Methods The Chinese and a observational study consisted of a population-based cross-sectiona subsequent 1-year follow-up study of 171 subjects with the fasting cholesterol of 5.13-8.00 mmol/L. study of 456 plasma total Results In the cross-sectional analysis, plasma low-density lipoprotein cholesterol (LDL-c) had a significant and negative association with the erythrocyte membrane n-6:n-3 PUFAs ratio (P for trend=0.019) after adjusting for sex, age and total PUFA percentage. In the follow-up study, 171 subjects were categorized into quartiles by the changes of n-6:n-3 ratio in erythrocyte membrane (△=month 12-month 0). In the top quartile whose ratios of n-6:n-3 increased by an average of 1.25 during the follow-up, the LDL-c-lowering extent was 3.3 times of that in the lowest quartile whose ratios of n-6:n-3 decreased by an average of 1.13 (-1.07 mmol/L v.s. -0.32 mmol/L). The hsCRP decreased by 0.11 mg/dL in the lowest quartile while increasing by 0.10 mg/dL in the top quartile (P for difference=0.052). Conclusion Our results suggested that the balance between n-6 and n-3 fatty acids may optimize the cardiovascular benefits from dietary PUFAs.展开更多

The therapeutic effect of gamma-aminobutyric acid(GABA)on diabetes was spread as one of the alarming epidemics worldwide.The study aims to investigate the function of Lactobacillus brevis KLDS_(1.0727) and KLDS_(1.037...The therapeutic effect of gamma-aminobutyric acid(GABA)on diabetes was spread as one of the alarming epidemics worldwide.The study aims to investigate the function of Lactobacillus brevis KLDS_(1.0727) and KLDS_(1.0373) strains as glutamic acid decarboxylase 65(GAD65)carriers capable of generating GABA by comparing in vitro free and freeze-dried models and GABA intervention in vivo.PCR amplification of gad and in vitro i.e.,(growth rate,viability at different pH,bile tolerance,and survivability in simulated gastric juice)were performed.In vivo experiments were conducted in 7 groups of C57BL/6J mice.Each group was injected with streptozotocin(Cont_(STZ),INSSTZ,LAC1_(STZ),LAC_(1MFDSTZ),LAC_(2STZ),LAC_(2MFDSTZ))daily except for the control(Cont).One group was injected with insulin(INSSTZ).The body weight and hyperglycemia in the blood were assessed weekly,post-euthanasia blood plasma parameters,insulin,and histological examination were evaluated.Results indicated L.brevis strains demonstrated a great tolerance to bile and simulated gastric juice in vitro(P<0.05).Cont_(STZ) had the highest average glucose level(6.84±6.46)mmol/L while INS_(STZ) expressed dramatically decreed in glucose level and displayed a significant decline in the average of weekly blood glucose(−5.74±3.08)mmol/L.The lowest body weight(ContSTZ)was(19.30±0.25)g.Based on the blood plasma analysis,L.brevis strains improved good cholesterol properties,liver and kidney functions,where most of these parameters fall within the average the reference range and prevent the development of symptoms of type 1 diabetes in vivo.As recommended,L.brevis should be commonly distributed as a postbiotic GABA in pharmaceutical and nutritional applications.展开更多

This is the first study to investigate the release, distribution, and efficacy of hydroxycamptothecin...Hydroxycamptothecin is a potent antineoplastic agent that has shown efficacy against multiple tumor lines in vitro. This is the first study to investigate the release, distribution, and efficacy of hydroxycamptothecin which was incorporated into the biodegradable polymer Polylactic Acid (PLA), and implant into brain directly. In vitro release curve generated showed that a large initial release occurred over the first three days and was followed by a steady, but considerably slower rate of release over the next 25 days. After implanting the discs into 40 male SD rats, the animals were followed up to 28 days, where the concentration in brain tissue was far higher than that in peripheral blood at the each of the eight time-points evaluated, and it was also within the therapeutic range for C6 cells tested in vitro. The in vivoefficacy of the discs was evaluated with rats inoculated intracranially with C6 glioma and treated with hydroxycamptothecin polymer compared to intravenous as well as intratumoral injections;the median survival is 21.1, 13.9, 14.9 days, respectively. Given these data, we conclude that the biodegradable polymer PLA releases hydroxycamptothecin, producing tumoricidal levels in brain tissues and prolonging survival in a rat glioma model.展开更多

文摘目的:观察非甾体类抗炎药消炎痛和阿斯匹林对C57BL/6 和Balb/c小鼠胃酸分泌的影响及胃黏膜损害的效应. 方法:应用幽门结扎术,测定消炎痛(40 mg/kg)和阿斯匹林(250 mg/kg)i.p后C57BL/6和Balb/c小鼠总胃酸分泌量(mL),胃酸度([H+]mmol/L)以及3 h总酸度(μmol/h)并应用Lanza score评分观察药物对胃黏膜的损害程度.同时应用[14C]-氨基比林蓄积试验观察2种小鼠离体胃对组胺刺激的效应. 结果:在基础状态下,C57BL/6和Balb/c小鼠的总胃酸分泌量和总酸度显著不同(0.82±0.06 vs 2.32±0.18; P<0.0 001;2.9±0.4 vs 9.7±1.6,P=0.0 005),而胃酸度没有显著差别(P=0.377).在C57BL/6小鼠,腹腔内应用消炎痛后总胃酸分泌量和总酸度与基础水平相比差别显著(2.08±0.16 vs 0.82±0.06,P<0.0 001; 9.4±1.1 vs 2.9±0.4,P<0.0 001),而胃酸度也没有显著差别(P=0.11);腹腔内应用阿斯匹林后总胃酸分泌量,胃酸度以及总酸度分别为1.5±0.3、16.1±1.4 和8.7±2.7,与基础水平相比均有显著差异(P=0.03, 0.009,0.03);在Balb/c小鼠,腹腔内应用消炎痛后总胃酸分泌量,胃酸度以及总酸度分别为3.4±0.3、5.8±1.0和6.5±1.2,前二者与基础水平相比有显著差异(P=0.007,0.01);腹腔内应用阿斯匹林后总胃酸分泌量,胃酸度以及总酸度与基础水平相比均没有显著差异(P=0.15,0.15,0.7).应用消炎痛后,C57BL/6和Balb/c小鼠Lanza score分别为12.6±0.9和16.4±1.1 (P=0.03);应用阿斯匹林后,C57BL/6和Balb/c小鼠Lanza score为10.4±1.0和11.0±1.1(P=0.91).应用[14C]-氨基比林蓄积试验观察2种小鼠离体胃对组胺刺激的效应,发现C57BL/6小鼠对不同浓度组胺刺激的效应显著低于Balb/c小鼠(10-5mol/L:30.8±8.5 vs 120.8±23.0.P=0.006;10-4mol/L:45.1±7.1 vs 236.1±48.7.P=0.005;10-3mol/L:37.9±5.3 vs 199.4±35.2,P=0.002). 结论:C57BL/6和Balb/c小鼠的胃酸分泌生理功能存在显著差异.消炎痛和阿斯匹林对C57BL/6和Balb/c小鼠的胃酸分泌功能和胃黏膜损害产生不同的效应.
