BACKGROUND Cervical intraepithelial neoplasia(CIN)is an important precursor of cervical cancer.Early detection and treatment can reduce the incidence of cervical cancer.AIM To investigate the detection rate of human p...BACKGROUND Cervical intraepithelial neoplasia(CIN)is an important precursor of cervical cancer.Early detection and treatment can reduce the incidence of cervical cancer.AIM To investigate the detection rate of human papillomavirus(HPV)E6/E7 mRNA in cervical tissue of patients with different types of epithelial cell neoplasia(CIN)and its relationship with CIN progression and diagnosis.METHODS One hundred women with HPV infection detected by cervical exfoliation cytology between January 2022 and January 2023 were retrospectively selected.These patients were graded CIN based on colposcopy and cervical pathology.The positive expression rates of HPV E6/E7 mRNA and HPV[polymerase chain reaction(PCR)-reverse dot crossing]were compared among all groups.Patients with HPV E6/E7 mRNA expression in the grade 1 CIN group were followed up for 1 yr.The relationship between atypical squamous epithelium and high malignant epithelial neoplasia was investigated by univariate and multivariate analysis.RESULTS The diagnostic sensitivity,specificity,and sensitivity of PCR-reverse point hybrid ization technology for secondary CIN were 70.41%,70.66%,and 0.714,respectively.Sensitivity and specificity for secondary CIN were 752%and 7853%,respectively,the area under the curve value was 0.789.Logistic Multifactorial model analysis revealed that the HPV positive rates and the HPV E6/E7 mRNA positive rates were independent risk factors of CIN grade I(P<0.05).In CIN grade I patients with positive for HPV E6/E7 mRNA,in its orientation to grade CIN patients,in its orientation to grade CIN patients,at 69.2%,compared with patients negative for HPV E6/E7 mRNA(30.8%),significant difference(P<0.05).CONCLUSION HPV E6/E7 mRNA and HPV(PCR-reverse dot hybrid)positive expression have a close relationship with CINgrade disease progression and is an independent risk factor for high-grade CIN lesions.展开更多
The bonding interface of 7B52 Al alloy laminated composite (ALC) fabricated by hot rolling was investigated using optical microscopy (OM), transmission electron microscopy (TEM), scanning electron microscopy (...The bonding interface of 7B52 Al alloy laminated composite (ALC) fabricated by hot rolling was investigated using optical microscopy (OM), transmission electron microscopy (TEM), scanning electron microscopy (SEM), ultrasonic flaw detection (UFD), and bonding strength tests. The results show that metallurgical bonding is achieved at the interface after composite rolling. The TEM analysis and tensile tests indicate that the 7B52 ALC plate combines high strength of the hard individual layer and good toughness of the soft individual layer. However, UFD technology and SEM analysis prove that the defects (thick oxide films, acid washed residues, air, oil and coarse particles) existing in the bonding interface are harmful to the bonding strength. To sum up, the composite roiling process is suitable for 7B52 ALC plate, and the content and size of the defects should be controlled strictly. Advanced surface treatment of each individual layer would be beneficial to further improve the bonding quality.展开更多
To analyze the resistance of Lianjing 7 against rice black-streaked dwarf disease and find the molecular markers closely linked with the resistance gene, an F2:3 population derived from an RBSDD-resistant cultivar Li...To analyze the resistance of Lianjing 7 against rice black-streaked dwarf disease and find the molecular markers closely linked with the resistance gene, an F2:3 population derived from an RBSDD-resistant cultivar Lianjing 7 and an RBSDD-susceptible cultivar Peiai 64, was evaluated using natural infection methods and artificial inoculation by scoring the disease severity rating. The results showed Lianjing 7 was resistant to RBSDD, with the incidence between 5.5% and 12.25% in all tests. For the parents of Lianjing 7, only Zhongjingchuan-2 was resistant to RBSDD. Besides this, Lianjing 7 assumed an antixenosis and antibiosis score similar with susceptible variety Wuyujing 3, which indicates that so RBSDD-resistance of Lianjing 7 is mainly caused by the resistance specifically to RBSDV. In the F2:3 population of Lianjing 7/Peiai 64, the disease severity rates showed a continuous distribution with transgressive segregation, it indicated the resistance of Lianjing 7 was controlled by multi-genes. The marker RM287 was determined closely linked with the resistance locus via BSA method. To conclude, Lianjing 7 has a high resistance to RBSDD, which is conferred with by Zhongjingchuan-2, and the marker RM287 should be useful for breeding novel RBSDD-resistant cultivars via marker-assisted selection (MAS).展开更多

Previous research revealed the positive activity of matrix metalloproteinase 7(MMP7) on migration and myelin regeneration of Schwa nn cells(SCs). However, understanding of the molecular changes and biological activiti...Previous research revealed the positive activity of matrix metalloproteinase 7(MMP7) on migration and myelin regeneration of Schwa nn cells(SCs). However, understanding of the molecular changes and biological activities induced by increased amounts of MMP7 in SCs remains limited. To better understand the underlying molecular events, primary SCs were isolated from the sciatic nerve stump of newborn rats and cultured with 10 nM human MMP7 for 24 hours. The results of genetic testing were analyzed at a relatively relaxed threshold value(fold change ≥ 1.5 and P-value < 0.05). Upon MMP7 exposure, 149 genes were found to be upregulated in SCs, whereas 133 genes were downregulated. Gene Ontology analysis suggested that many differentially expressed molecules were related to cellular processes, single-organism processes, and metabolic processes. Kyoto Enrichment of Genes and Genomes pathway analysis further indicated the critical involvement of cell signaling and metabolism in MMP7-induced molecular regulation of SCs. Results of Ingenuity Pathway Analysis(IPA) also revealed that MMP7 regulates biological processes, molecular functions, cellular components, diseases and functions, biosynthesis, material metabolism, cell movement, and axon guidance. The outcomes of further analysis will deepen our comprehension of MMP7-induced biological changes in SCs. The porcine TSARG7 had been identified by cloning in silico but had not been confirmed experimentally. The full-length mRNA of porcine TSARG7 gene was sequenced and two splice variants were discovered. The full-length cDNA of TSARG7 variant 1 was 2 513 bp and variant 2 was 2 634 bp. The putative porcine TSARG7 proteins, which were located in the cytoplasm, encoded 458 and 456 amino acids, respectively. Real-time PCR analysis showed that TSARG7 gene was expressed in various tissues, but at different levels. The expression levels of this gene were higher in the skeletal muscle, heart, and testis than that in other tissues, suggesting that the TSARG7 gene played a role in procine skeletal muscle, heart, and testis functions.展开更多

For all the cases having been considered, a 1/16 segmentof the whole toric shell are used to calculate the linear critical buckling load (Pc.,,) under auniform and nonwhform external pressure. BACKGROUND The clinical manifestations of trisomy 7 mosaicism are diverse and nonspecific,so prenatal diagnosis is very difficult.CASE SUMMARY Two pregnant women with abnormal prenatal screening results were included....BACKGROUND The clinical manifestations of trisomy 7 mosaicism are diverse and nonspecific,so prenatal diagnosis is very difficult.CASE SUMMARY Two pregnant women with abnormal prenatal screening results were included.One was a 22-year-old woman(G1P0).At 31st week of gestation,ultrasound revealed that the posterior horn of the left lateral ventricle was 10 mm and the right renal pelvis had a separation of 7 mm.The other pregnant woman was 33 years old(G2P1L1A0),and her fetus was found to have a cardiac malformation at the 24th week of gestation.Copy number variation sequencing,whole-exome sequencing and karyotype analysis were carried out after amniocentesis,and both fetuses were diagnosed with trisomy 7 mosaicism.After parental counseling,one woman continued the pregnancy,and the other woman terminated the pregnancy.CONCLUSION In trisomy 7 mosaicism,the low proportion of trisomy does not lead to abortion,but can result in abnormal fetal development,which can be detected via ultrasound.Therefore,clinicians need to pay more attention to various aspects of fetal growth and development,combining with imaging,cellular,molecular genetics and other methods to perform comprehensive evaluations of fetuses to provide more reliable genetic counseling for pregnant women.展开更多

The results showed that the full-length CDS sequence ...In this study, full-length CDS sequence of IGFBP-7 gene was cloned from Liangshan semi-fine wool sheep with RT-PCR method and analyzed with bioinformatics methods. The results showed that the full-length CDS sequence of IGFBP-7 gene in Liangshan semi-fine wool sheep was 846 bp in length, encoding 282 amino acids. The CDS sequence shared 99%, 95% and 90% homology with bovine, human and rat, respectively; the amino acid sequence shared 98%, 93% and 89%, respectively. The GenBank accession number was FJ589640.1. The amino acid molecular weight of IGFBP-7 was 29.0 kD, and the theoretical isoelec- tric point (pl) was 8.25. The result of phylogenetic analysis showed that IGFBP-7 gone in Liangshan semi-fine wool sheep exhibited close phylogenetic relationships with bovine, goat and other mammals, and distant phylogenetic relationships with Danio rerio and Haliotis diversicolor. IGFBP-7 gene had uniformly distributed hy- drophobic and hydrophilic regions, harboring one signal peptide, two transmembrane regions, 16 phosphorylation sites, four N-glycosylation sites and one O-glyco- sylation site. The result of secondary structure analysis showed that the random coil, or-helix and β-sheet regions accounted for 64.89%, 19.86% and 15.25%, respectively. The result of tertiary structure analysis showed that IGFBP-7 harbors an IGFBP_N domain and an Ig-like domain. This study provided scientific basis for further investigating the function of IGFBP-7 gene in sheep.展开更多

A program written in Allanguage is used to derive the coefficients of displacement equations: therefore the amount of manual work is greatly decreased. The E6 and E7 genes in HPV-16 are the major virulent genes in HPV, and we wanted to investigate the polymorphism of E6 and E7 genes of HPV-16 originated from Jingjiang, Jiangsu province. In research, HPV-16 sample was collected, and the E6 and E7 genes fragments were amplified by PCR assay. The sequences of E6 and E7 genes were used for phylogenetic analysis by Mega 5.0 software. By comparison, the major mutations of E6 gene were T178G (41.67%) and G658A (17%), as well as C58G, T61A and G188C (14%), T61C and C656A (11%). The mutation of E7 gene was mainly C491A and T935A (23%);G514C, G937C and G519C (11%). The 68 nucleotide site deficiency (69.4%) of E6 and 535 nucleotide site deficiency (23%) of E7 were dominant. In our study, we did not find the relevance between the E6 and E7 genes mutations and pathologic severity. But we think the E6 and E7 genes mutation of HPV-16 might affect vaccine prevention and treatment effect.展开更多
Objective:To analyze the structural features of human P2X7R protein,meanwhile homology,protein-protein interaction,and cell epitope were predicted to provide theoretical basis for the diagnosis and treatment of relate...Objective:To analyze the structural features of human P2X7R protein,meanwhile homology,protein-protein interaction,and cell epitope were predicted to provide theoretical basis for the diagnosis and treatment of related diseases.Methods:Human P2X7R protein was analyzed and predicted by ProtParam、TMHMM、ProtScale and SignalP 5.0 Server et al.Results:Human P2X7R protein is composed of 595 amino acids,the molecular formula is C 3077 H 4749 N 835 O 871 S 40,with the theoretical isoelectric point PI of 8.62,the instability coefficient is 46.07,the total average hydrophilicity is-0.317,and the protein is an hydrophilic protein instead of secreted protein,no any signal peptide,but transmembrane region is contained,which belong to transmembrane protein.There are 157αhelices and 288 random coils in the secondary structure of human P2X7R protein,accounting for 26.39%and 48.40%of all amino acids,The coverage rate of the three-level structure prediction results is 100%,and the sequence consistency is 80.34%,and the ramachandran plot show that the predicted structure is relatively stability.The interaction of proteins show that there are a variety of proteins interacting with human P2X7R,and P2X7R was closely related to inflammation;this protein had many B and T cell epitopes,which could provide ideas for vaccine development.Conclusion:The results indicate that the analysis of human P2X7R protein that is based on bioinformatics method has certain reliability.P2X7R is involved in the development of inflammatory diseases,and the antigenic epitope of B,Th and CTL cells screened out by prediction will provide the treatment scheme for diseases.展开更多

文摘BACKGROUND The clinical manifestations of trisomy 7 mosaicism are diverse and nonspecific,so prenatal diagnosis is very difficult.CASE SUMMARY Two pregnant women with abnormal prenatal screening results were included.One was a 22-year-old woman(G1P0).At 31st week of gestation,ultrasound revealed that the posterior horn of the left lateral ventricle was 10 mm and the right renal pelvis had a separation of 7 mm.The other pregnant woman was 33 years old(G2P1L1A0),and her fetus was found to have a cardiac malformation at the 24th week of gestation.Copy number variation sequencing,whole-exome sequencing and karyotype analysis were carried out after amniocentesis,and both fetuses were diagnosed with trisomy 7 mosaicism.After parental counseling,one woman continued the pregnancy,and the other woman terminated the pregnancy.CONCLUSION In trisomy 7 mosaicism,the low proportion of trisomy does not lead to abortion,but can result in abnormal fetal development,which can be detected via ultrasound.Therefore,clinicians need to pay more attention to various aspects of fetal growth and development,combining with imaging,cellular,molecular genetics and other methods to perform comprehensive evaluations of fetuses to provide more reliable genetic counseling for pregnant women.

文摘In this study, full-length CDS sequence of IGFBP-7 gene was cloned from Liangshan semi-fine wool sheep with RT-PCR method and analyzed with bioinformatics methods. The results showed that the full-length CDS sequence of IGFBP-7 gene in Liangshan semi-fine wool sheep was 846 bp in length, encoding 282 amino acids. The CDS sequence shared 99%, 95% and 90% homology with bovine, human and rat, respectively; the amino acid sequence shared 98%, 93% and 89%, respectively. The GenBank accession number was FJ589640.1. The amino acid molecular weight of IGFBP-7 was 29.0 kD, and the theoretical isoelec- tric point (pl) was 8.25. The result of phylogenetic analysis showed that IGFBP-7 gone in Liangshan semi-fine wool sheep exhibited close phylogenetic relationships with bovine, goat and other mammals, and distant phylogenetic relationships with Danio rerio and Haliotis diversicolor. IGFBP-7 gene had uniformly distributed hy- drophobic and hydrophilic regions, harboring one signal peptide, two transmembrane regions, 16 phosphorylation sites, four N-glycosylation sites and one O-glyco- sylation site. The result of secondary structure analysis showed that the random coil, or-helix and β-sheet regions accounted for 64.89%, 19.86% and 15.25%, respectively. The result of tertiary structure analysis showed that IGFBP-7 harbors an IGFBP_N domain and an Ig-like domain. This study provided scientific basis for further investigating the function of IGFBP-7 gene in sheep.
文摘The displacement, velocity and acceleration analysis of the general spatial 7R mechanism is discussed in this paper, fused on the method proposed in Ref. [2], an input-output algebra equation of the 16th degree in the tan-half-angle of the output angular displacement is derived. The derivation process and computation are considerably simple. A program written in Allanguage is used to derive the coefficients of displacement equations: therefore the amount of manual work is greatly decreased. The results are verified by a numerical example. The researches of this paper and Ref. [5]found a base for establishing an expert system of spatial mechanism analysis in the future.
文摘Human papillomavirus 16 (HPV-16) is a major high-risk type causing cervical cancer. The E6 and E7 genes in HPV-16 are the major virulent genes in HPV, and we wanted to investigate the polymorphism of E6 and E7 genes of HPV-16 originated from Jingjiang, Jiangsu province. In research, HPV-16 sample was collected, and the E6 and E7 genes fragments were amplified by PCR assay. The sequences of E6 and E7 genes were used for phylogenetic analysis by Mega 5.0 software. By comparison, the major mutations of E6 gene were T178G (41.67%) and G658A (17%), as well as C58G, T61A and G188C (14%), T61C and C656A (11%). The mutation of E7 gene was mainly C491A and T935A (23%);G514C, G937C and G519C (11%). The 68 nucleotide site deficiency (69.4%) of E6 and 535 nucleotide site deficiency (23%) of E7 were dominant. In our study, we did not find the relevance between the E6 and E7 genes mutations and pathologic severity. But we think the E6 and E7 genes mutation of HPV-16 might affect vaccine prevention and treatment effect.

文摘Objective:To analyze the structural features of human P2X7R protein,meanwhile homology,protein-protein interaction,and cell epitope were predicted to provide theoretical basis for the diagnosis and treatment of related diseases.Methods:Human P2X7R protein was analyzed and predicted by ProtParam、TMHMM、ProtScale and SignalP 5.0 Server et al.Results:Human P2X7R protein is composed of 595 amino acids,the molecular formula is C 3077 H 4749 N 835 O 871 S 40,with the theoretical isoelectric point PI of 8.62,the instability coefficient is 46.07,the total average hydrophilicity is-0.317,and the protein is an hydrophilic protein instead of secreted protein,no any signal peptide,but transmembrane region is contained,which belong to transmembrane protein.There are 157αhelices and 288 random coils in the secondary structure of human P2X7R protein,accounting for 26.39%and 48.40%of all amino acids,The coverage rate of the three-level structure prediction results is 100%,and the sequence consistency is 80.34%,and the ramachandran plot show that the predicted structure is relatively stability.The interaction of proteins show that there are a variety of proteins interacting with human P2X7R,and P2X7R was closely related to inflammation;this protein had many B and T cell epitopes,which could provide ideas for vaccine development.Conclusion:The results indicate that the analysis of human P2X7R protein that is based on bioinformatics method has certain reliability.P2X7R is involved in the development of inflammatory diseases,and the antigenic epitope of B,Th and CTL cells screened out by prediction will provide the treatment scheme for diseases.
