An efficient,economical,and phosgene-free approach was developed for the preparation of l,4-dihydro-2H-3,l-benzoxazin-2-one from 2-aminobenzyl alcohol.In terms of its key features,this reaction uses the cheap and recyclable non-metal selenium as a catalyst instead of the noble metal palladium;carbon monoxide as a carbonylation agent instead of virulent phosgene or one of its derivatives;and oxygen as an oxidant.The selenium-catalyzed oxidative carbonylation reaction of2-aminobenzyl alcohol proceeded efficiently in a single pot in the presence of triethylamine to afford l,4-dihydro-2H-3,l-benzoxazin-2-one in 87%yield.Furthermore,the selenium catalyst was readily recovered and recycled,affording a product yield of 80%after five cycles. 展开更多

背景与目的:目前有关宫颈病变的DNA甲基化研究较多,但DNA甲基化作为宫颈病变的诊断及分流指标在临床实践中的报道较少。本研究拟探讨FAM19A4、PAX1及miRNA124-2基因启动子区甲基化在宫颈病变进展中早期诊断的价值。方法:收集2020年3月—... 背景与目的:目前有关宫颈病变的DNA甲基化研究较多,但DNA甲基化作为宫颈病变的诊断及分流指标在临床实践中的报道较少。本研究拟探讨FAM19A4、PAX1及miRNA124-2基因启动子区甲基化在宫颈病变进展中早期诊断的价值。方法:收集2020年3月—2022年3月在郑州大学第三附属医院同时行宫颈液基细胞学(thinprep cytologic test,TCT)和HPV检测的患者的宫颈细胞学标本共129例,采用甲基化特异性PCR(methylation-specific PCR,MSP)检测不同宫颈病变中FAM19A4、PAX1及miRNA124-2基因启动子区甲基化改变情况,采用受试者工作特征(receiver operating characteristic,ROC)曲线评估3个基因甲基化改变对宫颈病变的诊断价值。本研究经郑州大学第三附属医院伦理委员会批准(伦理审批编号:2023-135-01)。结果:根据病理学检查结果分为4组:未见上皮内病变或恶性细胞(no intraepithelial lesions or malignant lesions,NILM)组(42例)、低度鳞状上皮内病变(low grade squamous intraepithelial lesion,LSIL)组(28例)、高度鳞状上皮内病变(high grade squamous intraepithelial lesion,HSIL)组(36例)和鳞癌(squamous cervical cancer,SCC)组(23例)。随宫颈病变级别的增加,FAM19A4、PAX1及miRNA124-2基因甲基化检出率逐步增高,差异有统计学意义(P均<0.05)。在HSIL组FAM19A4、PAX1、miRNA124-2甲基化检出率分别为81.2%、80.5%和71.8%,在SCC组3种基因甲基化检出率均高达100.0%;细胞学诊断宫颈癌的ROC曲线下面积(area under curve,AUC)为0.731,诊断灵敏度和特异度分别为65.9%和80.4%;FAM19A4、PAX1及miRNA124-2基因单独诊断HSIL+(HSIL和SCC)时,PAX1甲基化诊断HSIL+的效能最高,AUC为0.925,灵敏度为92.8%,特异度为87.3%;两两联合诊断时,FAM19A4与PAX1联合诊断HSIL+的AUC为0.930,灵敏度为95.7%,特异度为87.1%;FAM19A4与miRNA124-2联合诊断HSIL+,AUC为0.895,灵敏度为97.6%,特异度为85.7%;PAX1联合miRNA124-2诊断HSIL+,AUC为0.928,灵敏度为95.7%,特异度为89.1%;PAX1、FAM19A4及miRNA124-2基因甲基化联合诊断HSIL+,AUC为0.928,灵敏度为100.0%,特异度为81.8%。结论:FAM19A4、PAX1及miRNA124-2基因启动子区甲基化诊断宫颈病变具有较高的灵敏度和特异度,有潜力成为宫颈病变早期诊断的新指标。 展开更多

BACKGROUND: To date, a complete protein expression profile of the midbrain substantia nigra in a mouse model of chronic Parkinson's disease, induced by 1-methyl-4-phenyl-1, 2, 3, 6-tetrahydropyridine (MPTP), does not exist. In addition, there are no reports of analysis of differential protein expression. OBJECTIVE: To separate and evaluate MPTP-induced differential protein expression through the use of proteomics in the substantia nigra of a mouse model of chronic Parkinson's disease. DESIGN: Randomized controlled animal study. SETTING: Department of Neurology, the First Affiliated Hospital, Chongqing Medical University. MATERIALS: Sixteen 8-10-week old, healthy, male, C57BL mice, weighing 20-25 g, and of clean grade, were provided by the Experimental Animal Center of Chongqing Medical University. The experimental animals were disposed according to ethical criteria. MPTP was provided by Sigma Company, USA; Pdquest 2D image analysis software and gelatum/irradiance image analysis system (ChemiDoc XRS) by Bio-Rad, USA; and Voyager DE-PROMALD1-TOF-MS mass spectroscopy analyzer by AB1 Company, USA. METHODS: This study was performed in Chongqing Neurological Laboratory between November 2006 and July 2007. Mice were randomly divided into model and control groups, with 8 mice in each group. Mice in the model group were received a subcutaneous injection of MPTP (25 mg&g), twice a week, for five successive weeks, to establish a chronic Parkinson's disease model. Mice in the control group received the same volume of a subcutaneous saline injection at the same time points. Mice were sacrificed by anesthesia to rapidly obtain the midbrain for protein separation of the substantia nigra. MAIN OUTCOME MEASURES: (1) 2-ED handbook (Bio-Rad Company) was referenced for two-dimensional electrophoresis, (2) PDQUEST8,0 analytical electrophoresis pattern was adopted to evaluate differential protein expression. (3) Peptide mass finger print map and data were retrieved on http://www.prospector.ucsf.edu to compare differential substantia nigral protein expression in the two groups. RESULTS: Two-dimensional gel electrophoresis of substantia nigra tissue indicated that there were 33 differential protein expressions between the two groups. Three new proteins were evaluated, including α -enolase, which exhibited regulated expression, tumor necrosis factor ligand superfamily member 4, and cyclin-dependent kinase inhibitor 1B. CONCLUSION: There are three proteins that exhibit differential expression in the substantia nigra- α -enolase, tumor necrosis factor ligand superfamily member 4, and cyclin-dependent kinase inhibitor 1B. 展开更多

The cis and trans isomers separation of 2-butene-1,4-diol and lafutidine were studied by HPLC on two kinds of chiral columns: (S,S)-Whelk-O 1 and ChiraSpher. The isomers of 2-butene-1,4-diol can be separated on both chiral columns while the isomers of lafutidine can only be resolved on ChiraSpher column. The influence of different type and amount of mobile phase modifier on the isomers separation was extensively studied. The resolution of cis and trans isomers of 2-butene-1,4-diol was 2.61on (S,S)-Whelk-O 1 column with hexane-ethanol (97:3, v/v) as the mobile phase. The resolution of lafutidine was 1.89 on ChiraSpher column with hexane-ethanol-THF-diethylamine (92:3:5:0.1, v/v/v/v) as the mobile phase. LC-MS methods were developed to identify the isomer peaks. 展开更多

In order to find leading compounds with an excellent fungicidal activity, the title compound 2-( 1,3-dithiolan-2- yl-idene)-1-phenyl-2-( 1,2,4-tfiazol-1-yl) ethanone was synthesized according to the biological isosterism and its structure was confirmed by means of IR, MS, ^1 H NMR and elemental analysis. The single crystal structure of the title compound was determined by X-ray diffraction. The preliminary biological test shows that the synthesized compound exhibits some biological activities. 展开更多

The structure was solved in monoclinic, space group P21/c with a = 9.573(3), b = 19.533(7), c = 9.876(3), β = 92.35(4)°, V = 1845.2(10)3, T = 85(2) K, Z = 4, R = 0.040 and wR = 0.089 for 6424 observed reflections with I2σ(I). 展开更多

The novel crystal of the title compound 2-(((5-(((5,7-dimethyl-[1,2,4]triazolo-[1, 5-a]-pyrimidin-2-yl)thio)methyl)-4-phenyl-4H-1,2,4-triazol-3-yl)thio)methyl)-4H-chromen-4-one methanol solvate(C27H25N7O3S2, Mr = 559.66) has been prepared and its structure was determined by single-crystal X-ray diffraction. The crystal belongs to the monocline system, space group P21/c with a = 11.9879(9), b = 14.0743(10), c = 15.9706(11)A, β = 98.509(2)°, V = 2664.9(3)A3, Z = 4, Dc = 1.395 g/cm^3, F(000) = 1168, μ = 0.116 mm-1, MoKa radiation(λ = 0.71073), R = 0.0652, and wR = 0.1425 for 5220 observed reflections with I 〉 2σ(I). X-ray diffraction analyses reveal that the molecule adopts a C-shape. The planes of the benzopyrone and triazolopyrimidine were nearly parallel to each other, with a dihedral angle of 1.21(3)°. Intramolecular and intermolecular hydrogen bonds together with π-π interations are found to exist in the structure. The results of MTT assay indicate that the title compound displays excellent antiproliferative activity against two human cancer cell lines. 展开更多

UV and 1H NMR spectre TG-DTA, molar conductance and X-ray powder diffraction . Two inorganic-organic coordination polymers, [Cd(TBA)2]·3H2O (1) and [Cu(TBA)2]·2H2O (2) have been synthesized by reaction of the metal ions of Cd(II) and Cu(II) with the ligand HTBA [HTBA = 4- (1H-1, 2, 4-triazol-1-yl)benzoic acid] under solvothermal condition. The compounds were characterized by single-crystal X-ray diffraction, elemental analysis, IR spectroscopy, X-ray powder diffraction (XRPD), and thermo gravimetric analysis (TGA). The structural analyses reveal the two independent polymers exhibit different structures. Complex 1 exhibits a 3D framework with 1D channels, and complex 2 shows a 2-fold interpenetrating 3D diamond net. 展开更多

Background: The plasminogen activator inhibitor-1 (PAI-1) is a puissant antifibrinolytic factor;plasma PAI-1 level is high in type 2 diabetes. 4G/5G polymorphism of PAI-1 gene is a major genetic determinant of plasma PAI-1 levels, with 4G carriers having high PAI-1 level than 5G, theses pose the question about relation T2 patients and those polymorphisms. The aim of this study was to determine the relationship between the polymorphisms &minus;675 4G/5G and &minus;844 G/A of PAI-1 gene and type 2 diabetes mellitus. Methods: A case control study of 491 diabetic and 400 healthy controls. Genotyping of the polymorphism &minus;675 4G/5G was done by PCR-ASA (polymerase chain reaction, allele specific amplification), and the polymorphism &minus;844 G/A was done with PCR-RFLP (restriction fragment length polymorphism), the allelic frequency is calculated with hardy-Weinberg law, the statistic analysis was done by SPSS version 10. Results: Higher frequencies of The genotypes 4G/4G (p = 0.01) and 4G/5G of polymorphism &minus;675 4G/5G were seen in diabetic (p = 0.05) and higher frequencies of 5G/5G was seen in controls (p &minus;844 G/A was seen in diabetics and G/G was seen in controls (p = 0.01). Conclusion: Our study found association between 4G allele of &minus;675 4G/5G and A allele of &minus;844 G/A of PAI-1 gene and having type 2 diabetes mellitus in Tunisian population. 展开更多
