AIM: To investigate the antifibrotic effects of bone morphogenetic protein-7 (BMP-7) on Schistosoma japonicum (S. japonicum )-induced hepatic fibrosis in BALB/C mice. METHODS: Sixty BALB/C mice were randomly divided into three groups, including a control group (group A, n = 20), model group (group B, n = 20) and BMP-7 treated group (group C, n = 20). The mice in group B and group C were abdominally infected with S. japonicum cercariae to induce a schistosomal hepatic fibrosis model. The mice in group C were administered human recombinant BMP-7. Liver samples were extracted from mice sacrificed at 9 and 15 wk after modeling. Hepatic histopathological changes were assessed using Masson's staining. Transforming growth factor-beta 1 (TGF-β1), alpha-smooth muscle actin (α-SMA), phosphorylated Smad2/3 (pSmad2/3) and Smad7 protein levels and localization were measured by Western blotting and immunohistochemistry, respectively, and their mRNA expressions were detected by reverse transcriptionpolymerase chain reaction (RT-PCR). RESULTS: The schistosomal hepatic fibrosis mouse model was successfully established, as the livers of mice in group B and group C showed varying degrees of typical schistosomal hepatopathologic changes such as egg granuloma and collagen deposition. The degree of collagen deposition in group C was higher than that in group A (week 9: 22.95±6.66vs 2.02±0.76; week 15: 12.84±4.36 vs 1.74±0.80; P<0.05), but significantly lower than that in group B (week 9: 22.95±6.66 vs 34.43±6.96; week 15: 12.84±4.36 vs 18.90±5.07;P<0.05) at both time points. According to immunohistochemistry data, the expressions of α-SMA, TGF-β1 and pSmad2/3 protein in group C were higher than those in group A (α-SMA: week 9: 21.24±5.73 vs 0.33±0.20; week 15: 12.42±4.88 vs 0.34±0.27; TGF-β1: week 9: 37.00±13.74 vs 3.73±2.14; week 15: 16.71±9.80 vs 3.08±2.35; pSmad2/3: week 9: 12.92±4.81 vs 0.83±0.48; week 15: 7.87±4.09 vs 0.90±0.45; P<0.05), but significantly lower than those in group B (α-SMA: week 9: 21.24±5.73 vs 34.39±5.74; week 15: 12.42±4.88 vs 25.90±7.01; TGF-β1: week 9: 37.00±13.74 vs 55.66±14.88; week 15: 16.71±9.80 vs 37.10±12.51; pSmad2/3: week 9: 12.92±4.81 vs 19.41±6.87; week 15: 7.87±4.09vs 13.00±4.98;P<0.05) at both time points; the expression of Smad7 protein in group B was higher than that in group A and group C at week 9 (8.46±3.95 vs 1.00±0.40 and 8.46±3.95 vs 0.77±0.42; P<0.05), while there were no differences in Smad7 expression between the three groups at week 15 (1.09±0.38 vs 0.97±0.42 vs 0.89±0.39; P>0.05). Although minor discrepancies were observed, the results of RT-PCR and Western blotting were mainly consistentwith the immunohistochemical results. CONCLUSION: Exogenous BMP-7 significantly decreased the degree of hepatic fibrosis in both the acute and chronic stages of hepato-schistosomiasis, and the regulatory mechanism may involve the TGF-β/Smad signaling pathway. 展开更多

Improved Osteointegration of Ti-6Al-4V-implants of Different Surface Texture by the Use of Bone Morphogenetic Protein-3 (BMP-3)

Half of altogether 60 cylindrical implant devices made of titanium-aluminum-vanadium alloy ( Ti-6Al-4V) were plusna-sprayed with a hydroxyapatite-couting and the other half had a corundum blasted porous surface. 15 implants of each group of the titanium test buplants were coated with 230 μg porcine, high-purified BMP- 3-precipitute per implant. In each case a BMP- 3-couted and an uncoated control-device were implanted into the femoral part of the putellofemoral joint of the right and left leg of 30 adult giant rabbits. Histomorphological and histomorphometrical we found in both groups with BMP- 3-coated test devices an improved osteointegrution. Stutistical evaluation using the t-test for matched samples showed 5 weeks after surgery a significant higher volume of tony formed bone of the BMP- 3-coated corundum- blasted or hydroxyapathe- coated Ti- 6Al- 4 V test devices compared to the non-couted controls of the same t)pe (p 〈 0.01, t-test for matched samples). In both implant groups with BMP-couting a synergetic effect was verifiable although the bone ongrowth in the hydroxyaputite coated implants was more extensive than in the corundum blasted implants. Light microscopy demonstrated osteointegrution without connective tissue membrane around the surface of the implants. Our results indicate that composite metal implants,as used in endoprosthetics and implantology , are suitable carriers for BMP- 3 and im proved fixation of the implants can be achieved. The hydroxyapatite surface is superior to the corundum-blasted surface with regards to the observed parameters because of its pronounced bioactivity and its osteoconductive characteristics. 展开更多

目的:探究异氟醚通过调节骨发生形态蛋白4(BMP4)及其下游Smad蛋白对自发性高血压大鼠(SHR)脑损伤的作用机制。方法:采用随机数字表法将48只SHR分为模型组、异氟醚组、阳性药物组、异氟醚+LDN193189组,健康大鼠作为健康对照组,每组12只... 目的:探究异氟醚通过调节骨发生形态蛋白4(BMP4)及其下游Smad蛋白对自发性高血压大鼠(SHR)脑损伤的作用机制。方法:采用随机数字表法将48只SHR分为模型组、异氟醚组、阳性药物组、异氟醚+LDN193189组,健康大鼠作为健康对照组,每组12只。各组大鼠分别放入氧气箱中,健康对照组、模型组、阳性药物组大鼠通入特殊气体(30%O_(2)、70%N_(2))1 h,异氟醚组、异氟醚+LDN193189组通入混杂2%异氟醚的特殊气体1 h,通过麻醉气体检测仪检测异氟醚含量,确保异氟醚浓度始终保持在2%。通气结束后,异氟醚+LDN193189组大鼠腹腔内注射BMP4/Smad通路抑制剂溶液20μL(LDN193189,5 mg/kg),阳性药物组大鼠腹腔内注射缬沙坦溶液20μL(10 mg/kg),健康对照组、模型组、异氟醚组大鼠腹腔内注射等量的生理盐水。氧气箱通气实验及注射每日1次,持续10 d。无创血压检测仪检测尾动脉收缩压变化;神经学行为评分评价行为功能;脑含水量检测脑水肿程度;原位末端转移酶标记法(TUNEL)染色和尼氏染色观察海马组织病理学变化及神经元损伤情况;荧光定量聚合酶链式反应(PCR)及蛋白免疫印迹(Western Blot)法检测海马组织BMP4、Smad2 mRNA及蛋白表达水平。结果:与模型组比较,异氟醚组、阳性药物组、异氟醚+LDN193189组大鼠海马神经细胞凋亡减少,尼氏小体增多,血压、神经学评分、脑组织含水量显著降低,BMP4、Smad2 mRNA及蛋白表达量显著升高(P<0.05);与异氟醚组比较,异氟醚+LDN193189组大鼠神经细胞凋亡增多,尼氏小体急剧减少,血压、神经学评分、脑组织含水量显著升高,BMP4、Smad2 mRNA及蛋白表达量显著降低(P<0.05);异氟醚组与阳性药物组各项数据差异均无统计学意义(P>0.05)。结论:异氟醚可通过促进BMP4/Smad信号通路减轻SHR脑损伤。 展开更多

目的:探讨帕立骨化醇对肾性骨病(ROD)大鼠骨代谢及转化生长因子-β(TGF-β)/骨形态发生蛋白-7(BMP-7)/Smad通路的影响。方法:将90只大鼠随机分为6组:对照组、模型组、帕立骨化醇低剂量(0.2μg/kg)组、帕立骨化醇中剂量(0.4μg/kg)组、... 目的:探讨帕立骨化醇对肾性骨病(ROD)大鼠骨代谢及转化生长因子-β(TGF-β)/骨形态发生蛋白-7(BMP-7)/Smad通路的影响。方法:将90只大鼠随机分为6组:对照组、模型组、帕立骨化醇低剂量(0.2μg/kg)组、帕立骨化醇中剂量(0.4μg/kg)组、帕立骨化醇高剂量(0.8μg/kg)组、骨化三醇(10μg/kg)组,每组15只,对照组大鼠以普通饲料喂养,其余各组大鼠用含有腺嘌呤的饲料喂养,诱导建立ROD模型。分组进行药物治疗后,测定各组大鼠肾功能指标血尿素氮(BUN)与血肌酐(Scr)水平、血钙与血磷水平、股骨骨密度(BMD)、股骨生物力学指标最大载荷、弹性模量与屈服载荷、血清炎症因子IL-6、IL-17水平;采用蛋白免疫印迹法检测骨组织TGF-β/BMP-7/Smad通路蛋白表达情况。再次取45只大鼠随机分为3组:帕立骨化醇(0.8μg/kg)组、TGF-β抑制(LY2157299,150 mg/kg)组、帕立骨化醇(0.8μg/kg)+TGF-β抑制(LY2157299,150 mg/kg)组,每组15只,同样方法建立ROD模型。分组以药物治疗后,测定各组大鼠肾功能指标与股骨生物力学指标水平。结果:与对照组比较,模型组大鼠血钙水平、BMD、弹性模量、最大载荷、屈服载荷、骨组织TGF-β/BMP-7/Smad通路蛋白TGF-β及BMP-7表达、p-Smad3/Smad3显著降低(P<0.05),BUN与Scr水平、血磷水平、血清IL-6与IL-17水平显著升高(P<0.05)。与模型组比较,帕立骨化醇低、中、高剂量组和骨化三醇组大鼠血钙水平、BMD、弹性模量、最大载荷、屈服载荷、骨组织TGF-β/BMP-7/Smad通路蛋白TGF-β及BMP-7表达、p-Smad3/Smad3均升高,BUN与Scr水平、血磷水平、血清IL-6与IL-17水平均降低,且帕立骨化醇各组之间呈剂量依赖性(P<0.05),帕立骨化醇高剂量组和骨化三醇组比较,大鼠各指标差异无统计学意义(P>0.05)。与帕立骨化醇+TGF-β抑制组比较,帕立骨化醇组大鼠肾功能指标BUN、Scr与血磷水平降低(P<0.05),血钙水平、BMD、弹性模量、最大载荷、屈服载荷升高(P<0.05);TGF-β抑制组大鼠肾功能指标BUN、Scr与血磷水平升高(P<0.05),血钙水平、BMD、弹性模量、最大载荷、屈服载荷降低(P<0.05)。结论:帕立骨化醇可通过激活TGF-β/BMP-7/Smad信号通路抑制炎症反应,改善ROD大鼠肾功能及骨代谢异常,降低血磷水平,提高血钙水平及骨密度,修复骨生物力学,改善骨病症状。 展开更多

Aim To determine the effect of local simvastatin application on the mRNA expression level of transforming growth factor-β1 (TGF-β1), bone morphogenetic protein-2 (BMP-2) and vascular endothelial growth factor (VEGF) in the tooth sockets of rat. Methodology Forty-eight male Wistar rats were randomly divided into experimental and control groups (n=24). Polylactic acid/polyglycolic acid copolymer carriers, with or without simvastatin, were implanted into extraction sockets of right mandibular incisors. The expression of TGF-β1, BMP-2 and VEGF mRNA was determined by in situ hybridization in the tooth extraction socket at five days, one week, two weeks and four weeks after implantation. Results The fusiform stroma cells in the tooth extraction socket began to express TGF-β1, BMP-2 and VEGF mRNA in both experimental and control groups from one week after tooth extraction until the end of experiment. The expression of TGF-131 and BMP-2 mRNA in the experimental group was significantly up-regulated after one, two and four weeks, and expression of VEGF mRNA was significantly increased after one and two weeks compared with that in the control group. Conclusion The findings indicate that local administration of simvastatin can influence alveolar bone remodeling by regulating the expression of a school of growth factors which are crucial to osteogenesis in the tooth extraction socket. 展开更多

Background: Autologous periosteal grafting is used as treatment for articular cartilage defect. Objective: To study the effect of bone morphogenetic protein (BMP) on articular cartilage regeneration following periosteal grafting. Methods: 16 healthy 15 week-old New Zealand white rabbits of both sexes (32 knees) were randomly divided into experimental group (group A) and control group (group B). A4.0 mmdiameter full-thickness articular cartilage defect was created in the femoral intercondylar fossa in all rabbits. Following this, a4.0 mmdiameter section of the periosteum was harvested from the anteromedial part of the upper tibial bone. In group A (eight rabbits, 16 knees), the cartilage defect was covered with periosteum, into which 20 μg BMP and 20% Pluronic were injected. In group B (eight rabbits, 16 knees), the cartilage defect was covered with periosteum, into which the same dosage of 0.9% NS (Normal saline) and 20% Pluronic were injected. All rabbits were sacrificed at 4, 8, and 12 weeks postoperatively, the cartilage defect areas were examined macroscopically and microscopically, and the morphology of the chondrocytes and collagen fibers were examined by scanning electron microscopy. Results: The filling of the defects with regenerated tissue was observed in both the group. The most notable improvement was that the cartilage regeneration in group A was obviously superior to that in group B, with the total histological score in group A significantly higher. 骨质疏松性椎体压缩性骨折患者血清BMP-2,Runx2水平对术后骨折延迟愈合的预测价值

目的探讨骨质疏松性椎体压缩性骨折(Osteoporotic vertebral compressibility fracture,OVCF)患者血清骨形态发生蛋白-2(Bone morphogenetic protein-2,BMP-2)、核心结合因子2(Runt-related transcription factor 2,Runx2)水平对术后骨... 目的探讨骨质疏松性椎体压缩性骨折(Osteoporotic vertebral compressibility fracture,OVCF)患者血清骨形态发生蛋白-2(Bone morphogenetic protein-2,BMP-2)、核心结合因子2(Runt-related transcription factor 2,Runx2)水平对术后骨折延迟愈合的预测价值。方法选取2020年1月-2023年4月徐州医科大学附属淮安医院骨科收治的260例OVCF患者为研究对象,均采用经皮椎体成形术(Percutaneous vertebroplasty,PVP)或椎体后凸成形术(Percutanous kyphoplasty,PKP)治疗,根据术后3个月骨折愈合情况分延迟愈合组、正常愈合组。分析OVCF患者术后骨折延迟愈合的影响因素及血清BMP-2,Runx2水平对术后骨折延迟愈合的预测价值。绘制决策曲线和临床影响曲线,评价血清BMP-2,Runx2水平对术后骨折延迟愈合的价值。结果260例患者术后无失访,22例出现骨折延迟愈合,发生率为8.46%(22/260)。延迟愈合组年龄、吸烟比率、合并糖尿病比率、合并甲状腺疾病比率、有椎体裂隙征比率大于正常愈合组,骨密度T值、术前和术后3个月血清BMP-2,Runx2水平小于正常愈合组,差异有统计学意义(P<0.05)。合并糖尿病、有椎体裂隙征是OVCF患者术后骨折延迟愈合的独立危险因素,骨密度T值、BMP-2,Runx2是OVCF患者术后骨折延迟愈合的独立保护因素(P<0.05)。BMP-2,Runx2联合预测OVCF患者术后骨折延迟愈合对应AUC值为0.856。阈值在0.10~0.63范围内,BMP-2,Runx2联合评估模型评估OVCF患者术后骨折延迟愈合的净受益率优于单独检测。在阈值概率为0.26时,被该模型划分入高风险的人数与真阳性人数基本达成一致。结论OVCF患者血清BMP-2,Runx2水平降低,二者联合检测对术后骨折延迟愈合具有较高的预测效能。 展开更多

OBJECTIVE:To investigate the effects of acupotomy on the subchondral bone absorption and mechanical properties in rabbits with knee osteoarthritis(KOA).METHODS:The rabbits were divided into blank control,model,acupotomy and electroacupuncture(EA)groups,with 12 rabbits in each.Modified Videman's method was used to prepare KOA model.The acupotomy and EA group were given indicated intervention for 3 weeks.The behavior of rabbits in each group was recorded.Subsequently,cartilage–subchondral bone units were obtained and morphological changes were observed by optical microscope and micro computed tomography.Compression test was used to detect the mechanical properties of subchondral bone,Western blot and real-time polymerase chain reaction(RT-PCR)were applied to detect the expression of bone morphogenetic protein 2-Smad1(BMP2-Smad1)pathway in subchondral bone.RESULTS:Compared with the control group,rabbits in the KOA group showed lameness,knee pain,and cartilage degradation;the subchondral bone showed active resorption,the mechanical properties decreased significantly and the BMP2-Smad1 pathway downregulated significantly.Both acupotomy and EA intervention could increase the thickness of trabecular bone(Tb.Th),the bone volume fraction(BV/TV)and the thickness of subchondral bone plate,reduce the separation of trabecular bone(Tb.Sp),improve the maximum load and elastic modulus of subchondral bone,and effectively delay cartilage degeneration in KOA rabbits.This process may be achieved through upregulation the related proteins of BMP2-Smad1 pathway.The maximum load and elastic modulus of subchondral bone in the acupotomy group were slightly better than those in the EA group.CONCLUSIONS:Acupotomy could effectively protect cartilage by inhibiting abnormal bone resorption and improving mechanical properties of subchondral bone thorough the related proteins of BMP2-Smad1 pathway in KOA rabbits. 展开更多

目的探讨骨化三醇胶囊(Cal)通过骨形态发生蛋白2(BMP-2)/SMAD/Runt相关转录因子2(Runx2)通路对大鼠胫骨骨折愈合的影响及相关机制。方法将52只SD大鼠随机分为对照组(Cont)、胫骨骨折组、Cal组(灌胃10 ng/mL Cal,连续4周)、Cal+Noggin(BM... 目的探讨骨化三醇胶囊(Cal)通过骨形态发生蛋白2(BMP-2)/SMAD/Runt相关转录因子2(Runx2)通路对大鼠胫骨骨折愈合的影响及相关机制。方法将52只SD大鼠随机分为对照组(Cont)、胫骨骨折组、Cal组(灌胃10 ng/mL Cal,连续4周)、Cal+Noggin(BMP特异抑制剂)组(灌胃10 ng/mL Cal+经皮注射50 ng/mL Noggin,连续4周)。采用X射线、全自动生化分析仪、micro CT、HE染色、Western blot分析治疗效果及可能机制。结果4周后,胫骨骨折组骨折线明显,Cal组骨折线几乎不可见,断端有大量骨痂,Cal+Noggin组骨折愈合较差。胫骨骨折组骨密度、骨体积分数、ALP、OCN、PINP水平及BMP-2、Runx2、p-SMAD1/5/8/SMAD1/5/8表达高于Cont组,骨小梁分离度低于Cont组(P<0.05)。较于胫骨骨折组,Cal组骨密度、骨体积分数、ALP、OCN、PINP水平及BMP-2、Runx2、p-SMAD1/5/8/SMAD1/5/8表达增高,骨小梁分离度降低(P<0.05)。较于Cal组,Cal+Noggin组骨密度、骨体积分数、ALP、OCN、PINP水平及BMP-2、Runx2、p-SMAD1/5/8/SMAD1/5/8表达降低,骨小梁分离度增加(P<0.05)。结论Cal可能通过激活BMP-2/SMAD/Runx2通路,改善新生骨痂骨密度及微结构,促进胫骨骨折愈合。 展开更多

目的:通过实验研究观察肾衰泻浊丸对腺嘌呤致慢性肾衰竭(CRF)大鼠BMP-7/Smads/TGF-β1信号转导通路的影响,探讨肾衰泻浊丸延缓CRF进展及抗肾间质纤维化的机制。方法:采用腺嘌呤致CRF大鼠模型,观察实验大鼠治疗后血清BUN、Scr的变化及肾... 目的:通过实验研究观察肾衰泻浊丸对腺嘌呤致慢性肾衰竭(CRF)大鼠BMP-7/Smads/TGF-β1信号转导通路的影响,探讨肾衰泻浊丸延缓CRF进展及抗肾间质纤维化的机制。方法:采用腺嘌呤致CRF大鼠模型,观察实验大鼠治疗后血清BUN、Scr的变化及肾脏病理变化;采用免疫组织化学法检测肾组织中TGF-β1,Smad6及BMP-7蛋白表达;采用Western Blot法检测肾组织内的Smad6、BMP-7蛋白的表达,采用半定量RT-PCR检测肾组织内的TGF-β1 mRNA的表达水平。结果:肾衰泻浊丸能够明显降低CRF大鼠血清BUN、Scr水平,与对照组比较差异有统计学意义;能够减轻肾小管间质损伤;能够降低肾组织中TGF-β1蛋白的表达,增加Smad6,BMP-7蛋白的表达,与对照组比较差异有统计学意义(P<0.05);能够下调CRF大鼠肾组织中TGF-β1 mRNA的表达,与对照组比较差异有统计学意义(P<0.05)。结论:肾衰泻浊丸可能是通过升高Smad6,BMP-7蛋白的表达,降低TGF-β1的蛋白表达,从而抑制了TGF-β1信号向细胞核内转导的通路;肾衰泻浊丸通过影响BMP-7/Smads/TGF-β1信号通路的转导而减轻肾间质纤维化可能是其延缓CRF进展的机制之一。 展开更多

目的探讨骨形态蛋白2(BMP-2)/Smads信号通路在低剂量X射线促成骨细胞分化及矿化中的作用。方法将成骨细胞分为照射组及对照组,照射组给予单次0.1 Gy X射线照射,对照组处于相同的环境下不进行X射线照射。照射后24、48、72 h,采用CCK-8法... 目的探讨骨形态蛋白2(BMP-2)/Smads信号通路在低剂量X射线促成骨细胞分化及矿化中的作用。方法将成骨细胞分为照射组及对照组,照射组给予单次0.1 Gy X射线照射,对照组处于相同的环境下不进行X射线照射。照射后24、48、72 h,采用CCK-8法检测细胞增殖活性,采用对硝基苯磷酸二钠法检测细胞碱性磷酸酶(ALP)活性,采用茜素红染色剂进行细胞矿化染色、十六烷基吡啶试剂进行定量;利用RT-PCR检测BMP-2、Smad1、Smad5、Smad4、成骨相关因子2(Runx2)、锌指结构转录因子(Osterix)及骨钙素(OCN)的mRNA表达;另取成骨细胞分为0.1 Gy组、0.1 Gy拮抗剂组、control组及control拮抗剂组。0.1 Gy组、control组处理同上,两个拮抗剂组分别在0.1 Gy组及control组处理的基础上加入浓度为10μg/m L的头蛋白(NOGGIN)拮抗剂0.5 mg,照射72 h后,分别采用Western blotting及ELISA法检测上述各指标蛋白的表达,并进行各组间的比较。结果照射组与对照组各时间成骨细胞增殖活性比较差异无统计学意义(P均>0.05)。与对照组比较,照射后48 h,照射组ALP活性升高(P<0.05);照射后72 h,照射组细胞矿化定量增加(P<0.05);照射后48 h,照射组BMP-2、Smad1、Smad5、Smad4、Runx2、Osterix及OCN的mRNA表达上调(P均<0.05);照射后72 h,照射组Smad1 mRNA相对表达量高于对照组(P<0.05)。照射后72 h,0.1 Gy组BMP-2、Smad1、Smad4、Smad5、Runx2、Osterix及OCN的蛋白相对表达量高于control组(P均<0.05);0.1 Gy拮抗剂组与control拮抗剂组上述各指标蛋白表达均下降,两组间各指标蛋白表达差异无统计学意义(P均>0.05)。结论 BMP-2蛋白在低剂量X射线照射的成骨细胞中表达升高,其可通过其自分泌/旁分泌方式激活细胞Smads信号通路进而调控下游成骨基因的表达,从而促进成骨细胞分化与矿化。 展开更多
