Neuronal regulated cell death in aging-related neurodegenerative diseases:key pathways and therapeutic potentials

Regulated cell death(such as apoptosis,necroptosis,pyroptosis,autophagy,cuproptosis,ferroptosis,disulfidptosis)involves complex signaling pathways and molecular effectors,and has been proven to be an important regulat... Regulated cell death(such as apoptosis,necroptosis,pyroptosis,autophagy,cuproptosis,ferroptosis,disulfidptosis)involves complex signaling pathways and molecular effectors,and has been proven to be an important regulatory mechanism for regulating neuronal aging and death.However,excessive activation of regulated cell death may lead to the progression of aging-related diseases.This review summarizes recent advances in the understanding of seven forms of regulated cell death in age-related diseases.Notably,the newly identified ferroptosis and cuproptosis have been implicated in the risk of cognitive impairment and neurodegenerative diseases.These forms of cell death exacerbate disease progression by promoting inflammation,oxidative stress,and pathological protein aggregation.The review also provides an overview of key signaling pathways and crosstalk mechanisms among these regulated cell death forms,with a focus on ferroptosis,cuproptosis,and disulfidptosis.For instance,FDX1 directly induces cuproptosis by regulating copper ion valency and dihydrolipoamide S-acetyltransferase aggregation,while copper mediates glutathione peroxidase 4 degradation,enhancing ferroptosis sensitivity.Additionally,inhibiting the Xc-transport system to prevent ferroptosis can increase disulfide formation and shift the NADP^(+)/NADPH ratio,transitioning ferroptosis to disulfidptosis.These insights help to uncover the potential connections among these novel regulated cell death forms and differentiate them from traditional regulated cell death mechanisms.In conclusion,identifying key targets and their crosstalk points among various regulated cell death pathways may aid in developing specific biomarkers to reverse the aging clock and treat age-related neurodegenerative conditions. 展开更多

The complex morphological,anatomical,physiological,and chemical mechanisms within the aging brain have been the hot topic of research for centuries.The aging process alters the brain structure that affects functions and cognitions,but the worsening of such processes contributes to the pathogenesis of neurodegenerative disorders,such as Alzheimer's disease.Beyond these observable,mild morphological shifts,significant functional modifications in neurotransmission and neuronal activity critically influence the aging brain.Understanding these changes is important for maintaining cognitive health,especially given the increasing prevalence of age-related conditions that affect cognition.This review aims to explore the age-induced changes in brain plasticity and molecular processes,differentiating normal aging from the pathogenesis of Alzheimer's disease,thereby providing insights into predicting the risk of dementia,particularly Alzheimer's disease. 展开更多

With the rapidly aging human population,age-related cognitive decline and dementia are becoming increasingly prevalent worldwide.Aging is considered the main risk factor for cognitive decline and acts through alterations in the composition of the gut microbiota,microbial metabolites,and the functions of astrocytes.The microbiota–gut–brain axis has been the focus of multiple studies and is closely associated with cognitive function.This article provides a comprehensive review of the specific changes that occur in the composition of the gut microbiota and microbial metabolites in older individuals and discusses how the aging of astrocytes and reactive astrocytosis are closely related to age-related cognitive decline and neurodegenerative diseases.This article also summarizes the gut microbiota components that affect astrocyte function,mainly through the vagus nerve,immune responses,circadian rhythms,and microbial metabolites.Finally,this article summarizes the mechanism by which the gut microbiota–astrocyte axis plays a role in Alzheimer’s and Parkinson’s diseases.Our findings have revealed the critical role of the microbiota–astrocyte axis in age-related cognitive decline,aiding in a deeper understanding of potential gut microbiome-based adjuvant therapy strategies for this condition. 展开更多

Age-related driving mechanisms of retinal diseases and neuroprotection by transcription factor EB-targeted therapy

Retinal aging has been recognized as a significant risk factor for various retinal disorders,including diabetic retinopathy,age-related macular degeneration,and glaucoma,following a growing understanding of the molecular underpinnings of their development.This comprehensive review explores the mechanisms of retinal aging and investigates potential neuroprotective approaches,focusing on the activation of transcription factor EB.Recent meta-analyses have demonstrated promising outcomes of transcription factor EB-targeted strategies,such as exercise,calorie restriction,rapamycin,and metformin,in patients and animal models of these common retinal diseases.The review critically assesses the role of transcription factor EB in retinal biology during aging,its neuroprotective effects,and its therapeutic potential for retinal disorders.The impact of transcription factor EB on retinal aging is cell-specific,influencing metabolic reprogramming and energy homeostasis in retinal neurons through the regulation of mitochondrial quality control and nutrient-sensing pathways.In vascular endothelial cells,transcription factor EB controls important processes,including endothelial cell proliferation,endothelial tube formation,and nitric oxide levels,thereby influencing the inner blood-retinal barrier,angiogenesis,and retinal microvasculature.Additionally,transcription factor EB affects vascular smooth muscle cells,inhibiting vascular calcification and atherogenesis.In retinal pigment epithelial cells,transcription factor EB modulates functions such as autophagy,lysosomal dynamics,and clearance of the aging pigment lipofuscin,thereby promoting photoreceptor survival and regulating vascular endothelial growth factor A expression involved in neovascularization.These cell-specific functions of transcription factor EB significantly impact retinal aging mechanisms encompassing proteostasis,neuronal synapse plasticity,energy metabolism,microvasculature,and inflammation,ultimately offering protection against retinal aging and diseases.The review emphasizes transcription factor EB as a potential therapeutic target for retinal diseases.Therefore,it is imperative to obtain well-controlled direct experimental evidence to confirm the efficacy of transcription factor EB modulation in retinal diseases while minimizing its risk of adverse effects. 展开更多

目的 探讨T淋巴细胞核仁形成区嗜银蛋白 (Ag NORs)在恶性肿瘤诊断及疗效观察中的临床价值。方法 制作银染T淋巴细胞涂片 ,显微镜下选取转化良好的T淋巴细胞 ,利用KL型肿瘤免疫图像分析系统检测其Ag NORs含量 (Ag NORs面积与细胞核面... 目的 探讨T淋巴细胞核仁形成区嗜银蛋白 (Ag NORs)在恶性肿瘤诊断及疗效观察中的临床价值。方法 制作银染T淋巴细胞涂片 ,显微镜下选取转化良好的T淋巴细胞 ,利用KL型肿瘤免疫图像分析系统检测其Ag NORs含量 (Ag NORs面积与细胞核面积的比值 ,即I /S % )。结果 检测 6 6例正常人、117例恶性肿瘤患者和 2 5例良性肿瘤患者 ,发现 :(1)正常人的T淋巴细胞Ag NORs含量与良性肿瘤患者和恶性肿瘤患者均有显著性差异 (P <0 .0 5 ) ,其均值分别为 :7.0 9± 0 .79、6 .31± 0 .75、5 .4 4± 1.13。 (2 )正常人和良性肿瘤患者的T淋巴细胞AgNORs含量与鼻咽癌、食管癌、乳腺癌、肝癌患者之间有显著性差异(P <0 .0 5 )。鼻咽癌、食管癌、乳腺癌、肝癌患者T淋巴细胞AgNORs检测的阳性率分别为 6 2 .86 %、71.4 3%、6 6 .6 7%、70 %。 (3)观察 6 7例鼻咽癌患者放疗过程的不同阶段 ,发现放疗中和放疗结束患者的T淋巴细胞Ag NORs均值分别为 4 .38± 1.16和 4 .16± 1.2 7,较放疗前 (均值 5 .5 5± 1.2 0 )低 ,但放疗一年后有所上升 (均值 5 .75± 0 .4 8)。 (4)观察 2 8例食管癌患者的不同治疗阶段 (术前、术后七天和术后一月 ) ,发现其T淋巴细胞AgNORs值没有显著性差异 (P >0 .0 5 ) ,分别为 :5 .2 3± 1.16、5 .11± 展开更多

采用PHA活体注射结合秋水仙素培养,取点带石斑鱼全肾,低渗处理,空气干燥制片法制作染色体标本,对染色体进行Giemsa染色、C带及Ag NORs等系列研究。结果表明:(1)点带石斑鱼2n=48,核型组成为48t,NF=48,没有异型性染色体分化;(2)在最小一... 采用PHA活体注射结合秋水仙素培养,取点带石斑鱼全肾,低渗处理,空气干燥制片法制作染色体标本,对染色体进行Giemsa染色、C带及Ag NORs等系列研究。结果表明:(1)点带石斑鱼2n=48,核型组成为48t,NF=48,没有异型性染色体分化;(2)在最小一对染色体的着丝粒与其染色体臂之间,靠近着丝粒部位有明显的次缢痕;(3)在间期核中,通过银染表现出核仁的数目为1~4个,1个核仁的间期核数目最高,多达55%,4个核仁的间期核数目占2%;(4)50%有丝分裂中期相能观察到Ag NORs,Ag NORs主要出现在第24对同源染色体上,第5对同源染色体也可观察到,但其它染色体上则没有;(5)Ag NORs的数目在不同的细胞中表现出多态性,数目为1~4个,出现4个Ag NORs的频率最低(6.8%),出现2个Ag NORs的频率最高(58.1%);(6)第24对同源染色体近着丝粒的臂内具次缢痕,是Ag NORs所在的区域,该区域分布有大量的结构异染色质,即Ag NORs与C带强阳性呈现严格的同步对应;(7)点带石斑鱼所有染色体着丝粒为阳性C带,而且第24对染色体几乎整个染色体臂都呈C带阳性,着色强度与该对染色体上的着丝粒C带相同。最后讨论了核型演化规律和Ag NORs、C带的发生机制。 展开更多
