Lytic Characteristics and Identification of Two Alga-lysing Bacterial Strains 被引量:4

All previously reported bacterial species which are capable of lysing harmful algae have been isolated from coastal environments in which harmful algae blooms have occurred. Due to the low concentration of alga-lysing bacteria in an algal bloom, it is difficult to isolate the alga-lysing bacteria by existing methods. In this paper, two algae-lysing bacterial strains, P01 and P03, have been isolated from a biosystem immobilized on a sponge that was highly effective in removing algae and microcystins. Their lysing modes and effects on Microcystis aeruginosa have been studied. The results show that the degradation processes of these two strains for M. aeruginosa accorded with a first-order reaction model when the chlorophylla concentration was in the range from 0 to 1000 μgL-1. The degradation rate constants were 0.1067, 0.1274 and 0.2792 for P01 and0.0683, 0.0744 and 0.028 97 for P03, when the bacterial densities were 8.6 × 105, 8.6 × 106 and 8.6 × 107cells mL-1 respectively. Moreover, the two bacterial strains had favourable lytic effects not only on M. aeruginosa , but also on Chlorella and Scene-desmus. Their lytic effect on M. aeruginosa did not require physical cell to cell contact, but proceeded by the production of an extracellular product. The bacterial strains were identified as Bacillus species by PCR amplification of the 16S rRNA gene, BLAST analysis, and comparison with sequences in the GenBank nucleotide database. 展开更多

By processing S-wave data from the Fanshi-Huai’an-Taipusiqi DSS profile,which is a three-component,wide-angle reflection/refraction profile,and in the light of the results from P-wave interpretation,two-dimensional(2-D)structures of the crust and upper mantle are presented,including S-wave velocity Vs and the physical parameter of medium-Poisson’s ratio a.Taking other geological and geophysical information into account,and with reference to the results from petrophysical experiments at home and abroad,we carried out interpretation and inference with respect to deep crustal structure,tectonics,and lithologic characters.It has been concluded that in the upper and middle crust,a values are mostly not greater than 0.25,and rocks,which generally assume brittle,are mainly composed of granite; the rocks in the lower layer of the upper crust between Yangyuan-Huai’an containing inorganic CO2 itself releases carbon; for the rocks in the lower crust and crust-mantle transitional zone,which are comparatively 展开更多

In accordance with human activities, geomorphologic location, behavior and particular, the man-induced sediment disaster can be divided into 4 types: the drainage network, slope and gully, channel and plain-estuary-coastline. Each type includes erosion, transportation, accumulation, complexity and cascading. Based on human activity, geomorphology, sediment mechanics and catastrophology, the man-induced sediment disaster is characterized as follows: (1) accelerating tendency with geographical zoning back-ground; (2) non-order characters by blind action without special technical training; (3) gradually and sharply changing with human environment vibration; and (4) complexity and non-linear figure, etc. One of the reasons leading to man-induced sediment disaster is human environment vibration. 展开更多

S型控释氮肥运筹对水稻产量形成及氮素利用特征的影响

缓控释肥具有施用次数少、肥料利用率高的特点,S型热固性树脂包膜控释氮肥(S型控释氮肥)受土壤pH影响小、肥料稳定性好。本研究以S型控释氮肥为试材,分别设置了6个水稻品种的响应比较、4种氮肥施用量、5种氮肥施用时期及7种氮肥配施比例... 缓控释肥具有施用次数少、肥料利用率高的特点,S型热固性树脂包膜控释氮肥(S型控释氮肥)受土壤pH影响小、肥料稳定性好。本研究以S型控释氮肥为试材,分别设置了6个水稻品种的响应比较、4种氮肥施用量、5种氮肥施用时期及7种氮肥配施比例的4个盆栽试验,研究其对水稻产量形成、氮素吸收利用等的影响。结果表明:1)品种比较试验中,S型控释氮肥提高了6个水稻品种的产量,增幅在2.95%~18.57%,平均增加12.60%;产量提高的原因是穗数或每穗颖花数的增加;S型缓控释氮肥可显著提高供试水稻成熟期的氮素积累量、氮肥利用率,但氮素籽粒生产效率有所下降。2)控释肥运筹试验中,随着S型控释氮肥施用量和施用比例的增加,武运粳24号的产量呈先增加后下降的趋势,分别以每盆3g纯氮处理和控释氮肥占比80%处理产量增幅最大,作基肥时增产效应更显著;成熟期氮素积累量呈增加和先增加后下降的趋势,分别以每盆4.5 g纯氮处理和控释氮肥占比80%处理增幅最大,作基肥时增幅效应更显著;氮素籽粒生产效率和氮肥利用率呈降低趋势,氮素籽粒生产效率分别以控释氮肥占比0%和控释肥作保花肥最大,氮肥利用率分别以控释氮肥占比80%和控释肥作基肥时最大。3)控释肥对根系性状影响试验中,S型控释氮肥处理提高了武运粳24号水稻栽后30天单株不定根总长、根干重与吸收面积,提高了抽穗期单株不定根总长、根干重和根活力;单株不定根数、不定根总长、根干重、根活力的增加是产量、氮素积累量和氮肥利用率协同提高的根系基础。总之,S型控释氮肥可显著提高水稻的产量和氮肥利用率,作为基肥时最优。每盆3 g纯氮处理、控释氮肥占比80%左右时产量、成熟期氮素积累量、氮肥利用率协同增加,与其栽后30天、抽穗期根系性状优化有密切关系。 展开更多

After trace Sm^3+ ions and Gd^3+ ions doping, the emission intensity of red phosphors Y2O2S: Eu^3 + was enhanced and the voltage character (relation between emission intensity and excitation voltage) was improved while the other properties of physics and chemistry were not changed. The origins of enhancement and improvement are discussed. Probably the distortion and the defect of crystals are decreased by the substitution of Gd^3+ for Y^3+ instead of Eu^3+ for Y^3+ , and thus the Eu^3+ crystal field is improved, and radiationless process and energy loss resulted from crystal defect are weakened, which leads to increased luminescence intensity and voltage character improvement. The overlapping fluorescent spectra of Y2O2S: Sm^3+ emission and Y2O2S:Eu^3+ excitation as well as Eu^3 + excitation spectra transitions spectra lead to energy transfer from Sm^3 + sensitization of Sm^3+ ions fectively. containing Sm^3+ excitation the possibility of resonance ions to Eu^3+ ions, and the to Eu^3+ ions is achieved effectively. 展开更多

This article is devoted to the thermophysical and mechanical study of an eco-material, poto-poto. The objective is to study the influence of the addition of bamboo fibers on the thermophysical properties of the material, while verifying that it complies with the mechanical standards in Cameroon. A double study is therefore carried out: a mechanical characterization whose objective is to determine the mechanical properties (compressive strength, flexural strength and Young’s modulus) and a thermophysical characterization whose objective is to determine the thermal conductivity. For the thermophysical characterization, the asymmetric hot plane experimental setup based on 1D quadrupole modelling was developed and simulated for the estimation of thermal effusivity E and volume heat capacity ρCp. The obtained experimental results show that the apparent thermal conductivity of the developed materials decreases with increasing fibers. Although the reference material (0% fibers) has a much higher conductivity than the ordinary sand block (1.15 Wm<sup>-1</sup> K<sup>-1</sup>), the addition of fibers, already at 2%, contributes to decrease this conductivity (0.95 Wm<sup>-1</sup> K<sup>-1</sup>). From the point of view of thermal insulation and energy savings, the thermal conductivity results obtained show that the use of these materials with a maximum fibercontent of 6% would be a better thermal insulator than sand block or compressed earth brick. The compressive strength obtained is such that Rc > 0.6 MPa. All the materials developed meet the design standards when used as infill. 展开更多

俯冲带是地球上岩浆活动、高压—超高压变质作用、中深源地震、壳幔物质交换、元素循环和铜金大规模成矿的集中发生场所。富水流体不仅调控了这些地质作用,而且也深刻影响了全球C、S等挥发分的循环。当蚀变大洋岩石圈及上覆沉积物进入... 俯冲带是地球上岩浆活动、高压—超高压变质作用、中深源地震、壳幔物质交换、元素循环和铜金大规模成矿的集中发生场所。富水流体不仅调控了这些地质作用,而且也深刻影响了全球C、S等挥发分的循环。当蚀变大洋岩石圈及上覆沉积物进入俯冲带中深部(15~300 km),伴随由葡萄石-绿纤石相至超高压榴辉岩相的递进变质作用,含水矿物在不同深度的分解造成流体释放为一连续过程。除极端高地温梯度环境之外,大多数俯冲带洋壳释放的流体为富水流体。但,俯冲带不同深度所产生的流体特征有明显差异。出露于全球造山带的高压—超高压变质地体保存了分凝体、脉体、水压致裂角砾岩等流体作用的有力证据,绿辉石、石榴子石、绿帘石等矿物中原生流体包裹体为流体的直接记录。在俯冲带中等深度(<65 km),流体是溶质含量很低的含卤化物水溶液,可含CO_(3)^(2-)、SO_(4)^(2-)、HS^(-)等组分,其所含Si、Al、Ca、Mg、Fe、Na主量元素溶质相当于海水中固化物量的2~3倍,并具大离子亲石元素(LILE)、轻元素(B、Li)富集和高场强元素(HFSE)亏损的特点。当深度≥65 km时,流体转化为类似于超临界性质的溶液,含CH_(4)、C_(2)H_(6)、H_(2)S等挥发分,其所含主量元素溶质显著增加,微量元素除LILE等外,还负载相当量的HFSE和过渡族成矿元素。俯冲带65~100 km深度,为富水流体向超临界流体转变的区间。这种流体具有“亚超临界”性质,发生了氧化还原性质和元素溶解能力的渐变,并伴随硬柱石和角闪石的最终耗尽。高压变质岩石及相关脉体的O、Sr、Nd和金属同位素示踪研究表明俯冲带流体的源区多样,有蚀变基性洋壳、地幔橄榄岩和沉积物,并保存了海底热液蚀变作用的印迹。流体以脉冲方式沿网络状裂隙呈隧道式运移和传输,规模可达千米级,时间尺度在数月至数百年。目前通过高压—超高压变质岩的研究对俯冲带流体已经有了深入的了解,但展望未来,该领域依然有诸多争议和科学问题值得探索。 展开更多

通过对繁峙-怀安-太仆寺旗三分向深地震测深宽角反射\折射剖面的S波资料进行处理,结合P波处理结果,给出了地壳上地幔的S波速度VS及介质物性参数泊松比σ的二维结构;综合地质及其它地球物理资料,参照国内外岩石物理参数的实... 通过对繁峙-怀安-太仆寺旗三分向深地震测深宽角反射\折射剖面的S波资料进行处理,结合P波处理结果,给出了地壳上地幔的S波速度VS及介质物性参数泊松比σ的二维结构;综合地质及其它地球物理资料,参照国内外岩石物理参数的实验结果,对地壳深部结构、构造及岩性进行了解释和推测。其结论为:中上地壳大部分地区σ值不大于025,岩石主要由花岗岩组成,整体呈脆性;其中阳原-怀安间的上地壳下层岩体,含有来自上地幔的无机CO2气体并还原释放出碳;下地壳及壳幔过渡层σ值大部分大于026,整体呈相对塑性;下地壳岩性在怀安以北地区为中酸性麻粒岩、怀安至阳原为英角闪片麻岩、阳原以南为石英岩;莫霍\过渡层主要由剖面两端的基性麻粒岩-闪岩和剖面中部地区的石英云母片岩-中性麻粒岩所组成。根据σ值的横向变化并结合震相特征,推断了阳原及张北附近的超壳断裂带和怀安的深断裂带。结合地震活动性,对地震较为集中发生的阳原上地壳下层进行了解释,认定是由于该岩体较脆硬且“漂浮”在下部的较塑性岩体之上,受超壳断裂带、区域应力场及上地幔物质上涌的影响,岩体容易在σ值显著变化的部位产生错动、滑动,引发地震,且地震多集中于σ值较小的脆硬岩体一侧。 展开更多

【目的】克隆甘蔗(Saccharum officinarum)S-腺苷蛋氨酸脱羧酶(S-adenosylmethionine decarboxylase,SAMDC)基因,并进行序列特征、原核表达和不同逆境胁迫下表达特性分析。【方法】通过对甘蔗茎全长cDNA文库测序和分析,获得SAMDC基因cDN... 【目的】克隆甘蔗(Saccharum officinarum)S-腺苷蛋氨酸脱羧酶(S-adenosylmethionine decarboxylase,SAMDC)基因,并进行序列特征、原核表达和不同逆境胁迫下表达特性分析。【方法】通过对甘蔗茎全长cDNA文库测序和分析,获得SAMDC基因cDNA全长,命名为Sc-SAMDC,并进行序列分析。随后将编码S-腺苷蛋氨酸脱羧酶的cDNA片段克隆到原核表达载体pET29a(+)中,构建融合表达质粒,转化至Esherichia coli BL21(DE3)中进行表达。最后利用定量PCR技术分析甘蔗幼苗中该基因在不同外源胁迫下的表达特性。【结果】序列分析显示,甘蔗Sc-SAMDC基因(GenBank Accession number:GQ246459)cDNA全长1968bp,存在3个读码框(袖珍读码框tORF、上游读码框uORF和主读码框mORF),mORF长1200bp,编码399个氨基酸的SAMDC酶原,预测分子量为43.6kD,该酶原含有两个高度保守的功能结构域(酶原剪切位点和PEST结构域)。原核表达产物经SDS-PAGE表明,Sc-SAMDC以融合蛋白形式表达,相对分子量约为50kD。定量PCR分析表明,Sc-SAMDC基因在聚乙二醇(PEG)、NaCl、水杨酸(SA)和H2O2外源胁迫下表达特性不同,受PEG和NaCl的诱导,受SA和H2O2的抑制。【结论】本研究成功地克隆了Sc-SAMDC基因并在原核生物中进行了表达研究,分析了该基因的表达特性,为其进一步的生物学功能研究及其应用奠定了基础。 展开更多
