The survey was based ...The basic goal of this research was to determine the impact of the presence of bacterial (CFU) and somatic cells count content (SCC) in quality of fresh milk in some small cattle farms in Kosovo. The survey was based on existing standards for milk quality in Kosovo placed under administrative guidance MA-no. 20/2006. The study was based on fresh milk analysis of 150 farms performed during the period September-December 2012, which was obtained in 9 different localities (collection points) of the Kosovo. Our study reveals that CFU and SCC in fresh milk were significantly affected by a number of factors, as: sampling period (repetition), locality, breed, and time of sampling (evening or/and morning). According to the results for CFU and SCC, there were big differences between the farms (milk collection points) included in the study (P < 0.0403) and (P < 0.0293). The results show that small size breed like Busha and its crosses tend to be less exposed to SCC/mL in milk (72.840) and (293.592), compared to Black Holstein (613.462), Simmental (521.519) and Brown Swiss (418.44). Milk produced in evening tended to be of better quality (259.854 CFU/mL) compared to the one from morning milking (576.689 CFU/mL). Fresh milk quality analyzed in the third repetition was better for about 33.3% compared with the repletion first. For CFU and SCC, the analyses show that about 74.7% and 64.7% of milk produced belongs to extra quality, while lower quality of milk of category three is 12.0% and 23.3%, respectively. Considering that about 85% of milk produced in Kosovo comes from small-scale dairy farms, the current study sets out that small-scale milk production system cannot be neglected by interest parties in dairy sector and needs permanent following up and improvement.展开更多

A total of 2256 milk test day records from 379 Bardhoka (BAR), Sharri sheep (SHA), Kosovo sheep (KOS) and Balusha (BAL) ewes in eight herds across the country were collected and analyzed through a period April-October 2010. The general linear model and Duncan’s test were used to analyze the effect of different variables on presence of CFU and SCC in fresh milk. The effect of all variables was considered as a fixed. The overall results show that farm (P 0422), respectively, had a significant effect on presence of SCC. For CFU farm (P 0422) and breed (P 1211.17 to 6425.76 CFU/mL and 846.07 to 2043.15 SCC/mL milk, respectively. Although the rate of fresh milk contamination with SCC tends to be relatively low compare with CFU, by all means both variables should not be underestimated bearing in mind that a high rate of them on fresh milk are negatively correlated with farmer’s profit, consumer food safety and overall animal health.展开更多

However, bacterial profiles in rumen original digesta, liquid and solid fractions vary from each other and need to be better established.Methods: To compare bacterial profiles in each fraction, samples of rumen digesta from six cows fed either a high fiber diet(HFD) or a high energy diet(HED) were collected via rumen fistulas. Rumen digesta was then squeezed through four layers of cheesecloth to separate liquid and solid fractions. The bacterial profiles of rumen original digesta, liquid and solid fractions were analyzed with High-throughput sequencing technique.Results: Rumen bacterial diversity was mainly affected by diet and individual cow(P 〉 0.05) rather than rumen fraction. Bias distributed bacteria were observed in solid and liquid fractions of rumen content using Venn diagram and LEf Se analysis. Fifteen out of 16 detected biomarkers(using LEf Se analysis) were found in liquid fraction, and these 15 biomarkers contributed the most to the bacterial differences among rumen content fractions.Conclusions: Similar results were found when using samples of original rumen digesta, rumen liquid or solid fractions to assess diversity of rumen bacteria; however, more attention should be draw onto bias distributed bacteria in different ruminal fractions, especially when liquid fraction has been used as a representative sample for rumen bacterial study.展开更多
Objective:To investigate the in vitro antimicrobial potential of Thermopsis turcica Kit Tan,Vural&Kckdk against periodoutopathogenie bacteria,its antioxidant activity and cytotoxic effect on various cancer...Objective:To investigate the in vitro antimicrobial potential of Thermopsis turcica Kit Tan,Vural&Kckdk against periodoutopathogenie bacteria,its antioxidant activity and cytotoxic effect on various cancer cell lines.Methods:In vitro antimicrobial activities of elhanol.methanol,ethyl acetate(ElAc,n-hexane and water extracts of Thermopsis turcica herb against periodontopathogenic bacteria,Aggregatibacter actinnmycelemconilans ATCC 29523 and Porphyromonas gingivalis ATCC 33277 were tested by agar well diffusion,minimum inhibitory concentration(MIC)and minimal bactericidal concentration(MBC),Antioxidant properties of the extracts were evaluatod by 1,1-diphenyl-2-picryl-hydrazyl radical scavenging activity and p-carotene bleaching methods.Amounts of phenolic contents of the extracts were also analysed by using the Folin-Ciocalteu reagent.Additionally,cytotoxic activity of the extracts on androgcn-insensilivc prostate cancer,androgen—sensitive prostate cancer,chronic myelogenous leukemia and acute promyelocytic leukemia bunian cancer cell lines were determined by 3-4,5-dimelhylthiazol-2-yh-2,5-diphenyltclrazolium bromide assay.Human gingival fibroblast cells were used as a control.Results:Our data showed that ELAc extract had the highest antimicrobial effect on Aggregatibacter actinomycetemitans(MIC:1.562 mg/ml_MHC:3.124 mg/ml.)and Porph yromonas gingiralis(MIC:0.781 mg/mL,MBC:1.562 mg/mL).In antioxidant assays.ElAc extract exhibited also the highest radical scavenging activity[IC_(50)=(30.0±0.3)μg/ml.]and the highest inhibition[(74.35±0.30)%]|against lineloic acide oxidation.The amount of phenolic content of it was also the highest[(l62.5±l.2)μg/mg gallic acid].In cytotoxic assay,only etbanol[IC_(50)=(80.00±1.21)μg/ml.]and EtAc extract[IC_(50)=(70.0±0.9)μg/ml]were toxic on acute promyeloeytic leukemia cells at 20—100μg/mL.P<0.05>.However,no toxic effect was observed on human gingival fibroblast cells.Cunclusions:According to our findings,owing to its antioxidant and cytotoxic potential,EtAc exlrael might include anticancer agents for acute promyelocytic leukemia.展开更多
