[Objective] This study aimed to construct DNA vaccine of foot-and-mouth disease (FMD).[Method] Plasmid carriers plESZP1 and pUTK3CP1 with PRV were constructed for FMDV P1 gene expression.Mice were immunized,and thei...[Objective] This study aimed to construct DNA vaccine of foot-and-mouth disease (FMD).[Method] Plasmid carriers plESZP1 and pUTK3CP1 with PRV were constructed for FMDV P1 gene expression.Mice were immunized,and their antibody level was detected.The two eukaryotic expression plasmids constructed were transfected into Vero cells.PCR,IFA and Westem-blot were carried out to detect the transcription and expression of the objective gene.Balb/C mice were intramuscularly inoculated with the DNA plasmid which expressed the target gene correctly,and the antibody level in mice was detected by the means of ELISA and serum neutralization (SN).[Result] DNA plasmid carrying P1 gene which encodes FMDV capsid protein caused specific body fluid immunoreaction in mice,and the antibody level of anti-FMDV had no difference in the mice induced by the two recombinant plasmids.[Conclusion] This study lays a foundation for evaluating the genetically modified vaccine by immunizing animals with recombinant PRV containing the FMDV P1 gene and recombinant virus.展开更多

Experimental measurements of the edge fluctuations, velocities of the toroidal, the poloidal flow and electric field have been carried out on both of SOL and the boundary region of HL-1M for Ohmic, biased H-mode, Lower Hybrid Current Drive (LHCD), Supersonic Molecular Beam Injection (MBI), Multi-shot Pellet Injection (MPI), Neutral Beam Injection (NBI), Ion Cyclotron Resonance Heating (ICRH) and Electric Cyclotron Resonance Heating (ECRH) discharges. The results show that the suppressions of the fluctuations are related to poloidal rotations produced by different discharge modes in the improved particle confinement property, simultaneously the change of the radial and poloidal electric field is generated and becomes more negative at the Tokamak plasma edge, and the sheared poloidal flow is related to the reduction in fluctuation level, and the poloidal velocity is mainly dominated by the E × B drift.展开更多

Objectives: Assess the impact of parents’ level of knowledge, from their psychosocial experience and from the quality of life on the glycemic control of children, adolescents and young adults with type 1 (DT1) diabetes in Brazzaville. Patients and Methods: This was an analytical cross-sectional study with prospective data collection during 7 months at Brazzaville University Hospital CHU, involving 103 parents. We studied socio-demographic variables, the balance of children’s diabetes by glycated hemoglobin, parents’ knowledge of diabetes by questionnaire “Michigan Diabetes Research and Training Center’s Revised Diabetes Knowledge Test”. The assessment of anxiety and depression through the “Hospital Anxiety and Depression” (HAD) score. Parental stress was assessed by the “Pediatric Inventory for Parents” (PIP). Results: We reported certain anxiety symptomatology in 60 parents (58.3%). Certain depression was reported in 63 parents (61.2%), and a lesser degree of parental distress. These psychosocial disorders in the parents did not influence the glycemic balance of the children. Fear of hypoglycemia has been found stable over time if not treated. Uncooked cornstarch has been found to reduce the risk of hypoglycemia but has not been studied in relation to fear of hypoglycemia. The aims of this study were to through clinical data, self-reported measures and clinical interviews explore subjects’ experience of using uncooked cornstarch before bedtime and their beliefs and experiences of fear of hypoglycemia. Methods: Mixed methods with both quantitative and qualitative data were used. Self-reported measures of hypoglycemia and fear of hypoglycemia were compared to subjects’ responses during a clinical interview. The interviews were analyzed with a functional behavior analytical approach. Results: A total of five subjects took part in the study. One subject perceived the uncooked cornstarch helpful in reducing hypoglycemia. Several subjects could recall frightening hypoglycemic episodes triggering their fear. Three out of the five subjects reported avoidance behaviors such as excessive self-monitoring of blood glucose or overeating related to fear of hypoglycemia. Conclusions: The uncooked cornstarch was found appetizing but was not perceived as having an effect on BG or hypoglycemia frequency. Objective:To assess emotional fluctuations,physical and mental health status,and indicators closely related to red blood cells,such as RIO kinase 3(Riok3),MAX interactor 1(Mxi1),and microRNA 191(miR191),in participant...Objective:To assess emotional fluctuations,physical and mental health status,and indicators closely related to red blood cells,such as RIO kinase 3(Riok3),MAX interactor 1(Mxi1),and microRNA 191(miR191),in participants with different levels of red blood cells.Methods:Participants who underwent physical examinations at Dongfang Hospital between April and October 2019 were divided into healthy,blood deficiency,and anemia groups(30 individuals in the healthy and blood deficiency group respectively,and 13 in the anemia group).The physical and mental conditions of the participants were evaluated through questionnaires,and emotional fluctuations were assessed through an emotion-inducing experiment,in which participants watched video segments designed to induce specific emotions.Relative expression levels of miR191,Riok3,and Mxi1 from venous blood samples were also determined.Results:The main psychological factors identified in the anemia and blood deficiency groups were obsessive-compulsive symptoms,depression,anxiety,and other negative emotions.Relative gene expression levels indicated that miR191 was upregulated and Riok3 and Mxi1 were downregulated in both the blood deficiency and anemia groups.Regarding the emotional score of disgust on video stage,the main effect was significant(F=335.58,P<.001),which showed that watching the three videos caused participants to have a dominant emotion,and there is a difference on group(F=5.35,P=.01),with higher disgust scores in the anemia and blood deficiency groups.The symptoms of blood deficiency and anemia,such as weakness in limbs were significantly negatively correlated with Riok3 and Mxi1 expression(r=-0.38 and-0.31 respectively),but was significantly positively correlated with miR191 expression(r=0.29).Conclusion:We determined that a close relationship exists between red blood cell levels and emotional status.Our findings suggest that individuals with anemia and blood deficiency are more likely to experience psychological problems and negative emotions,particularly disgust.We also demonstrate that emotional regulation is related to mir191-Riok3-Mxi1 pathway activity,identifying these pathway components are potential targets for genetic therapies in combination with psychological therapy.展开更多

文摘为了促进广谱高效大豆埃氏慢生根瘤菌Y63-1菌株的高效生产与推广应用,本研究对其培养基进行了筛选与优化。首先比较了Y63-1在YMA、TY、SM、PA和BSE 5种根瘤菌基本培养基中的生长速度,结果表明Y63-1在TY基本培养基中生长最快。以TY为基本培养基进行单因素碳源及无机盐利用试验,结果表明葡萄糖是Y63-1生长的最佳碳源,CaCl_(2)为必要的培养基成分,Rh微量元素对菌株的生长有很大的促进作用。进一步对蛋白胨、葡萄糖、酵母粉及Rh微量元素4种组分进行正交优化,获得了适宜Y63-1生长的最佳培养基,配方(1 L)为:8 g蛋白胨、10 g葡萄糖、3 g酵母粉、0.1 g CaCl_(2)·6H_(2)O、3 mL Rh微量元素,pH7.0。此培养基也能显著提升USDA110的生长速率,可以广泛应用于慢生根瘤菌菌剂的大规模生产。
