胃癌患者EB病毒感染情况及其对癌组织p53、Bcl-2表达的影响

目的探究胃癌患者人类疱疹(EB)病毒感染情况及其对癌组织p53、B淋巴细胞瘤-2(Bcl-2)表达的影响。方法选取新乡市中心医院2019年8月至2023年1月收治的76例胃癌患者,采用原位杂交法检测患者癌组织及癌旁组织EB病毒编码小分子RNA表达,采用... 目的探究胃癌患者人类疱疹(EB)病毒感染情况及其对癌组织p53、B淋巴细胞瘤-2(Bcl-2)表达的影响。方法选取新乡市中心医院2019年8月至2023年1月收治的76例胃癌患者,采用原位杂交法检测患者癌组织及癌旁组织EB病毒编码小分子RNA表达,采用逆转录聚合酶链式反应(RT-PCR)法检测患者癌组织及癌旁组织p53、Bcl-2 mRNA表达,分析EB病毒感染与胃癌临床病理特征及癌组织p53、Bcl-2表达的关系。结果76例胃癌患者癌组织标本中EB病毒阳性率为27.63%(21/76),邻近癌旁组织标本中EB病毒阳性率为4.29%(3/76),胃癌患者癌组织EB病毒阳性率高于癌旁组织(P<0.05);与EB病毒阴性的胃癌患者比,EB病毒阳性患者中病灶位于近端胃、组织浸至浆膜层、有淋巴结转移的占比较多(P<0.05);胃癌患者癌组织p53及Bcl-2 mRNA表达均高于癌旁组织(P<0.05);EB病毒感染胃癌患者癌组织p53及Bcl-2 mRNA表达均高于未感染胃癌患者(P<0.05)。结论EB病毒感染与胃癌患者近端胃病变、组织浸至浆膜层及有淋巴结转移有关,EB病毒可能通过驱动宿主p53基因甲基化来上调p53表达,与Bcl-2协同促进癌细胞生长,这些可能为临床提供胃癌诊疗评估因子及免疫治疗新靶点。 展开更多

目的:探讨新的凋亡抑制基因livin在宫颈癌组织中的表达,及其与bc1-2和p53表达的相关性。方法:应用免疫组织化学链霉菌抗生物素蛋白-过氧化酶连接法(SP法),检测livin、bcl-2、p53基因在lO例正常宫颈组织及57例宫颈癌组织中的表达。结果:L... 目的:探讨新的凋亡抑制基因livin在宫颈癌组织中的表达,及其与bc1-2和p53表达的相关性。方法:应用免疫组织化学链霉菌抗生物素蛋白-过氧化酶连接法(SP法),检测livin、bcl-2、p53基因在lO例正常宫颈组织及57例宫颈癌组织中的表达。结果:Livin基因在正常宫颈组织中不表达,57例宫颈癌组织中,40例表达阳性,占70.1%。Livin基因表达阳性率与宫颈癌的病理分级和临床分期有关。宫颈癌组织中p53蛋白表达阳性、阴性者中,livin基因表达阳性率分别为76.7%(33/43)、21.4%(3/14),两者比较,差异有统计学意义(P<0.05);bcl-2蛋白表达阳性、阴性者中,livin基因表达阳性率分别为87.8%(36/41)、25%(4/16),两者比较,差异有统计学意义(P<0.05)。结论:Livin基因的异常表达而引起的细胞凋亡抑制,在宫颈癌的发生中起一定作用,其表达与宫颈癌组织中p53、bc1-2蛋白的异常表达密切相关。 展开更多

Objective: To evaluate the anti-tumor effects of SeO2 and its mechanisms on three human lung cancer cell lines. Methods: Three lung cancer cells A549, GLC-82 and PG were treated with 3-30 μmol/L SeO2. Flow cytometry was used to detect apoptosis, and analyze the changes of expression of p53 and Bcl-2, as well as ROS and Ca2+ level within cells. Results:SeO2 markedly inhibited cell proliferation and viability, and prompted apoptosis after 48 h treatment. SeO2 at 10 μmol/L induced 47.8% apoptosis in A549 cells, 40.8% in GLC-82 cells, 18.2% in PG cells. SeO2 at 30 μmol/L induced 37.8% apoposis in PG cells,but did not increase apoptotic raes in other two cells. SeO2 could down-regulate the mean fluorescent intensity of Bcl-2 from 65.8 to 9.6 in A549, but not in GLC-82 and in PG cells, up-regulate wild type p53 level in all three cells. SeO2 decreased the ROS and Ca2+ level markedly within three tested cells. Conclusion: SeO2 showed anti-tumor effect via apoptosis pathway in three lung cancer cell lines. The decrease of ROS and Ca2+ level within cells as well as regulation of Bcl-2 and p53 expression may play important roles in above apoptotic procedure. 展开更多

[目的]观察SO2对大鼠肺细胞凋亡相关基因mRNA和蛋白表达的影响。[方法]运用荧光实时定量RT- PCR和免疫组化技术研究了不同浓度SO2(0、14、28、56 mg/m3)吸入对大鼠肺细胞p53、bax、bcl-2三种细胞凋亡相关基因mRNA和蛋白表达的影响。[结果]肺中p53和bax mRNA表达水平及bax/bcl-2比值呈剂量依赖性增加,在低浓度SO2(14mg/m3)吸入条件下,p53、bax mRNA表达水平及bax/bcl-2比值虽然有升高趋势,但统计学上未见显著差异, 在SO2浓度为28.00和56.00mg/m3时均为显著增加(与对照组相比,p53:在14mg/m3为1.07倍,在28mg/m3为1.23倍, 在56 mg/m3为1.39倍;bax:在14 mg/m3为1.12倍,在28 mg/m3为1.77倍,在56 mg/m3为2.26倍;bax/bcl-2比值:在 14 mg/m3为1.13倍,在28 mg/m3为2.19倍,在56 mg/m3为3.01倍);而bcl-2 mRNA表达水平呈剂量依赖性降低,在低浓度SO2(14mg/m3)吸入条件下,bcl-2 mRNA水平虽然有降低趋势,但统计学上未见显著差异,在SO2浓度为28.00和 56.00mg/m3时显著降低(与对照组相比,bcl-2:在14mg/m3为0.99倍,在28mg/m3为0.78倍,在56mg/m3为0.73倍)。免疫组化的实验结果表明吸入SO2后大鼠肺中p53和bax蛋白表达水平呈剂量依赖性增加,而’bcl-2蛋白表达水平呈剂量依赖性降低。[结论] SO2可以改变凋亡相关基因的表达,表明SO2可诱导大鼠肺细胞凋亡,这可能与一些凋亡相关疾病的发生有关。这些机制的阐明有助于对SO2毒作用机制的理解和所致疾病的治疗。 展开更多

目的 探讨人肾透明细胞癌中 ki- 6 7、bcl- 2、bax、p5 3表达及癌细胞自发凋亡率的临床意义。方法 选取 4 8例档存的肾透明细胞癌石蜡包埋标本 ,采用免疫组织化学方法检测癌细胞中 ki- 6 7、bcl- 2、bax、p5 3的表达 ,采用缺口末端标... 目的 探讨人肾透明细胞癌中 ki- 6 7、bcl- 2、bax、p5 3表达及癌细胞自发凋亡率的临床意义。方法 选取 4 8例档存的肾透明细胞癌石蜡包埋标本 ,采用免疫组织化学方法检测癌细胞中 ki- 6 7、bcl- 2、bax、p5 3的表达 ,采用缺口末端标记法 ( TU NEL )检测癌细胞的自发凋亡率。结果  ki- 6 7表达率 ( PI)为 0 .15 %~ 36 .4 % ,随TNM分期的增高而增高。癌细胞自发凋亡率 ( AI)为 0 .2 %~ 15 .4 % ,随 TNM分期的增高有上升趋势。各因子之间关系分析 :1高 bax表达组的癌细胞凋亡率高 ;2 p5 3阴性组的 AI高 ,bax高 ,PI低 ;3未发现 bcl- 2与其他因子间有相关关系。预后分析 :PI、AI、p5 3、bax、bcl- 2 / bax、TNM是预后因素 ,PI、p5 3是独立预后因素。结论 细胞增生状态、细胞凋亡状态以及凋亡调控因子的表达情况是影响肾透明细胞癌发生。 展开更多

TP53BP2基因真核表达载体的构建及在人胚肾Expi293F细胞中蛋白表达、纯化及活性鉴定

目的构建人肿瘤抑制因子p53结合蛋白2(tumor suppressor p53-binding protein 2,TP53BP2)的重组真核表达载体,转染人胚肾Expi293F细胞,获得高纯度的重组人全长TP53BP2蛋白并对其进行生物学活性鉴定。方法利用UniProt网站查询TP53BP2基... 目的构建人肿瘤抑制因子p53结合蛋白2(tumor suppressor p53-binding protein 2,TP53BP2)的重组真核表达载体,转染人胚肾Expi293F细胞,获得高纯度的重组人全长TP53BP2蛋白并对其进行生物学活性鉴定。方法利用UniProt网站查询TP53BP2基因序列,并进行Expi293F表达系统序列优化,通过同源重组连接至pcDNA3.1(+)-P2Ae GFP载体并进行双酶切和测序鉴定,通过转染试剂聚乙烯亚胺(polyethylenimine,PEI)将pcDNA3.1(+)-P2A-eGFPTP53BP2质粒瞬时转染至Expi293F细胞,荧光显微镜观察转染效率,收集实验组及对照组细胞,利用免疫印记试验(Western blot,WB)检测TP53BP2重组蛋白表达水平。通过His标签纯化试剂盒及Superdex 20010/300GL层析柱进行蛋白纯化,十二烷基硫酸钠聚丙烯酰胺凝胶电泳(sodium dodecyl sulfate polyacrylamide gel electrophoresis,SDSPAGE)对纯化后重组蛋白进行鉴定。利用免疫共沉淀(Co-immunoprecipitation,Co-IP)检测重组人全长TP53BP2蛋白与p65蛋白结合情况。利用免疫荧光(immunofluorescence,IF)检测重组人全长TP53BP2蛋白与p65蛋白共定位。利用表面等离子体共振(surface-plasmon resonance,SPR)技术,检测纯化后的重组人全长TP53BP2蛋白与TP53BP2抗体的相互作用。结果经测序和双酶切鉴定,重组质粒pcDNA3.1(+)-P2A-eGFP-TP53BP2构建成功。经荧光显微镜观察结果显示转染效率约为60%,WB结果表明TP53BP2蛋白在Expi293F细胞中过表达,证明转染成功。SDS-PAGE结果表明纯化后重组蛋白纯度在90%以上,证明纯化成功。Co-IP结果表明,TP53BP2重组蛋白可与p65蛋白相互作用。IF结果表明,His标签蛋白、TP53BP2蛋白及p65蛋白存在共定位,表明三者之间存在相互作用。SPR结果表明,纯化的重组人TP53BP蛋白与TP53BP2抗体具有较好的结合活性。以上结果均证明重组人全长TP53BP2蛋白具有生物学活性。结论成功构建了TP53BP2基因真核表达载体并在人胚肾Expi293F细胞中成功表达出具有生物学活性的重组人全长TP53BP2蛋白,为进一步研究TP53BP2的结构和功能奠定了基础。 展开更多

AIM: To analyze the protein expression essential for apoptosis in liver steatosis. METHODS: The expression of proapoptotic proteins p53, Bax, and antiapoptotic Bcl-2 in hepatocytes with steatosis (SH) and without steatosis (NSH) was evaluated in 84 patients at various stages of non-alcoholic fatty liver disease (NAFLD). RESULTS: Immunohistochemical staining of liver tissue showed the activation of p53 protein in SH and NSH with increased liver steatosis, diminished Bcl-2 and slightly decreased Bax protein. Positive correlation was found between the stage of liver steatosis with p53 expression in SH (r = 0.54, P < 0.01) and NSH (r = 0.49, P < 0.01). The antiapoptotic protein Bcl-2 was diminished together with the advancement of liver steatosis, especially in non-steatosed hepatocytes (r =0.43, P < 001). CONCLUSION: Apoptosis is one of the most important mechanisms leading to hepatocyte elimination in NAFLD. The intensification of inflammation in NAFLD induces proapoptotic protein p53 with the inhibition of antiapoptotic Bcl-2. 展开更多

AIM: To investigate the interrelationship between H pylori and Epstein-Barr virus (EBV) infection in the gastric carcinogenesis having in focus the p53 mutation and the c-Myc, Bcl-2 and Bax expression. METHODS: seventy-one gastric carcinoma tissues were assessed by polymerase chain reaction (PCR) for H pylori and in situ hybridization for EBV. c-Myc, Bcl-2 and Bax expression were detected by immunohistochemistry and single-stranded conformational polymorphism (SSCP) for p53 mutation. RESULTS: The positivity rates for H pylori and EBV were 94.4% and 8.45%, respectively. The majority of the cases displayed only the H pylori presence. All EBV positive cases were also H pylori positive. None infectious agent was observed in 5.55% of the cases. The intestinal type tumor was more frequent in the co-infected and non-infected groups. The female predominated in the non-infected group showing statistical significance (70.4% vs 29.6%, P=0.039). The Bcl-2 was only detected in the group exclusively infected by H pylori. However, c-Myc and Bax were detected in the three groups but with a low frequency in the co-infected group. Mutation of p53 was present in all groups, with the highest frequencies in the H pylori positive groups. CONCLUSION: The frequency of H pylori infection in gastric carcinomas was high. The presented data indicated that gastric carcinogenesis has different pathways depending of the presence of the two investigated infectious agents, suggesting a possible involvement of H pylori with apoptotic process. The low expression of c-Myc and Bax in the EBV-positive groups suggests that EBV may inhibit the expression of these proteins. Nevertheless, p53 mutation shows to be a relevant alteration, independent of both infectious agents. 展开更多

METHODS: Thirty-eight patients, with premalignant gastric lesions including 18 colonic-type intestinal metaplasia (IM) and 20 mild or moderate dysplasia, were randomly divided into a treatment group (n = 19) receiving folic acid 10 mg thrice daily and a control group (n = 19) receiving sucralfate 1 000 mg thrice daily for 3 mo. All patients underwent endoscopies and four biopsies were taken prior to treatment and repeated after concluding therapy. Folate concentrations in gastric mucosa were measured with chemiluminescent enzyme immunoassay. Epithelial apoptosis and the expression of Bcl-2 and p53 protein in gastric mucosa were detected with flow cytometric assay. RESULTS: The mean of folate concentration in gastric mucosa was 9.03±3.37 μg/g wet wt in the folic acid treatment group, which was significantly higher than 6.83±3.02 μg/g wet wt in the control group. Both the epithelial apoptosis rate and the tumor suppressor p53 expression in gastric mucosa significantly increased after folic acid treatment. In contrast, the expression of Bcl-2 oncogene protein decreased after folic acid therapy. CONCLUSION: These data indicate that folic acid may play an important role in the chemoprevention of gastric carcinogenesis by enhancing gastric epithelial apoptosis in the patients with premalignant lesions. 展开更多

METHODS: The intensity of telomerase activity, apoptosis, p53 and bcl-2 expression in GISTs were detected by telomeric repeat amplification protocol, in situ end-labeling technique, and immunohistochemistry, respectively. RESULTS: The positive rates of telomerase activity of malignant GIST, potential malignant GIST and benign GIST were 85% (17/20), 22.8% (2/9) and 0 (0/9), respectively. The apoptosis indices of malignant GIST, potential malignant GIST, and benign GIST were 11.7±5.4, 30.2±5.6 and 45.2 ±7.2, respectively. The intensity of telomerase activity and apoptosis were related to the biological characteristics of GISTs (85% vs 22.8%, 0, 0; P 〈 0.01 or 11.7±5.4 vs 30.2±5.6, 45.2±7.2, 72.1±9.3; P 〈 0.05). The intensity of telomerase activity was negatively correlated with cellular apoptosis (22.9±8.4 vs 9.5±5.7, P 〈 0.01). The intensity of telomerase activity was positively correlated with/753, bcl-2 expression (40.0% vs 78.9%, 40.0% vs 84.2%; P 〈 0.05). CONCLUSION: The detection of telomerase activity, apoptosis and its control genes in GIST will be helpful for the discrimination of the malignant and benign GIST and evaluation of the prognosis. 展开更多

METHODS: Cell culture, MTT, Electromicroscopy, agarose gel electrophoresis, lucigenin, Western blot and electrophoretic mobility shift assay (EMSA) analysis were employed to investigate the effect of JTE-522 on cell proliferation and apoptosis in AGS cells and related molecular mechanisms. RESULTS: JTE-522 inhibited the growth of AGS cells and induced the apoptosis. Lucigenin assay showed the generation of ROS in cells under incubation with JTE-522. The increased ROS generation might contribute to the induction of AGS cells to apoptosis. EMSA and Western blot revealed that NF-kB activity was almost completely inhibited by preventing the degradation of IkBalpha. Additionally, by using Western blot we confirmed that the level of bcl-2 was decreased, whereas p53 showed a great increase following JTE-522 treatment. Their changes were in a dose-dependent manner. CONCLUSION: These findings suggest that reactive oxygen species, NF-kB, p53, bcl-2 and caspase-3 may play an important role in the induction of apoptosis in AGS cells after treatment with JTE-522. 展开更多

Objective: This study was designed to detect the expression of bcl-2 and p53 proteins in colorectal carcinomas and to determine their association with the patient survival and stage of the diseases. Methods: Immunohistochemistry method was used to detect the expression of bcl-2 and p53 proteins in 93 cases of colorectal carcinoma. The stain results were obtained by analyzing the clinic-pathological characteristics of patients. Results: Fifty-seven percent (53/93) of the colorectal carcinomas were bcl-2 protein positive. The positive rate of bcl-2 protein in lymph node involvement cases was lower (15/37) than the cases without node involvement (38/58, P<0.01). The positive rate of p53 protein was 43% (40/93) in colon-rectum carcinomas. No significant correlation was observed between p53 protein expression and clinic-pathological manifestations (P>0.05) but the survival was significantly worse (P=0.0001) in the p53 protein positive cases. Neither bcl-2 nor p53 alone was correlated with stage of the disease. When combined bcl-2/p53 status was analyzed, a group with bcl-2(+) and p53(?) had the best prognosis. This group was significantly associated with earlier Dukes’ stages (P=0.1763). In multivariate Cox regression analysis, lymph node involvement and p53 protein expression were two independent factors correlated with survival time. Conclusion: The expression of bcl-2 and p53 represent biological characteristics of colorectal carcinomas. Assessment of both bcl-2 and p53 status may be valuable in predicting the prognosis of patients. 展开更多
