After breeding,Black-necked Grebes(Podiceps nigricollis)perform a moult-migration to autumn hypersaline staging sites,where they moult the flight feathers and forage on superabundant brine shrimp(Artemia spp.)before l...After breeding,Black-necked Grebes(Podiceps nigricollis)perform a moult-migration to autumn hypersaline staging sites,where they moult the flight feathers and forage on superabundant brine shrimp(Artemia spp.)before leaving for wintering areas.During the stay in moulting sites,the grebes experience changes in organs and muscle size(atrophy,hypertrophy),and almost double their body mass,which has been suggested to act as an insurance against a collapse in prey availability in late autumn.During two years we collected blood samples from hundreds of individuals at one of the most important European moulting sites(the Odiel marshes,SW Spain),which is a highly polluted area due to mining drainage and chemical industry.We assessed the potential effect of moulting stage,day of the year and body condition on 16 blood biochemical parameters.Because of the changes in prey availability and body composition of grebes,we expected some physiological adjustments during moult.Elevated levels of cholesterol suggested that birds in active moult increased foraging effort to face the costs of moulting.There was increased amount of lactate dehydrogenase,corresponding to periods of breast muscle atrophy.Birds in active moult augmented protein ingestion,likely to account for the requirements of feather growth.We also show that the probability of fasting due to low prey availability increased late in the moulting season,as demonstrated by an increase in plasmaβ-hydroxy-butyrate.展开更多

However, clinically applying M4 faces ...Conditionally replication adenovirus M4, which was constructed in our lab, was proved to have good clinical application prospect for its good antitumor and antimetastasis effect. However, clinically applying M4 faces many problems. One of the most important is the safety of M4. In this study, we investigated the safety of M4 by comparing with Adv-TK, which was proved to be safe in Ⅰ–Ⅲ phase clinical trials. M4 and Adv-TK were injected into mice via the tail vein separately, and the mice were sacrificed at the indicated time. Blood was collected for biochemical tests, the liver was harvested for hematoxylin and eosin (H&E) staining and viral quantification, and splenic lymphocytes were separated for adenovirus specific cellular immune response. Our results showed that M4 had no obvious effect on mouse general symptoms. A transient reversible infiltration of inflammatory cells in collect abbacy was only observed in M4 group, and a transient slight increase in Cr level was detected both after M4 and Adv-TK injection. The adenovirus specific cellular immune response induced by M4 was similar to that by Adv-TK, and the distribution and metabolism of M4 in the mouse liver were also similar to those of Adv-TK. It was concluded that conditionally replication adenovirus M4 had the same safety as Adv-TK. The study provides safety basis for the coming clinical trials of M4.展开更多
The onsite next generation sequencing(NGS)of Ebola virus(EBOV)genomes during the 2013–2016 Ebola epidemic in Western Africa provides an opportunity to trace the origin,transmission,and evolution of this virus.Herein,...The onsite next generation sequencing(NGS)of Ebola virus(EBOV)genomes during the 2013–2016 Ebola epidemic in Western Africa provides an opportunity to trace the origin,transmission,and evolution of this virus.Herein,we have diagnosed a cohort of EBOV patients in Sierra Leone in 2015,during the late phase of the outbreak.The surviving EBOV patients had a recovery process characterized by decreasing viremia,fever,and biochemical parameters.EBOV genomes sequenced through the longitudinal blood samples of these patients showed dynamic intra-host substitutions of the virus during acute infection,including the previously described short stretches of 13 serial TNC mutations.Remarkably,within individual patients,samples collected during the early phase of infection possessed Ts at these nucleotide sites,whereas they were replaced by Cs in samples collected in the later phase,suggesting that these short stretches of TNC mutations could emerge independently.In addition,up to a total of 35 nucleotide sites spanning the EBOV genome were mutated coincidently.Our study showed the dynamic intra-host adaptation of EBOV during patient recovery and gave more insight into the complex EBOV-host interactions.展开更多
