平升高是HDCP患者发生妊娠结局不良的独立危险因素(P<0.05)。血清PECAM-1、E-选择素和TSP-1水平对HDCP患者发生妊娠结局不良具有较高的诊断效能,3项指标联合检测的灵敏度为82.2%,特异度为96.0%,曲线下面积为0.945,明显高于PECAM-1(Z=2.157,P=0.031)、E-选择素(Z=3.617,P<0.05)和TSP-1(Z=3.326,P=0.001)单项检测。结论血清PECAM-1、E-选择素和TSP-1是反映HDCP病情严重程度的指标,是HDCP患者发生妊娠结局不良的独立影响因素,3项指标联合检测对HDCP患者发生妊娠结局不良具有较高的诊断效能。 展开更多

Gamma-ray spectrometer (GRS) is one of the main payloads on the Chang'E-1 (CE-1) lunar probe, mainly aimed to detect the elemental abundances and distributions on the lunar surface. At 03:58 on 28 November 2007, it performed the first observation of the lunar gamma rays. As of 24 October 2008, 2105 h of effective gamma rays spectra had been acquired by CE-1 GRS, which covers the whole surface of the moon. This paper mainly describes the data processing procedures and methods of deriving the elemental abundances by using the CE-1 GRS time series corrected spectra: first, to bin data into pixels for mapping; then, to perform a background deduction of the cumulative spectra and obtain a peak area of the elements; and finally, to use the elemental abundances inversion model to produce the elemental abundances. Based on these processing methods, the global abundance maps of U, K, and Th at a 5°×5° equal-area pixel are acquired by CE-1 GRS data. The paper gives a preliminary analysis of the uncertainties of the elemental abundances. 展开更多

小冰川对气候变化非常敏感,监测与定量评估此类冰川变化有助于理解冰川对气候变化的响应幅度与机制。本研究结合多源遥感数据(卫星遥感与无人机航测),分析了近50年来青藏高原念青唐古拉山廓琼岗日1号冰川面积变化趋势,定量评估了该冰川... 小冰川对气候变化非常敏感,监测与定量评估此类冰川变化有助于理解冰川对气候变化的响应幅度与机制。本研究结合多源遥感数据(卫星遥感与无人机航测),分析了近50年来青藏高原念青唐古拉山廓琼岗日1号冰川面积变化趋势,定量评估了该冰川近期的冰面高程变化幅度与空间分布。结果表明,1968—2021年廓琼岗日小型冰斗冰川的面积从(1.444±0.013)km^(2)缩减至(0.712±0.001)km^(2),萎缩幅度达到50.7%,冰川末端退缩平均速率约为(6.23±0.71)m·a^(-1)。基于2020—2021年高精度无人机航测数据发现,廓琼岗日1号冰川冰面平均高程差达到(-2.41±0.69)m,冰川末端高程变化大于3 m,中部的冰面高程下降幅度在1.5~3 m之间。研究还发现冰川表面河道对冰面高程空间变化起着重要作用,该冰川表面共发育有13条表面河道,2020—2021年河道向西北方向偏移约2 m。冰面河道的向下侵蚀与侧向消融导致末端冰面高程变化呈现显著的空间差异。 展开更多

目的:探讨lncRNA ZFPM2-AS1靶向miR-519e-5p对脊索瘤U-CH1细胞生物学行为的影响。方法:体外培养U-CH1细胞,将ZFPM2-AS1的小干扰RNA以及miR-519e-5p的模拟物分别转入U-CH1细胞中,并随机分组:si-NC组、si-ZFPM2-AS1组、miR-NC组、miR-519e... 目的:探讨lncRNA ZFPM2-AS1靶向miR-519e-5p对脊索瘤U-CH1细胞生物学行为的影响。方法:体外培养U-CH1细胞,将ZFPM2-AS1的小干扰RNA以及miR-519e-5p的模拟物分别转入U-CH1细胞中,并随机分组:si-NC组、si-ZFPM2-AS1组、miR-NC组、miR-519e-5p组、si-ZFPM2-AS1+anti-miR-NC组、si-ZFPM2-AS1+anti-miR-519e-5p组;实时荧光定量聚合酶链式反应(RT-qPCR)检测U-CH1细胞中lncRNA ZFPM2-AS1与miR-519e-5p表达量;MTT、流式细胞术、划痕实验、Transwell实验分别检测U-CH1细胞增殖、凋亡、迁移、侵袭;双荧光素酶报告实验检测lncRNA ZFPM2-AS1与miR-519e-5p的靶向调控关系;蛋白印迹(western blotting)法分析蛋白E-cadherin、N-cadherin表达。结果:与si-NC组、miR-NC组比较,si-ZFPM2-AS1组、miR-519e-5p组细胞活力、划痕愈合率、侵袭细胞数及蛋白N-cadherin水平下降,凋亡率、E-cadherin水平增高(P<0.05);lncRNA ZFPM2-AS1可负向调控miR-519e-5p表达;si-ZFPM2-AS1+anti-miR-519e-5p组与si-ZFPM2-AS1+anti-miR-NC组比较,细胞活力、划痕愈合率、侵袭细胞数及蛋白N-cadherin水平增高,凋亡率、E-cadherin水平下降(P<0.05)。结论:下调lncRNA ZFPM2-AS1可促进U-CH1细胞凋亡并阻滞增殖、迁移、侵袭,其机制与上调miR-519e-5p表达有关。 展开更多

目的 探讨妊娠期高血压疾病(HDP)孕妇血清E-选择素(E-selection)、富亮氨酸重复序列相互作用蛋白1(LRRFIP1)水平对不良分娩结局的预测价值。方法 选择2020年6月至2022年6月秦皇岛市妇幼保健院接收的200例HDP孕妇为HDP组,根据疾病严重程... 目的 探讨妊娠期高血压疾病(HDP)孕妇血清E-选择素(E-selection)、富亮氨酸重复序列相互作用蛋白1(LRRFIP1)水平对不良分娩结局的预测价值。方法 选择2020年6月至2022年6月秦皇岛市妇幼保健院接收的200例HDP孕妇为HDP组,根据疾病严重程度,分为妊娠期高血压组(69例)、子痫前期组(90例)和重度子痫前期组(41例);根据是否发生不良分娩结局,分为分娩不良组(128例)和分娩良好组(72例)。选取同期产检正常者100例为对照组。采用酶联免疫吸附法检测血清E-selection、LRRFIP1水平;采用受试者工作特征曲线分析血清E-selection、LRRFIP1水平对HDP孕妇不良分娩结局的预测价值。结果 HDP组孕妇血清中E-selection水平为55.96±4.08ng/mL、LRRFIP1为7.86±1.29ng/mL,均高于对照组,差异有统计学意义(P<0.05)。妊娠期高血压组、子痫前期组、重度子痫前期组血清E-selection、LRRFIP1水平比较,差异均有统计学意义(P<0.05)。与分娩良好组相比,分娩不良组孕妇血清中E-selection、LRRFIP1水平上升,差异有统计学意义(P<0.05)。与对照组相比,HDP组不良分娩结局的总发生率较高,差异有统计学意义(P<0.05)。血清E-selection、LRRFIP1水平预测HDP孕妇不良分娩结局的曲线下面积(AUC)分别为0.712和0.709,两者联合预测的AUC高于单独预测(Z=5.662、5.828,P<0.05)。结论 HDP孕妇血清E-selection、LRRFIP1水平升高,与HDP病情程度关系密切,均有望作为不良分娩结局的预测因子。 展开更多

A map of the average atomic number of lunar rock and soil can be used to differentiate lithology and soil type on the lunar surface.This paper establishes a linear relationship between the average atomic number of lunar rock or soil and the flux of position annihilation radiation(0.512-Me V gamma-ray) from the lunar surface.The relationship is confirmed by Monte Carlo simulation with data from lunar rock or soil samples collected by Luna(Russia) and Apollo(USA) missions.A map of the average atomic number of the lunar rock and soil on the lunar surface has been derived from the Gamma-Ray Spectrometer data collected by Chang'e-1,an unmanned Chinese lunar-orbiting spacecraft.In the map,the higher average atomic numbers(ZA > 12.5),which are related to different types of basalt,are in the maria region;the highest ZA(13.2) readings are associated with Sinus Aestuum.The middle ZA(~12.1) regions,in the shape of irregular oval rings,are in West Oceanus Procellarum and Mare Frigoris,which seems to be consistent with the distribution of potassium,rare earth elements,and phosphorus as a unique feature on the lunar surface.The lower average atomic numbers(ZA < 11.5)are found to be correlated with the anorthosite on the far side of the Moon. 展开更多

目的探索碱性核蛋白1(basonuclin 1,BNC1)基因对肺鳞状细胞癌细胞迁移和侵袭能力的影响及其作用机制。方法利用生物信息学分析在线数据库中BNC1基因在肺鳞状细胞癌患者中的表达规律及对生存的影响;采用免疫组织化学对组织芯片进行染色... 目的探索碱性核蛋白1(basonuclin 1,BNC1)基因对肺鳞状细胞癌细胞迁移和侵袭能力的影响及其作用机制。方法利用生物信息学分析在线数据库中BNC1基因在肺鳞状细胞癌患者中的表达规律及对生存的影响;采用免疫组织化学对组织芯片进行染色并分析BNC1的蛋白水平;在癌症细胞系百科全书(Cancer Cell Line Encyclopedia,CCLE)数据库中筛选BNC1高表达和低表达的肺鳞状细胞癌细胞系;对敲低和过表达的肺鳞状细胞癌细胞H226、SK-MES1进行划痕愈合实验、Transwell迁移和侵袭实验,检测细胞的迁移与侵袭能力的变化;采用RT-qPCR验证10个文献来源的有关迁移与侵袭高度相关的基因;采用Western blot对筛选的调控基因进一步验证。结果生物信息学分析发现BNC1高表达于肺鳞状细胞癌组织且与较低生存率相关(P=0.047);肺鳞状细胞癌组织中BNC1的表达高于癌旁组织(P<0.05);H226细胞系BNC1表达较高,而SK-MES1细胞系BNC1表达较低;敲低BNC1能够抑制肺鳞状细胞癌细胞的迁移和侵袭(P<0.05),过表达BNC1可增强肺鳞状细胞癌细胞的迁移和侵袭(P<0.05);在mRNA层面筛选BNC1下游调控分子,发现E-钙粘蛋白(E-cadherin,CDH1)在BNC1过表达时减低(P<0.05),在BNC1敲低时升高(P<0.05),Western blot验证结果与mRNA表达一致(P<0.05)。结论BNC1可能是肺鳞状细胞癌患者的预后不良基因,BNC1基因可能通过下调CDH1的表达,促进肺鳞状细胞癌细胞的迁移和侵袭。 展开更多

Piezoelectric semiconductors(PSs)possess both semiconducting properties and piezoelectric coupling effects,making them optimal building blocks for semiconductor devices.PS fiber-like structures have wide applications in multi-functional semiconductor devices.In this paper,a one-dimensional(1D)theoretical model is established to describe the piezotronic responses of a PS fiber under gradient temperature changes.The theoretical model aims to explain the mechanism behind the resistance change caused by such gradient temperature changes.Numerical results demonstrate that a gradient temperature change significantly affects the physical fields within the PS fiber,and can induce changes in its surface resistance.It provides important theoretical guidance on the development of piezotronic devices that are sensitive to temperature effects. 展开更多

目的分析Ⅰ型子宫内膜癌患者中缺氧诱导因子-1(HIF-1)、E-钙黏蛋白(E-cad)的表达及其相关性。方法选取子宫内膜非典型增生病变患者、子宫内膜健康者、Ⅰ型子宫内膜癌患者各30例,所有受试者均接受HIF-1、E-cad检测,观察其检测结果。结果... 目的分析Ⅰ型子宫内膜癌患者中缺氧诱导因子-1(HIF-1)、E-钙黏蛋白(E-cad)的表达及其相关性。方法选取子宫内膜非典型增生病变患者、子宫内膜健康者、Ⅰ型子宫内膜癌患者各30例,所有受试者均接受HIF-1、E-cad检测,观察其检测结果。结果Ⅰ型子宫内膜癌患者HIF-1阳性率83.33%(25/30)高于子宫内膜非典型增生病变患者36.67%(11/30)子宫内膜健康者10.00%(3/30),E-cad阳性率33.33%(10/30)低于子宫内膜非典型增生病变患者93.33%(28/30)、子宫内膜健康者96.67%(29/30),差异有统计学意义(P<0.05)。分析得知,Ⅰ型子宫内膜癌患者E-cad阳性表达与病理学分期、肌层浸润程度、淋巴结转移有关(P<0.05),HIF-1阳性表达均与组织学分级、病理学分期、肌层浸润程度、淋巴结转移有关(P<0.05)。Spearman分析显示,Ⅰ型子宫内膜癌中E-cad与HIF-1成负相关(r=-0.561,P<0.05)。结论E-cad、HIF-1均在子宫内膜癌病变中有参与性,联合检测E-cad、HIF-1在判定Ⅰ型子宫内膜癌患者和病变侵袭性方面具有临床价值。 展开更多

Changes in hemeoxygenase-1 and superoxide dismutase in the peri-hematomal brain tissues of rats following intracerebral hemorrhage

BACKGROUND: The mechanism of intracerebral hemorrhage (ICH)-induced hemorrhagic brain injury is very complicated, involving the position-occupying effect of cephalophyma, ischemic factors, the toxic effect of hematoma components, the destruction of blood-brain barrier, etc. The expression and effect of hemeoxygenase-1 (HO-1) in the cerebrovascular disease has been paid close attention. OBJECTIVE: To observe the expression of HO-1 and change of superoxide dismutase (SOD) in the peri-hematomal brain tissue of rats following ICH. DESIGN: Randomized controlled animal experiment. SETTING: Department of Neurology, Yijishan Hospital Affiliated to Wannan Medical College. MATERIALS: Forty healthy male SD rats, of clean grade, weighing from 250 to 300 g, were provided by Qinglongshan Animal Farm of Nanjing. The involved 40 rats were randomized into sham-operation group (n =5) and ICH group (n =35), and ICH group was divided into 7 subgroups with 5 rats in each: ICH 6, 12, 24, 48, 72, 100 and 168 hours groups. Rabbit anti-rat HO-1 immunohistochemial kit ( Boster Co., Ltd., Wuhan) and SOD kit (Jiancheng Bioengineering Institute, Nanjing)were used in this experiment. METHODS: This experiment was carried out in the Department of Neurology, Yijishan Hospital Affiliated to Wannan Medical College Between April and July 2005. In the ICH group: Autologous blood of rats was injected into the head of caudate nucleus to create ICH animal models. In the sham-operation group, the same amount of normal saline was injected into the head of caudate nucleus of rats. The brains of rats in each group were harvested at different time points. The hematoma-side brain tissue was cut open in the coronal plane taking hematomal region as center, and the posterior part was fixed with 100 g/L neutral formaldehyde. 100 mg brain tissue was taken from anterior part. The number of positive cells in HO-1 and SOD activity in peri-hematomal brain tissue at different time after ICH were detected by immunohistochemical method and xanthine oxidation method respectively. MAIN OUTCOME MEASURES: ① The expression of HO-1 in the peri-hematomal brain tissue of rats in two groups following ICH.② The expression of SOD activity in the peri-hematomal brain tissue of rats in two groups following ICH. RESULTS: ①The number of HO-1 positive cells in the peri-hematomal brain tissue of rats in two groups following ICH 6, 12, 24, 48, 72, 120 and 168 hours was (11.03±2.01),(16.47±2.98),(25.50±5.65),(51.57±7.05),(47.33±4.73),(26.57±5.12),(7.63±2.17) cells/high-fold visual field , respectively; The number of HO-1 positive cells in the ICH 12-120 hours groups was significantly higher than that of sham-operation group [(6.07±1.85)cells/high-fold visual field, P < 0.01]; The HO-1 positive cells were the most in the ICH 48 hours group and were still expressed a little in the ICH 168 hours group. ② The SOD in the brain tissue of rats at ICH 6, 12, 24, 48, 72, 120 and 168 hours was (404.46±8.14),(396.84±10.97),(387.74±5.32),(356.21±9.27),(307.95±10.15),(357.48±11.28) and (402.98±7.23) kNU/g, respectively; The SOD activity of ICH 12 to 120 hours groups was significantly lower than that of sham-operation group [(415.47±11.44) kNU/g,P < 0.01], and that of ICH 72 hours group was the lowest. There was no significant difference of SOD activity between ICH 168 hours group and sham-operation group (P > 0.05). CONCLUSION: Following ICH, the expression of HO-1 in peri-hematomal brain tissue of rats in two groups is obviously increased, but the antioxidant ability of brain tissue is decreased. The changes of both maybe play an important role in the formation of ICH-induced hemorrhagic brain injury. 展开更多
