The halopyrimidine 5-bromo-2′-deoxyuridine(BrdU)is an exogenous marker of DNA synthesis.Since the introduction of monoclonal antibodies against BrdU,an increasing number of methodologies have been used for the immunodetection of this synthesized bromine-tagged base analogue into replicating DNA.BrdU labeling is widely used for identifying neuron precursors and following their fate during the embryonic,perinatal,and adult neurogenesis in a variety of vertebrate species including birds,reptiles,and mammals.Due to BrdU toxicity,its incorporation into replicating DNA presents adverse consequences on the generation,survival,and settled patterns of cells.This may lead to false results and misinterpretation in the identification of proliferative neuroblasts.In this review,I will indicate the detrimental effects of this nucleoside during the development of the central nervous system,as well as the reliability of BrdU labeling to detect proliferating neuroblasts.Moreover,it will show factors influencing BrdU immunodetection and the contribution of this nucleoside to the study of prenatal,perinatal,and adult neurogenesis.Human adult neurogenesis will also be discussed.It is my hope that this review serves as a reference for those researchers who focused on detecting cells that are in the synthetic phase of the cell cycle. 展开更多

经由食物链长期摄入氯霉素残留物会给人体带来各种危害和疾病。因此,建立快速、准确且高灵敏的食品中氯霉素检测方法具有重要意义。采用溶胶-凝胶法制备了表面氨基化的SiO_(2)纳米颗粒,并用X射线衍射、透射电镜和红外光谱对其进行了表... 经由食物链长期摄入氯霉素残留物会给人体带来各种危害和疾病。因此,建立快速、准确且高灵敏的食品中氯霉素检测方法具有重要意义。采用溶胶-凝胶法制备了表面氨基化的SiO_(2)纳米颗粒,并用X射线衍射、透射电镜和红外光谱对其进行了表征。然后,利用酰胺键和碱基互补配对作用在SiO_(2)纳米颗粒表面依次组装互补DNA链和氯霉素适配体,成功构建了无标记化学发光氯霉素适配体传感器。该传感器制备简便,其中的氯霉素适配体既可作为生物识别元件,又可与苯甲酰甲醛反应产生化学发光信号,无需标记信号分子。该传感器的氯霉素检出限为3.26×10^(-9) mol·L^(-1),且选择性较好。将该传感器应用于实际样品中进行加标,回收率在94.67%~103.00%之间,表明该适体传感器可用于食品中氯霉素的检测。 展开更多

建立一种3-氨基-2-恶唑烷酮(3-amino-2-oxazolidinone,AOZ)夹心免疫检测方法。通过1,6-己二醇连接2-硝基-4-羧基苯甲醛和生物素合成针对AOZ的新型衍生试剂。在样品前处理时加入衍生试剂可对AOZ进行衍生,衍生产率为89%。在酶标板上包被抗... 建立一种3-氨基-2-恶唑烷酮(3-amino-2-oxazolidinone,AOZ)夹心免疫检测方法。通过1,6-己二醇连接2-硝基-4-羧基苯甲醛和生物素合成针对AOZ的新型衍生试剂。在样品前处理时加入衍生试剂可对AOZ进行衍生,衍生产率为89%。在酶标板上包被抗AOZ单克隆抗体并以辣根过氧化物酶标记的亲和素或抗生物素抗体作为第二结合物可实现AOZ的夹心酶联免疫吸附检测(enzyme-linked immunosorbnent assay,ELISA)。从实际应用的角度,得到双抗夹心和抗体-亲和素夹心模式下两个表位的极限距离,分别为12Å和13Å,理想距离分别为16Å和17Å。在双抗夹心和抗体-亲和素夹心模式下的检出限分别达到1.8 pg/mL和0.8 pg/mL(以AOZ质量浓度计),相对于竞争ELISA,其灵敏度最高提高了25倍。平均回收率为73%~85%,平均相对标准偏差为9.0%。 展开更多

基于磁共振动脉自旋标记成像的2型糖尿病患者大脑灌注特征网络研究

目的利用MR动脉自旋标记(arterial spin labeling,ASL)分析2型糖尿病(type 2 diabetes mellitus,T2DM)患者大脑微循环血流灌注及灌注特征模式的改变,并分析这些变化与生化指标间的相关性。材料与方法选取就诊于我院符合T2DM诊断标准的患... 目的利用MR动脉自旋标记(arterial spin labeling,ASL)分析2型糖尿病(type 2 diabetes mellitus,T2DM)患者大脑微循环血流灌注及灌注特征模式的改变,并分析这些变化与生化指标间的相关性。材料与方法选取就诊于我院符合T2DM诊断标准的患者28人,健康对照(healthy control,HC)组26人,进行ASL成像,进行基于体素水平的分析及主成分分析,比较两组间大脑血流量(cerebral blood flow,CBF)及灌注特征网络的改变。结果与HC组比较,糖尿病患者两侧中央旁小叶、左侧补充运动区、两侧扣带回中部、左侧岛盖部额下回、左侧颞中回、左侧颞下回等区域的灌注明显较低(P<0.05,GRF多重比较校正)。与疾病相关的两个灌注网络的方差组分占总方差的比率分别为17.6%和11.7%(位于95%置信区间)具有显著性。第一个灌注网络特征表达值与空腹血糖明显正相关(r=0.32,P=0.001),以第二个灌注特征网络为模板提取的糖尿病组CBF与患者空腹血糖负相关(r=0.12,P=0.03)。结论糖尿病患者部分脑区血流灌注降低,基于主成分的灌注特征能够区分T2DM患者与HC,其灌注模式的改变反映了大脑血流灌注的重塑,为糖尿病微血管病变的早期诊断及干预提供了参考依据。 展开更多

层位追踪是地震资料解释中一项基础且重要的工作,常规智能层位追踪方法的精度难以满足实际生产需求。为此,提出了一种基于U^(2)-Net的高精度多套层位追踪方法。首先,设计一种充填标签的制作方法,遍历地震数据每个像素点,判断当前像素点... 层位追踪是地震资料解释中一项基础且重要的工作,常规智能层位追踪方法的精度难以满足实际生产需求。为此,提出了一种基于U^(2)-Net的高精度多套层位追踪方法。首先,设计一种充填标签的制作方法,遍历地震数据每个像素点,判断当前像素点所在位置并为其划分一个层位区域;对于穿过断层的层位,则自动搜寻相邻层位,实现非全区层位、断层等复杂条件下的地震反射层位及不整合面的充填标签的制作;然后,利用充填标签,采用U^(2)-Net网络模型对F3数据体和M工区地震资料进行训练。与U-Net+PPM网络模型相比,U^(2)-Net网络模型的预测精度更高,稳定性更好,泛化性更强,训练时间更短,且预测复杂地区的地震反射层位的准确率和平均交并比都大于95%。该方法可以较好地适应低信噪比地震资料的层位追踪。 展开更多

The health effects of ambient PM 2.5 and its potential mechanisms have generated considerable interest.In vitro cell studies and ex vivo animal experiments may not accurately determine the characteristics of PM 2.5 particles.To better understand their detailed mechanisms,we performed an in vivo study using single photon emission tomography(SPECT)imaging.To mimic the PM 2.5 particles,SiO2 nanoparticles modified by ethylene carbonate or polyvinyl pyrrolidone were labeled with 131I.After administration via inhalation,in vivo SPECT imaging of the radiolabeled particles in sprague dawley rats was performed.It was found that radioactivity accumulated in the lungs and trachea 6 and 24 h after administration.In addition,significant radioactivity was observed in the abdomen,including the liver and kidneys.The results were also confirmed by ex vivo autoradiography.This study revealed that in vivo SPECT imaging could be an effective method for investigating the properties of PM 2.5 particles. 展开更多

针对我国胆固醇-3,4-^(13)C_(2)产品制备方法空白,以乙酸苯酯-1,2-^(13)C_(2)和4-胆甾烯-3-酮为起始原料,合成胆固醇-3,4-^(13)C_(2)。总体标记合成路线采用胆固醇基础结构拆分后引入标记砌块再重构目标化合物结构的方案,其中4-胆甾烯-3... 针对我国胆固醇-3,4-^(13)C_(2)产品制备方法空白,以乙酸苯酯-1,2-^(13)C_(2)和4-胆甾烯-3-酮为起始原料,合成胆固醇-3,4-^(13)C_(2)。总体标记合成路线采用胆固醇基础结构拆分后引入标记砌块再重构目标化合物结构的方案,其中4-胆甾烯-3-酮通过开环氧化、合环得到标记前体,乙酸苯酯-1,2-^(13)C_(2)作为^(13)C标记引入砌块与标记前体通过乙酰化反应得到乙酰化标记中间体,最后乙酰化标记中间体再经过碳环重构、酯化、还原反应得到^(13)C二标记的类固醇激素类化合物胆固醇-3,4-^(13)C_(2)。其中还原反应显示出一定的不对称合成现象。以投入的乙酸苯酯-1,2-^(13)C_(2)的物质的量计算反应总产率为16.2%。所得终产品经MS、NMR和HPLC表征确认,液相色谱纯度≥96%,液相质谱检测丰度≥98 atom%^(13)C,该产品可用于临床质谱、生物医药等领域检测用稳定同位素内标试剂以及临床质谱用疾病筛查试剂盒原料。 展开更多

目的探讨三维动脉自旋标记(3D-ASL)技术联合ABCD_(2)评分在前循环短暂性脑缺血发作(TIA)中的应用价值。方法选择2021年5月至2022年12月在牡丹江医学院附属红旗医院确诊为前循环TIA的患者64例,完成各项磁共振检查,其中双时相3D-ASL标记... 目的探讨三维动脉自旋标记(3D-ASL)技术联合ABCD_(2)评分在前循环短暂性脑缺血发作(TIA)中的应用价值。方法选择2021年5月至2022年12月在牡丹江医学院附属红旗医院确诊为前循环TIA的患者64例,完成各项磁共振检查,其中双时相3D-ASL标记后延迟时间(PLD)分别为1.5、2.5 s,完成ABCD_(2)评分并进行为期90 d的随访。对3D-ASL图像进行分析,测量脑血流量(CBF)并计算相对脑血流量(rCBF),分析3D-ASL、ABCD_(2)评分对TIA患者病情及预后的评估价值。结果PLD=1.5 s CBF值、rCBF值均小于PLD=2.5 s;rCBF值与ABCD_(2)评分呈负相关;3D-ASL对TIA患者异常检出率高于MRA;3D-ASL异常组脑梗死发生率高于正常组;不同ABCD_(2)评分风险组脑梗死发生率比较,差异有统计学意义(P<0.05);ROC曲线分析结果显示,3D-ASL联合ABCD_(2)评分预测TIA患者90 d预后的曲线下面积(AUC)最大为0.907。结论双时相3D-ASL能够发现TIA患者细微灌注改变,反映代偿情况,可作为TIA患者进一步检查的方式,联合ABCD_(2)评分可以有效预测TIA患者90 d预后,具有较大的临床应用价值。 展开更多

目的观察聚肌苷酸聚胞苷酸(Poly-IC)处理对高血脂大鼠脑缺血/再灌注(I/R)损伤后Bcl-2和Bax表达的影响,同时检测脑梗死灶、血脑屏障通透性和行为损伤症状,探讨其对高血脂合并脑I/R损伤是否发挥神经保护作用。方法高血脂大鼠随机分为脑I/... 目的观察聚肌苷酸聚胞苷酸(Poly-IC)处理对高血脂大鼠脑缺血/再灌注(I/R)损伤后Bcl-2和Bax表达的影响,同时检测脑梗死灶、血脑屏障通透性和行为损伤症状,探讨其对高血脂合并脑I/R损伤是否发挥神经保护作用。方法高血脂大鼠随机分为脑I/R组、Poly-IC预处理组、Poly-IC后处理组和假手术(Sham)组,每组20只。0~4分神经行为评分记录各组大鼠的神经行为表现,伊文思蓝染色法检测各组大鼠血脑屏障通透性,TTC染色法观察各组大鼠脑组织梗死情况,TUNEL染色法测神经细胞的凋亡情况,Western blotting观察Bcl-2和Bax相对表达水平。结果与sham组比较,I/R组神经行为损伤症状严重,评分明显升高(P<0.05)。而Poly-IC预处理和后处理组评分较I/R组均明显降低(P<0.05)。伊文思蓝染色结果显示,I/R组较sham组血脑屏障通透性明显增高(P<0.05),而Poly-IC预处理或后处理可明显改善血脑屏障通透性(P<0.05)。脑梗死体积检测结果显示,sham组未见梗死灶均匀红染,而I/R组脑组织可见白色梗死区。与I/R组比较,Poly-IC预处理和后处理组梗死灶体积均明显缩小(P<0.05)。I/R组大鼠大脑皮质内细胞凋亡指数较sham组显著升高(P<0.05),而Poly-IC预处理和后处理组大鼠大脑皮质内细胞凋亡指数较I/R组均明显降低(P<0.05)。与sham组比较,I/R组Bax表达明显升高(P<0.05),Bcl-2表达明显降低(P<0.05)。与I/R组比较,Poly-IC预处理和后处理组Bax表达均明显降低(P<0.05),Bcl-2表达明显升高(P<0.05)。结论Poly-IC预处理或后处理可改善神经行为损伤症状,减轻血脑屏障损害,缩小脑梗死体积,降低神经细胞凋亡指数,对高血脂大鼠脑I/R损伤起到神经保护作用;该保护作用可能与Bcl-2和Bax表达水平改变有关。 展开更多

BACKGROUND Type 2 diabetes mellitus(T2DM) has been strongly associated with an increased risk of developing cognitive dysfunction and dementia.The mechanisms of diabetes-associated cognitive dysfunction(DACD) have not been fully elucidated to date.Some studies proved lower cerebral blood flow(CBF) in the hippocampus was associated with poor executive function and memory in T2DM.Increasing evidence showed that diabetes leads to abnormal vascular endothelial growth factor(VEGF) expression and CBF changes in humans and animal models.In this study,we hypothesized that DACD was correlated with CBF alteration as measured by three-dimensional(3D) arterial spin labeling(3D-ASL) and VEGF expression in the hippocampus.AIM To assess the correlation between CBF(measured by 3D-ASL and VEGF expression) and DACD in a rat model of T2DM.METHODS Forty Sprague-Dawley male rats were divided into control and T2DM groups.The T2DM group was established by feeding rats a high-fat diet and glucose to induce impaired glucose tolerance and then injecting them with streptozotocin to induce T2DM.Cognitive function was assessed using the Morris water maze experiment.The CBF changes were measured by 3D-ASL magnetic resonance imaging.VEGF expression was determined using immunofluorescence.RESULTS The escape latency time significantly reduced 15 wk after streptozotocin injection in the T2DM group.The total distance traveled was longer in the T2DM group;also,the platform was crossed fewer times.The percentage of distance in the target zone significantly decreased.CBF decreased in the bilateral hippocampus in the T2DM group.No difference was found between the right CBF value and the left CBF value in the T2DM group.The VEGF expression level in the hippocampus was lower in the T2DM group and correlated with the CBF value.The escape latency negatively correlated with the CBF value.The number of rats crossing the platform positively correlated with the CBF value.CONCLUSION Low CBF in the hippocampus and decreased VEGF expression might be crucial in DACD.CBF measured by 3D-ASL might serve as a noninvasive imaging biomarker for cognitive impairment associated with T2DM. 展开更多

目的探讨动脉自旋标记(arterial spin labeling,ASL)成像定量评估2型糖尿病(type 2 diabetes mellitus,T2DM)肾脏血流灌注与肾功能损伤的相关性和临床应用价值。材料与方法前瞻性招募不同程度肾功能损伤的T2DM患者,根据有无蛋白尿及估... 目的探讨动脉自旋标记(arterial spin labeling,ASL)成像定量评估2型糖尿病(type 2 diabetes mellitus,T2DM)肾脏血流灌注与肾功能损伤的相关性和临床应用价值。材料与方法前瞻性招募不同程度肾功能损伤的T2DM患者,根据有无蛋白尿及估算肾小球滤过率(estimated glomerular filtration rate,eGFR)值将其分组:A组,T2DM患者,无蛋白尿;B组,出现蛋白尿且eGFR≥60 mL/(min·1.73 m^(2)),即糖尿病肾病(diabetic kidney disease,DKD)Ⅰ~Ⅱ期;C组,15 mL/(min·1.73 m^(2))≤eGFR<60 mL/(min·1.73 m^(2)),即DKDⅢ~Ⅳ期。同时纳入健康志愿者作为对照组。行双肾常规MRI平扫及ASL扫描获取双侧肾皮质肾血流量(renal blood flow,RBF)值,同时收集肾功能及尿常规等生化指标,分析各组肾皮质RBF值的差异性,以及相关指标的相关性、敏感性及最佳诊断阈值等。结果左肾与右肾肾皮质RBF值差异无统计学意义(P均>0.05)。四组受试者肾皮质RBF值总体差异具有统计学意义(P均<0.001)。进一步两两比较:与对照组相比,A、B、C组肾皮质RBF值明显降低(P均<0.05),对照组为(174.28±23.89)mL/(100 g·min),A组降低为(159.66±28.54)mL/(100 g·min),B组降低为(142.16±19.49)mL/(100 g·min),C组最低为(122.55±18.59)mL/(100 g·min),降低比例依次为8%、18%、30%。与A、B两组比较,C组肾皮质RBF值明显降低(P均<0.05),降低比例分别为23%、14%。B组与A组相比,肾皮质RBF值降低(P均<0.05),降低比例为11%。肾皮质RBF值与血清肌酸酐(serum creatinine,Scr)呈负相关(r=-0.429,P<0.001),而与eGFR呈正相关(r=0.377,P<0.001)。肾皮质RBF值鉴别健康志愿者与T2DM的AUC值为0.651(95%CI:0.520-0.768);鉴别T2DM与DKD的AUC值为0.734(95%CI:0.619-0.829);鉴别早期DKD与中晚期DKD的AUC为0.760(95%CI:0.580-0.891)。结论ASL成像可无创定量评估肾脏血流灌注变化,且根据RBF减低可有效评估T2DM患者病情进展程度,有望成为无创评估T2DM患者肾功能损害的有效影像检查方法。 展开更多

Our objective was to synthesize and optimize a dual-labeled trastuzumab-based imaging agent that can be used to validate an optical imaging agent with potential use in identifying tumor metastases in human epidermal growth factor receptor 2 (HER2) positive breast cancer patients. Methods: [111In]-DTPA-trastuzumab-IRDye 800 was synthesized by a three-step procedure. Purity, stability, immunoreactivity, internalization and biodistribution were explored in HER2+ SKBR-3 cells. Biodistribution of [111In]-DTPA-trastuzumab-IRDye 800 was performed in a SKBR-3 xenograft model. Results: [111In]-DTPA-trastuzumab-IRDye 800 demonstrated high purity by both chemical and fluorometric determinations. Both flow cytometry and the Lindmo assay demonstrated a high binding affinity of [111In]-DTPA-trastuzumab-IRDye 800 to HER2-overexpressing cells. The dual-labeled conjugate was stable in PBS, but not in serum after 24 h at 37 ℃. Larger molecules (〉150 kD) were seen after a 24 h-incubation in human serum. Biodistribution studies revealed tumor-specific accumulation of [111In]-DTPA- trastuzumab-IRDye 800 in SKBR-3 tumors, and tumor uptakes at 24 and 48 h were (12.42±1.72)% and (9.96±1.05) %, respectively, following intravenous administration. The tumor-to-muscle ratio was 9.13±1.68 at 24 h, and increased to 12.79±2.13 at 48 h. Liver and kidney showed marked uptake of the dual-labeled imaging agent. Conclusions: [111In]-DTPA-trastuzumab-IRDye 800 is an effective diagnostic biomarker that can be used to validate dual-labeled, molecularly targeted imaging agents and can allow these agents to be translated into clinical practice for identifying HER2+ lesions. 展开更多

Objective: To investigate the effect of fluorescent dye labeling on the targeting capabilities of 111 In- (DTPA).-trastuzumab-(IRDye 800)m. Methods: Trastuzumab-based conjugates were synthesized and conjugated with diethylenetriaminepentaacetic acid (DTPA) at molar ratios of I, 2, 3 and 5 and with a fluorescent dye (IRDye 800CW) at molar ratios of 1, 3 and 5. Immunoreactivity and internalization were assessed on SKBR-3 cells, overexpressing human epidermal growth factor receptor 2. The stability in human serum and phosphate-buffered saline (PBS) was evaluated. The biodistribution of dual-labeled conjugates was compared with that of 111In-(DTPA)2-trastuzumab in a SKBR-3 xenograft model to evaluate the effect of dye-to-protein ratio. Results: All trastuzumab-based conjugates exhibited a high level of chemical and optical purity. Flow cytometry results showed that increasing dye-to-protein ratios were associated with decreased immunoreactivity. Stability studies revealed that the conjugate was stable in PBS, while in human serum, increased degradation and protein precipitation were observed with increasing dye-to-protein ratios. At 4 h, the percentages of internalization of dual-labeled conjugates normalized by dye-to-protein ratio (m) were 24.88%±2.10%, 19.99%±0.59%, and 17.47%±1.26% for "m" equal to 1, 3, and 5, respectively. A biodistribution study revealed a progressive decrease in tumor uptake with an increase in the dye-to-protein ratios. The liver, spleen and kidney showed a marked uptake with increased dye-to-protein ratios, particularly in the latter. Conclusions: With non-specific-site conjugation of the fluorescent dye with a protein based on imaging agent, the increase in dye-to-protein ratios negatively impacted the immunoreactivity and stability, indicating a reduced tumor uptake. 展开更多
