Here, we report that AOF1 (KDM1B under the new nomenclature), a mammalian protein related to LSD1, also possesses histone demethylase activity with specificity for H3K4mel and H3K4me2. Like LSD1, the highly conserved SWIRM domain is required for its enzymatic activity. However, AOF1 differs from LSD1 in several aspects. First, AOF1 does not appear to form stable protein complexes containing histone deacetylases. Second, AOF1 is found to localize to chromosomes during the mitotic phase of the cell cycle, whereas LSD1 does not. Third, AOF1 represses transcription when tethered to DNA and this repression activity is independent of its demethylase activity. Structural and functional analyses identified its unique N-terminal Zf-CW domain as essential for the demethylase activity-independent repression function. Collectively, our study identifies AOF1 as the second histone demethylase in the family of flavin-dependent amine oxidases and reveals a demethylase-independent repression function of AOF1.展开更多

Converters rely on passive filtering as a crucial element due to the high-frequency operational characteristics of power electronics.Traditional filtering methods involve a dual inductor-capacitor(LC)cell or an induct...Converters rely on passive filtering as a crucial element due to the high-frequency operational characteristics of power electronics.Traditional filtering methods involve a dual inductor-capacitor(LC)cell or an inductor-capacitor-inductor(LCL)T-circuit.However,capacitors are susceptible to wear-out mechanisms and failure modes.Nevertheless,the necessity for monitoring and regular replacement adds to an elevated cost of ownership for such systems.The utilization of an active output power filter can be used to diminish the dimensions of the LC filter and the electrolytic dc-link capacitor,even though the inclusion of capacitors remains an indispensable part of the system.This paper introduces capacitorless solid-state power filter(SSPF)for single-phase dc-ac converters.The proposed configuration is capable of generating a sinusoidal ac voltage without relying on capacitors.The proposed filter,composed of a planar transformer and an H-bridge converter operating at high frequency,injects voltage harmonics to attain a sinusoidal output voltage.The design parameters of the planar transformer are incorporated,and the impact of magnetizing and leakage inductances on the operation of the SSPF is illustrated.Theoretical analysis,supported by simulation and experimental results,are provided for a design example for a single-phase system.The total harmonic distortion observed in the output voltage is well below the IEEE 519 standard.The system operation is experimentally tested under both steady-state and dynamic conditions.A comparison with existing technology is presented,demonstrating that the proposed topology reduces the passive components used for filtering.展开更多
The importance of reducing ship resistance is growing considerably as a result of the increase in atmospheric emissions and the drive towards green shipping through decarbonization.Until this point,Energy Saving Devic...The importance of reducing ship resistance is growing considerably as a result of the increase in atmospheric emissions and the drive towards green shipping through decarbonization.Until this point,Energy Saving Devices(ESD),in particular,Hull Vane®(HV),have been widely applied as a potential technique for reducing wave-making resistance for vessels with higher Froude Number(Fr).The advantages of HV for a medium-speed vessel,where the wave-making component accounts for almost 50%of total resistance,have yet to be investigated.This study presents the computational analysis of the KCS model(1∶75.5);for a particular trim condition by using the VOF method and RANS solver.The hull acts as a candidate vessel for the class of medium-speed characteristics.A total of 36 numerical simulations were carried out to study the changes in resistance and motion characteristics of the vessel with and without HV.To validate the numerical setup,the experimental work of Hou et al(2020)on the DTMB hull was used.The effectiveness of HV can be comprehended by the reduction percentage in total resistance,trim,sinkage,and transom wave height,in comparison to bare hull condition.The reduction in total resistance extends up to 6%for Fr=0.32 with configuration 2 with negative AoF.The CFD results indicate that there is a reduction in trim up to 57%for the maximum speed with a corresponding Fr=0.34 with a positive angle of foil(AoF).The trim correction effect is increasing with the depth of submergence of HV.Concerning sinkage,there occurs nearly a 31%reduction for Fr=0.34 with a positive AoF.There exists a substantial reduction in the height of the transom wave with the inclusion of HV,the results of which are discussed in detail.From the presented results,retrofitting the Hull Vane®is effective in the selected speed range but pronouncing as the speed of the vessel increases.展开更多
