Methods:The analgesic effect was determined by acetic acid-induced writhing test in mice. While the anti-inflammatory activity was determined by egg albumin-induced oedema of the rat paw.Phytochemical screening was done by standard procedures. Results:Triumfetta rhomboidea leaf extract(50 -400 mg/kg) caused a statistically significant inhibition on the egg albumin-induced eodema or inflammation in Wister albino rats with P【0.001(ANOVA).This effect was higher than the observed effect with Piroxicam(0.5 mg/kg) which was used as a standard. The effect was also dose-dependent.Furthermore,Triumfetta rhomboidea extract caused a statistically significant reduction in the number of acetic acid-induced writhing in mice, with P【0.001(ANOVA).These effects were also does-dependent and greater than the analgesic effects by paracetamol which was used as a reference drug.Phytochemical screening revealed the presence of flavonoids, steroids, triterpenoids alkaloids, tannins and saponins in Truimfetta rhomboidea leaf extract. Conclusion:Triumfetta rhomboidea can be recommended for acute inflammatory disorders and diseases associated with pains. This also supports its traditional use as an anti-snake bite and anti-cancer or anti-tumor agent.Further study is on the way to find out the mechanism of its action and also to isolate, identify and characterize the active principle responsible for these effects in this plant.展开更多

Methods: The anti-inflammatory and analgesic activities were evaluated, using the carrageenan-induced rat paw edema model and the acetic acid writhing test in mice. The gastroprotective activity was determined using HCl/Et OH induced gastric ulcers in rats. The purification and structure elucidation of compound(s) from the more effective fraction were determined by chromatographic and spectroscopic methods and in comparison with data reported in the literature. Results: The fraction F-Et OH showed an important antiinflammatory activity associated with significant analgesic and gastroprotective properties. The purification and structure elucidation of compound(s) from this fraction lead to the identification of one diterpenoid and four sterols. Conclusions: These results suggested that components from the active fraction can be used to treat various anti-inflammatory diseases.展开更多

Merr. 70% ethanol extract with a cellular model of LPS-stimulated RAW264.7 cell. Methods RAW264.7 cells were treated with different concentrations of ethanol fraction (25,50 and 100 g · L^-1 ) and stimu- the conditioned media was collected and analyzed. The quantity of ni- lated with LPS (10 μg· L^-1) for 24 hours, tric oxide (NO) was assayed by Griess reagent. The production of inflammatory mediators was determined by en- zyme-linked immunosorbent assay (ELISA), such as prostaglandin E2 (PGE2), tumor necrosis factor ot (TNF-ot) interleukin- 1 β (IL-1 β) and interleukin 6 (IL-6) in cell supernatant. The concentrations of inflammatory medi- 9 ators were calculated according to the standard curves generated by each of the recombinant cytokines provided with LPS can induce RAW264.7 cells to promote the pro- the ELISA kits. Results Compared with the control group, TNF-α, IL-1β and IL-6. Compared with the duction of inflammatory mediators (P 〈 0.01 ) , including NO, PGE2, model group, ethanol fraction significantly suppressed LPS induced release of inflammatory mediators such as nitric NO, PGE2, TNF-α, IL-1β and IL-6 in a good dose dependent manner (P 〈 0.05, P 〈 0.01 ). Conclusions Eth- anol fraction could significantly inhibit the production of LPS-induced inflammatory response in RAW264.7 cells, and its anti-inflammatory effect may be related to reduce the production of inflammatory mediators NO, PGE2, TNF-α, IL-1 β and IL-6. These results demonstrate that the ethanol fraction is the bioactive component of Disporum can- toniense (Lour.) Merr. , and the ethanol fraction will be further developed as a herbal remedy for preventive and/ or curative purposes in various inflammatory diseases.展开更多

The Food and Drug Administration has classified these factors in category B, which means that they do not demonstrate a fetal risk. However, during pregnancy fetuses are exposed to high anti-tumor necrosis factor(TNF) levels that are measurable in their plasma after birth. Since antibodies can transfer through the placenta at the end of the second and during the third trimesters, it is important to know the safety profile of these drugs, particularly for the fetus, and whether maintaining relapse of the disease compensates for the potential risks of fetal exposure. The limited data available for the anti-TNF drugs to date have not demonstrated any significant adverse outcomes in the pregnant women who continued their therapy from conception to the first trimester of gestation. However, data suggest that antiTNFs should be discontinued during the third trimester, as they may affect the immunological system of the newborn baby. Each decision should be individualized, based on the distinct characteristics of the patient and her disease. Considering all the above, there is a need for more clinical studies regarding the effect of antiTNF therapeutic agents on pregnancy outcomes.展开更多

The study aimed to elucidate the mechanisms underlying the neuroprotective effects of syringaldehyde on ischemic brain cells. Rat models of cerebral ischemia were intraperitoneally administered syringaldehyde. At 6 and 24 hours after syringaldehyde administration, cell damage in the brain of cerebral ischemia rats was obviously reduced, superoxide dismutase activity and nuclear respiratory factor 1 expression in the brain tissue were markedly increased, malondi-adehyde level was obviously decreased, apoptosis-related cysteine peptidase caspase-3 and -9 immunoreactivity was obviously decreased, and neurological function was markedly improved. These ifndings suggest that syringaldehyde exerts neuroprotective effects on cerebral ischemia injury through anti-oxidation and anti-apoptosis.展开更多

Thus, not surprisingly NSAIDs, along with antiinfectious agents, list on the top for causes of DrugInduced Liver Injury (DILI). The incidence of liver disease induced by NSAIDs reported in clinical studies is fairly uniform ranging from 0.29/100 000 [95% confidence interval (CI): 0.17-051] to 9/100 000 (95% CI: 6-15). However, compared with these results, a higher risk of liver-related hospitalizations was reported (3-23 per 100 000 patients). NSAIDs exhibit a broad spectrum of liver damage ranging from asymptomatic, transient, hyper-transaminasemia to fulminant hepatic failure. However, under-reporting of asymptomatic, mild cases, as well as of those with transient liver-tests alteration, in conjunction with reports non-compliant with pharmacovigilance criteria to ascertain DILI and flawed epidemiological studies, jeopardize the chance to ascertain the actual risk of NSAIDs hepatotoxicity. Several NSAIDs, namely bromfenac, ibufenac and benoxaprofen, have been withdrawn from the market due to hepatotoxicity; others like nimesulide were never marketed in some countries and withdrawn in others. Indeed, the contro-versy concerning the actual risk of severe liver disease persists within NSAIDs research. The present work intends (1) to provide a critical analysis of the dissimilar results currently available in the literature concerning the epidemiology of NSAIDS hepatotoxicity; and (2) to review the risk of hepatotoxicity for each one of the most commonly employed compounds of the NSAIDs family, based on past and recently published data.展开更多

文摘BACKGROUND Depression is a common and serious psychological condition, which seriously affects individual well-being and functional ability.Traditional treatment methods include drug therapy and psychological counseling;however, these methods have different degrees of side effects and limitations. In recent years, nonconvulsive electrotherapy(NET)has attracted increasing attention as a noninvasive treatment method. However, the clinical efficacy and potential mechanism of NET on depression are still unclear. We hypothesized that NET has a positive clinical effect in the treatment of depression, and may have a regulatory effect on serum inflammatory factors during treatment.AIM To assess the effects of NET on depression and analyze changes in serum inflammatory factors.METHODS This retrospective study enrolled 140 patients undergoing treatment for depression between May 2017 and June 2022,the observation group that received a combination of mindfulness-based stress reduction(MBSR)and NET treatment(n=70)and the control group that only received MBSR therapy(n=70).The clinical effectiveness of the treatment was evaluated by assessing various factors, including the Hamilton Depression Scale(HAMD)-17,self-rating idea of suicide scale(SSIOS),Pittsburgh Sleep Quality Index(PSQI),and levels of serum inflammatory factors before and after 8 wk of treatment. The quality of life scores between the two groups were compared.Comparisons were made using t and χ^(2) tests.RESULTS After 8 wk of treatment, the observation group exhibited a 91.43%overall effectiveness rate which was higher than that of the control group which was 74.29%(64 vs 52,χ^(2)=7.241;P<0.05).The HAMD,SSIOS,and PSQI scores showed a significant decrease in both groups. Moreover, the observation group had lower scores than the control group(10.37±2.04 vs 14.02±2.16,t=10.280;1.67±0.28 vs 0.87±0.12,t=21.970;5.29±1.33 vs 7.94±1.35,t=11.700;P both<0.001).Additionally,there was a notable decrease in the IL-2,IL-1β,and IL-6 in both groups after treatment.Furthermore, the observation group exhibited superior serum inflammatory factors compared to the control group(70.12±10.32 vs 102.24±20.21,t=11.840;19.35±2.46 vs 22.27±2.13,t=7.508;32.25±4.6 vs 39.42±4.23,t=9.565;P both<0.001).Moreover,the observation group exhibited significantly improved quality of life scores compared to the control group(Social function:19.25±2.76 vs 16.23±2.34;Emotions:18.54±2.83 vs 12.28±2.16;Environment:18.49±2.48 vs 16.56±3.44;Physical health:19.53±2.39 vs 16.62±3.46;P both<0.001)after treatment.CONCLUSION MBSR combined with NET effectively alleviates depression, lowers inflammation(IL-2,IL-1β,and IL-6),reduces suicidal thoughts, enhances sleep, and improves the quality of life of individuals with depression.
文摘Objective:To investigate the analgesic and anti-inflammatory effects of the methanolic extract of the leaves of Triumfetta rhomboidea on mice and rats respectively. And to screen the phytochemical constituent of the extract. This also supports its traditional use as an anti-snake bite and anti-cancer or anti-tumor agent.Further study is on the way to find out the mechanism of its action and also to isolate, identify and characterize the active principle responsible for these effects in this plant.
文摘Objective: To explore anti-inflammatory activities of organic extract and its semi-purified fractions(ethanol, acetone, methanol/dichloromethane) from the Mediterranean gorgonian Eunicella singularis. Conclusions: These results suggested that components from the active fraction can be used to treat various anti-inflammatory diseases.

文摘[Objectives] This study aimed to investigate the anti-inflammatory effects of Baeckea frutescence L.in vitro and in vivo.[Methods]The anti-inflammatory activity of B.frutescence was measured by mouse peritoneal capillary permeability test, mouse auricle swelling test and rat cotton granuloma test. The RAW264.7 macrophage model was stimulated by lipopolysaccharide(LPS),and the effects of B.frutescence on tumor necrosis factor-α(TNF-α) and interleukin-6(IL-6) were detected by enzyme-linked immunosorbent assay(ELISA),and the relative expression of inducible nitric oxide synthase(i NOS) and cyclooxygenase-2(COX-2) proteins was detected by Western blot. Thus, the in vitro anti-inflammatory activity of B.frutescence was studied.[Results] B.frutescence significantly inhibited mouse peritoneal capillary permeability, inhibited xylene-induced mouse auricle swelling and rat cotton granuloma, significantly inhibited TNF-α and IL-6 levels, and down-regulated the expression of i NOS and COX-2 proteins in cells.[Conclusions]B.frutescence showed good anti-inflammatory activity both in vitro and in vivo, and the mechanism may be related to the release inhibition of inflammatory factors TNF-α and the regulation of COX-2.
文摘Aim To evaluate the anti-inflammatory effects of ethanol fraction prepared from Disporum cantoniense (Lour.) Merr. These results demonstrate that the ethanol fraction is the bioactive component of Disporum can- toniense (Lour.) Merr. , and the ethanol fraction will be further developed as a herbal remedy for preventive and/ or curative purposes in various inflammatory diseases.

文摘Treatment of inflammatory bowel disease has significantly improved since the introduction of biological agents, such as infliximab, adalimumab, certolizumab pegol, and golimumab. Considering all the above, there is a need for more clinical studies regarding the effect of antiTNF therapeutic agents on pregnancy outcomes.
文摘There are few studies on the neuroprotective effects of syringaldehyde in a rat model of cerebral ischemia. These ifndings suggest that syringaldehyde exerts neuroprotective effects on cerebral ischemia injury through anti-oxidation and anti-apoptosis.
文摘Non-steroidal anti-inflammatory drugs (NSAIDs) constitute a family of drugs, which taken as a group, represents one of the most frequently prescribed around the world. The present work intends (1) to provide a critical analysis of the dissimilar results currently available in the literature concerning the epidemiology of NSAIDS hepatotoxicity; and (2) to review the risk of hepatotoxicity for each one of the most commonly employed compounds of the NSAIDs family, based on past and recently published data.

文摘Objective:To investigate therapeutic effects of Widman flap operation on treating severe chronic periodontitis and influence of it on inflammatory factors. Methods:A total of 129 cases of patients with severe chronic periodontitis treated in the dental department of our hospital from Nov2014 to Mar2016 were selected as investigate subjects. They were randomly divided to be 63 cases of control group and 66 cases of observation group. For control group, basic treatment was provided. For observation group, improved Widman flap operation was provided. Compared with the effects between two groups of patients, and analyzed the variation of each inflammatory factor before and after treatment. Results:Before treatment, no significant difference showed on PLI, SBI, CAL, PD indexes between two groups of patients. 6 weeks after treatment, PLI, SBI, PD in observation group were significantly decreased comparing with prior treatment, and PLI, SBI were significantly lower than control group at the same phase. 12 weeks after treatment, PLI in control group were significantly decreased comparing with prior treatment;PLI, SBI, CAL, PD in observation group were significantly decreased comparing with prior treatment, and were significantly lower than control group at the same phase. Before treatment, no significant difference showed on IL-1β, IL-6, TNF-α, CRP levels between two groups of patients. After treatment, 4 inflammatory factors in control group showed no significant variation comparing with the same group before treatment, 4 inflammatory factor levels in observation group were significantly lower than which before treatment, IL-1β, IL-6, TNF-α were significantly lower than control group at the same phase. Conclusion:Improving extent of modified Widman flap operation to periodontal indexes and inflammatory factors in patients with severe periodontitis were better than the basic treatment for control group. It could be a choice for treatment of severe periodontitis.
文摘BACKGROUND The incidence of chronic wounds is rising due to an aging population and lifestyle changes in our country. In addition,as the disease spectrum evolves, chronic wounds have become common clinical issues that seriously threaten health and impose significant social and economic burdens.AIM To investigate how depression, anxiety, peripheral blood inflammatory factors, and stress levels affect therapeutic outcomes in patients with chronic wounds.METHODS Retrospectively collected clinical data from 110 patients with chronic wounds treated at Changde Hospital,Xiangya School of Medicine,Central South University(The First People’s Hospital of Changde City)between January 2021 and December 2023,categorizing them into effective and ineffective groups based on treatment effects.Differences between both groups were analyzed using univariate analysis, independent risk factors identified via logistic regression, and their predictive value assessed through receiver operating characteristic analysis.RESULTS Following treatment,95 cases were classified as the effective group(cured or improved),while 15 cases with improvement formed the ineffective group.Significant differences between both groups were noted in wound area, infection status, daily bed time,Hamilton Anxiety Rating Scale(HAMA)scores,Hamilton Depression Rating Scale(HAMD)scores, and levels of interleukin-6,tumor necrosis factor-alpha,and superoxide dismutase(P<0.05).Logistic regression analysis identified a wound area≥7 cm^(2),HAMA≥9 scores, and HAMD≥8 scores were independent risk factors for ineffective treatment in patients with chronic wounds(P<0.05).The receiver operating characteristic curve analysis revealed that the area under the curve for ineffective treatment based on wound area,HAMA,and HAMD was 0.767,0.805,and 0.768 respectively.CONCLUSION Wound size, anxiety, and depression are significant factors influencing the therapeutic outcomes in patients with chronic wounds that require careful attention, alongside the development of appropriate strategies.
文摘During the last few years, condensed thienopyrimidine derivatives have received considerable attention. The therapeutic importance of thienopyrimidines prompted us to synthesize some of spiro(benzothieno[2,3-d]pyrimidine-4-one) derivatives. Some of the novel benzothino-pyrimidine derivatives 3a, 9b, 10b, 11a, 11b, and 11c showed considerable potent anti-inflammatory and analgesic activity of superior G.I.T. safety profile in experimental rats in comparing to indomethacin and tramadol as reference drugs.
