Sensitive detection and precise quantitation of trace-level crucial biomarkers in a complex sample matrix has become an important area of research.For example,the detection of high-sensitivity cardiac troponin I(hs-cT...Sensitive detection and precise quantitation of trace-level crucial biomarkers in a complex sample matrix has become an important area of research.For example,the detection of high-sensitivity cardiac troponin I(hs-cTnI)is strongly recommended in clinical guidelines for early diagnosis of acute myocardial infarction.Based on the use of an electrode modified by single-walled carbon nanotubes(SWCNTs)and a Ru(bpy)32+-doped silica nanoparticle(Ru@SiO2)/tripropylamine(TPA)system,a novel type of electrochemiluminescent(ECL)magneto-immunosensor is developed for ultrasensitive detection of hs-cTnI.In this approach,a large amount of[Ru(bpy)3]2+is loaded in SiO2(silica nanoparticles)as luminophores with high luminescent efficiency and SWCNTs as electrode surface modification material with excellent elec-trooxidation ability for TPA.Subsequently,a hierarchical micropillar array of microstructures is fabricated with a magnet placed at each end to efficiently confine a single layer of immunomagnetic microbeads on the surface of the electrode and enable 7.5-fold signal enhancement.In particular,the use of transparent SWCNTs to modify a transparent ITO electrode provides a two-order-of-magnitude ECL signal amplifi-cation.A good linear calibration curve is developed for hs-cTnI concentrations over a wide range from 10 fg/ml to 10 ng/ml,with the limit of detection calculated as 8.720 fg/ml(S/N 3).This ultrasensitive immunosensor exhibits superior detection performance with remarkable sta-=bility,reproducibility,and selectivity.Satisfactory recoveries are obtained in the detection of hs-cTnI in human serum,providing a potential analysis protocol for clinical applications.展开更多

An alcohol dehydrogenase (ADH)-coexisted solidstate electrochemiluminescence (ECL) biosensor for sensitive detection of the p53 gene was developed. The electrode modified by multiwalled carbon nanotubes, Ru(bpy...An alcohol dehydrogenase (ADH)-coexisted solidstate electrochemiluminescence (ECL) biosensor for sensitive detection of the p53 gene was developed. The electrode modified by multiwalled carbon nanotubes, Ru(bpy)]2+3 and polypyrrole ( MWNTs-Ru (bpy) ]2+3 -PPy ) was prepared to adsorb the ssDNA by electrostatic interactions. Then, the ssDNA recognized the gold nanoparticles (AuNPs)-labeled p53 gene and produced the AuNPs-dsDNA electrode with the AuNPs layer. The AuNPs layer adsorbed the ADH molecules for producing the ECL signal. Thus, the biosensor was based on coupling enzyme substrate reaction with solid-state ECL detection, and it displayed good sensitivity and specificity. The detection limit of the wild type p53 sequence (wtp53) is as low as 0. 1 pmol/L and the discrimination is up to 57. 1% between the wtp53 and the muted type p53 sequence (mtp53). The amenability of this method to the analyses of p53 from normal and cancer cell lysates is demonstrated. The signal of wtp53 in the MGC-803 gastric cancer cell lysates turns out to be about 61.8% that of the wtp53 in the GES-1 normal gastric mucosal cell lysates, and the concentration of the wtp53 is found to decrease about 59 times. The method is highly complementary to enzyme-linked immunosorbent assay (ELISA), and it holds promise for the diagnosis and management of cancer.展开更多

ICCD(Intensified charge coupled device) was used to get the images of ECL of Ru(bpy) 3 Cl 2 ·6H 2 O and tripropylamine(TPA) on the electrodes. The results showed that the ECL was concentrated on the edge of the electrode. For the perforated platinum electrode, the ECL intensity was found to be dependent both on the electrode edge and the edge of the holes on the disk electrode. The ECL intensity of Ru(bpy) 3 Cl 2 ·6H 2 O measured on the four hole disk electrode was double higher than that measured on the simple disk electrode and the detection limit was increased approximately by three orders of magnitude from 1 fmol/L to 1 amol/L.展开更多

Methods: Genomic DNAs from 460 infertile me...Aim: To determine the deletion junctions of infertile men in Taiwan with azoospermia factor region c (AZFc) deletions and to evaluate the genotype/phenotype correlation. Methods: Genomic DNAs from 460 infertile men were examined. Bacterial artificial chromosome clones were used to verify the accuracy of polymerase chain reaction. Deletion junctions of the AZFc region were determined by analysis of sequence-tagged sites and gene-specific markers. Results: Complete AZFc deletions, including BPY2, CDY1 and DAZ genes, were identified in 24 men. The proximal breakpoints were clustered between sY1197 and sY1192, and the distal breakpoints were clustered between sY1054 and sYl125 in all but one of the 24 men. The testicular phenotypes of men with complete AZFc deletion varied from oligozoospermia, to hypospematogenesis, to maturation arrest. Conclusion: We identified a group of infertile men with uniform deletion junctions of AZFc in the Taiwan population.
