Evidence showed that N6-methyladenosine(m^(6)A)modification plays a pivotal role in influencing RNA fate and is strongly associated with cell growth and developmental processes in many species.However,no information r...Evidence showed that N6-methyladenosine(m^(6)A)modification plays a pivotal role in influencing RNA fate and is strongly associated with cell growth and developmental processes in many species.However,no information regarding m^(6)A modification in Eimeria tenella is currently available.In the present study,we surveyed the transcriptome-wide prevalence of m^(6)A in sporulated oocysts and unsporulated oocysts of E.tenella.Methylated RNA immunoprecipitation sequencing(MeRIP-seq)analysis showed that m^(6)A modification was most abundant in the coding sequences,followed by stop codon.There were 3,903 hypermethylated and 3,178 hypomethylated mRNAs in sporulated oocysts compared with unsporulated oocysts.Further joint analysis suggested that m^(6)A modification of the majority of genes was positively correlated with mRNA expression.The mRNA relative expression and m^(6)A level of the selected genes were confirmed by quantitative reverse transcription PCR(RT-qPCR)and MeRIP-qPCR.GO and KEGG analysis indicated that differentially m^(6)A methylated genes(DMMGs)with significant differences in mRNA expression were closely related to processes such as regulation of gene expression,epigenetic,microtubule,autophagy-other and TOR signaling.Moreover,a total of 96 DMMGs without significant differences in mRNA expression showed significant differences at protein level.GO and pathway enrichment analysis of the 96 genes showed that RNA methylation may be involved in cell biosynthesis and metabolism of E.tenella.We firstly present a map of RNA m^(6)A modification in E.tenella,which provides significant insights into developmental biology of E.tenella.展开更多
BACKGROUND Cervical intraepithelial neoplasia(CIN)is an important precursor of cervical cancer.Early detection and treatment can reduce the incidence of cervical cancer.AIM To investigate the detection rate of human p...BACKGROUND Cervical intraepithelial neoplasia(CIN)is an important precursor of cervical cancer.Early detection and treatment can reduce the incidence of cervical cancer.AIM To investigate the detection rate of human papillomavirus(HPV)E6/E7 mRNA in cervical tissue of patients with different types of epithelial cell neoplasia(CIN)and its relationship with CIN progression and diagnosis.METHODS One hundred women with HPV infection detected by cervical exfoliation cytology between January 2022 and January 2023 were retrospectively selected.These patients were graded CIN based on colposcopy and cervical pathology.The positive expression rates of HPV E6/E7 mRNA and HPV[polymerase chain reaction(PCR)-reverse dot crossing]were compared among all groups.Patients with HPV E6/E7 mRNA expression in the grade 1 CIN group were followed up for 1 yr.The relationship between atypical squamous epithelium and high malignant epithelial neoplasia was investigated by univariate and multivariate analysis.RESULTS The diagnostic sensitivity,specificity,and sensitivity of PCR-reverse point hybrid ization technology for secondary CIN were 70.41%,70.66%,and 0.714,respectively.Sensitivity and specificity for secondary CIN were 752%and 7853%,respectively,the area under the curve value was 0.789.Logistic Multifactorial model analysis revealed that the HPV positive rates and the HPV E6/E7 mRNA positive rates were independent risk factors of CIN grade I(P<0.05).In CIN grade I patients with positive for HPV E6/E7 mRNA,in its orientation to grade CIN patients,in its orientation to grade CIN patients,at 69.2%,compared with patients negative for HPV E6/E7 mRNA(30.8%),significant difference(P<0.05).CONCLUSION HPV E6/E7 mRNA and HPV(PCR-reverse dot hybrid)positive expression have a close relationship with CINgrade disease progression and is an independent risk factor for high-grade CIN lesions.展开更多

The results show that nanometer M6 C particles discontinuously precipitate in the γ matrix or along the γ/γ′ interface of the alloy during high temperature tensile creep. Thereinto, the amount of fine M6 C carbide increases as creep goes on, and the coherent interfaces of M6 C phase precipitating from the γ matrix are {100} and {111} planes. The thermodynamics analysis indicates that the solubility of element carbon in the matrix decreases when the alloy is deformed by the axial tensile stress during creep, so as to cause the carbon segregating in the regions of stress concentration and combining with carbide-forming elements M(W, Co), which promotes the fine M6 C carbide to precipitate from the γ matrix.展开更多

The solution for the forward displacement analysis(FDA) of the general 6-6 Stewart mechanism(i.e., the connection points of the moving and fixed platforms are not restricted to lying in a plane) has been extensive...The solution for the forward displacement analysis(FDA) of the general 6-6 Stewart mechanism(i.e., the connection points of the moving and fixed platforms are not restricted to lying in a plane) has been extensively studied, but the efficiency of the solution remains to be effectively addressed. To this end, an algebraic elimination method is proposed for the FDA of the general 6-6 Stewart mechanism. The kinematic constraint equations are built using conformal geometric algebra(CGA). The kinematic constraint equations are transformed by a substitution of variables into seven equations with seven unknown variables. According to the characteristic of anti-symmetric matrices, the aforementioned seven equations can be further transformed into seven equations with four unknown variables by a substitution of variables using the Grobner basis. Its elimination weight is increased through changing the degree of one variable, and sixteen equations with four unknown variables can be obtained using the Grobner basis. A 40th-degree univariate polynomial equation is derived by constructing a relatively small-sized 9 × 9 Sylvester resultant matrix. Finally, two numerical examples are employed to verify the proposed method. The results indicate that the proposed method can effectively improve the efficiency of solution and reduce the computational burden because of the small-sized resultant matrix.展开更多
Providing alternative PNT service to GNSS-challenged users will be an important function of next-generation NGSO broadband satellite communication systems.Herein,a packet-based PNT service architecture in NGSO broadba...Providing alternative PNT service to GNSS-challenged users will be an important function of next-generation NGSO broadband satellite communication systems.Herein,a packet-based PNT service architecture in NGSO broadband systems is proposed in which a primary satellite and selected assistant satellites work together to provide PNT service to requesting users.Its positioning performance bounds are mathematically formulated by rigorously analyzing the bounds constrained by different waveforms.Simulations are conducted on different configurations of Walker Delta MEO constellations and Walker Star LEO constellations for corroboration,revealing the following:(1)Both MEO and LEO constellations achieve sub-meter-level positioning precision given enough satellites.(2)Compared to the GNSS Doppler-based velocity estimation method,the position advance based velocity estimation algorithm is more precise and applicable to the PNT service in NGSO broadband systems.(3)To provide PNT service to users in GNSS-challenged environments,the primary and each assistant satellite need only∼0.1‰of the time of one downlink beam.展开更多

[ Method] Peripheral lymphocytas were isolated from a hybrid pig ( Duroc x Landrace x Shandong local pig) and stimulated with canavalin A in vitro. With total RNA as templates, the IL-2 and IL-6 cDNA were amplified by the RT-PCR and sequenced, respectively. [Result] The sequence of IL-2 gene had 100.0% nucleotide homology to the published IL-2 sequences. The sequence of IL-6 gene had 99.8% -100.0% nucleotide homology to the published IL-6 sequences, and the amino acid homology was 99.1% -100.0%. [ Conclusion] NO great variation of the IL-2 gene and IL-6 gene is observed in the hybdd pig, and these results provide a basis for research about functions of IL-2 and IL-6 protein.展开更多

The interruption of auto-expansion ...A new magnetic hydro-dynamics model for nozzle arc emphasizing the interaction of arc with PTFE (polytetrafluorethylene) vapour is established based on the conservation equations. The interruption of auto-expansion circuit breaker is simulated numerically by finite element method and the influence of PTFE vapour on the arc is analysed with this model. The results reveal that the flow field inside the arc chamber is determined by the arc current, the arcing time, the nozzle arc and the clogging of its thermal boundary. The establishment of quenching pressure relies on both SF6 gas and PTFE vapour that absorbed arc energy in the nozzle. The PTFE vapour leads to an increase in the pressure of nozzle arc obviously, and a decrease in the temperature of arc. But it enhances the temperature of arc at zero current and slows down the decreasing rate of arc temperature as the current decreases.展开更多
Lonicerae japonicaeFlos (LJF) is widely used in traditional Chinese medicines for the treatment of various diseases, which is now in great demand every year and has a broad development prospect. However, the flowering...Lonicerae japonicaeFlos (LJF) is widely used in traditional Chinese medicines for the treatment of various diseases, which is now in great demand every year and has a broad development prospect. However, the flowering phase of common LJF varieties is so short, which seriously restricts the development of LJF industry. As a new cultivated kind of Lonicerae japonicae Flos, “Hua Jin 6” has characteristics in long flowering phase and conveniently picking, which makes it have a broad development prospect. The aim of this study is to provide scientific guidance for its suitable harvest period by measuring yield and quality of “Hua Jin 6” from different harvest time. Studies show that flower size had a slowly rising trend from the first day to the seventh day, and then slowly declined or kept stable. There were no significant differences of total phenolic acid contents in different samples from different days, but contents of total flavonoids were on the rise and up to maximum in the ninth day. The contents of total iridoids had an increasing tendency from the first day to the fifth day and then kept relatively stable in other days. We demonstrated that the quality of “Hua Jin 6” is relatively stable and suitable for harvesting in all flower buds white stage in term of HPLC fingerprints. Our findings can make it possible to select the suitable time for different harvest purpose.展开更多
Rho-related GTPase from plants(ROP)proteins play an essential role in plant stress resistance.In this study,the full-length GlROP6 gene was cloned based on G.littoralis transcriptome sequencing data acquired in respon...Rho-related GTPase from plants(ROP)proteins play an essential role in plant stress resistance.In this study,the full-length GlROP6 gene was cloned based on G.littoralis transcriptome sequencing data acquired in response to salt stress.The protein sequence,conserved domains,secondary structure,three-dimensional structure,phylogenetic relationships,and expression pattern of the GlROP6 gene were systematically analysed.Our results showed that the full-length GlROP6 gene had an open reading frame of 606 bp,which encoded 201 amino acid residues with a relative molecular weight of 22.23463 kDa and a theoretical isoelectric point of 9.06.Amino acid sequence analyses indicated that the structure of the GlROP6 protein was conserved,and included five G-box motifs(G1–G5),an effector binding region,a Rho insert region and a C-terminal hypervariable region.According to our phylogenetic analysis,the GlROP6 protein was closely related to the ROP protein of Daucus carota subsp.Sativus.Our quantitative real-time PCR results revealed that GlROP6 was highly expressed in flower,and GlROP6 expression was significantly upregulated in G.littoralis roots treated with NaCl.This study will facilitate investigations into the function of GlROP genes in response to salt stress in G.littoralis.展开更多
