[Objectives]This study was conducted to breed special pig breeds resistant to diarrhea by using modern biotechnology.[Methods]From Guizhou local breeds,such as Nuogu pigs,Kele pig,Yorkshire pigs and Duroc pigs,190 sam...[Objectives]This study was conducted to breed special pig breeds resistant to diarrhea by using modern biotechnology.[Methods]From Guizhou local breeds,such as Nuogu pigs,Kele pig,Yorkshire pigs and Duroc pigs,190 samples were collected for the analysis of anti-diarrhea gene.[Results]The anti-diarrhea genotype frequency of Kele pigs was 70.00%,which was higher than that of Nuogu pigs(67.37%)and Yorkshire pigs(Yorkshire pigs and Duroc pigs)(50.59%).The favorable anti-diarrhea gene of all Nuogu pigs,Kele pigs,and Yorkshire pigs and Duroc pigs was G,with gene frequencies of 0.7355,0.8368 and 0.8500,respectively,and the frequencies of allele A were 0.2645,0.1632 and 0.1500,respectively.In the process of generation selection,combination selection of GG♂×GG♀,GG♂×GA♀,GA♂×GG♀and GA♂×GA♀was conducted,and GG individuals were selected while gradually phasing out GA and AA individuals.The anti-diarrhea genotypes of 98 pigs in the offspring were tested,and it was found that the frequency of genotype GG was greatly improved,and the frequencies in Nuogu pigs,Kele pigs,Yorkshire pigs and Duroc pigs were increased to 73.91%,81.82%,85.25%and 66.67%respectively,thus forming a special anti-diarrhea breed.[Conclusions]This study provides a basis for selecting excellent breeding pigs,establishing core populations and screening resistance genes in the core populations and their offspring.展开更多

The inhibitory effects of the extracts of the 4 Tibetan veterinary drugs, Veronica ciliate Fisch, Usnea diffracta Vain, Sophoraflavescens var. flavescens, Lamiophlomis rotata ( Benth. ) Kudo on 14 common diarrheagenic bacteria were detected by K-B diffusion method and micro-broth dilution method. Optimization was performed on prescriptions of the 4 Tibetan veterinary drugs using orthogonal design software, and a mouse bacterial diarrhea model was established with a clinical isolate, Salmonella blegdam. According to the LD50 value of the optimal prescription of the 4 Tibetan veterinary drugs, the established mouse bacterial diarrhea model was treated in 3 dose groups, i. e. , the high, middle and low dose groups (0. 060, 0. 030 and 0. 020 g/ml, respectively). The results showed that the 4 Tibetan veterinary drugs had very good inhibitory effects on most of the tested diarrheagenic bacteria, and among them, U. diffracta had better inhibitory effects on all the tested bacteria. The optimal prescription of the d Tibetan veterinary drugs (high dose, 0.06 g/ml) exhibited very good inhibitory effect on mouse diarrhea, indicating that the prescription has very good anti-diarrhea effect, which is beneficial to the clinical treatment of such disease and the development of Tibetan veterinary drugs.展开更多

Methods: Se...Objective: To investigate whether Ershen Pill(ESP,二神丸) could alleviate the symptom of Pi(Spleen)-Shen(Kidney) yang deficiency(PSYD)-induced diarrhea in rat model and explore its anti-diarrhea mechanism. Methods: Seventy-five Sprague-Dawley rats were divided into 5 groups by a random number table, including control, positive, model, low-dose(LD) and high-dose(HD) ESP groups, 15 rats in each group. All the rats, except those in the control group, were developed PSYD induced-diarrhea based on its pathology and etiology. The rats in positive, LD and HD ESP groups were treated with Shenling Baizhu Pill(参苓白术丸), LD(1.05 g/kg) or HD(3.50 g/kg) ESP petroleum ether extract once a day for 2 weeks, respectively. Body weight change and diarrhea index were measured. The histology scores of the kidney were evaluated via hematoxylin and eosin(HE) staining. Aquaporin-3(AQP3) expression in the colon was analyzed by immunofluorescence, quantitative reverse transcription-polymerase chain reaction(qRT-PCR) and Western blot, respectively. Results: Compared with the model group, oral administration of LD and HD ESP prevented body weight loss and inhibited diarrhea after 2-week treatment(P<0.05). Kidney deterioration was impeded, and the histology score in LD and HD ESP groups were 8.2 and 10.5, respectively, which were both higher than those in the model group(P<0.05). In addition, ESP treatment alleviated rat colitis, and HD ESP significantly improved the AQP3 positive staining intensity in the colon tissue compared with the model group. The result from Western blot revealed that AQP3 protein synthesis in colon tissue of LD and HD ESP groups increased by 2.1-and 5.9-fold compared with the model group(P<0.05). qR T-PCR result showed that AQP3 gene expression in the HD ESP group was also up-regulated by 2.5-fold normalized to the model group(P<0.05). Conclusion: ESP extract effectively alleviates the symptoms of PSYD and relieves PSYD-induced diarrhea by improving AQP3 synthesis in the colon.展开更多
