BACKGROUND Toxic epidermal necrolysis(TEN)is a life-threatening dermatological emergency mainly induced by drug hypersensitivity reactions.Standard management includes discontinuation of culprit drug and application of immunomodulatory therapy.However,mortality remains high due to complications like septic shock and multiorgan failures.Innovative approaches for skin care are crucial.This report introduces borneol-gypsum,a traditional Chinese drug but a novel dressing serving as an adjuvant of TEN therapy,might significantly improve skin conditions and patient outcomes in TEN.CASE SUMMARY A 38-year-old woman diagnosed with eosinophilic granulomatosis with polyangiitis experienced gangrenous complications and motor nerve involvement.After initial treatment of high-dose corticosteroids and cyclophosphamide,symptom of foot drop improved,absolute eosinophil counts decreased,while limb pain sustained.Duloxetine was added to alleviate her symptom.Subsequently,TEN developed.Additional topical application of borneol-gypsum dressing not only protected the skin lesions from infection but also significantly eased localized pain.This approach demonstrated its merit in TEN management by promoting skin healing and potentially reducing infection risks.CONCLUSION Borneol-gypsum dressing is a promising adjuvant that could significantly improve TEN management,skin regeneration,and patient comfort. 展开更多

In this review, the mechanism of borneol in promoting permeation by destroying the highly ordered lipid structure of the lipid layer and by destroying the hydrogen-bond network was described. The application of borneol in promoting the transdermal absorption of the active ingredients of traditional Chinese medicine and chemical drugs was introduced. The application of borneol as a natural ingredient added to functional cosmetics was summarized, and its effects on skin-spot treatment, acne skin care, eczema skin care, skin repair and anti-oxidation were introduced. Finally, the possible problems in the application of borneol in cosmetics were put forward, and the application prospect of borneol in the development of cosmetics was given. 展开更多

Hyperforin is an antidepressant drug that has unstable therapeutic effects, due to its poor ability to cross the blood-brain barrier. Borneol and electroacupuncture have both been found to increase the permeability of the blood-brain barrier. As such, the current study examined the distribution of hyperforin in the rat brain, and the effects on the brain distribution of hyperforin of borneol alone (orally administered), and borneol combined with electroacupuncture treatment. High-performance liquid chromatography technology and pharmacokinetic analysis revealed that treatment with borneol alone (300, 600 mg/kg) increased peak concentration and the area under the curve for hyperforin in the brain. In addition, the bioavailability of hyperforin in rat brain increased by 42.7%. However, increasing the dose of borneol dose did not appear to increase the distribution of hyperforin in the brain. Importantly, applying electroacupuncture at Baihui (GV 20) or Yamen (GV 15) appeared to enhance the brain-delivery effects of borneol, although this effect was weak. Overall, our results indicated that borneol alone or combined with electroacupuncture can provide promising strategies for brain-targeted delivery in central nervous system therapy. 展开更多

The present study collected cerebrospinal fluid samples from the corpus striatum in rats treated with borneol, moschus, storax, and acorus tatarinowii using brain microdialysis technology. Levels of excitatory neurotransmitters aspartic acid and glutamate, as well as inhibitory neurotransmitters glycine and ^-aminobutyric acid, were measured in samples using reversed-phase high-performance liquid chromatography, phosphate gradient elution, and fluorescence detection. Results showed that concentrations of all four amino acid neurotransmitters significantly increased in the corpus striatum following treatment with borneol or moschus, but effects due to borneol were more significant than moschus. Acorus tatarinowii treatment increased ^-aminobutyric acid expression, but decreased glutamate concentrations. Storax increased aspartic acid concentrations and decreased glycine expression. Results demonstrated that borneol and moschus exhibited significant effects on con amino acid neurotransmitter expression; storax exhibited excitatory effects and acorus tatarinowii resulted in inhibitory effects. 展开更多

In this study, we show that the percutaneous absorption and brain distribution of tetramethylpyrazine(TMP) is enhanced when combined with borneol(BN) in a microemulsionbased transdermal therapeutic system(ME-TTS). The formulation of the TMP and BN microemulsion(TEM-BN-ME) was optimized in skin permeation studies in vitro following a uniform experimental design. Male Sprague-Dawley rats were used for the in vivo pharmacokinetic and tissue distribution studies of TMP-BN-ME-TTS. In the pharmacokinetic study, the TMP-BN-ME-TTS treated rats had significantly higher( P < 0.05) C max and AUC of TMP than the TMP-ME-TTS treated rats, indicating that BN improves the rate and extent of TMP percutaneous absorption. In the tissue distribution study, the AUC of TMP in brain was significantly higher in the TMP-BN-ME-TTS group( P < 0.05), indicating that BN facilitates the distribution of TMP in brain. In summary, BN enhanced the percutaneous absorption and brain distribution of TMP in a microemulsion-based transdermal therapeutic system. 展开更多

Successful chemotherapy with paclitaxel(PTX)is impeded by multidrug resistance(MDR)in tumor cells.In this study,lipid-albumin nanoassemblies co-loaded with borneol and paclitaxel(BOR/PTX LANs)were prepared to circumvent MDR in C6 glioma cells.The physiochemical properties including particle size,encapsulation efficiency and morphology were evaluated in vitro.Quantitative and qualitative investigations of cellular uptake were carried out in C6 glioma cells.The cytotoxicity of the BOR/PTX LANs was determined by MTT assay.After that,the tumor targeting was also evaluated in C6 glioma bearing mice by in vivo imaging analysis.BOR/PTX LANs have a higher entrapment efficiency(90.4±1.2%),small particle size(107.5±3.2 nm),narrow distribution(P.I.=0.171±0.02).The cellular uptake of PTX was significantly increased by BOR/PTX LANs compared with paclitaxel loaded lipidalbumin nanoassemblies(PTX LANs)in quantitative research.The result was further confirmed by confocal laser scanning microscopy qualitatively.The cellular uptake was energy-,timeand concentration-dependent,and clathrin-and endosome/lysosome-associated pathways were involved.The BOR/PTX LANs displayed a higher cytotoxicity agaist C6 glioma cells in comparion with PTX LANs and Taxol.Moreover,the encapsulation of BOR in LANs obviously increased the accumulation of the drug in tumor tissues,demonstrating the tumor targeted ability of BOR/PTX LANs.These results indicated that BOR/PTX LANs could overcome MDR by combination of drug delivery systems and P-gp inhibition,and shown the potential for treatment of gliomas. 展开更多

The number of people with Alzheimer’s disease(AD)is increasing annually,with the nidus mainly concentrated in the cortex and hippocampus.Despite of numerous efforts,effective treatment of AD is still facing great challenges due to the blood brain barrier(BBB)and limited drug distribution in the AD nidus sites.Thus,in this study,using vinpocetine(VIN)as a model drug,the objective is to explore the feasibility of tackling the above bottleneck via intranasal drug delivery in combination with a brain guider,borneol(BOR),using nanoemulsion(NE)as the carrier.First of all,the NE were prepared and characterized.In vivo behavior of the NE after intranasal administration was investigated.Influence of BOR dose,BOR administration route on drug brain targeting behavior was evaluated,and the influence of BOR addition on drug brain subregion distribution was probed.It was demonstrated that all the NE had comparable size and similar retention behavior after intranasal delivery.Compared to intravenous injection,improved brain targeting effect was observed by intranasal route,and drug targeting index(DTI)of the VIN–NE group was 154.1%,with the nose-to-brain direct transport percentage(DTP)35.1%.Especially,remarkably enhanced brain distribution was achieved after BOR addition in the NE,with the extent depending on BOR dose.VIN brain concentration was the highest in the VIN-1-BOR-NE group at BOR dose of 1 mg/kg,with the DTI reaching 596.1%and the DTP increased to 83.1%.BOR could exert better nose to brain delivery when administrated together with the drug via intranasal route.Notably,BOR can remarkably enhance drug distribution in both hippocampus and cortex,the nidus areas of AD.In conclusion,in combination with intranasal delivery and the intrinsic brain guiding effect of BOR,drug distribution not only in the brain but also in the cortex and hippocampus can be enhanced significantly,providing the perquisite for improved therapeutic efficacy of AD. 展开更多

Objective: To explore the effect of Bismuth Subgallate/Borneol (Suile<sup>TM</sup> BSB) healing dressing combined with autologous platelet-rich gel (APG) in the treatment of diabetic foot ulcer (DFU). Methods: A total of 120 patients with DFU hospitalized in the Changsha Central Hospital from August 2020 to September 2021 were selected and randomly divided into an experimental group (BSB + APG, n = 60) and a control group (BSB, n = 60) according to random number table method. The total therapeutic effect, healing time, hospital stay, level indexes of various inflammatory factors before and after treatment and ulcer area were observed in the two groups. Results: The total effect of the control group was worse than that of the experimental group, and the data between the two groups were significant (P 0.05);after treatment, the levels of inflammatory factors including WBC, CRP, IL-6 and TNF-α in the control group were higher than those in the experimental group, and there was significant difference between the two groups (P 0.05);after 14 days of treatment, the ulcer area in the control group was larger than that in the experimental group, and the data between the two groups were significant (P Conclusion: BSB combined with APG can achieve better therapeutic effect, reduce the inflammatory reaction of patients, and promote wound healing in the treatment of patients with diabetic foot ulcer. 展开更多

The result of the experiment shows that the yield and purity are excellent with an extraction pressure of 11 MPa, an extracting temperature of 40&deg;C, a carbon dioxide flow rate of 25 L&middot;h<sup>-</sup><sup>1</sup> and an extraction time of 20 minutes. A gas chromatography mass spectrometry (GC-MS) method has been developed and fully validated for the simultaneous determination of natural borneol (NB) and its metabolite, camphor, in rat plasma. Following a single liquid-liquid extraction, the analytes were separated using an HP-5MS capillary column (0.25 mm ? 30 m ? 0.25μm) and analyzed by MS in the selected ion monitoring mode. Selected ion monitor (m/z) of borneol, camphor and internal standard was 95, 95 and 128, respectively. Linearity, accuracy, precision and extraction recovery of the analytes were all satisfactory. The method was successfully applied to pharmacokinetic studies of NB after oral administration to Wistar rats. 展开更多

Stroke is one of the leading causes of disability and death globally.It occurs when a major artery is occluded in the brain and leads to death of cells within the injured tissue.(+)-Borneol,a simple bicyclic monoterpene extracted from traditional Chinese medicine,is widely used in various types of diseases.However,no study has proved the effects of(+)-borneol on functional recovery from permanent ischemic stroke and the mechanism is still unknown.Here,we report that in the rat model of permanent cerebral ischemia,we found that(+)-borneol(1.0 mg/kg) significantly ameliorated infarct size and neurological scores via reducing the expression of inducible nitric oxide synthase(iNOS)and tumor necrosis factor-alpha(TNF-α) in a dose dependent manner.Notably,(+)-borneol showed long-term effects on the improvement of sensorimotor functions in the photothrombotic model of stroke,which decreased the number of foot faults in the grid-walking task and forelimb asymmetry scores in the cylinder task,at least in part through reducing loss of dendritic spines in the length,brunch number and density.These findings suggest that(+)-borneol could serve as a therapeutic target for ischemic stroke. 展开更多

Protective effect of borneol combined with safflower on neurovascular unit in rats with ischemic stroke

Background:Compatibility is a characteristic of the clinical application of traditional Chinese medicine,often leading to enhanced therapeutic effects.In the treatment of cerebral ischemia,blood-activating and open orifices herbs are frequently used individually;however,their combination is not commonly practiced.This study aims to investigate the impact of combining safflower and borneol as examples of open orifices herbs and blood-activating herbs on the neurovascular unit in rats with ischemic stroke.The objective is to determine whether this combination exhibits superior therapeutic efficacy compared to using borneol or safflower alone while exploring its underlying mechanism.These findings may provide novel insights for clinical treatments.Methods:SD male rats were randomly divided into 6 groups:sham operation group,model group,borneol group(0.1 g/kg),safflower group(5 g/kg),borneol combined with safflower group(0.1 g/kg+5 g/kg)and nimodipine group(0.01 g/kg).The middle cerebral artery cerebral ischemia(MCAO)model were prepared after continuous intragastric administration for 7 days in each group,the neurological function of each group were scored 24h after operation,and water content in brain tissue were measured by weighing method.The activity of superoxide dismutase(SOD)and the contents of nitric oxide(NO)and malondialdehyde(MDA)in brain tissue and serum were determined by spectrophotometry,and the mRNA expressions of matrix metalloproteinase 2(MMP-2),tight junction protein 1(ZO-1),vascular endothelial growth factor(VEGF)and brain-derived neurotrophic factor(BDNF)were detected by Real time PCR.Result:Compared with the model group,the group treated with borneol combined with safflower exhibited a significant decrease in the neural function score of MCAO rats(P<0.01).Additionally,it led to a reduction in brain tissue water content(P<0.01),elevated SOD activity,and reduced levels of NO and MDA in both serum and brain tissue(P<0.01 or P<0.05).Moreover,this treatment resulted in a decrease in the mRNA expression of MMP-2 and an increase in ZO-1 in brain tissue,along with an increase in the mRNA expression of VEGF and BDNF(P<0.01).Conclusion:Borneol combined with safflower demonstrates a protective effect on the neurovascular unit in rats with ischemic stroke.This effect is likely associated with increased SOD activity,reduced MDA and NO content in both serum and brain tissue of MCAO rats,and a decrease in MMP-2 mRNA expression in brain tissue,coupled with an increase in ZO-1,VEGF,and BDNF mRNA expression.These effects were superior to those observed with borneol or safflower administered alone. 展开更多

目的建立同时测定砂仁中樟脑、龙脑、乙酸龙脑酯含量的QuEChERS/气相色谱-质谱(QuEChERS/GC-MS)法。方法样品用无水乙醇提取,经N-丙基乙二胺、十八烷基硅烷键合硅胶、石墨化炭黑和硫酸镁吸附净化处理。色谱柱为HP-5 MS型色谱柱(30 m... 目的建立同时测定砂仁中樟脑、龙脑、乙酸龙脑酯含量的QuEChERS/气相色谱-质谱(QuEChERS/GC-MS)法。方法样品用无水乙醇提取,经N-丙基乙二胺、十八烷基硅烷键合硅胶、石墨化炭黑和硫酸镁吸附净化处理。色谱柱为HP-5 MS型色谱柱(30 m×0.25 mm,0.25μm),程序升温,流速为1.0 mL/min,分流进样,分流比为20∶1,电子轰击离子源,选择性监测、全扫描模式,定量特征离子质荷比为95。比较QuEChERS/GC-MS法和气相色谱-氢火焰离子化检测器(GC-FID)法含量测定结果的相对相差值,考察方法的可靠性。结果樟脑、龙脑、乙酸龙脑酯的质量浓度分别在50~2000μg/mL、5~200μg/mL、100~5000μg/mL范围内与峰面积线性关系良好(R2≥0.997,n=6);检测限均为0.005 mg/kg,定量限均为0.015 mg/kg;精密度、稳定性、重复性试验的RSD均低于3.0%;50%,100%,200%水平的加样回收率为93.37%~105.37%,RSD为1.38%~2.85%(n=6)。QuEChERS/GC-MS法和GC-FID法测定樟脑、龙脑、乙酸龙脑酯含量的相对相差值分别为4.76%,4.64%,3.67%。结论该方法操作简便、灵敏度高、重复性好、结果准确,可用于砂仁中樟脑、龙脑、乙酸龙脑酯的含量测定。 展开更多

目的制备紫杉醇-天然冰片复合物,同时探究紫杉醇-天然冰片复合物亚微乳的处方和制备工艺及体外抗肿瘤效果。方法采用研磨法制备紫杉醇-天然冰片复合物并经红外光谱(FT-IR)和差式扫描量热(DSC)分析鉴定所得固体复合物;采用两步高压乳匀... 目的制备紫杉醇-天然冰片复合物,同时探究紫杉醇-天然冰片复合物亚微乳的处方和制备工艺及体外抗肿瘤效果。方法采用研磨法制备紫杉醇-天然冰片复合物并经红外光谱(FT-IR)和差式扫描量热(DSC)分析鉴定所得固体复合物;采用两步高压乳匀法制备复合物亚微乳,应用单因素考察和正交实验优化处方及制备工艺,通过噻唑蓝(MTT)比色法实验、细胞克隆形成实验、细胞划痕实验,考察该制剂对HCT-116细胞的作用效果。结果复合物中紫杉醇3312.76 cm^(-1)和3513.92 cm^(-1)的红外光谱吸收峰消失,且DSC分析显示在154.56℃处出现一个明显的新的吸收峰,表明紫杉醇可能与天然冰片偶联,形成新的复合物。复合亚微乳最佳处方为10%脂肪酸甘油酯,原料药0.44%[紫杉醇-天然冰片(1∶3)],3%混合乳化剂[蛋黄卵磷脂-泊洛沙姆188(1∶2)],2.0%甘油,0.3%油酸;最佳工艺为80℃乳化,60 MPa高压均质10次,再于100℃水浴灭菌45 min。细胞MTT实验中其半数抑制浓度(IC 50)为0.75μg·mL^(-1),细胞克隆实验结果中,空白对照组、紫杉醇原料药及紫杉醇-天然冰片亚微乳的划痕愈合面积分别为(36.44±3.35)%、(13.59±9.28)%、(8.30±4.09)%(P<0.05);平板克隆实验中,空白对照组,紫杉醇原料药组,亚微乳组细胞克隆率分别为(37.92±0.729)%、(9.16±1.335)%、(3.36±1.065)%(P<0.05)。结论紫杉醇-天然冰片亚微乳注射液处方及工艺合理,质量稳定。细胞实验表明,该亚微乳注射剂能明显抑制HCT-116细胞的增殖和迁移。 展开更多

为了探究龙脑樟叶粉对攸县麻鸭繁殖性能、血清免疫、抗氧化和肠道健康指标的影响,试验按体重相近原则将100日龄健康的攸县麻鸭公鸭、母鸭各分为2个组,分别为对照组和试验组,每组各6个重复,每个重复6只。对照组饲喂基础日粮,试验组根据... 为了探究龙脑樟叶粉对攸县麻鸭繁殖性能、血清免疫、抗氧化和肠道健康指标的影响,试验按体重相近原则将100日龄健康的攸县麻鸭公鸭、母鸭各分为2个组,分别为对照组和试验组,每组各6个重复,每个重复6只。对照组饲喂基础日粮,试验组根据前期龙脑樟粉在肉鸡、蛋鸡日粮中的适宜添加浓度,选择在基础日粮中添加3%龙脑樟叶粉,预试期7 d,正试期42 d。试验结束后统计繁殖性能指标,每个重复随机选择2只鸭进行屠宰,测定免疫性能、抗氧化指标和肠道健康指标。结果表明:与对照组相比,母鸭试验组的卵巢重和卵巢指数均显著提高(P<0.05),公鸭试验组的肝脏重极显著降低(P<0.01),肝脏指数显著降低(P<0.05);母鸭试验组的血清免疫球蛋白A(IgA)质量浓度显著提高(P<0.05);公鸭试验组的空肠和十二指肠杯状细胞数显著提高(P<0.05);公母鸭试验组盲肠食糜中的大肠杆菌、公鸭试验组的乳酸杆菌、母鸭试验组的双歧杆菌的菌落数均极显著降低(P<0.01)。说明日粮中添加3%龙脑樟叶粉可提升攸县麻鸭公鸭肠道健康水平、母鸭免疫性能,促进母鸭卵巢发育,同时降低公母鸭肠道有益与有害菌菌落数量。 展开更多

水溶性冰片磷酸酯前药的设计、合成及生物活性评价

对冰片进行结构优化,以改善冰片的溶解性,促进其在脑卒中治疗方面的进一步应用。利用磷酸酯修饰原理,设计并合成冰片前药BP-3。使用蒸发光散射检测器(ELSD)测定BP-3的溶解性,在小鼠血浆中测试原药释放的程度与速度,并在短暂性大脑中动... 对冰片进行结构优化,以改善冰片的溶解性,促进其在脑卒中治疗方面的进一步应用。利用磷酸酯修饰原理,设计并合成冰片前药BP-3。使用蒸发光散射检测器(ELSD)测定BP-3的溶解性,在小鼠血浆中测试原药释放的程度与速度,并在短暂性大脑中动脉闭塞(tMCAO)小鼠模型中评估BP-3的神经保护作用。结果显示,BP-3在20 mg/mL时可以完全溶解于生理盐水;在小鼠血浆中,2 h内释放约40%的冰片;在tMCAO小鼠模型中,TTC染色结果显示BP-3可以有效减少梗死面积;尼氏染色结果表明,BP-3可以改善神经元损伤;握力和抓力测试结果阐明,BP-3可以减少损伤带来的运动能力降低;转棒测试结果证明,BP-3可以促进小鼠运动能力的恢复。BP-3具有良好的水溶性、适宜的原药释放速率以及优异的神经保护作用,具备广阔的药物开发前景。 展开更多
