In order to study the significance of flow cytometry immunophenotyping in the diagnosis of acute leukemia, CD45/SSC gating multiparameter flow cytometry (FCM) was utilized to analyze the immunophenotypes of 139 cases...In order to study the significance of flow cytometry immunophenotyping in the diagnosis of acute leukemia, CD45/SSC gating multiparameter flow cytometry (FCM) was utilized to analyze the immunophenotypes of 139 cases of acute leukemia. 139 cases of acute leukemia were enrolled in our hospital from April 1998 to April 2000. Morphological analysis and FCM immunophenotypic tests were conducted on all cases. Our results showed that CD45/SSC gating multiparameter flow cytometry immunophenotyping could reflect the origin of leukemic cells specifically. It is one of the important methods for the diagnosis of ALL, AML, and HAL. CD45/SSC gating multiparameter FCM analysis is a good technique for immunophenotyping. FCM immunophenotypic analysis can help improve the diagnosis and classification of acute leukemia, and extend the use of FCM in clinical practice.展开更多

Here we review the achieve- ments of the HLDA Workshops and provide links to information on CD molecules and antibodies against them, including the 93 new CDs assigned in the 8th Workshop. We consider what remains to be achieved (including an estimate of the number of leucocyte surface molecules still to be discovered), and how the field can best move forward.展开更多
Effective vaccination in duces memory T cells,which protect the host against pathogen re-infecti ons.Therefore,detection of memory T cells is essential for evaluating vaccine efficacy,which was originally dependent on...Effective vaccination in duces memory T cells,which protect the host against pathogen re-infecti ons.Therefore,detection of memory T cells is essential for evaluating vaccine efficacy,which was originally dependent on cytokine induction assays.Currently,two isoforms of CD45 tyrosine phosphatase,CD45RO expression and CD45RA exclusion(CD45RO^(+)/CD45RA^(-)) are used extensively for detecting memory T cells in cattle.The CD45RO^(+)/CD45RA^(-) markers were first established in humans around three decades ago,and were adopted in cattle soon after.However,in the last two decades,some published data in humans have challenged the initial paradigm,and required multiple markers for identifying memoryT cells.On the contrary,memoryT cell detection in cattle still mostly relies on CD45RO^(+)/CD45RA^(-)despite some con troversial evidence.In this review,we summarized the current literature to exami ne if CD45RO^(+)/CD45RA^(-)are valid markers for detecting memoryT cells in cattle.It seems CD45RA and CD45RO(CD45RA/RO)as markers for identifyi ng bovine memoryT cells are questi on able.展开更多
Circulating tumor cells(CTCs)are precursors of distant metastasis in a subset of cancer patients.A better understanding of CTCs heterogeneity and how these CTCs survive during hematogenous dissemination could lay the ...Circulating tumor cells(CTCs)are precursors of distant metastasis in a subset of cancer patients.A better understanding of CTCs heterogeneity and how these CTCs survive during hematogenous dissemination could lay the foundation for therapeutic prevention of cancer metastasis.It remains elusive how CTCs evade immune surveillance and elimination by immune cells.In this study,we unequivocally identified a subpopulation of CTCs shielded with extracellular vesicle(EVs)-derived CD45(termed as CD45^(+)CTCs)that resisted T cell attack.A higher percentage of CD45^(+)CTCs was found to be closely correlated with higher incidence of metastasis and worse prognosis in cancer patients.Moreover,CD45^(+)tumor cells orchestrated an immunosuppressive milieu and CD45^(+)CTCs exhibited remarkably stronger metastatic potential than CD45−CTCs in vivo.Mechanistically,CD45 expressing on tumor surfaces was shown to form intercellular CD45-CD45 homophilic interactions with CD45 on T cells,thereby preventing CD45 exclusion from TCR-pMHC synapse and leading to diminished TCR signaling transduction and suppressed immune response.Together,these results pointed to an underappreciated capability of EVs-derived CD45-dressed CTCs in immune evasion and metastasis,providing a rationale for targeting EVs-derived CD45 internalization by CTCs to prevent cancer metastasis.展开更多
