We experimentally realize the dual-wavelength bad cavity laser for the first time. As the Cs cell temperature is kept between 118℃ and 144℃, both the 1359nm and 147Ohm lasing outputs of dual-wavelength bad cavity laser are detected. The laser output power of dual-wavelength bad cavity laser is measured when changing the 455 nm pumping laser frequency and power at 127℃ Cs cell temperature. Both the 1359 nm laser and the 1470 nm laser are working at the deep bad cavity regime, and the ratio between the linewidth of cavity mode and the laser gain bandwidth a ≈ 40 for 1359nm and 1470nm lasers. The 147Ohm laser linewidth is measured to be 407.3Hz. The dual-wavelength bad cavity laser operating on atomic transitions demonstrated here has a potential in the application as a stable optical local oscillator, even an active optical frequency standard directly in the future. 展开更多

BACKGROUND: Phycocyanin can relieve decrease of mitochondrial membrane potential through reducing production of active oxygen so as to protect neurons after hypoxia/reoxygenation. OBJECTIVE: To observe the effect of phycocyanin on activity of PC12 cells and mitochondrial membrane potential after hypoxia/reoxygenation. DESIGN: Randomized controlled study SETTING : Cerebrovascular Disease Institute of Affiliated Hospital, Medical College of Qingdao University MATERIALS: The experiment was carried out at the Key Laboratory of Prevention and Cure for cerebropathia in Shandong Province from October to December 2005. PC12 cells, rat chromaffin tumor cells, were provided by Storage Center of Wuhan University; phycocyanin was provided by Ocean Institute of Academia Sinica; Thiazoyl blue tetrazolium bromide (MTT) and rhodamine 123 were purchased from Sigma Company, USA; RPMI-1640 medium, fetal bovine serum and equine serum were purchased from Gibco Company, USA. METHODS: ① Culture of PC12 cells: PC12 cells were put into RPMI-1640 medium which contained 100 g/L heat inactivation equine serum and 0.05 volume fraction of fetal bovine serum and incubated in CO2 incubator at 37℃. Number of cells was regulated to 4 × 10^5 L 1, and cells were inoculated at 96-well culture plate. The final volume was 100μL. ② Model establishing and grouping: Cultured PC12 cells were randomly divided into three groups: phycocyanin group, model control group and non-hypoxia group. At 24 hours before hypoxia, culture solution in phycocyanin group was added with phycocyanin so as to make sure the final concentration of 3 g/L , but cells in model control group did not add with phycocyanin. Cells in non-hypoxia group were also randomly divided into adding phycocyanin group (the final concentration of 3 g/L) and non-adding phycocyanin group. Cells in model control group and phycocyanin group were cultured with hypoxia for 1 hour and reoxygenation for 1, 2 and 3 hours; meanwhile, cells in non-hypoxia group were cultured with oxygen and were measured at 1 hour after hypoxia/reoxygenation. ③ Detecting items: At 1, 2 and 3 hours after reoxygenation, absorbance (A value) of PC12 cells was measured with MTT technique so as to observe activity and quantity of cells. Fluorescence intensity of PC12 cells marked by rhodamine 123 was measured with confocal microscope in order to observe changes of mitochondrial membrane potential. MAEN OUTCOME MEASURES: Comparisons between quantity and activity of PC12 cells and mitochondria membrane potential at 1, 2 and 3 hours after reoxygenation. RESULTS: ① Effect of phycocyanin on quantity and activity of PC12 cells: A value was 0.924±0.027 in adding phycocyanin group and 0.924±0.033 in non-adding phycocyanin group. A value was lower in model control group and phycocyanin group than that in non-hypoxia group at 1, 2 and 3 hours after reoxygenation (0.817±0.053, 0.838±0.037, 0.875±0.029; 0.842±0.029, 0.872±0.025, 0.906±0.023, P 〈 0.05). A value was higher in phycocyanin group than that in model control group at 1, 2 and 3 after culture (P 〈 0.05). With culture time being longer, A value was increased gradually in phycocyanin group and model control group after reoxygenation (P 〈 0.05). ~ Effect of phycocyanin on mitochondrial membrane potential of PC12 cells: Fluorescence intensity was 2.967±0.253 in adding phycocyanin group and 2.962±0.294 in non-adding phycocyanin group. Fluorescence intensity was lower in model control group and phycocyanin group than that in non-hypoxia group at 1, 2 and 3 hours after hypoxia/reoxygenation (1.899±0.397, 2.119±0.414, 2.287±0.402; 2.191±0.377, 2.264±0.359, 2.436±0.471, P 〈 0.05); but it was higher in phycocyanin group than that in model control group at 1, 2 and 3 after reoxygenation (P 〈 0.05). With culture time being longer, fluorescence intensity was increased gradually in phycocyanin group and model control group after reoxygenation (P 〈 0.05). CONCLUSION: Phycocyanin and reoxygenation can protect PC12 cells after hypoxia injury through increasing mitochondrial membrane potential and cellular activity, and the effect is improved gradually with prolonging time of reoxygenation. 展开更多

Correlation of cognitive function with acetylcholinesterase activity and P300 event-related potential of patients with type 2 diabetes mellitus

Changes in structure of cholinergic neuron system of brain and receptor expression after brain injury can cause cognitive impairment. OBJECTIVE" To comparatively observe the intelligence quotient (IQ), latent period and wave amplitude of P300 event-related potential and the difference of activity of acetylcholinesterase (ACHE) in blood and cerebrospinal fluid between patients with type 2 diabetes mellitus and with non-diabetes mellitus, and analyze the correlation of IQ of cognitive impairment patients with diabetes mellitus with AChE activity, latent period and wave amplitude of P300 event-related potential in cerebrospinal fluid. DESIGN: Correlation analysis of contrast observation SETTING: Department of Endocrinology, Affiliated Hospital of Binzhou Medical College PARTICIPANTS: Totally 32 patients with type 2 diabetes mellitus who received the treatment in the Department of Endocrinology, Affiliated Hospital of Binzhou Medical College between April 2004 and April 2005 were recruited, serving as diabetes mellitus group. They, including 19 male and 13 female, aged 49 to 73 years, with disease course of 4 to 11 years, all met the diagnostic criteria of diabetes mellitus revised by World Health Organization in 1999. Another 30 patients with non-diabetes mellitus who homeochronously underwent lumbar anesthesia in the Department of Surgery and Department of Gynecology were recruited, serving as non-diabetes mellitus group. The 30 patients included 18 male and 12 female, and their age ranged from 46 to 71 years. Informed consents of detected items were obtained from the involved patients. METHODS: ① Evaluation,on IQ: The IQ of involved subjects was evaluated with Chinese Version of the Wechsler Adult Intelligence Scale revised by Gong Yao-xian (WAIS-RC). WAIS-RC included 6 verbal subscales and 5 performance subscales. The test scores of the 11 subscales integrated into the scores of the whole scale, and the scores on the WAIS-RC included verbal IQ (VlQ), performance IQ (PIQ) and full scale IQ (FIQ). FIQ ≤79 scores indicated low IQ and FIQ≤69 indicated intelligence impairment. ② Detection of P300 wave: P300 wave was detected with evoked potential instrument (MYTOPRO, Italian), and data of latent period and amplitude of P300 event-related potential were automatically shown by computer. ③ Detection of AChE activity in blood and cerebrospinal fluid: Activity of AChE of blood and cerebrospinal fluid was measured with biochemical methods by using CORNING-560 autoanalyzer.④Correlation analysis: Correlation of FIQ with AChE of cerebrospinal fluid and P300 wave of patients with type 2 diabetes mellitus was analyzed, t test was used in intergroup comparison and linear correlation analysis for relevant treatment. MAIN OUTCOME MEASURES: ① Comparison of IQ, latent period and wave amplitude of P300 wave as well as the activity of AChE between two groups. ② Analysis on the correlation of FIQ of patients with type 2 diabetes mellitus with AChE of cerebrospinal fluid and P300 wave. RESULTS: Thirty-two patients with diabetes mellitus and 30 non-diabetes mellitus participated in the result analysis. ①Comparison of IQ, latent period and wave amplitude of P300 wave as well as the activity of AChE between two groups: The scores of VIP, PIQ and FIQ of patients with type 2 diabetes mellitus were (97.4±10.4). (92.6±8.4) and (95.2±9.7) scores, respectively; and those of patients with non-diabetes mellitus were (104.7±9.6), (102.5±8.5)and(102.7±8.9) scores, respectively, and P 〈 0.05-0.01 was set in intergroup comparison. The latent period of P300 wave at points Fz , Cz and Pz of patients with type 2 diabetes mellitus was (370.8±41.8).(371.5±39.1)and (375.1±43.1) ms, respectively, and that of patients with non-diabetes mellitus was ( 332.1 ±28.3 ), (335.7 ±29.4)and (339.7 ±27.3) ms, respectively, and P 〈 0.01 was set in intergroup comparison; Wave amplitude of P300 of patients with type 2 diabetes mellitus was (8.6±4.1),(8.6±4.0) and (7.7±4.0) μV, respectively and that of patients with non-diabetes mellitus was (11.9±4.1),(11.5±4.4) and (10.9±5.0) μV, respectively , and P 〈 0.05-0.01 was set in intergroup comparison; The level of AChE in blood and cerebrospinal fluid of patients with type 2 diabetes mellitus was (235.61 ±50.34)and (17.89±4.46) μkat/L, respectively, which was significantly higher than that of patients with non-diabetes mellitus [(205.03±44.15)and (14.63±0.48) μkat /L, respectively], and P 〈 0.05-0.01 was set in the intergroup comparison. ② Correlation of FIQ value of patients with type 2 diabetes mellitus with AChE of cerebrospinal fluid and P300 wave: The value of FIQ was significantly negatively correlated with the AChE activity of cerebrospinal fluid (r=-0.588 1, P 〈 0.01 ), significantly negatively correlated with the latent period at points Fz. C and Pz of P300 wave (r= -0.700 5, -0.689 4, -0.688 5, P 〈 0.01 ), and significantly positively correlated with the amplitude at points Fz . Cz and Pz of P300 wave(r= 0.607 4,0.616 1,0.592 0,P 〈 0.01 ). CONCLUSION: ① Cognitive impairment of patients with type 2 diabetes mellitus might be related to the increase of activity of AChE in cerebrospinal fluid. ②Combined application of examination of P300 wave and evaluation of IQ is more useful in deciding the state of cognitive function of patients with type 2 diabetes mellitus. 展开更多

Utilization of Lithium Slag as An Admixture in Blended Cements: Physico-mechanical and Hydration Characteristics 被引量:12

Physical and mechanical properties variations of lithium slag were systematically investigated by three different ways such as physical, chemical activation, physical-chemical combined activation. Mechanisms of the cementitious properties and hydration process of lithium slag composite cement were studied by XRD and SEM. The results showed that specific surface area increased from 254 to 700 m2/kg while median particle size decreased from 14.97 to 8.45 urn with the increase of grinding time. Physical, chemical activation and combined activation improved the strength and hydration degree of lithium slag composite cement. Compared with original lithium slag, the flexural strength and compressive strength of mortars were improved significantly with the increase of grinding time. A higher strength of the cement with the lithium slag was attained; The sample with 10% lithium slag got the highest strength when the grinding time was 10 min; the compressive strength was higher than OPC at 28 days, which increased by 12.3%. When the Na2SO4 content was 0.6%, the compressive strength increased by 1.4%; when the Al2(SO4)3·18H2O content was 0.4%, the compressive strength increased by 5.8% at 28 days. Compared with the late strength, the improving degree of early strength was larger with the incorporation of activator. The results of XRD and SEM were consistent with the results of mechanical properties; it is also evident that lithium slag composite cement hydration products were mainly AFt, Ca(OH)2, CaSO4·2H2O, and C-S-H gel. 展开更多

A highly charged manned spacecraft threatens the life of an astronaut and extravehicular activity, which can be effectively reduced by controlling the spacecraft surface charging. In this article, the controlling of surface charging on Chinese Space Station (CSS) is investigated, and a method to reduce the negative potential to the CSS is the emission electron with a hollow cathode plasma eontactor. The analysis is obtained that the high voltage (HV) solar array of the CSS collecting electron current can reach 4.5 A, which can be eliminated by emitting an adequate electron current on the CSS. The theoretical analysis and experimental results are addressed, when the minimum xenon flow rate of the hollow cathode is 4.0 sccm, the emission electron current can neutralize the collected electron current, which ensures that the potential of the CSS can be controlled in a range of less than 21 V, satisfied with safety voltage. The results can provide a significant reference value to define a flow rate to the potential controlling programme for CSS. 展开更多

The absolute configuration was determined by single-crystal X-ray diffraction of the key intermediate(2S,3S,E)-ethyl 4-(3,4-dimethoxybenzylidene)-3-(3,4-dimethoxyphenyl)-5-oxotetrahydrofuran-2-carboxylate. The crystal belongs to tetragonal system, space group P41 with a = 13.0665(2), b = 13.0665(2), c = 13.4735(2)A, V = 2300.4(6) A^3, Z = 4, Dc = 1.278 g/cm^3, μ(CuKα) = 0.801 mm^-1, F(000) = 936, S = 1.050, R = 0.0364 and wR(I 〉 2σ(I)) = 0.1071. X-ray diffraction results showed that the molecular structure is stabilized totally by Van der Waals force. The antibacterial activity tests showed that the title compound possesses slight synergistic effect against MRSA and Acinetobacter baumanii. 展开更多

Archaea has been shown to be the dominant group in trench sediments. However, the activity potentials and detailed diversity of these communities as well as their inter-trench variations are still not known. In this study, we combined datasets from two pairs of primers to investigate at high resolution the structure and activity potentials of the archaeal communities in vertically sectioned sediment cores taken from the deepest points of the Mariana (10,853 m) and Mussau (7011 m) trenches. The compositions of the potentially active communities revealed, via 16S ribosomal RNA gene (rDNA) and RNA (rRNA), significant differences between samples. Marine Group I (MGI), with nine identified subgroups, was the most dominant class in the active archaeal communities of the two trenches. Significantly different species composition and vertical variations were observed between the two trenches. Vertical transitions from aerobic MGI α to anaerobic MGI η and υ subgroups were observed in MST but not in MT sediments, which might be related to the faster microbial oxygen consumption in MST. These results provide a better understanding on archaeal activity and diversity in trench sediments. 展开更多

Acidobacteria is one of the most dominant and abundant phyla in soil,and was believed to have a wide range of metabolic and genetic functions. Relatively little is known about its community structure and elevational diversity patterns. We selected four elevation gradients from 1000 to 2800 m with typical vegetation types of the northern slope of Shennongjia Mountain in central China. The vegetation types were evergreen broadleaved forest,deciduous broadleaved forest,coniferous forest and sub-alpine shrubs. We analyzed the soil acidobacterial community composition,elevational patterns and the relationship between Acidobacteria subdivisions and soil enzyme activities by using the 16 S rRNA meta-sequencing technique and multivariate statistical analysis. The result found that 19 known subdivisions as well as an unclassified phylotype were presented in these forest sites,and Subdivision 6 has the highest number of detectable operational taxonomic units(OTUs). A significant single peak distribution pattern(P 〈 0.05) between the OTU number and the elevation was observed. The Jaccard and Bray–Curtis index analysis showed that the soil Acidobacteria compositional similarity significantly decreased(P 〈 0.01) with the increase in elevation distance. Mantel test analysis showed the most of the soil Acidobacteria subdivisions had the significant relationship(P 〈 0.01) with different soil enzymes. Therefore,soil Acidobacteria may be involved in different ecosystem functions in global elemental cycles. Partial Mantel tests and CCA analysis showed that soil pH,soil temperature and plant diversity may be the key factors in shaping the soil Acidobacterial community structure. 展开更多

Soil microorganisms play a key role in soil organic matter dynamics, nutrient cycling, and soil fertility maintenance in forest ecosystems, and they are influenced by stand age and soil depth. However, few studies have simultaneously considered these two factors. In this study, we measured soil microbial biomass carbon (SMBC), soil microbial biomass nitrogen (SMBN), soil basal respiration (SBR) rate, and potential extracellular enzyme activity (EEA) in soil to a depth of 60 cm under 10-, 30-, and 40-year-old Scots pine (Pinus sylvestris var. mongolica) stands (Y10, Y30, and Y40, respectively) in plantations in northern China in 2011. Soil water content (SWC), soil pH, soil organic carbon (SOC), and soil total nitrogen (STN) were also measured to explore their effects on soil microbial indices across different stand ages and soil depths. Our results showed that SMBC, SMBN, and the SBR rate were generally higher for the Y30 stand than for the Y10 and Y40 stands. Potential EEA, except forα-glucosidase, decreased significantly with increasing stand age. Soil organic carbon,STN, SWC, and soil pH explained 67%of the variation in soil microbial attributes among the three stand ages. For the same stand age, soil microbial biomass and the SBR rate decreased with soil depth. Lower microbial biomass, lower SBR rate, and lower EEA for the mature Y40 stand indicate lower substrate availability for soil microorganisms, lower soil quality, and lower microbial adaptability to the environment.
