Hypoglycaemic effect of Berberis vulgaris L.in normal and streptozotocin-induced diabetic rats 被引量:6

Objective:To achieve a primary pharmacological screening contained in the aqueous extract of Berberis vulgaris(B.vulgaris) and to examine the hypoglycaemic effect and biochemical parameters of aqueous and saponins extract on groups of rats rendered diabetic by injection of streptozotocin.Methods:The phytochemicol tests to detect the presence of different compounds were based on the visual observation of color change or formation of precipitate after the addition of specific reagents.Diabetes was induced in rats by intraperitoneal(i.p.) injection of streptozotocin(STZ) at a dose of 65 mg/kg bw.The fasting blood glucose levels were estimated by glucose oxidase-peroxidase reactive strips(Dextrostix,Bayer Diagnostics).Blood samples were taken by cutting the tip of the tail.Serum cholesterol and serum triglycerides were estimated by enzymatic DHBS colorimetric method.Results:Administration of 62.5 and 2S.0 mg/kg of saponins and aqueous extract respectively in normal rats group shows a significant hypoglycemic activity(32.33%and 40.17%respectively) during the first week.However,diabetic group treated with saponin extract produced a maximum fall of 73.1%and 76.03%at day 1 and day 21 compared to the diabetics control.Also,blood glucose levels of the diabetic rats treated with aqueous extract showed decrease of 78.79%on the first day and the effect remains roughly constant during 3 week. Both extracts also declined significantly biochemical parameters(20.77%-49.00%).The control in the loss of body weight was observed in treated diabetic rats as compared to diabetic controls. Conclusions:These results demonstrated significant antidiabetic effects and showed that serum cholesterol and serum triglycerides levels were decreased,significantly,consequently this plant might be of value in diabetes treatment. 展开更多

A new species,Berberis viridiflora X.H.Li,and a new variety,Berberis sanguinea Franch.var.viridisepala X.H.Li,L.C.Zhang & W.H.Li are described and illustrated from Baoxing County,a biodiversity hotspot located on the eastern edge of Hengduan Mountains in Sichuan Province,Southwest China,Both new taxa resemble B.sanguinea Franch.var.sanguinea,but B.viridiflora differs by the greenish flowers,and the petals being truncate,obtuse,or undulate at apex;while B.sanguinea var.viridisepala differs by the greenish or yellowish green flowers.Morphological features of the pollen grains of B.sanguinea and the two new taxa are revealed by scanning election microscope.B.sanguinea var.sanguinea displays obvious similarities with a sympatric congener,Berberis multiovula T.S.Ying in the morphology of flowers,stems and leaves,especially its ovule number varies greatly from 2 to 9,thus,B.multiovula characterized by the5-ovuled ovary is reduced to a synonymy of B.sanguinea var.sanguinea.On the basis of field surveys and study of herbarium specimens,a total of 16 species and varieties of Berberis are recorded from Baoxing County,and a key is provided.Among the 16 taxa,most phenotypic variations in Chinese Berberis can be displayed,including the habit of plants,most morphological variations of stems,branches and leaves,all types of inflorescences,all color types of flowers,and nearly all types of the shape and color of fruits.Baoxing County and its adjacent Sichuan Giant Panda Sanctuaries possibly constitute an active diversification center of Berberis in eastern Hengduan Mountains of China. 展开更多

Puccinia striiformis Westend.f.sp.tritici Erikss.(Pst)infects wheat and causes stripe rust.The rust is heteroecious with wheat as the primary uredinial and telial host and barberry(Berberis spp.)as the alternate pycnial and aecial host.More than 40 Berberis species have been identified as alternate hosts for Pst,and most of these are Chinese Berberis species.However,little is known about Berberis species or their geographic distributions in the Yunnan-Guizhou plateau in southwestern China.The Yunnan-Guizhou plateau is considered to be an important and relatively independent region for the evolution of the wheat stripe rust pathogen in China because the entire disease cycle can be completed within the region.In this study,we conducted a survey of barberry plants in the Yunnan-Guizhou plateau and identified the eight Pst-susceptible Berberis species under controlled conditions,including B.julianae,B.tsienii,B.veitchii,B.wilsonae,B.wilsonae var.guhtzunica,B.franchetiana,B.lepidifolia and B.pruinosa.These species are reported here for the first time to serve as alternate hosts for the wheat stripe rust pathogen under controlled conditions. 展开更多

Berberis species medicinal plants in Pakistan are endangered, high-value with important eco-cultural, commercial and livelihood roles in mountain communities. To assess the geographical distribution of Berberis species across the Karakoram Mountain Ranges in Pakistan, we used IUCN Red List Categories and Criteria (2001) to calculate the extent of occurrence (EOO, 〈100 km^2) and the area of occupancy (AOO, 〈10 km^2) of Berberis pseudumbellata subsp, pseudumbellata and B. pseudumbellata subsp. gilgitica. Overgrazing and habitat loss were key population- limiting factors. The two subspecies had contrasting responses to temperature, elevation, precipitation and insect susceptibility. B. pseudumbellata subsp, gilgitica is endemic to Gilgit-Baltistan and grows in single-cropping zone (ar- eas 〉 200 m a.s.1.). Status evaluation revealed that both subspecies meet the criteria set for critically endangered species. Prolonged disregard of its declining population trend might lead to its extinction; therefore, integrated con- servation efforts are necessary. 展开更多

Objective:To evaluate the antiviral activity and phytochemicals of selected plant extracts and their effect on the mitogen-activated protein kinase(MAPK)signaling pathway modulated by hepatitis C virus(HCV)nonstructural protein 5 A(NS5A).Methods:A total of ten plant extracts were initially screened for their toxicities against Hep G2 cells.The non-toxic plants were tested for their inhibitory effect on the expression of HCV NS5A at both m RNA and protein levels using real-time PCR and Western blotting assays,respectively.The differential expression of the genes associated with MAPK pathway in the presence of NS5A gene and plant extract was measured through real-time PCR.Subsequently,the identification of secondary metabolites was carried out by phytochemical and HPLC analysis.Results:The phytochemical profiling of Berberis lyceum revealed the presence of alkaloids,phenols,saponins,tannins,flavonoids,carbohydrates,terpenoids,steroids,and glycosides.Similarly,quercetin,myricetin,gallic acid,caffeic acid,and ferulic acid were identified through HPLC analysis.The methanolic extract of Berberis lyceum strongly inhibited HCV RNA replication with an IC50 of 11.44μg/m L.RT-PCR and Western blotting assays showed that the extract reduced the expression of HCV NS5A in a dosedependent manner.Berberis lyceum extract also attenuated NS5A-induced dysregulation of the MAPK signaling pathway.Conclusions:Our findings suggest that Berberis lyceum extract strongly inhibits HCV propagation by reducing HCV NS5A-induced perturbation of MAPK signaling. 展开更多

Don, an unexplored medicinal plant belonging to the family Berberidaceae, is a large deciduous shrub found in Western Himalaya between 1800-3000 m. Chemical profiling of fruit, leaf, root and stem was done by direct analysis in real time mass spectrometry followed by multivariate analysis for discrimination among the plant parts. The bioactive compounds, including magnofiorine, berberine, jatrorrhizine, thalifendine/berberrubine, demethyleneberberine, reticuline, 8-oxoberberine, N-methyltetrahydroberberine, tetrahydropalmatine, tetrahydroberberine and palmatine, were identified by their exact mass measurement and the corresponding molecular formula of each compound. A comparative study of distribution pattern for all these bioactive alkaloids showed qualitative and quantitative variations in different parts of B. petiolaris. Principal component analysis clearly dis- criminated each part of B. petiolaris plant. 展开更多

[Method] Broth microdilution method and checkerboard dilution method were used and the reference strain E. coli ATCC25922 and clinical sample separation strains of E. coli and Salmonella were selected to determine the minimum inhibitory concentration( MIC) and fractional inhibitory concentration( FIC). [Result] The MICs of Berberis julianae Schneid and Cinnamomum camphora( L.) Presl. for E. coli and Salmonella were both 1 mg /mL,and that of other antibiotics for the two strains were in the quality control range provided by NCCLS. [Conclusion] The synergistic effect or additive effect were observed when they were applied with the commonly used antibiotics. 展开更多

Barberry has played a prominent role in herbal healing for more than 2500 years. Most of the berberis species have medicinal uses because of the presence of alkaloid “berberine” an isoquinoline alkaloid. The root and inner bark are also used for yellow dyeing of clothes. The objective of the present study was to evaluate the seed dormancy, effect of stratification, light conditions and planting bed quality on seed germination of berberis manipurana seeds. A set of freshly processed seeds were sowed in three light conditions i.e., normal light (~5000 lux), poly house ca. 50% shade (~2500 lux) and poly house ca. 75% shade (~1250 lux). In the laboratory, half of the collected seeds were packed in plastic bags, labeled and stored at 4°C (stratification treatment);another half was stored at room temperature of 25°C. Stratified seeds from both the conditions (4°C and 25°C) were sowed in the seed bed at 15 days interval till 120th day (0 - 120 days) to check the viability and germination behaviors. Highest germination rate was observed from seeds stored at 4°C, which registered 82.5% (1.5) germination in the poly-bags against 70% (2.5) from seeds stored at 25°C. While 69% (2.0) and 56% (1.5) seeds germinated in the seeds beds from seeds stratified at 4°C and 25°C respectively. 展开更多

The dried roots of Berberis heteropoda Schrenk have traditionally been used to treat acute gastroenteritis and dysentery. The aim of this study was to confirm the antibacterial activity of an extract of Berberis heteropoda Schrenk root in vitro and its therapeutic effects on rats with diarrhea-predominant irritable bowel syndrome(D-IBS) in vivo, as well as to identify the related signaling pathways. A water extract of Berberis heteropoda Schrenk root(BHS) inhibited the growth of S. aureus, E. coli, P. aeruginosa and S. faecalis. BHS potentially damaged the structure of the bacterial cell membrane and decreased the activity of some membranous enzymes, eventually killing the S. aureus, E. coli, P. aeruginosa and S. faecalis bacteria. Oral administration of BHS(low, middle and high dose group, L, M and H) significantly alleviated the abdominal pain, diarrhea, and depression-like symptoms of D-IBS rats, and the efficacy index ranged from 30% to 60%, indicating that the BHS treatment was effective. BHS(L, M and H) alleviated the abnormal pathological changes in the brain, as evidenced by HE staining. The expression of CHAT, 5-HT, C-FOS and CGRP was reduced by the BHS treatment(L, M and H). Our findings provide novel insights into the use of the natural product BHS to inhibit pathogenic bacteria by destroying the bacterial structure, indicating that BHS possesses certain biological activities. Furthermore, BHS has the potential to alleviate diarrhea, abdominal pain and depression-like behaviors in D-IBS rats by regulating the brain-gut peptide levels. 展开更多

Berberis amurensis(Berberidaceae)is a traditional Chinese medicine,which is often used to treat hypertension,inflammation,dysentery and enteritis.It contains alkaloids,mainly including berberine,berbamine,magnoflorine,jatrorrhizine and palmatine.Berberis amurensis extracts(BAEs)is often orally taken.Oral herbs might be metabolized by intestinal bacteria in the small intestine.However,the interaction between the herb and the gut microbiota is still unknown.In the current study,UPLC/Q-TOFMS/MS combined with Metabolitepilot and Peakview software was used to identify the metabolites of BAEs in anti-biotic cocktail induced pseudo germ-free rats and normal rats.As a result,a total of 46 metabolites in normal rats were detected and its main metabolic pathways include demethylation,dehydrogenation,methylation,hydroxylation,sulfation and glucuronidation.Only 29 metabolites existed in pseudo germ-free rats.Dehydrogenated metabolites(M29,M30,M34 and M36),methylated metabolites(M33,M41 and M46)and other metabolites were not detected in pseudo germ-free rats.The result implied that the intestinal bacteria have an influence on the metabolism of BAEs.Furthermore,this investigation might contribute to the understanding of the metabolism of BAEs,and further promote its clinical application. 展开更多

In the present study, an enviro-friendly synthesis of silver nanoparticles from Berberis lycium Royle leaf extract and their antibacterial efficacy against five pathogenic bacteria were investigated. This biosynthesis technique is proved to be advantageous over physical and chemical methods as no toxic chemicals are used. The structural and morphological characterization was made by UV-visible spectroscopy, scanning electron microscopy, and transmission electron microscopy. The synthesized nanoparticles were oval, rectangular, and spherical in shape, size ranges from 8 to 100 nm and exhibited an absorption peak at 458 nm. The biosynthesized silver nanoparticles have shown good antibacterial effect toward tested bacteria. It is believed that these biosynthesized silver nanoparticles can play a vital role in nano-based products in future. 展开更多

(BAR) augments penile erection using in vitro and in vivo experiments. Methods: The ex vivo study used corpus cavemosum strips prepared from adult male New Zealand White rabbits. In in vivo studies for intracavernous pressure (ICP), blood pressure, mean arterial pressure (MAP), and increase of peak ICP were continuously monitored during electrical stimulation of Sprague-Dawley rats. Results: Preconstricted with phenylephrine (PE) in isolated endothelium- intact rabbit corus cavernosum, BAR relaxed penile smooth muscle in a dose-dependent manner, which was inhibited by pretreatment with NG-nitro-L-arginine methyl ester (L-NAME), a nitric oxide synthase inhibitor, and 1H-[1,2,4]-oxadiazole-[4,3-α]-quinoxalin-l-one, a soluble guanylyl cclase inhibitor. BAR significantly relaxed penile smooth muscles dose-dependently in ex vivo, and this was inhibited by pretreatment with L-NAME 1H-[1,2,4]- oxadiazole-[4,3-α ]-quinoxalin-l-one. BAR-induced relaxation was significantly attenuated by pretreatment with tetraethylammonium (TEA, P〈0.01), a nonselective K+ channel blocker, 4-aminopyridine (4-AP, P〈0.01), a voltage-dependent K+ channel blocker, and charybdotoxin (P〈0.01), a large and intermediate conductance Ca2. sensitive-K+ channel blocker, respectively. BAR induced an increase in peak ICP, ICP/MAP ratio and area under the curve dose dependently. Conclusion: BAR augments penile erection via the nitric oxide/cyclic guanosine monophosphate system and Ca2. sensitive-K+ (BK+ and IK+) channels in the corpus cavemosum. 展开更多

Berberine content was dete... [Objectives] To establish the quality standard of barberry branches. [Methods] Microscopic identification and thin layer identification were used to qualitatively identify barberry branches. Berberine content was determined by HPLC method, and the content of water, total ash, acid insoluble ash and extract was detected according to the method of the Chinese Pharmacopoeia (2020 edition). [Results] The microscopic identification showed that the features were obvious, and stone cells, cork cells, epidermal cells, stomata, fibers and catheter with reticulated pores could be found. Berberine was detected in barberry branches by thin layer chromatography, and the characteristic spots were separated clearly. Moisture, total ash, and acid insoluble ash content shall not exceed 13%, 6%, and 3%, respectively, and extract content shall not be less than 10%. Berberine hydrochloride (C_(20)H_(18)ClNO_(4)) should be calculated in the branches of Berberis wilsonae Hemsley and Berberis aggregata C. K. Schneid., and berberine (C_(20)H_(17)NO_(4)) content should not be less than 0.05%. The linear relationship was good in the range of 0.002-0.240 mg/mL ( R^(2)=0.999 5). [目的]研究喀什小檗花色素及其喷雾干燥制取微胶囊的抗氧化活性,以期为工业中应用微胶囊花色素提供一定的指导。[方法]采用DPPH自由基清除法和ABTS自由基清除法测定体外抗氧化活性,研究花色素在不同状态下清除DPPH自由基和ABTS自由基的作用。[结果]不同样品对DPPH自由基的清除能力依次为花色素>微胶囊花色素;不同样品对ABTS自由基的清除能力依次为花色素>微胶囊花色素。花色素微胶囊化前后对DPPH自由基和ABTS自由基均有一定的清除效果,但均低于维生素E的清除能力。[结论]花色素抗氧化能力较好,微胶囊花色素因为微胶囊化后花色素含量降低,协同作用降低,所以其抗氧化能力有一定程度的降低,但是花色素微胶囊化后便于贮存,由于花色素被包埋而隔离外界环境,延长了保存时间。 展开更多

黑果小檗根不同极性部位抑制肠痉挛和抗炎活性的体外研究及组分初探

目的通过体外实验探究黑果小檗根(Berberis heteropoda Schrenk roots)不同极性部位的肠道解痉作用和抗炎活性;通过成分对比分析,初步推测潜在活性组分。方法(1)用60%乙醇和水分别提取黑果小檗根的主要成分,再对乙醇提取物依次进行系统... 目的通过体外实验探究黑果小檗根(Berberis heteropoda Schrenk roots)不同极性部位的肠道解痉作用和抗炎活性;通过成分对比分析,初步推测潜在活性组分。方法(1)用60%乙醇和水分别提取黑果小檗根的主要成分,再对乙醇提取物依次进行系统溶剂萃取(石油醚、二氯甲烷、乙酸乙酯和饱和正丁醇),得到不同极性萃取物。(2)采用高效凝相色谱法(HPLC法)对不同极性提取物中代表性组分进行定量分析并比较其含量差异。(3)通过小鼠离体结肠平滑肌实验,研究不同提取物对正常和痉挛状态下肠道肌肉张力的影响,筛选具有抑制肠痉挛作用的活性部位。(4)采用脂多糖(LPS)诱导RAW 264.7细胞建立体外炎症细胞模型,通过不同提取物干预后测定细胞上清液中一氧化氮(NO)和白介素-6(IL-6)含量,筛选抗炎活性部位。(5)采用半制备液相色谱法对潜在活性部位进行成分分离,并用核磁共振法和质谱法进行成分定性。结果(1)HPLC成分对比分析表明,饱和正丁醇萃取物中木兰花碱、非洲防己碱、药根碱的含量高于其他萃取物,二氯甲烷萃取物中小檗胺、阿魏酸、巴马汀和小檗碱的含量高于其他萃取物。此外,发现乙酸乙酯萃取物中含有明显区别于其他萃取物的的未知峰(峰面积占比60%)。(2)小鼠结肠张力的体外实验结果表明,二氯甲烷萃取物对正常小鼠结肠张力有明显的抑制作用。此外,二氯甲烷萃取物可缓解乙酰胆碱和氯化钡引起的肠痉挛。(3)炎症细胞模型干预实验的结果表明,乙酸乙酯萃取物的抗炎作用明显优于其他萃取物。(4)从乙酸乙酯萃取物中分离得到了先前发现的未知峰化合物,结果鉴定为阿魏酸-4-O-葡萄糖苷。结论研究发现黑果小檗根二氯甲烷部位同比其他部位的肠道解痉作用最强,同时乙酸乙酯部位是发挥抗炎作用的主要活性部位,以此丰富了黑果小檗根已知的药理作用。此外,从黑果小檗根中首次分离得到了化合物:阿魏酸-4-O-葡萄糖苷,为黑果小檗根的物质基础研究提供了参考。 展开更多

Population Diversity of Puccinia graminis is Sustained Through Sexual Cycle on Alternate Hosts 被引量:2

A high degree of virulence diversity has been maintained in the population of Puccinia graminis f. sp. tritici (Pgt) in northwestern United States. Although Berberis vulgaris is present in the region and Pgt has been isolated from aecial infections on B. vulgaris, the population is too diverse to be explained by the limited presence of B. vulgaris alone. Since 2008, we have isolated P. graminis from aecial infections on fruits of Mahonia repens and Mahonia aquifolium from northwestern United States. These two native woody shrub species, widely distributed in western North America, were once classified as resistant to P. graminis based on artificial inoculations. By isolating P. graminis from aecia, we established that M. repens and M. aquifolium along with B. vulgaris (albeit infrequent) serve as the alternate hosts ofP. graminis in the region. The isolates of P. graminis from Mahonia of North America had diverse virulence patterns and most of the isolates could be differentiated on Morocco, Line E, Chinese Spring, Little Club, LMPG-6, Rusty, and other genotypes that are considered to be universally susceptible to most Pgt isolates. This discovery explained the persistence of virulence diversity of Pgt observed in isolates derived from uredinia on cereal crops in the region. In addition to cereal crops, uredinial stage of the P. graminis population is sustained by wild grasses, especially Elymus glaucus, a native grass sharing the same habitat with the rusted Mahonia spp. Although virulence to some important stem rust resistance genes was observed in some isolates derived from Mahonia of North America when tested against single stem rust resistance gene stocks, the overall virulence is very limited in these isolates. This is likely a result of limited selection pressure on the rust population. In contrast to northwestern United Sates, the Pgt population in east of the Rocky Mountains of North America has declined steadily with a single race, QFCSC, being predominant in the last decade. This decline is likely due to a combination of factors, of which a lack of sexual recombination in the region is perhaps the most important one. 展开更多

以冰川红叶小檗和紫叶小檗当年生枝条为试验材料,于12月和3月测定其7个处理温度(4℃、-10℃、-20℃、-30℃、-35℃、-40℃、-50℃)下的相对电导率、电阻抗参数值以及3个处理温度(-10℃、-20℃、-40℃)下的萌芽率,并对电导法、电阻抗图... 以冰川红叶小檗和紫叶小檗当年生枝条为试验材料,于12月和3月测定其7个处理温度(4℃、-10℃、-20℃、-30℃、-35℃、-40℃、-50℃)下的相对电导率、电阻抗参数值以及3个处理温度(-10℃、-20℃、-40℃)下的萌芽率,并对电导法、电阻抗图谱法及恢复生长法的测定结果进行相关性分析。结果表明,不同低温处理下的冰川红叶小檗当年生枝条的相对电导率均低于紫叶小檗;12月份冰川红叶小檗当年生枝条用相对电导率,胞外电阻率(r e)及低频电阻率(r 1)拟合的半致死温度分别为-34.84℃、-31.46℃和-29.47℃,紫叶小檗拟合的半致死温度分别为-28.37℃、-27.12℃和-26.07℃。3月份冰川红叶小檗当年生枝条用相对电导率,胞外电阻率(r e)及低频电阻率(r1)拟合的半致死温度分别为-31.94℃、-29.94℃和-27.98℃,紫叶小檗拟合的半致死温度分别为-26.84℃、-26.08℃和-25.43℃,证明冰川红叶小檗的半致死温度均低于紫叶小檗的半致死温度;12月份不同低温处理下的当年生枝条冰川红叶小檗的萌芽率为85.79%、74.33%和14.77%,紫叶小檗的萌芽率为75.74%、57.65%和8.06%;3月份不同低温处理下的当年生枝条冰川红叶小檗的萌芽率为88.80%、77.77%和16.16%,紫叶小檗的萌芽率为78.92%、68.37%和12.38%。由此表明,冰川红叶小檗的萌芽率均高于紫叶小檗,冰川红叶小檗的抗寒性高于紫叶小檗。 展开更多
