A novel environmental friendly catalyst, H4SiW12O40/PAn was prepared and identified by FT-IR, XRD and TG/DTA. The optimal synthetic protocol was a PAn to H4SiW12O40 mass ratio of 1 : 1, a 20 mL of methanol and a reaction time of 2 h at reflux. It was used in catalytic synthesis of ten important ketals and acetals with a high catalytic activity. With an aldehyde/ketone to glycol molar ratio of 1 : 1.5 and a 1 wt% catalyst loading, the yields of ketals and acetals could reach 60.0%- 93.8 % after 1 h. 展开更多

目的调查学龄前孤独症谱系障碍(autism spectrum disorder,ASD)儿童血清叶酸和维生素B_(12)(vitamin B_(12),VB_(12))水平及其与神经发育水平的关联。方法招募324例2~6岁ASD儿童和318例2~6岁健康儿童,采用化学发光免疫分析法测定血清叶... 目的调查学龄前孤独症谱系障碍(autism spectrum disorder,ASD)儿童血清叶酸和维生素B_(12)(vitamin B_(12),VB_(12))水平及其与神经发育水平的关联。方法招募324例2~6岁ASD儿童和318例2~6岁健康儿童,采用化学发光免疫分析法测定血清叶酸和VB_(12)水平,采用社交反应量表和儿童孤独症评定量表评估ASD儿童的核心症状,采用格塞尔发育量表评估神经发育水平。结果ASD儿童的血清叶酸及VB_(12)水平均显著低于健康儿童(P<0.05)。ASD儿童血清叶酸水平与大运动和精细运动发育商呈正相关(P<0.05),血清VB_(12)水平与适应性行为、精细运动、语言发育商呈正相关(P<0.05)。2~<4岁ASD儿童血清叶酸水平与各能区发育商均呈正相关(P<0.05),血清VB_(12)水平与语言发育商呈正相关(P<0.05);ASD男童血清VB_(12)水平与语言、个人-社会发育商呈正相关(P<0.05)。结论学龄前ASD儿童血清叶酸和VB_(12)水平较健康儿童低,且与神经发育水平相关,尤其<4岁ASD儿童。因此,保持正常的血清叶酸和VB_(12)水平可能有益于ASD儿童,特别是<4岁ASD儿童的神经发育。 展开更多

Fe取代的Pt/H-ZSM-12催化剂的合成及其正十二烷加氢异构化性能研究

在长链烷烃的加氢异构反应中,H-ZSM-12分子筛独特的孔道结构有利于多支链异构体的生成,但其较强的酸性加剧了裂解副反应的发生。以Fe同晶取代分子筛骨架中的Al,可以有效降低分子筛的酸强度。为了提高异构产物的选择性,采用水热法合成了F... 在长链烷烃的加氢异构反应中,H-ZSM-12分子筛独特的孔道结构有利于多支链异构体的生成,但其较强的酸性加剧了裂解副反应的发生。以Fe同晶取代分子筛骨架中的Al,可以有效降低分子筛的酸强度。为了提高异构产物的选择性,采用水热法合成了Fe取代度(以n(Fe):n(Al+Fe)计)分别为0%、50%和100%的Z12-Al、Z12-Al-Fe和Z12-Fe分子筛。紫外可见吸收光谱(UV-Vis)表征结果表明,Z12-Al-Fe和Z12-Fe中引入的Fe绝大多数以[FeO_(4)]的结构存在于分子筛骨架中。红外吸收光谱(FT-IR)和X-射线光电子能谱(XPS)表征结果表明,H-Z12-Al-Fe和H-Z12-Fe骨架中的Fe与Si—OH结合形成了Si—OH—Fe键。氨气程序升温脱附(NH_(3)-TPD)和吡啶红外(Py-IR)表征结果表明,随着分子筛中Fe含量的增加,其强酸位的酸强度逐渐降低,总Bronsted酸的数量明显减少。在分子筛载体上负载足量的金属Pt后,以正十二烷(n-C_(12))为模型化合物评价了双功能催化剂的加氢异构性能(反应压力为2.0 MPa,n(H_(2)):n(n-C_(12))为6.0)。结果表明,Pt/H-Z12-Fe上Bronsted酸强度的降低和酸性位数量的减少使其催化活性低于Pt/H-Z12-Al,但弱Bronsted酸性对裂解反应的抑制使其在90%左右的n-C_(12)转化率下获得了最高的总异构选择性(85.7%)和多支链异构体选择性(53.8%),比Pt/H-Z12-Al分别高出18.6%和9.7%。此外,Pt/H-Z12-Fe在120 h的连续反应中表现出了优异的稳定性。 展开更多

外源添加VB_(12)对肺炎克雷伯氏菌代谢3-羟基丙酸的影响

为了探究外源添加维生素B12(Vitamin B12,VB_(12))对Klebsiella pneumoniae HD79和K.pneumoniae HD79-T利用甘油还原途径生产3-羟基丙酸(3-hydroxypropionic acid,3-HP)的影响以及摸索VB_(12)对K.pneumoniae HD79和K.pneumoniae HD79-T... 为了探究外源添加维生素B12(Vitamin B12,VB_(12))对Klebsiella pneumoniae HD79和K.pneumoniae HD79-T利用甘油还原途径生产3-羟基丙酸(3-hydroxypropionic acid,3-HP)的影响以及摸索VB_(12)对K.pneumoniae HD79和K.pneumoniae HD79-T的阈值上限,将不同浓度(0.01、0.02、0.03、0.04、0.05 g/L)的VB_(12)添加到K.pneumoniae HD79及K.pneumoniae HD79-T的发酵培养基中,利用HPLC检测其底物消耗及产物产生情况、qRT-PCR检测还原途径相关基因的mRNA表达情况以及酶联免疫试剂盒检测代谢相关酶活性。结果表明,VB_(12)对K.pneumoniae HD79和K.pneumoniae HD79-T的阈值为0.01 g/L和0.03 g/L。与未添加VB_(12)相比,菌株K.pneumoniae HD79和K.pneumoniae HD79-T的3-HP产量分别提高了24.39%,8.86%;醛脱氢酶基因puuC表达量分别提高了2.49倍和1.68倍;ALDH、GDHt和PDOR的酶活力分别提高了50.24%、40.36%和18.29%,及30.49%、37.84%和13.56%。说明通过外源添加辅酶因子VB_(12)对肺炎克雷伯氏菌高产3-HP是可行策略。 展开更多

孕期叶酸、维生素B_(12)与妊娠期糖尿病的关系

目的了解孕期女性叶酸和维生素B_(12)的膳食摄入和体内代谢状况,及其与妊娠期糖尿病(GDM)的关系。方法本研究为基于母子营养与健康队列的横断面研究,以山东省高密市人民医院、北京市海淀区妇幼保健院和怀柔区妇幼保健院为研究现场,遵循... 目的了解孕期女性叶酸和维生素B_(12)的膳食摄入和体内代谢状况,及其与妊娠期糖尿病(GDM)的关系。方法本研究为基于母子营养与健康队列的横断面研究,以山东省高密市人民医院、北京市海淀区妇幼保健院和怀柔区妇幼保健院为研究现场,遵循自愿原则募集孕24~30周的孕妇进入研究。采用3天24小时膳食回顾法进行膳食调查,计算孕妇每日叶酸、维生素B_(12)及相关营养素摄入情况;采集孕妇空腹静脉血,血清和红细胞叶酸检测采用微粒子化学发光法,血清同型半胱氨酸检测采用酶循环法,血清维生素B_(12)的检测方法为化学发光法。同期利用口服葡萄糖耐量试验进行妊娠期糖尿病的诊断。结果本研究共纳入368名孕妇,妊娠期糖尿病患病率为39.9%。膳食叶酸摄入、维生素B_(12)摄入在非GDM组的中位数分别为543.00μg DFE和3.01μg,在GDM组的中位数分别为360.90μg DFE和2.58μg;非GDM组的孕妇红细胞叶酸均值(969.64±288.57 ng/mL)高于GDM组(962.86±280.28 ng/mL),血清叶酸、血清维生素B_(12)和血清同型半胱氨酸在非GDM组的中位数分别为14.26 ng/mL、223.00 pmol/L、4.72μmol/L,在GDM组的中位数分别为12.93 ng/mL、233.00 pmol/L和4.89μmol/L。多因素logistic分析结果显示,孕期膳食碳水化合物摄入不足增加GDM发生风险(OR=2.19,95%CI:1.17~4.09),血清叶酸过量(>20 ng/mL)与GDM低风险有关(OR=0.55,95%CI:0.30~1.00)。结论血清叶酸含量较高的孕妇患妊娠期糖尿病的可能性更低。 展开更多

妊娠期糖尿病患者炎症因子、FA和Vit B-12水平检测及临床意义分析

目的研究分析妊娠期糖尿病患者炎症因子、叶酸(Folic Acid,FA)、维生素B12(Vitamin B-12,Vit B-12)的检测结果。方法回顾性选取2020年1月—2022年1月福清市妇幼保健院的60例妊娠期糖尿病患者的临床资料为观察组,同期60例健康孕妇的临床... 目的研究分析妊娠期糖尿病患者炎症因子、叶酸(Folic Acid,FA)、维生素B12(Vitamin B-12,Vit B-12)的检测结果。方法回顾性选取2020年1月—2022年1月福清市妇幼保健院的60例妊娠期糖尿病患者的临床资料为观察组,同期60例健康孕妇的临床资料为对照组。两组均行疾病指标(空腹血糖、空腹胰岛素)、炎症因子[C反应蛋白(C-reactive Protein,CRP)、肿瘤坏死因子-α(Tumor Necrosis Factor-α,TNF-α)、白介素-6(Interleukin-6,IL-6)、同型半胱氨酸(Homocysteine,HCY)]、FA、Vit B-12检测,比较两组检测结果差异,分析上述指标的临床意义。结果观察组空腹血糖、空腹胰岛素、血清CRP、TNF-α、IL-6、HCY水平较对照组高,观察组FA、Vit B-12水平较对照组低,差异有统计学意义(P均<0.05)。高HCY亚组患者的FA、Vit B-12较低HCY亚组低,差异有统计学意义(P均<0.05)。妊娠期糖尿病患者的HCY与FA、Vit B-12呈负相关性(r=-0.598、-0.617,P均<0.05)。结论妊娠期糖尿病患者病发后有明显的炎症因子水平上升、FA和Vit B-12水平下降现象,其中患者HCY水平的上升与FA、Vit B-12水平的下降明显相关。 展开更多

目的探讨孕妇及脐带静脉血中VitB_(12)、叶酸及同型半胱氨酸(HCY)的含量与胎儿生长受限(FGR)的相关性。方法选取2019年1月1日至2022年12月31日在郑州市妇幼保健院产科分娩的84例FGR患儿作为FGR组,另选择同期在我院分娩的84例正常新生儿... 目的探讨孕妇及脐带静脉血中VitB_(12)、叶酸及同型半胱氨酸(HCY)的含量与胎儿生长受限(FGR)的相关性。方法选取2019年1月1日至2022年12月31日在郑州市妇幼保健院产科分娩的84例FGR患儿作为FGR组,另选择同期在我院分娩的84例正常新生儿作为对照组。比较两组新生儿生长指标、孕妇及脐带静脉血中VitB_(12)、叶酸及HCY含量,采用Pearson相关性检验分析孕妇及脐带静脉血血清VitB_(12)、叶酸及HCY含量与新生儿生长指标的相关性。结果FGR组出生体质量、出生身长、出生头围分分别为(2.18±0.46)kg、(45.89±1.14)cm、(27.65±0.87)cm,均明显低于对照组的(3.42±0.51)kg、(50.37±1.59)cm、(33.06±0.96)cm,差异均有统计学意义(P<0.05);FGR组孕妇静脉血血清VitB_(12)、叶酸的含量分别为(54.28±11.69)ng/L、(3.69±1.43)μg/L,脐带静脉血分别为(45.32±10.71)ng/L、(5.22±1.65)μg/L,均明显低于对照组孕妇静脉血的(417.24±57.25)ng/L、(16.77±3.74)μg/L和脐带静脉血的(278.38±58.35)ng/L、(18.35±3.80)μg/L,差异均有统计学意义(P<0.05);FGR组孕妇静脉血中血清HCY含量为(47.98±7.99)μmol/L,脐带静脉血中血清HCY含量为(28.29±4.87)μmol/L,均高于对照组孕妇静脉血的(8.58±2.51)μmol/L和脐带静脉血的(8.95±2.55)μmol/L,差异均有统计学意义(P<0.05);经Pearson相关性检验分析结果显示,孕妇静脉血中血清VitB_(12)、叶酸、HCY含量与脐带静脉血中的含量呈正相关(P<0.05);孕妇及脐带静脉血中血清VitB_(12)、叶酸含量与新生儿出生体质量、出生身长和出生头围呈正相关(P<0.05),而孕妇及脐带静脉血HCY含量与新生儿出生体质量、出生身长和出生头围呈负相关(P<0.05)。结论孕妇及脐带静脉血低VitB_(12)、叶酸含量及高HCY含量可能引起FGR发生,临床可根据孕妇静脉血血清VitB_(12)、叶酸及HCY含量推测胎儿是否缺乏VitB_(12)、叶酸,以及HCY含量是否过高,并及时采取干预措施,减少FGR的发生。 展开更多

目的研究广西地区壮族正常听力人群线粒体基因12S rRNA的突变携带率以及突变特点,为临床防聋及治聋提供参考。方法采用晶芯十五项遗传性耳聋基因检测试剂盒(微阵列芯片法)对广西地区150例壮族正常听力的受试者进行线粒体基因12S rRNA的... 目的研究广西地区壮族正常听力人群线粒体基因12S rRNA的突变携带率以及突变特点,为临床防聋及治聋提供参考。方法采用晶芯十五项遗传性耳聋基因检测试剂盒(微阵列芯片法)对广西地区150例壮族正常听力的受试者进行线粒体基因12S rRNA的2个突变位点检测,对确诊的阳性结果进行Sanger测序分析。结果150例受试者中,检出线粒体基因12S rRNA 1555 A>G突变者1例(携带率为0.67%),经Sanger测序证实为1555 A>G突变,未发现1494C>T位点突变。结论在广西地区壮族人群中开展线粒体基因12S rRNA突变筛查具有重要意义,可以为突变携带者及其母系亲属提供合理的用药指导、遗传咨询,是预防药物性聋的有效措施。 展开更多

鞣花酸的抗氧化性及其对PC-12细胞氧化损伤的保护作用研究

通过鞣花酸清除自由基、抑制亚油酸自氧化、还原铁离子(Fe^(3+))能力实验研究其抗氧化活性;以过氧化氢(H2O2)诱导PC-12细胞建立氧化损伤模型,探索鞣花酸对PC-12细胞氧化应激损伤的保护作用,建立鞣花酸抗氧化活性与对细胞氧化应激损伤之... 通过鞣花酸清除自由基、抑制亚油酸自氧化、还原铁离子(Fe^(3+))能力实验研究其抗氧化活性;以过氧化氢(H2O2)诱导PC-12细胞建立氧化损伤模型,探索鞣花酸对PC-12细胞氧化应激损伤的保护作用,建立鞣花酸抗氧化活性与对细胞氧化应激损伤之间的联系。结果表明,鞣花酸对羟基自由基(·OH)、超氧阴离子自由基(·O_(2)^(-))、ABTS^(+)·自由基清除的半抑制浓度分别为92.14、36.70、47.91μg/mL;对亚油酸自氧化的抑制率高达90%;对铁离子具有很强的还原能力。鞣花酸对H2O2诱导的PC-12细胞氧化损伤具有良好的保护作用,且与抗氧化活性之间具有良好的相关系。鞣花酸对细胞氧化损伤的保护作用可能与提高细胞的抗氧化能力有关。 展开更多

By RT-PCR method, the full-length cDNAs of △^12 fatty acid desaturase gene were isolated from peanut (Arachis hypogaea L.) genotypes with normal and high ratio of oleic to linoleic acid, which were designated AhFAD2B and AhFAD2B', respectively. Sequence alignment of their coding regions revealed that an extra A was inserted at the position +442 bp of AhFAD2B' sequence of high oleic acid genotypes, which resulted in the shift of open reading frame and a truncated protein AhFAD2B', with the loss of one histidine box involved in metal ion complex required for the reduction of oxygen. Analysis of transcript level showed that the expression of △^12 fatty acid desaturase gene in high oleic acid genotype was slightly lower than that in normal genotype. The enzyme activity experiment of yeast (Saccharomyces cerevisiae) cell transformed with AhFAD2B or AhFAD2B' proved that only AhFAD2B gene product showed significant △^12 fatty acid desaturase activity, but AhFAD2B' gene product did not. These results suggested that the change of AhFAD2B' gene sequence resulted in lower activity or deactivation of △^12 fatty acid desaturase in high oleic acid genotype. 展开更多

A reversed phase high performance liquid chromatography (HPLC) method was established for the simultaneous determination of 12, 13-dihydroxyeuparin and glycyrrhizic acid in Yanyanfang mixture. A Grace Apollo Cl8 column (250 mm × 4.6 mm, 5 μm) was used as the stationary phase and the mobile phase was composed of acetonitrile and aqueous phosphoric acid (0.2%, v/v). Gradient elution was carried out at the flow rate of 1.0 mL/min and the column temperature was 30 ℃. An ultraviolet (UV) detector was used with a selected wavelength of 240 nm. Calibration curves were linear within the concentration range of 4.6-45.75 μg/mL for 12, 13-dihydroxyeuparin (r〉0.9999) and 106.9-1068.9μg/mL for glycyrrhizic acid (r〉0.9999), respectively. Recoveries were 102.18% for 12, 13-dihydroxyeuparin and 101.17% for glycyrrhizic acid. The method developed could be applied to the simultaneous determination of 12, 13- dihydroxyeuparin and glycyrrhizic acid in Yanyanfang mixture. 展开更多

Synthesis of acetic acid by direct oxidation of ethylene on Pd-H4SiW12O40-based catalysts was studied in a fixed-bed integral reactor and a pulse differential reactor. From the performance of the catalysts with different compositions and configurations, it is proposed that acetic acid is predominantly produced via an intermediate of acetaldehyde. This can be easily confirmed by comparing the product distributions in the integral and the differential reactors. The active sites for acetic acid formation are considered to exist mainly at the boundaries between the H4SiW12O40 and the Pd particles. The Pd-based catalysts reduced by H2/N2 have higher activities than those reduced by hydrazine, as explained by the degree of Pd dispersion obtained from the characteristics of hydrogen chemical adsorption. It was found that the Pd-Se-SiW12/SiO2 catalyst with selenium tetrachloride as a precursor was more active than that with potassium selenite, and that the acetic acid yield can be greatly increased by adding a suitable amount of dichloroethane (C2H4C12/C2H4 mole ratio=0.03) to the reactants. 展开更多

The cDNA of the delta-12 fatty acid desaturase gene, IgFAD2, was cloned from the marine microalgae Isochrysis galbana, a species capable of producing docosahexaenoic acid. Sequence analysis indicated that the open reading frame measured a length of 1 158 bp and encoded 386 amino acids with a predicted molecular weight of 42.8 kDa and an isoelectric point of 9.2. Computational analysis of the protein sequence of IgFAD2 showed typical features of membrane-bound desaturase such as three conserved histidine boxes along with four membranespanning regions that were universally present among plant desaturases. Quantitative real-time PCR results showed that the abundance of IgFAD2 transcript was significantly upregulated under different environmental stresses including low temperature(15℃), high salinity(salinity of 62 and 93), and nitrogen starvation(220 μmol/L). Heterologous expression indicated that yeast cells transformed with a plasmid construct containing IgFAD2 could convert endogenous oleic acid(18:1^(?9), OA) into linoleic acid(18:2^(?9, 12), LA). These findings confirm that I. galbana IgFAD2 plays important roles in the biosynthetic pathways of unsaturated fatty acids. 展开更多

The freshwater microalga Haematococcus pluvialis accumulates large amounts of fatty acids in response to adverse conditions.However,the key fatty acid desaturase genes in H.pluvialis remain unknown.In this study,we cloned and functionally characterized aΔ12 fatty acid desaturase gene,and designated it as HpFAD2.The open reading frame of HpFAD2 consisted of 1137 base pairs and encoded 378 amino acids.The deduced polypeptide showed 70%identity to other endoplasmic reticulumΔ12 fatty acid desaturases,whereas it had only 44%identity to plastidΔ12 fatty acid desaturases.The PSORT algorithm and phylogenetic analysis further confirmed its affiliation to the endoplasmic reticulumΔ12 fatty acid desaturases.Heterologous expression was performed in Saccharomyces cerevisiae cells transformed with the recombinant plasmid pYES2-HpFAD2.Two additional fatty acids(C16:2 and C18:2)were detected in the yeast transformants.The results indicatedΔ12 desaturation activity and substrate preference for C18:1 over C16:1.The transcriptional levels of H.pluvialis HpFAD2 at different growth stages were measured by quantitative polymerase chain reaction(PCR),indicating that the HpFAD2 transcriptional levels were significantly higher in red cells than those in green cells.Our study brings more insight into the fatty acid biosynthetic pathway of H.pluvialis. 展开更多

In order to solve the serious leaching problem of supported heteropoly acid catalysts in polar reaction media,12-molybdophosphoric acid encapsulated in the supercage of Cs+-exchanged Y zeolite was prepared by the "ship in the bottle" synthesis.The influence of ion-exchange conditions and the synthesis parameters on the encapsulation of PMo12 were investigated.The obtained solid sample was characterized by X-ray diffraction(XRD),31P magic angle spin nuclear magnetic resonance(MAS NMR) and Fourier Transform Infrared Spectroscopy(FT-IR),and its catalytic activity in the esterification of acetic acid and n-butanol was tested.The ion-exchange time,concentration of aqueous Cs+ solution,pH value,and amount of Mo added in the synthesis mixture were revealed to influence the encapsulation very remarkably.Under the optimal conditions,12-molybdophosphoric acid could be successfully encapsulated in the supercage of CsY zeolite,and the samples showed considerable catalytic activity and excellent reusability in the esterification reaction. 展开更多

The effects of PLA concentration, stirring speed, injection rate of core and shell solutions, and polyvinyl alcohol(PVA) concentration on phase change properties were investigated. The thermal properties of microP CMs were characterized by differential scanning calorimetry(DSC).
