<Abstract>factors,such as cigarette smoking,in esophageal squamous cell carcinoma(ESCC)in northeastern Iran,a region with a high incidence of ESCC.METHODS:The expression of p53 and p21 proteins was investigated ...<Abstract>factors,such as cigarette smoking,in esophageal squamous cell carcinoma(ESCC)in northeastern Iran,a region with a high incidence of ESCC.METHODS:The expression of p53 and p21 proteins was investigated immunohistochemically in tumor tissue from 80 ESCC patients and in 60 available paraffinembedded blocks of adjacent normal specimens from the cases,along with normal esophageal tissue from 80 healthy subjects.RESULTS:Positive expression of p53 protein was detected in 56.2%(45/80)of ESCC cases,and in none of the normal esophageal tissue of the control group(P<0.001).Furthermore,73.8%(59/80)of ESCC cases and 43.8%(35/80)of controls had positive expression of p21 protein(P<0.001).Cigarette smoking was significantly associated with p53 over-expression in ESCC cases(P=0.010,OR=3.64;95%CI:1.32-10.02).p21 over-expression was associated with poorer clinical outcome among the ESCC patients(P=0.009).CONCLUSION:Over-expression of p53 in association with cigarette smoking may play a critical role in ESCC carcinogenesis among this high-risk population of northeastern Iran.Furthermore,p21 over-expression was found to be associated with poor prognosis,specifically in the operable ESCC patients.展开更多

虽然水生生物大多是变温动物并且温度对化合物的毒性有非常重要的作用,但生态毒理学实验一般在实验室标准温度(20℃)或者生物最适温度下进行。我们调查了适应温度(20℃、24℃和28℃)对镉暴露(0,20和40μg·L^(-1))中大型溞生理和亚...虽然水生生物大多是变温动物并且温度对化合物的毒性有非常重要的作用,但生态毒理学实验一般在实验室标准温度(20℃)或者生物最适温度下进行。我们调查了适应温度(20℃、24℃和28℃)对镉暴露(0,20和40μg·L^(-1))中大型溞生理和亚细胞水平的作用,实验终点包括镉的急性毒性(48 h-EC50)、大型溞的21 d存活率、21 d累积繁殖数(21-d CR)、第一次生育时间(time to first brood,TFB)、能量储存、电子转移活性(electron transport system activity,ETS)、热应激蛋白hsp70和金属硫蛋白(MT)。结果显示:适应温度升高时大型溞的镉耐性降低,20℃、24℃和28℃下的48 h-EC50分别为(40.2±9.9)μg·L^(-1)、(15.6±2.7)μg·L^(-1)和(10.7±1.9)μg·L^(-1);28℃下,大型溞21 d存活率分别比对照(20℃,0μg·L^(-1)镉暴露)降低23%(20μg·L^(-1)镉暴露)和87%(40μg·L^(-1)镉暴露)并且大型溞的21 d累积繁殖数(21-d CR)显著降低。大型溞的能量储存随适应温度的升高而降低,24℃和28℃时分别降低了46%和62%(0μg·L^(-1)镉暴露),48%和60%(20μg·L^(-1)镉暴露)、80%和91%(40μg·L^(-1)镉暴露)。而镉浓度只有达到40μg·L^(-1)时,对大型溞的能量储存有明显影响。适应温度和镉暴露对电子转移系统活性(ETS)没有显著影响。28℃的适应温度和40μg·L^(-1)的镉暴露都能诱导大型溞热应激蛋白hsp70和金属硫蛋白(MT)显著上升,但适应温度(20℃~28℃)和镉暴露(0~40μg·L^(-1))的结合对hsp70先诱导后抑制,MT则一直被诱导上升。总之,生物在不同温度下对毒物的反应存在差异,将不同适应温度下的毒性数据整合到生态危险性评价能够保证生物获得更充分的保护;此外,hsp70和MT的变化是环境压力的综合反应,作为某种污染物的生物标志物时需要综合考虑环境因素。展开更多
[目的]探讨微小RNA 21(micro RNA-21,miR-21)和细胞程序性死亡蛋白(programmed cell death protein 4,PDCD4)在Ⅱ期食管鳞癌术后患者癌组织中表达,分析其与临床病理特征的关系及对预后的影响。[方法]收集胸中段Ⅱ期食管癌术后患者标本97...[目的]探讨微小RNA 21(micro RNA-21,miR-21)和细胞程序性死亡蛋白(programmed cell death protein 4,PDCD4)在Ⅱ期食管鳞癌术后患者癌组织中表达,分析其与临床病理特征的关系及对预后的影响。[方法]收集胸中段Ⅱ期食管癌术后患者标本97例,将样本组织脱蜡提取总RNA;应用实时免疫荧光定量聚合酶链反应(RT-PCR)法检测所有标本癌组织、癌旁组织miR-21的表达情况;免疫组化法检测测癌组织和癌旁组织PDCD4蛋白表达水平。[结果]miR-21在Ⅱ期食管鳞癌术后患者癌组织中的表达较癌旁组织升高(t=12.06,P<0.01),PDCD4在癌组织中的表达较癌旁组织降低(t=8.36,P<0.01)。miR-21和PDCD4在癌组织中的表达呈负相关(r=-0.41,P<0.01),在癌旁组织中的表达呈正相关(r=0.51,P<0.01)。癌组织中miR-21的表达与食管鳞癌的浸润深度、淋巴结转移、内镜下病变长度呈正相关(r=0.23,P=0.02;r=0.22,P=0.03;r=0.38,P<0.01)、与分化程度呈负相关(r=-0.60,P<0.01),PDCD4蛋白的表达与食管鳞癌的浸润深度、淋巴结转移、内镜下病变长度负相关(r=-0.21,P=0.04;r=-0.24,P=0.01;r=-0.37,P<0.01)、与分化程度呈正相关(r=0.53,P<0.01),二者与年龄、性别、位置不相关(P>0.05)。COX回归模型发现Ⅱ期食管鳞癌患者肿瘤浸润深度、淋巴结转移、分化程度、内镜下病变长度、miR-21表达、PDCD4表达均为影响患者PFS的因素(χ~2=10.36,P=0.01;χ~2=7.89,P<0.01;χ~2=6.48,P=0.02;χ~2=5.32,P=0.03;χ~2=9.56,P=0.01;χ~2=5.40,P=0.02)。miR-21对于术后3年PFS、OS预测曲线下面积分别为73.5%、81.0%(P<0.01);PDCD4对于术后3年PFS、OS预测曲线下面积分别为77.3%、73.2%(P<0.01)。miR-21、PDCD4高、低表达组PFS差异有统计学意义(χ~2=19.6,P<0.01;χ~2=11.45,P<0.01),OS差异也有统计学意义(χ~2=27.84,P<0.01;χ~2=21.35,P<0.01)。[结论]Ⅱ期食管鳞癌术后患者癌组织与癌旁正常组织中miR-21、PDCD4蛋白表达存在明显差异。高表达miR-21提示预后较差,而高表达PDCD4提示预后较好。展开更多
