背景:目前对于M2型巨噬细胞衍生外泌体的研究多集中于促进伤口愈合及成骨细胞的增殖和分化,而很少有研究关注其对小胶质细胞表型的调控作用。目的:探讨M2型巨噬细胞衍生外泌体对于小胶质细胞的表型调控作用及分子机制。方法:①提取骨髓... 背景:目前对于M2型巨噬细胞衍生外泌体的研究多集中于促进伤口愈合及成骨细胞的增殖和分化,而很少有研究关注其对小胶质细胞表型的调控作用。目的:探讨M2型巨噬细胞衍生外泌体对于小胶质细胞的表型调控作用及分子机制。方法:①提取骨髓原代巨噬细胞,用50 ng/m L白细胞介素4刺激巨噬细胞24 h促进巨噬细胞M2型极化,流式细胞术和细胞免疫荧光鉴定M2型巨噬细胞标志物CD206;②提取和鉴定M2型巨噬细胞衍生外泌体;③将小胶质细胞BV2随机分为3组:对照组、脂多糖组、治疗组,对照组不做处理,脂多糖组加入500 ng/m L脂多糖干预24 h,治疗组同时加入500 ng/m L脂多糖和25μg/m L M2型巨噬细胞衍生外泌体干预24 h,ELISA检测培养上清中肿瘤坏死因子α和白细胞介素10的分泌量,q RT-PCR检测细胞中诱导型一氧化氮合酶、精氨酸酶1、白细胞介素1β和白细胞介素10的m RNA表达,Western blot检测诱导型一氧化氮合酶、精氨酸酶1的蛋白表达以及核因子κB信号通路相关蛋白表达。结果与结论:①ELISA结果显示,与对照组相比,脂多糖组肿瘤坏死因子α分泌明显增多;与脂多糖组相比,治疗组肿瘤坏死因子α的分泌减少而白细胞介素10的分泌增多;②q RT-PCR结果显示,与对照组相比,脂多糖组白细胞介素1β、诱导型一氧化氮合酶m RNA表达升高;与脂多糖组相比,治疗组白细胞介素1β、诱导型一氧化氮合酶m RNA表达降低,白细胞介素10、精氨酸酶1 m RNA表达升高;③Western blot结果显示,与对照组相比,脂多糖组诱导型一氧化氮合酶蛋白表达升高;与脂多糖组相比,治疗组诱导型一氧化氮合酶蛋白表达降低,而精氨酸酶1蛋白表达升高;(4)与对照组相比,脂多糖组核因子κB信号通路中P65、p-IκB-α蛋白表达降低;与脂多糖组相比,治疗组P65、p-IκB-α蛋白表达升高。结果表明:M2型巨噬细胞衍生外泌体可以显著抑制脂多糖诱导小胶质细胞的炎症反应,促进抗炎因子白细胞介素10的表达,抑制促炎因子肿瘤坏死因子α、白细胞介素1β的表达,促进小胶质细胞表型由M1型向M2型极化,其机制可能与M2型巨噬细胞衍生外泌体抑制核因子κB信号通路激活有关。 展开更多

Postoperative cognitive dysfunction is a seve re complication of the central nervous system that occurs after anesthesia and surgery,and has received attention for its high incidence and effect on the quality of life of patients.To date,there are no viable treatment options for postoperative cognitive dysfunction.The identification of postoperative cognitive dysfunction hub genes could provide new research directions and therapeutic targets for future research.To identify the signaling mechanisms contributing to postoperative cognitive dysfunction,we first conducted Gene Ontology and Kyoto Encyclopedia of Genes and Genomes pathway enrichment analyses of the Gene Expression Omnibus GSE95426 dataset,which consists of mRNAs and long non-coding RNAs differentially expressed in mouse hippocampus3 days after tibial fracture.The dataset was enriched in genes associated with the biological process"regulation of immune cells,"of which Chill was identified as a hub gene.Therefore,we investigated the contribution of chitinase-3-like protein 1 protein expression changes to postoperative cognitive dysfunction in the mouse model of tibial fractu re surgery.Mice were intraperitoneally injected with vehicle or recombinant chitinase-3-like protein 124 hours post-surgery,and the injection groups were compared with untreated control mice for learning and memory capacities using the Y-maze and fear conditioning tests.In addition,protein expression levels of proinflammatory factors(interleukin-1βand inducible nitric oxide synthase),M2-type macrophage markers(CD206 and arginase-1),and cognition-related proteins(brain-derived neurotropic factor and phosphorylated NMDA receptor subunit NR2B)were measured in hippocampus by western blotting.Treatment with recombinant chitinase-3-like protein 1 prevented surgery-induced cognitive impairment,downregulated interleukin-1βand nducible nitric oxide synthase expression,and upregulated CD206,arginase-1,pNR2B,and brain-derived neurotropic factor expression compared with vehicle treatment.Intraperitoneal administration of the specific ERK inhibitor PD98059 diminished the effects of recombinant chitinase-3-like protein 1.Collectively,our findings suggest that recombinant chitinase-3-like protein 1 ameliorates surgery-induced cognitive decline by attenuating neuroinflammation via M2 microglial polarization in the hippocampus.Therefore,recombinant chitinase-3-like protein1 may have therapeutic potential fo r postoperative cognitive dysfunction. 展开更多

The electrocatalytic oxidation of biomass-derived furfural(FF)feedstocks into 2-furoic acid(FA)holds immense industrial potential in optics,cosmetics,polymers,and food.Herein,we fabricated Co O/Ni O/nickel foam(NF)and Cu_(2)O/Ni O/NF electrodes via in situ pulsed laser irradiation in liquids(PLIL)for the bifunctional electrocatalysis of oxygen evolution reaction(OER)and furfural oxidation reaction(FOR),respectively.Simultaneous oxidation of NF surface to NiO and deposition of CoO and/or Cu_(2)O on NF during PLIL offer distinct advantages for enhancing both the OER and FOR.CoO/NiO/NF electrocatalyst provides a consistently low overpotential of~359 m V(OER)at 10 m A/cm^(2),achieving the maximum FA yield(~16.37 m M)with 61.5%selectivity,79.5%carbon balance,and a remarkable Faradaic efficiency of~90.1%during 2 h of FOR at 1.43 V(vs.reversible hydrogen electrode).Mechanistic pathway via in situ electrochemical-Raman spectroscopy on CoO/NiO/NF reveals the involvement of phase transition intermediates(NiOOH and CoOOH)as surface-active centers during electrochemical oxidation.The carbonyl carbon in FF is attacked by hydroxyl groups to form unstable hydrates that subsequently undergo further oxidation to yield FA products.This method holds promise for large-scale applications,enabling simultaneous production of renewable building materials and fuel. 展开更多

Memristors as non-volatile memory devices have gained numerous attentions owing to their advantages in storage,in-memory computing, synaptic applications, etc. In recent years, two-dimensional(2D) materials with moderate defects have been discovered to exist memristive feature. However, it is very difficult to obtain moderate defect degree in 2D materials, and studied on modulation means and mechanism becomes urgent and essential. In this work, we realized memristive feature with a bipolar switching and a configurable on/off ratio in a two-terminal MoS_(2) device(on/off ratio ~100), for the first time, from absent to present using laser-modulation to few-layer defect-free MoS_(2)(about 10 layers), and its retention time in both high resistance state and low resistance state can reach 2×10^(4) s. The mechanism of the laser-induced memristive feature has been cleared by dynamic Monte Carlo simulations and first-principles calculations. Furthermore, we verified the universality of the laser-modulation by investigating other 2D materials of TMDs. Our work will open a route to modulate and optimize the performance of 2D semiconductor memristive devices. 展开更多

MoS_(2)targets were irradiated by infra-red(IR)pulsed laser in a high vacuum to determine hot plasma parameters,atomic,molecular and ion emission,and angular and charge state distributions.In this way,pulsed laser deposition(PLD)of thin films on graphene oxide substrates was also realized.An Nd:YAG laser,operating at the 1064 nm wavelength with a 5 ns pulse duration and up to a 1 J pulse energy,in a single pulse or at a 10 Hz repetition rate,was employed.Ablation yield was measured as a function of the laser fluence.Plasma was characterized using different analysis techniques,such as time-of-flight measurements,quadrupole mass spectrometry and fast CCD visible imaging.The so-produced films were characterized by composition,thickness,roughness,wetting ability,and morphology.When compared to the MoS_(2)targets,they show a slight decrease of S with respect to Mo,due to higher ablation yield,low fusion temperature and high sublimation in vacuum.The pulsed IR laser deposited Mo Sx(with 1<x<2)films are uniform,with a thickness of about 130 nm,a roughness of about 50 nm and a higher wettability than the MoS_(2)targets.Some potential applications of the pulsed IR laser-deposited Mo Sx films are also presented and discussed. 展开更多

目的探讨X连锁RNA结合基序蛋白(RBMX)对膀胱癌细胞(1376细胞和UC-3细胞)的增殖、迁移、侵袭的影响以及其在糖酵解中的作用。方法采用慢病毒表达系统和siRNA干扰技术,分别构建RBMX过表达和敲低的膀胱癌细胞模型(1376细胞和UC-3细胞)。采... 目的探讨X连锁RNA结合基序蛋白(RBMX)对膀胱癌细胞(1376细胞和UC-3细胞)的增殖、迁移、侵袭的影响以及其在糖酵解中的作用。方法采用慢病毒表达系统和siRNA干扰技术,分别构建RBMX过表达和敲低的膀胱癌细胞模型(1376细胞和UC-3细胞)。采用RT-qPCR和Westernblotting分别在mRNA水平和蛋白水平上检测细胞模型是否构建成功。通过EdU增殖实验和克隆形成实验检测过表达和敲低RBMX后细胞的生长和集落形成能力,同时通过Transwell实验分析过表达和敲低RBMX后对细胞迁移、侵袭能力的影响;随后,采用Westernblotting检测过表达和敲低RBMX后糖酵解关键蛋白PKM1(M1型丙酮酸激酶)和PKM2(M2型丙酮酸激酶)的表达变化;最后,利用葡萄糖和乳酸检测试剂盒分析过表达和敲低RBMX对膀胱癌细胞糖酵解的影响。结果RT-qPCR和Westernblotting结果显示,过表达RBMX膀胱癌细胞的mRNA和蛋白表达水平显著高于阴性对照组(P<0.05),敲低RBMX膀胱癌细胞的mRNA和蛋白表达水平显著低于阴性对照组(P<0.05)。过表达RBMX明显抑制膀胱癌细胞的增殖、克隆形成、迁移和侵袭,敲低RBMX则作用相反。Westernblotting实验结果显示,过表达RBMX使PKM1表达上升,PKM2表达下降,敲低RBMX则作用相反。葡萄糖消耗及乳酸生成实验表明,过表达RBMX均能抑制膀胱癌细胞葡萄糖消耗及乳酸生成(P<0.05),敲低RBMX均能促进膀胱癌细胞葡萄糖消耗及乳酸生成(P<0.05)。结论RBMX通过下调PKM2抑制膀胱癌的发生发展和糖酵解能力,有望成为膀胱癌诊断和治疗的潜在分子靶标。 展开更多

The chemical composition, microstructure and micro-hardness of the as-deposited GSM were investigated. The results show that the TA2 part of exhibiting near-equiaxed grains was Widmanst?tten α-laths microstructure. The TA15 part containing large columnar grains was fine basket-weave microstructure. The graded zone was divided into four deposited layers with 3000 μm in thickness. As the distance from the TA2 part increases, the alloy element contents and the β phase volume fraction increase, the α phase volume fraction decreases and the microstructure shows the evolution from Widmanst?tten α-laths to basket-weave α-laths gradually. The micro-hardness increases from the TA2 part to the TA15 part due to the solid solution strengthening and grain boundary strengthening. 展开更多

A Y2O3 particle enhanced Ni/TiC composite coating was fabricated in-situ on a TC4 Ti alloy by laser surface cladding. The phase component, microstructure, composition distribution and properties of the composite layer were investigated. The composite layer has graded microstructures and compositions, due to the fast melting followed by rapid solidification and cooling during laser cladding. The TiC powders are completely dissolved into the melted layer during melting and segregated as fine dendrites when solidified. The size of TiC dendrites decreases with increasing depth. Y2O3 fine particles distribute in the whole clad layer. The Y2O3 particle enhanced Ni/TiC composite layer has a quite uniform hardness along depth with a maximum value of HV1380, which is 4 times higher than the initial hardness. The wear resistance of the Ti alloy is significantly improved after laser cladding due to the high hardness of the composite coating. 展开更多

超参数调优是神经网络建模的关键问题。针对传统的超参数调优方法存在的问题,该文提出了一种基于m×2正则化交叉验证的超参数调优方法。目的是给出一种适用于复杂模型、大数据集背景下的计算开销较小且稳健的超参数调优方法。该方... 超参数调优是神经网络建模的关键问题。针对传统的超参数调优方法存在的问题,该文提出了一种基于m×2正则化交叉验证的超参数调优方法。目的是给出一种适用于复杂模型、大数据集背景下的计算开销较小且稳健的超参数调优方法。该方法的思想是从完整的数据集上选取少部分数据进行调优,避免模型在数据集较大时非常耗时的超参数调优难题;在m×2交叉验证的基础上设置正则化条件均衡训练集与验证集之间的分布差异,从而减少分布不一致带来的性能波动;使用信噪比作为调优的优化目标,从而可以综合考虑模型性能评价指标的均值和方差;并采用正交设计选择相关性较低的超参数组合以提高调优效率。以命名实体任务为例进行实验,在CoNLL 2003数据集上的实验结果显示,提出的调优方法能够选到和网格搜索性能上没有显著差异的超参数组合,且调优时间可显著降低约66%。 展开更多

The coating microstructure and the interface between bond coating and ceramic coating were characterized by SEM, XRD and EDS. The results show that two-layer substructure exists in the ceramic coating: one layer evolving from fully melted region where the sintered grains grow fully; another layer resembling the liquid-phase-sintered structure consisting of three-dimensional net where the melted Al2O3 particles are embedded in the TiO2-rich matrix. The mechanism of the two-layer substructure formation is also explained in terms of the melting and flattening behavior of the powders during laser cladding processing. The spinel compounds NiAl2O4 and acicular compounds Cr2O3 are discovered in the interface between bond coating and ceramic coating. It proves that the chemical reactions in the laser cladding process will significantly enhance the coating adhesion. 展开更多

目的探讨白介素(IL)-35在巨噬细胞中的表达及其对前列腺癌(PCa)细胞增殖、侵袭和上皮间质转化(EMT)的影响和机制。方法检测巨噬细胞标记分子CD68、M1型标记分子肿瘤坏死因子-α(TNF-α)、IL-1β和M2型标记分子CD206、IL-10在人单核/巨... 目的探讨白介素(IL)-35在巨噬细胞中的表达及其对前列腺癌(PCa)细胞增殖、侵袭和上皮间质转化(EMT)的影响和机制。方法检测巨噬细胞标记分子CD68、M1型标记分子肿瘤坏死因子-α(TNF-α)、IL-1β和M2型标记分子CD206、IL-10在人单核/巨噬细胞系THP-1细胞诱导分化后的M0、M1与M2型巨噬细胞中的表达。使用不同类型的巨噬细胞-源条件培养液(CM)处理PCa细胞系PC-3,并将其分为:M0-CM组、M1-CM组、M2-CM组和预先使用IL-35中和抗体(IL-35 NA)干预的M2-CM处理组(M2-CM+IL-35 NA组)。检测各组中PC-3细胞的增殖活力、侵袭、EMT,以及细胞内JAK/STAT信号通路p-JAK2/JAK2与p-STAT3/STAT3比值的差异。结果与未刺激的THP-1细胞比较,M0、M1和M2型巨噬细胞中CD68 mRNA表达升高(P<0.01);与M0型巨噬细胞比较,TNF-α、IL-1βmRNA在M1型巨噬细胞中升高(P<0.01),而CD206、IL-10 mRNA在M2型巨噬细胞中升高(P<0.01);与M0-CM组比较,M2-CM组PC-3细胞增殖活力、侵袭、EMT和细胞内p-JAK2/JAK2、p-STAT3/STAT3比值升高(P<0.05),M2-CM+IL-35 NA组PC-3细胞的增殖活力、侵袭、EMT和细胞内p-JAK2/JAK2、p-STAT3/STAT3比值均显著降低(P<0.05),M1-CM组PC-3细胞的增殖活力、侵袭、EMT和细胞内p-JAK2/JAK2、p-STAT3/STAT3比值均无显著改变(P>0.05);与M2-CM组比较,M1-CM组和M2-CM+IL-35 NA组增殖活力、侵袭、EMT和细胞内p-JAK2/JAK2、p-STAT3/STAT3比值降低(P<0.05)。结论M2型巨噬细胞可通过分泌IL-35来促进PC-3细胞的增殖、侵袭和EMT,这可能与IL-35能上调JAK2/STAT3信号通路的活化相关。 展开更多

目的超声造影联合血清纤维胶凝蛋白-3(Ficolin-3)、叉头框蛋白M1(FOXM1)对2型糖尿病(T2DM)下肢动脉病变的诊断价值分析。方法选取2022年2月至2023年2月我院收治的T2DM患者114例,以下肢动脉血管造影检查为金标准,将患者分为下肢动脉病变... 目的超声造影联合血清纤维胶凝蛋白-3(Ficolin-3)、叉头框蛋白M1(FOXM1)对2型糖尿病(T2DM)下肢动脉病变的诊断价值分析。方法选取2022年2月至2023年2月我院收治的T2DM患者114例,以下肢动脉血管造影检查为金标准,将患者分为下肢动脉病变组23例(病变组)和无下肢动脉病变组91例(T2DM组),另选取同期于本院进行体检的健康志愿者91例为对照组。对患者及健康志愿者进行血清Ficolin-3、FOXM1及超声造影检查;ROC曲线分析血清Ficolin-3、FOXM1对T2DM下肢动脉病变的诊断价值;采用四格表法分析血清Ficolin-3、FOXM1、超声造影及3项联合对T2DM下肢动脉病变的诊断价值。结果病变组、T2DM组血清Ficolin-3水平显著高于对照组,且病变组血清Ficolin-3水平显著高于T2DM组(均P<0.05);病变组、T2DM组血清FOXM1水平显著低于对照组,且病变组血清FOXM1水平显著低于T2DM组(均P<0.05);血清Ficolin-3诊断T2DM下肢动脉病变的曲线下面积为0.868,敏感度为86.96%,特异度为75.82%,最佳截断值为35.12μg/ml;血清FOXM1诊断T2DM下肢动脉病变的曲线下面积为0.854,敏感度为82.61%,特异度为78.02%,最佳截断值为0.57;血清Ficolin-3、FOXM1检查结果与金标准均具有中度一致性(Kappa=0.480、0.481,均P<0.001);超声造影检查结果显示,T2DM下肢动脉病变的阳性检出率为73.91%,与金标准具有较高一致性(Kappa=0.631,P<0.001);3项联合检测的敏感度、漏诊率均显著优于超声造影单独诊断,准确度显著优于Ficolin-3、FOXM1单独诊断,差异均有统计学意义(均P<0.05)。结论血清Ficolin-3、FOXM1联合超声造影检查在T2DM下肢动脉病变诊断中的应用价值较高,进一步提升了诊断的敏感度、准确度,减少了误诊率。 展开更多

老年心力衰竭并发肺炎患者血清FOXM1和IGF2表达水平及与预后价值研究

目的探究血清叉头盒蛋白M1(forkhead box protein M1,FOXM1)和胰岛素样生长因子2(insulin-like growth factor 2,IGF2)表达对老年心力衰竭合并肺炎患者预后的预测价值。方法将邯郸市中心医院2021年3月~2022年6月收治的126例老年心力衰... 目的探究血清叉头盒蛋白M1(forkhead box protein M1,FOXM1)和胰岛素样生长因子2(insulin-like growth factor 2,IGF2)表达对老年心力衰竭合并肺炎患者预后的预测价值。方法将邯郸市中心医院2021年3月~2022年6月收治的126例老年心力衰竭并发肺炎患者设为病例组,并根据随访情况将122例患者分为预后不良组(n=33)和预后良好组(n=89),另选取该院同期126例健康体检者为对照组。检测两组(病例组和对照组)血清FOXM1和IGF2水平,检测病例组用力肺活量(forced vital capacity,FVC)和第一秒用力呼容积(forced expiratory volume in one second,FEV1)。采用Spearman分析法分析老年心力衰竭并发肺炎患者血清FOXM1和IGF2水平与心功能分级的相关性;受试者工作特征(receiver operating characteristic,ROC)曲线分析血清FOXM1和IGF2水平对老年心力衰竭并发肺炎患者预后的预测价值。结果与对照组比较,病例组血清FOXM1(2.39±0.55 vs 1.06±0.21)和IGF2(71.33±7.96pg/ml vs 47.82±5.14pg/ml)水平明显较高,差异有统计学意义(t=25.358,27.581,均P<0.05);与预后良好组比较,预后不良组血清FOXM1(3.87±1.06 vs 1.95±0.51)和IGF2水平(85.88±9.54pg/ml vs 69.14±8.73pg/ml)明显较高,差异具有统计学意义(t=13.453,9.174,均P<0.05);预后良好组和预后不良组心功能分级比较差异有统计学意义(χ^(2)=7.120,P<0.05),且与预后不良组比较,预后良好组FEV1(1.24±0.32L vs 1.08±0.25L)和FEV1/FVC(55.46%±5.77%vs 52.30%±5.38%)明显较高,差异有统计学意义(t=2.592,2.735,均P<0.05);老年心力衰竭并发肺炎患者血清FOXM1水平和IGF2水平与心功能分级呈显著正相关(r=0.496,0.517,均P<0.05)。ROC曲线结果显示,血清FOXM1单独预测老年心力衰竭并发肺炎患者预后的曲线下面积(area under the curve,AUC)为0.854(95CI%:0.779~0.912),其敏感度、特异度分别为75.76%,86.52%,最佳截断值为2.75;IGF2单独预测老年心力衰竭并发肺炎患者预后的AUC为0.874(95CI%:0.802~0.927),其敏感度、特异度分别为72.73%,85.39%,最佳截断值为78.30 pg/ml;二者联合预测老年心力衰竭并发肺炎患者预后的AUC显著大于血清FOXM1和IGF2单独诊断的AUC(Z=2.413,2.737,P=0.006,0.016)。结论血清FOXM1和IGF2水平在老年心力衰竭并发肺炎患者中升高,且二者联合检测对患者预后具有较高的预测价值。 展开更多

目的探讨瞬时受体电位M7通道(TRPM7)和B细胞迁移基因2(BTG2)在宫颈癌组织中的表达及临床意义。方法选取2018年5月至2020年5月石家庄市妇幼保健院收治的110例宫颈癌患者作为研究对象,术中取其肿瘤组织及癌旁组织,根据随访情况分为预后良... 目的探讨瞬时受体电位M7通道(TRPM7)和B细胞迁移基因2(BTG2)在宫颈癌组织中的表达及临床意义。方法选取2018年5月至2020年5月石家庄市妇幼保健院收治的110例宫颈癌患者作为研究对象,术中取其肿瘤组织及癌旁组织,根据随访情况分为预后良好组与预后不良组。采用免疫组化法检测癌旁组织和肿瘤组织中TRPM7、BTG2蛋白表达,分析TRPM7、BTG2蛋白表达水平与患者临床病理特征的关系,比较预后不良组与预后良好组肿瘤组织中TRPM7、BTG2蛋白表达,采用Cox回归分析宫颈癌患者预后的影响因素,采用Kaplan-Meier曲线分析TRPM7、BTG2水平与宫颈癌患者预后的关系。结果与癌旁组织比较,肿瘤组织中TRPM7蛋白表达水平升高,BTG2蛋白表达水平降低(P<0.05)。TRPM7、BTG2蛋白表达水平与肿瘤分化程度、淋巴结转移、恶性肿瘤国际临床病理(TNM)分期有关(P<0.05)。与预后良好组比较,预后不良组肿瘤组织中TRPM7蛋白表达水平升高,BTG2蛋白表达水平降低(P<0.05)。TRPM7、TNM分期(Ⅲ期)、肿瘤低分化程度、淋巴结转移是宫颈癌患者预后的危险因素(P<0.05),BTG2是宫颈癌患者预后的保护因素(P<0.05)。TRPM7高表达组3年生存率低于TRPM7低表达组(P<0.05);BTG2高表达组3年生存率高于BTG2低表达组(P<0.05)。结论宫颈癌患者肿瘤组织中TRPM7蛋白表达上调,BTG2蛋白表达下调,均与宫颈癌预后有关。 展开更多
