In order to relieve the experience dependence and improve the design, a new Multidisciplinary Design Optimization (MDO) method "Bi-Level Integrated System Collaborative Optimization (BLISCO)" is applied to the conceptual design of an HOV, which consists of hull module, resistance module, energy module, structure module, weight module, and the stability module. This design problem is defined by 21 design variables and 23 constraints, and its objective is to maximize the ratio of payload to weight. The results show that the general performance of the HOV can be greatly improved by BLISCO.展开更多

Fully human antibodies have minimal immunogenicity and safety profiles.At present,most potential antibody drugs in clinical trials are humanized or fully human.Human antibodies are mostly generated using the phage dis...Fully human antibodies have minimal immunogenicity and safety profiles.At present,most potential antibody drugs in clinical trials are humanized or fully human.Human antibodies are mostly generated using the phage display method(in vitro)or by transgenic mice(in vivo);other methods include B lymphocyte immortalization,human–human hybridoma,and single-cell polymerase chain reaction.Here,we describe a structure-based computer-aided de novo design technology for human antibody generation.Based on the complex structure of human epidermal growth factor receptor 2(HER2)/Herceptin,we first designed six short peptides targeting the potential epitope of HER2 recognized by Herceptin.Next,these peptides were set as complementarity determining regions in a suitable immunoglobulin frame,giving birth to a novel anti-HER2 antibody named "HF,"which possessed higher affinity and more effective anti-tumor activity than Herceptin.Our work offers a useful tool for the quick design and selection of novel human antibodies for basic mechanical research as well as for imaging and clinical applications in immune-related diseases,such as cancer and infectious diseases.展开更多

The application of the design mindset over a broader scope of disciplines such as business and technology results in creative people from overlapping disciplines who are flexible, curious, and imaginative; have a tolerance for ambiguity and provide meaningful frameworks for difficult situations. The design process becomes a secondary skill-set that compliments another primary knowledge/skill base. With this model, business disciplines could apply design thinking to the understanding of fundamental economic drivers and technology/engineering/manufacturing disciplines could apply design thinking over deep process expertise in implementation. What about the work of the traditional design disciplines? Following the model described above for business and technology, design disciplines should apply "design thinking" on top of some foundational skill set that comes from the area of human values. According to the traditional design theory, the FBS design model pays more attention to the function and stru cture of the product. But this model still couldn’t strengthen the relation bet ween product appearance design and human-machine design effectively. This paper adopt converse design thinking and presents an improved design thinking methodo logy based on C: FBS for product appearance design and give a general summarizat ion for the features, methods and technology based on human-machine interaction and interface. Meanwhile it also combines with the behavior design of product r elated IT fields and constructs a new outline to improve the design of product a ppearance supported by the technology of computer aided design. This paper introduced three of the important challenges in this relationship, which have, thus far, been only partially or implicitly acknowledged in design and planning. The aim was to elucidate the key disregarded and neglected aspects in the human-nature relationship that need to be considered in different phases of the design and planning of future projects. This paper will thus inform the designers and planners about the previously overlooked but important challenges in the human-nature relationship, which must be moved to the foreground of the design and planning processes. These challenges include the definition of the term “nature”, the reciprocal effects among scales and adhering to or going beyond environmental sustainability. As the main conclusion, the paper shows how these challenges can improve the mutually supportive interaction between human beings and nature in the field of design and planning. Specifically speaking, the paper discussed how to convert the overlooked challenges in the human-nature relationship into potentials in the design and planning process. The main applied methods for this paper were a review of the related literature and the appropriate analysis.展开更多

Human detection is important in many applications and has attracted significant attention over the last decade. The Histograms of Oriented Gradients (HOG) as effective local descriptors are used with binary sliding wi...Human detection is important in many applications and has attracted significant attention over the last decade. The Histograms of Oriented Gradients (HOG) as effective local descriptors are used with binary sliding window mechanism to achieve good detection performance. However, the computation of HOG under such framework is about billion times and the pure software implementation for HOG computation is hard to meet the real-time requirement. This study proposes a hardware architecture called One-HOG accelerator operated on FPGA of Xilinx Spartan-6 LX-150T that provides an efficient way to compute HOG such that an embedded real-time platform of HW/SW co-design for application to crowd estimation and analysis is achieved. The One-HOG accelerator mainly consists of gradient module and histogram module. The gradient module is for computing gradient magnitude and orientation;histogram module is for generating a 36-D HOG feature vector. In addition to hardware realization, a new method called Histograms-of-Oriented-Gradients AdaBoost Long-Feature-Vector (HOG-AdaBoost-LFV) human classifier is proposed to significantly decrease the number of times to compute the HOG without sacrificing detection performance. The experiment results from three static image and four video datasets demonstrate that the proposed SW/HW (software/hardware) co-design system is 13.14 times faster than the pure software computation of Dalal algorithm.展开更多
