亮菌多糖对AOM/DSS炎症相关性结肠癌小鼠模型及其肠道菌群的影响 被引量:2

目的:探讨亮菌多糖对氧化偶氮甲烷(AOM)/葡聚糖硫酸钠(DSS)诱导的炎症相关性结肠癌小鼠模型及其肠道菌群的影响。方法:将18只6~8周龄小鼠通过随机数字法分为对照组(Control)、模型组(Model),亮菌多糖干预组(ATPS60 mg/kg)。Model和ATPS... 目的:探讨亮菌多糖对氧化偶氮甲烷(AOM)/葡聚糖硫酸钠(DSS)诱导的炎症相关性结肠癌小鼠模型及其肠道菌群的影响。方法:将18只6~8周龄小鼠通过随机数字法分为对照组(Control)、模型组(Model),亮菌多糖干预组(ATPS60 mg/kg)。Model和ATPS组一次性腹腔注射AOM 10 mg/kg,对照组注射等量生理盐水,Model和ATPS组以饮用1.8%DSS水溶液5 d,普通水14 d为个一周期,进行三个循环周期。对照组全程饮用普通饮用水。Model组饮用自由水期间ATPS组小鼠每天进行灌胃处理;71天实验结束。收集小鼠结肠并观察其长度和出血情况,记录肿瘤大小和数目,并对结直肠进行HE和Ki-67染色。Elisa检测Control、Model和ATPS组arrb1、IL-10和TNF-α的表达情况。蛋白免疫印迹法(Western blot,WB)检测各组arrb1蛋白表达情况。同时,采集小鼠粪便,进行DNA提取和菌群测序。结果:AOM/DSS处理后小鼠体重明显下降,与Control组相比Model组小鼠结肠长度缩短明显,P=0.002。ATPS干预一定程度上可以抑制小鼠结肠长度的缩短,P=0.046,差别具有统计学意义。病理组织结果显示,对照组小鼠结肠隐窝形态结构正常,排列整齐,均匀分布。模型组小鼠结肠隐窝结构破坏明显,腺体结构扭曲变形,细胞核深染增大,结肠肿瘤形成。ATPS组可见结肠隐窝基本排列整齐,腺体扭曲变形有所改善,未见核大深染的异性细胞,提示ATPS存在抑制炎癌转化的药理作用。同时,Ki-67染色结果显示ATPS能够抑制结肠细胞异常增殖。Elisa和WB结果显示:Model组较Control组相比arrb1和TNF-α水平升高,IL-10水平降低。经过ATPS治疗后arrb1与TNF-α水平明显下降,IL-10水平表达升高。表明ATPS能够通过干预arrb1的表达治疗结肠炎相关性结肠癌。16S rDNA基因测序发现三组微生物区系丰富度和多样性存在显著差异。Control组、Model组和ATPS组共有的物种数量为69个,Model组特有的物种数量为11个,ATPS组特有的物种数量为6个。小鼠在经过AOM/DSS处理后普雷沃菌科_UCG-001菌属较对照组明显增多。ATPS组较Model组拟普雷沃菌属增加明显。结果发现普雷沃菌科_UCG-001是接受亮菌多糖的小鼠中最丰富的属,在小鼠炎癌转化发展过程中明显减少,表明该属的有益作用。结论:亮菌多糖能够通过调节arrb1和恢复AOM/DSS造成的Alpha多样性指数和群落构成变化,治疗慢性非可控性结肠炎,抑制慢性结肠炎炎癌转化。 展开更多

A high-performance LED-side-pumped two-rod Nd,Ce:YAG laser with continuous-wave(CW) and acousto–optical(A-O) Q-switched operation is demonstrated in this work. A symmetrically shaped flat–flat cavity with two identical LEDside-pumped laser modules is employed for power scalability. In the CW regime, the maximum output average power of laser at 1064 nm is 4.41 W, corresponding to a maximum optical conversion efficiency of 5.3% and a slope efficiency is 12.4%. In the active Q-switched regime, the pulse energy of laser reaches as high as 0.89 m J at a repetition rate of 800 Hz with a pulse width of 457.2 ns, the corresponding highest peak output power is 1.94 k W and the M~2 factor is measured to be about 8.8. To the best of the authors' knowledge, this is the first demonstration and the highest performance of a CW LED-side-pumped two-rod laser Nd,Ce:YAG with Watt-level output reported so far. 展开更多

目的:探究薏苡附子败酱散(Yiyi Fuzi Baijiang San,YYFZBJS)联合电针(electroacupuncture,EA)对AOM/DSS模型小鼠炎症相关性结直肠癌(colitis-associated colorectal cancer,CAC)的防治作用及对结直肠癌上皮间质转化(epithelial-mesenchy... 目的:探究薏苡附子败酱散(Yiyi Fuzi Baijiang San,YYFZBJS)联合电针(electroacupuncture,EA)对AOM/DSS模型小鼠炎症相关性结直肠癌(colitis-associated colorectal cancer,CAC)的防治作用及对结直肠癌上皮间质转化(epithelial-mesenchymal transition,EMT)相关蛋白表达情况的影响。方法:建立AOM/DSS诱导的CAC小鼠模型,将40只小鼠随机分为模型组、针药结合组、薏苡附子败酱散组、电针组和阿司匹林组,于实验第2周开始干预治疗,治疗过程中记录各组小鼠一般情况,统计体质量变化、疾病活动指数(disease activity index,DAI)评分及生存情况,于实验末比较各组小鼠结直肠长度、成瘤数量及肿瘤大小,通过HE染色观察结直肠组织及肿瘤组织的病理变化,免疫组织化学染色观察结直肠肿瘤Ki-67、PCNA、p-NF-κB、NF-κB、E-cadherin、ZO-1、Occludin、Vimentin和N-cadherin蛋白的表达。结果:与模型组相比,针药结合组小鼠体质量显著升高(P<0.05),DAI分值降低(P<0.05),生存率为87.5%,结直肠长度显著增加(P<0.01),直径小于2 mm的肿瘤数量减少(P<0.05),直径介于2~4 mm之间和大于4 mm的肿瘤数量显著减少(P<0.01),总成瘤数量明显下降(P<0.01),结直肠形态明显改善;肿瘤组织病理评分降低(P<0.01),肿瘤组织中Ki-67、PCNA、p-NF-κB、NF-κB、Vimentin和N-cadherin蛋白表达量降低(P<0.01),E-cadherin、ZO-1和Occludin蛋白表达量升高(P<0.05)。结论:薏苡附子败酱散联合电针对AOM/DSS模型小鼠的CAC发生发展有防治作用,其机制可能与下调NF-κB信号通路,减缓EMT进程有关。 展开更多

We demonstrate antimonene as a saturable absorber(SA) to generate an ultrafast mode-locked and Q-switched laser in the 2 μm wavelength region. The two antimonene-based SAs were prepared and inserted separately in a thulium–holmiumdoped fiber laser to produce the pulsed laser. Antimonene was coated onto a tapered fiber to generate soliton mode-locked pulses and used in thin-film form for the generation of Q-switched pulses. The mode-locking was stable within a pump power of 267 m W–511 m W, and the laser operated at a central wavelength of 1897.4 nm. The mode-locked laser had a pulse width of 1.3 ps and a repetition rate of 12.6 MHz, with a signal-to-noise ratio of 64 d B. Q-switched laser operation was stable at a wavelength of 1890.1 nm within a pump power of 312 m W–381 m W. With the increase in pump power from 312 m W to 381 m W, the repetition rate increased to a maximum of 56.63 k Hz and the pulse width decreased to a minimum value of 2.85 μs. Wide-range tunability of the Q-switched laser was also realized within the wavelength range of1882 nm–1936 nm. 展开更多

The results show that t... We demonstrate a high-energy and high-power pulse laser on a xenon lamp-pumped Er:YAP crystal. The laser performance and thermal focal lengths under different working frequencies are discussed. The results show that the thermal lens effect is gradually aggravated with the increase of working frequencies, and even working at 100 Hz, a single pulse energy of 234 m J can be achieved. A maximum average power of 41.5 W is achieved with a working frequency of 20 Hz and slope efficiency of 2.82%. This output power is much higher than other xenon lamp-pumped erbium laser devices.A Q-switched laser is demonstrated by using the TeO2crystal, the maximum output energies of 11.5 m J and 3.5 m J are obtained at 50 and 100 Hz, the corresponding peak powers are 93.4 k W and 17.2 kW, respectively.The laser wavelengths and beam quality factors are also characterized in the free-running and Q-switched modes. A higher pulse energy and peak power laser could be achieved further by improving the damage threshold of TeO2acousto-optical Q-switching. All the experimental results illustrate that the xenon lamp-pumped Er:YAP laser is a promising candidate for high-power and high-frequency mid-infrared laser devices. 展开更多

We report a high repetition frequency, high power stability and low laser noise laser-diode(LD) end-pumped Nd: YAG ceramic passively Q-switched laser at 1123 nm based on a Ti_(3)C_(2)T_(x)-polyvinyl alcohol(PVA) film as a saturable absorber(SA). A Brewster polarizer(BP) and a birefringent crystal(BC) are incorporated to enable frequency selection and filtering for the passively Q-switched 1123 nm pulsed laser to improve the power stability and reduce the noise. When the pump power is 5.1 W, an average output power of 457.9 m W is obtained, corresponding to a repetition frequency of 1.09 MHz,a pulse width of 56 ns, a spectral line width of 0.65 nm, a power instability of ±0.92%, and a laser noise of 0.89%.The successful implementation of the “Ti_(3)C_(2)T_(x)-PVA film passively Q-switching” combined with “frequency selection and filtering of BP + BC” technology path provides a valuable reference for developing pulsed laser with high repetition frequency, high stability and low noise. 展开更多

驱动甲烷厌氧氧化(AOM)过程的微生物不仅能同化吸收甲烷等碳氢化合物作为碳源,同时也能进行氮素的同化吸收,从而导致这些微生物的生物体出现显著的^(13)C、^(15)N亏损。目前还缺乏来自冷泉环境有机氮δ^(15)N数据来直接证实AOM参与氮的... 驱动甲烷厌氧氧化(AOM)过程的微生物不仅能同化吸收甲烷等碳氢化合物作为碳源,同时也能进行氮素的同化吸收,从而导致这些微生物的生物体出现显著的^(13)C、^(15)N亏损。目前还缺乏来自冷泉环境有机氮δ^(15)N数据来直接证实AOM参与氮的循环过程。本研究分析了来自墨西哥湾海底5个冷泉站位的自生碳酸盐岩的总氮(TN)、总无机氮(TIN)和总有机氮(TON)的含量及其δ^(15)N值,并结合前人获得的总无机碳和总有机碳含量和δ^(13)C数据进行研究,结果发现总氮的δ^(15)N与总有机碳的δ^(13)C只在部分站位表现出正相关性。然而,在排除无机氮的干扰后,总有机氮的δ^(15)N与总有机碳的δ^(13)C在所有站位均表现出正相关性。表明这种有机碳、氮同位素上的耦合与甲烷的活动密切相关,有望作为有效示踪过去海底甲烷渗漏及AOM作用的指标。 展开更多

原子磁力仪的基本工作原理是通过光泵浦将原子极化,并通过检测自旋极化的拉莫尔进动来完成磁场探测,其中用来极化原子的泵浦激光起到了重要作用。自由运转的激光器由于受环境的影响,其频率随时间抖动,从而导致原子磁力仪输出的波动。本... 原子磁力仪的基本工作原理是通过光泵浦将原子极化,并通过检测自旋极化的拉莫尔进动来完成磁场探测,其中用来极化原子的泵浦激光起到了重要作用。自由运转的激光器由于受环境的影响,其频率随时间抖动,从而导致原子磁力仪输出的波动。本文首先在理论上推导了Bell-Bloom磁力仪输出与泵浦光频率波动的关系,其次采用基于双AOM移频的差分探测法实现系统稳频,并自行设计了自动锁频算法且对其进行了实验验证,结果表明可使系统稳定在工作零点。稳频前频率的抖动均方根约为28.02 MHz,通过双AOM稳频模块后激光频率抖动均方根减小到0.12 MHz。通过激光频率的变化反推到铯原子磁力仪中,测得闭环的等效输出噪声为0.012 pT。 展开更多

目的观察硒对致癌剂氧化偶氮甲烷(azoxymethane,AOM)所致结肠癌模型大鼠肾上腺皮质束状带细胞组织化学的变化。方法随机将20只3周龄SPF级断乳雄性Sprague Dawley(SD)大鼠随机分4组:正常对照组、实验对照组、致癌剂前补硒组和致癌剂后补... 目的观察硒对致癌剂氧化偶氮甲烷(azoxymethane,AOM)所致结肠癌模型大鼠肾上腺皮质束状带细胞组织化学的变化。方法随机将20只3周龄SPF级断乳雄性Sprague Dawley(SD)大鼠随机分4组:正常对照组、实验对照组、致癌剂前补硒组和致癌剂后补硒组。用AOM(15mg/kg)每周腹腔注射,连续2周,诱导大鼠结肠癌的形成。亚硒酸钠(Na2SeO3)水溶液(4mg/L)分别在AOM前、后干预,并持续至实验结束。各组均于34周取大鼠肾上腺,用组织化学的方法,观察大鼠肾上腺皮质束状带细胞组织化学的变化,对脂类、琥珀酸脱氢酶(SDH)和3β-羟类固醇脱氢酶(3β-HSD)进行染色,并图像分析。结果亚甲基蓝染色光镜下观察,可见AOM腹腔注射的大鼠结肠黏膜出现异常隐窝(aberrant crypt,AC)和异常隐窝灶(aberrant crypt foci,ACF)。组织化学显示,与正常对照组相比,实验对照组大鼠肾上腺皮质束状带细胞的SDH和3β-HSD的MOD值明显增加(P<0.05),脂类的MOD值明显减少(P<0.05);硒干预的各组与实验对照组相比,大鼠肾上腺皮质束状带细胞的SDH和3β-HSD的MOD值均有增加,脂类的MOD值减少,其中致癌剂后补硒组SDH和3β-HSD的MOD值增加有统计意义(P<0.05)。结论硒可增强AOM所致大鼠结肠癌形成过程中肾上腺皮质束状带细胞的功能。 展开更多

The study of nonlinear optical responses in the mid-infrared(mid-IR)regime is essential for advancing ultrafast mid-IR laser applications.However,nonlinear optical effects under mid-IR excitation are rarely reported due to the lack of suitable nonlinear optical materials.The natural van derWaals heterostructure franckeite,known for its narrow bandgap and stability in air,shows great potential for developing mid-IR nonlinear optical devices.We have experimentally demonstrated that layered franckeite exhibits a broadband wavelength-dependent nonlinear optical response in the mid-IR spectral region.Franckeite nanosheets were prepared using a liquid-phase exfoliation method,and their nonlinear optical response was characterized in the spectral range of 3000 nm to 5000 nm.The franckeite nanosheets exhibit broadband wavelengthdependent third-order nonlinearities,with nonlinear absorption and refraction coefficients estimated to be about 10^(-7)cm/W and 10^(-11)cm^(2)/W,respectively.Additionally,a passively Q-switched fluoride fiber laser operating around a wavelength of 2800 nm was achieved,delivering nanosecond pulses with a signal-to-noise ratio of 43.6 dB,based on the nonlinear response of franckeite.These findings indicate that layered franckeite possesses broadband nonlinear optical characteristics in the mid-IR region,potentially enabling new possibilities for mid-IR photonic devices. 展开更多

Research on novel ultrafast photonic devices with wide adaptability has become important scientific technical means to realize both scheme innovation and performance breakthrough in fiber laser generation.As types of transition metal oxide,manganese dioxide(MnO_(2))materials exhibit remarkable properties including high photothermal stability,strong oxidation resistance,and excellent optical properties,making them promising candidate for utilization as modulation devices in nonlinear optics and ultrafast optics fields.We investigate the impact of MnO_(2)-based saturable absorber(SA)on the pulse characteristics.The experiment reveals that MnO_(2)-based SA supports effectively pulsed laser generation in wide pump power range and large dispersion parameter space with signal-to-noise ratio more than 85 dB.As far as we know,the pump power response range is outstanding among the most of the reported pulsed lasers,which is attributed to the large modulation depth of MnO_(2) SA.We also investigate the impact of dispersion on the characteristics of laser output,which is not involved in other similar works.This research indicates that MnO_(2) as a photonic device has vast potential in advanced ultrafast optics. 展开更多

Pyrite is one of the common authigenic minerals in marine sediments.Previous studies have shown that the morphological and isotopic characteristics of pyrite are closely related to the geochemical environment where it is formed.To better understand the for-mation mechanism of authigenic pyrite,we analyzed the isotopic composition,morphology,and distribution of pyrite in the sediment at 500m below the seafloor from Xisha Trough,South China Sea.Mineral morphologies were observed by scanning electron micros-copy and Raman spectrography.X-Ray computed tomography was applied to measure the particle size of pyrite.The size of pyrite crystals in the matrix sediment mainly ranged between 25 and 65µm(av.ca.40µm),although crystals were larger(av.ca.50μm)in the veins.The pyrites had a fine-grained truncated octahedral shape with occasionally well-developed growth steps,which implies the low growth rate and weak anaerobic oxidation of methane-sulfate reduction when pyrite was formed.Theδ^(34)S values of pyrites ranged from+20.8‰Vienna-defined Canyon Diablo Troilite(V-CDT)to+33.2‰V-CDT and from+44.8‰V-CDT to+48.9‰,which suggest two growth stages.In the first stage,with the continuous low methane flux,the pyrite possibly formed in an environment with good access to seawater.In the second stage,the pyrites mainly developed in sediment fractures and appeared in veins,probably due to the limited availability of sulfate.The less exposure of pyrite to the environment in the second stage was probably caused by sediment accumulation or perturbation.In this study,an episodic pyritization process was identified,and the paleoenvironment was reconstructed for the sediment investigated. 展开更多

Introduction: Acute otitis media is an acute inflammation of the mucosa of the middle ear cavities. It is often secondary to nasopharyngitis, which favors the passage of infection through the Eustachian tube to the middle ear. The aim of our study was to improve the management of AOM in the Paediatric Department of the Hospital National Ignace Deen (Conakry). Patients and Methods: This was a prospective descriptive study lasting 6 months from 01 July to 31 December 2011;the study covered 525 cases out of a total of 6276 children, i.e. a frequency of 8.36%. Results: The most affected age group was 6 to 11 months. Males predominated (69.71%). 82.29% had a history of recurrent rhinopharyngitis. The most frequent reason for consultation was incessant crying (66.29%). Rhinopharyngitis and malaria were the most commonly associated pathologies (87.62% and 39.62% respectively). 72.19% of our patients were admitted with congestive AOM and received medical treatment. We recorded one case of otomastoiditis which was treated surgically. Conclusion: AOM is more common in children aged between 6 and 24 months. Good collaboration between paediatricians and ENT specialists is essential to reduce the morbidity of AOM. 展开更多
