Evidence showed that N6-methyladenosine(m^(6)A)modification plays a pivotal role in influencing RNA fate and is strongly associated with cell growth and developmental processes in many species.However,no information r...Evidence showed that N6-methyladenosine(m^(6)A)modification plays a pivotal role in influencing RNA fate and is strongly associated with cell growth and developmental processes in many species.However,no information regarding m^(6)A modification in Eimeria tenella is currently available.In the present study,we surveyed the transcriptome-wide prevalence of m^(6)A in sporulated oocysts and unsporulated oocysts of E.tenella.Methylated RNA immunoprecipitation sequencing(MeRIP-seq)analysis showed that m^(6)A modification was most abundant in the coding sequences,followed by stop codon.There were 3,903 hypermethylated and 3,178 hypomethylated mRNAs in sporulated oocysts compared with unsporulated oocysts.Further joint analysis suggested that m^(6)A modification of the majority of genes was positively correlated with mRNA expression.The mRNA relative expression and m^(6)A level of the selected genes were confirmed by quantitative reverse transcription PCR(RT-qPCR)and MeRIP-qPCR.GO and KEGG analysis indicated that differentially m^(6)A methylated genes(DMMGs)with significant differences in mRNA expression were closely related to processes such as regulation of gene expression,epigenetic,microtubule,autophagy-other and TOR signaling.Moreover,a total of 96 DMMGs without significant differences in mRNA expression showed significant differences at protein level.GO and pathway enrichment analysis of the 96 genes showed that RNA methylation may be involved in cell biosynthesis and metabolism of E.tenella.We firstly present a map of RNA m^(6)A modification in E.tenella,which provides significant insights into developmental biology of E.tenella.展开更多
BACKGROUND Cervical intraepithelial neoplasia(CIN)is an important precursor of cervical cancer.Early detection and treatment can reduce the incidence of cervical cancer.AIM To investigate the detection rate of human p...BACKGROUND Cervical intraepithelial neoplasia(CIN)is an important precursor of cervical cancer.Early detection and treatment can reduce the incidence of cervical cancer.AIM To investigate the detection rate of human papillomavirus(HPV)E6/E7 mRNA in cervical tissue of patients with different types of epithelial cell neoplasia(CIN)and its relationship with CIN progression and diagnosis.METHODS One hundred women with HPV infection detected by cervical exfoliation cytology between January 2022 and January 2023 were retrospectively selected.These patients were graded CIN based on colposcopy and cervical pathology.The positive expression rates of HPV E6/E7 mRNA and HPV[polymerase chain reaction(PCR)-reverse dot crossing]were compared among all groups.Patients with HPV E6/E7 mRNA expression in the grade 1 CIN group were followed up for 1 yr.The relationship between atypical squamous epithelium and high malignant epithelial neoplasia was investigated by univariate and multivariate analysis.RESULTS The diagnostic sensitivity,specificity,and sensitivity of PCR-reverse point hybrid ization technology for secondary CIN were 70.41%,70.66%,and 0.714,respectively.Sensitivity and specificity for secondary CIN were 752%and 7853%,respectively,the area under the curve value was 0.789.Logistic Multifactorial model analysis revealed that the HPV positive rates and the HPV E6/E7 mRNA positive rates were independent risk factors of CIN grade I(P<0.05).In CIN grade I patients with positive for HPV E6/E7 mRNA,in its orientation to grade CIN patients,in its orientation to grade CIN patients,at 69.2%,compared with patients negative for HPV E6/E7 mRNA(30.8%),significant difference(P<0.05).CONCLUSION HPV E6/E7 mRNA and HPV(PCR-reverse dot hybrid)positive expression have a close relationship with CINgrade disease progression and is an independent risk factor for high-grade CIN lesions.展开更多

The results show that nanometer M6 C particles discontinuously precipitate in the γ matrix or along the γ/γ′ interface of the alloy during high temperature tensile creep. Thereinto, the amount of fine M6 C carbide increases as creep goes on, and the coherent interfaces of M6 C phase precipitating from the γ matrix are {100} and {111} planes. The thermodynamics analysis indicates that the solubility of element carbon in the matrix decreases when the alloy is deformed by the axial tensile stress during creep, so as to cause the carbon segregating in the regions of stress concentration and combining with carbide-forming elements M(W, Co), which promotes the fine M6 C carbide to precipitate from the γ matrix.展开更多

The solution for the forward displacement analysis(FDA) of the general 6-6 Stewart mechanism(i.e., the connection points of the moving and fixed platforms are not restricted to lying in a plane) has been extensive...The solution for the forward displacement analysis(FDA) of the general 6-6 Stewart mechanism(i.e., the connection points of the moving and fixed platforms are not restricted to lying in a plane) has been extensively studied, but the efficiency of the solution remains to be effectively addressed. To this end, an algebraic elimination method is proposed for the FDA of the general 6-6 Stewart mechanism. The kinematic constraint equations are built using conformal geometric algebra(CGA). The kinematic constraint equations are transformed by a substitution of variables into seven equations with seven unknown variables. According to the characteristic of anti-symmetric matrices, the aforementioned seven equations can be further transformed into seven equations with four unknown variables by a substitution of variables using the Grobner basis. Its elimination weight is increased through changing the degree of one variable, and sixteen equations with four unknown variables can be obtained using the Grobner basis. A 40th-degree univariate polynomial equation is derived by constructing a relatively small-sized 9 × 9 Sylvester resultant matrix. Finally, two numerical examples are employed to verify the proposed method. The results indicate that the proposed method can effectively improve the efficiency of solution and reduce the computational burden because of the small-sized resultant matrix.展开更多

[ Method] Peripheral lymphocytas were isolated from a hybrid pig ( Duroc x Landrace x Shandong local pig) and stimulated with canavalin A in vitro. With total RNA as templates, the IL-2 and IL-6 cDNA were amplified by the RT-PCR and sequenced, respectively. [Result] The sequence of IL-2 gene had 100.0% nucleotide homology to the published IL-2 sequences. The sequence of IL-6 gene had 99.8% -100.0% nucleotide homology to the published IL-6 sequences, and the amino acid homology was 99.1% -100.0%. [ Conclusion] NO great variation of the IL-2 gene and IL-6 gene is observed in the hybdd pig, and these results provide a basis for research about functions of IL-2 and IL-6 protein.展开更多

The interruption of auto-expansion circuit breaker is simulated numerically by finite element method and the influence of PTFE vapour on the arc is analysed with this model. The results reveal that the flow field inside the arc chamber is determined by the arc current, the arcing time, the nozzle arc and the clogging of its thermal boundary. The establishment of quenching pressure relies on both SF6 gas and PTFE vapour that absorbed arc energy in the nozzle. The PTFE vapour leads to an increase in the pressure of nozzle arc obviously, and a decrease in the temperature of arc. But it enhances the temperature of arc at zero current and slows down the decreasing rate of arc temperature as the current decreases.展开更多
Numerous studies currently compare the lipid metabolism in patients with cardiovascular disease(CVD)and healthy individuals to identify lipid markers for predicting CVD.In this study,multidimensional mass spectrometry...Numerous studies currently compare the lipid metabolism in patients with cardiovascular disease(CVD)and healthy individuals to identify lipid markers for predicting CVD.In this study,multidimensional mass spectrometry-based shotgun lipidomics was used to examine the serum lipidomics of participants in a clinical randomized controlled feeding trial undergoing olive oil(OO),camellia seed oil(CSO),and soybean oil(SO)dietary interventions.189 lipid molecules are identified,including 14 species of phosphatidylinositol,45 species of ethanolamine glycerols(PE),47 species of choline glycerophospholipids(PC),39 species of triacylglycerols(TAG),18 species of lysophosphatidylcholine,and 26 species of sphingomyelin.After screening,10 lipid markers are found,among which 18:2 fatty acid(FA),16:1 FA,C54:4/C55:11,C54:3/C55:10,and C52:3/C53:10 in TAG pool,p18:0/20:0 and a18:0/18:1 in PC pool,and p18:1/20:4 in PE pool have differential regulation in the SO group compared to OO and CSO.The d16:0/18:1 in PC pool and C52:2/C53:9 in TAG pool are differentially regulated by OO and CSO.The C52:2/C53:9 in TAG pool has a significant negative correlation with aspartate aminotransferase(r=-0.363,P=0.048)and high-density lipoprotein cholesterol(r=-0.519,P<0.01).This study provides a reference for researching the effect of dietary fat on blood lipid metabolism.展开更多

However, the flowering...Lonicerae japonicaeFlos (LJF) is widely used in traditional Chinese medicines for the treatment of various diseases, which is now in great demand every year and has a broad development prospect. However, the flowering phase of common LJF varieties is so short, which seriously restricts the development of LJF industry. As a new cultivated kind of Lonicerae japonicae Flos, “Hua Jin 6” has characteristics in long flowering phase and conveniently picking, which makes it have a broad development prospect. The aim of this study is to provide scientific guidance for its suitable harvest period by measuring yield and quality of “Hua Jin 6” from different harvest time. Studies show that flower size had a slowly rising trend from the first day to the seventh day, and then slowly declined or kept stable. There were no significant differences of total phenolic acid contents in different samples from different days, but contents of total flavonoids were on the rise and up to maximum in the ninth day. The contents of total iridoids had an increasing tendency from the first day to the fifth day and then kept relatively stable in other days. We demonstrated that the quality of “Hua Jin 6” is relatively stable and suitable for harvesting in all flower buds white stage in term of HPLC fingerprints. Our findings can make it possible to select the suitable time for different harvest purpose.展开更多
Rho-related GTPase from plants(ROP)proteins play an essential role in plant stress resistance.In this study,the full-length GlROP6 gene was cloned based on G.littoralis transcriptome sequencing data acquired in respon...Rho-related GTPase from plants(ROP)proteins play an essential role in plant stress resistance.In this study,the full-length GlROP6 gene was cloned based on G.littoralis transcriptome sequencing data acquired in response to salt stress.The protein sequence,conserved domains,secondary structure,three-dimensional structure,phylogenetic relationships,and expression pattern of the GlROP6 gene were systematically analysed.Our results showed that the full-length GlROP6 gene had an open reading frame of 606 bp,which encoded 201 amino acid residues with a relative molecular weight of 22.23463 kDa and a theoretical isoelectric point of 9.06.Amino acid sequence analyses indicated that the structure of the GlROP6 protein was conserved,and included five G-box motifs(G1–G5),an effector binding region,a Rho insert region and a C-terminal hypervariable region.According to our phylogenetic analysis,the GlROP6 protein was closely related to the ROP protein of Daucus carota subsp.Sativus.Our quantitative real-time PCR results revealed that GlROP6 was highly expressed in flower,and GlROP6 expression was significantly upregulated in G.littoralis roots treated with NaCl.This study will facilitate investigations into the function of GlROP genes in response to salt stress in G.littoralis.展开更多

The E6 and E7 genes in HPV-16 are the major virulent genes in HPV, and we wanted to investigate the polymorphism of E6 and E7 genes of HPV-16 originated from Jingjiang, Jiangsu province. In research, HPV-16 sample was collected, and the E6 and E7 genes fragments were amplified by PCR assay. The sequences of E6 and E7 genes were used for phylogenetic analysis by Mega 5.0 software. By comparison, the major mutations of E6 gene were T178G (41.67%) and G658A (17%), as well as C58G, T61A and G188C (14%), T61C and C656A (11%). The mutation of E7 gene was mainly C491A and T935A (23%);G514C, G937C and G519C (11%). The 68 nucleotide site deficiency (69.4%) of E6 and 535 nucleotide site deficiency (23%) of E7 were dominant. In our study, we did not find the relevance between the E6 and E7 genes mutations and pathologic severity. But we think the E6 and E7 genes mutation of HPV-16 might affect vaccine prevention and treatment effect.展开更多

This is a step before analyzing the Architectures of VoIPv6 in the current internet in order for it to cope with IPv6 traffic transmission requirements in general and specifically voice traffic, which is being attracting the efforts of research, bodes currently. These tests were conducted in the application level without looking into the network level of the network. VoIPv6 performance tests were conducted in the current tunneled and native IPv6 aiming for better end-to-end VoIPv6 performance. The results obtained in this study were shown in deferent codec's for different bit rates in Kilo bits per second, which act as an indicator for the better performance of G.711 compared with the rest of the tested codes.展开更多

The results showed that the resistance of Xikemai 6 against CYR31 was controlled by 2 pairs of dominant genes and a pair of recessive genes; the resistance against CYR32 was controlled by three pairs of dominant resistant genes( two pairs of genes performed cumulative effect); the resistance against CYR33 was controlled by a pair of dominant genes and a pair of recessive genes; the resistance against Su11-4 was controlled by a pair of dominant genes and a pair of recessive genes independently or collaboratively; the resistance against V26 was controlled by a pair of dominant genes independently. Due to good performance of Xikemai 6 in test and production,as well as years of resistance identification and genetic analysis,Xikemai 6 was proved to be an excellent cultivar with good resistance against stripe rust,and the inheritance of its resistance was stable,so Xikemai 6 could be used as a germplasm resource and resistance material with excellent comprehensive character. Molecular marker and localization could be further studied,to provide new resistance parents for disease-resistant breeding of wheat.展开更多
[ Objective] This paper aimed to investigate the origin, characteristics and molecular evolution of duck derived H4N6 subtype avian influ- enza virus (DK/SH/Y20/06) and enrich the epidemiologic data of the waterfowl...[ Objective] This paper aimed to investigate the origin, characteristics and molecular evolution of duck derived H4N6 subtype avian influ- enza virus (DK/SH/Y20/06) and enrich the epidemiologic data of the waterfowl origin AIV. [Method] The entire genome of DK/SH/Y20/06 was amplified and subjected to genome sequencing. The molecular software was used for sequence analysis and phylogenetic tree construction of DK/ SH/Y20/06 with some other reference sequences in GenBank. [Result] The results indicated that the amino acid sequence adjacent to HA cleav- age site was PEKASR ↓ GLF, which was the typical characteristics of the LPAIV. The phylogenetic analysis indicated that the HA gene of the isolate was derived from the Eurasian lineage in the eastern hemisphere. The NA gene was at the same branch with A/rnallard/Yan chen/2005( H4N6), sharing 98.3% sequence identity. The PB2, PB1, NP and PA gene of this isolate had genetically close relationships with H6 subtype AIV which is epidemic in China at present. The M gene fell into the same branch with A/environment/Korea/CSM05/2004( H3N1 ). The NS segment had the highest similarity with A/wild duck/Korea/YS44/2004(H1N2). The eight genes were not at the same branch and shared a low similarity with other H4N6 subtype avian influenza viruses isolated in North America. [Condusion] These data showed that DK/SH/Y20/06(H4N6) was possibly a re- combinant virus derived from H4N6 subtype, H6N2, H6N5, H3N1 and H1 N2 subtype AIV by complex gene recombination in duck.展开更多

Talaporfin standard curves were obtained i...N-aspartylchlorin e6 (talaporfin) concentrations in cancer cells, mouse liver tissues, and human plasma specimens were determined with fluorescence microtiter plate analysis. Talaporfin standard curves were obtained in each of the sample specimens of cells, mouse tissues, or human plasma including serial concentration of talaporfin. The correlation co-efficiencies (r2) of talaporfin standard curves were 0.99-1.00, and the CV less than 5%. Talaporfin incorporation into cells of human breast cancer cell line MCF-7 after incubating with 25 μg/mL talaporfin for up to 24 h revealed that the t-max of the drug incorporation was approximately 5 h, and the maximum drug concentration incorporated was 25 μg/107 cells. Talaporfin incorporation into MCF-7 cells was significantly decreased in the presence of 3 μg/mL cyc-losporine (p < 0.05). Balb/c nu/nu mice implanted human cholangiocarcinoma NOZ cells in liver were administered intravenously 5mg/mouse of talaporfin, and the tissues of normal liver and tumor, as well as plasma specimens, were analyzed for talaporfin concentrations. The mean (SD) of talaporfin concentration in plasma after 30 min of administration was 41.6 (2.3) μg/mL, while the level decreased to undetectable concentrations 2 h after administration. In contrast, the talaporfin concentrations in normal and tumor tissues after 30 min of administration were 1.1-7.8 μg/g tissue, and the level slightly increased or was almost maintained for up to 2-4 h after administration. The heparinized blood of the healthy subjects was incubated with 25 μg/mL talaporfin for up to 24 h. The plasma talaporfin concentration did not significantly change during the incubation, and thus talaporfin appears not to be incorporated into the blood cells. We established rapid and simple analysis procedures of talaporfin in biological specimens using a fluorescence microtiter plate assay. Using this assay procedure, the unique pattern of talaporfin disposition and pharmacokinetics were revealed in human cancer cells, liver tissues of tumor bearing mice, and human blood.展开更多

文摘BACKGROUND Cervical intraepithelial neoplasia(CIN)is an important precursor of cervical cancer.Early detection and treatment can reduce the incidence of cervical cancer.AIM To investigate the detection rate of human papillomavirus(HPV)E6/E7 mRNA in cervical tissue of patients with different types of epithelial cell neoplasia(CIN)and its relationship with CIN progression and diagnosis.METHODS One hundred women with HPV infection detected by cervical exfoliation cytology between January 2022 and January 2023 were retrospectively selected.These patients were graded CIN based on colposcopy and cervical pathology.The positive expression rates of HPV E6/E7 mRNA and HPV[polymerase chain reaction(PCR)-reverse dot crossing]were compared among all groups.Patients with HPV E6/E7 mRNA expression in the grade 1 CIN group were followed up for 1 yr.The relationship between atypical squamous epithelium and high malignant epithelial neoplasia was investigated by univariate and multivariate analysis.RESULTS The diagnostic sensitivity,specificity,and sensitivity of PCR-reverse point hybrid ization technology for secondary CIN were 70.41%,70.66%,and 0.714,respectively.Sensitivity and specificity for secondary CIN were 752%and 7853%,respectively,the area under the curve value was 0.789.Logistic Multifactorial model analysis revealed that the HPV positive rates and the HPV E6/E7 mRNA positive rates were independent risk factors of CIN grade I(P<0.05).In CIN grade I patients with positive for HPV E6/E7 mRNA,in its orientation to grade CIN patients,in its orientation to grade CIN patients,at 69.2%,compared with patients negative for HPV E6/E7 mRNA(30.8%),significant difference(P<0.05).CONCLUSION HPV E6/E7 mRNA and HPV(PCR-reverse dot hybrid)positive expression have a close relationship with CINgrade disease progression and is an independent risk factor for high-grade CIN lesions.

文摘The precipitation behavior of carbide in K416 B superalloy was investigated by means of creep measurement and microstructure observation. The results show that nanometer M6 C particles discontinuously precipitate in the γ matrix or along the γ/γ′ interface of the alloy during high temperature tensile creep. Thereinto, the amount of fine M6 C carbide increases as creep goes on, and the coherent interfaces of M6 C phase precipitating from the γ matrix are {100} and {111} planes. The thermodynamics analysis indicates that the solubility of element carbon in the matrix decreases when the alloy is deformed by the axial tensile stress during creep, so as to cause the carbon segregating in the regions of stress concentration and combining with carbide-forming elements M(W, Co), which promotes the fine M6 C carbide to precipitate from the γ matrix.

文摘[Objective] The aim of this study was to perform genome-wide analysis of glucose-6-phosphate dehydrogenase(G6PDH) and reveal its evolution in Eucalyptus grandsis.[Method] The gene character,protein sequence and phylogenetic tree of G6PDH gene were analyzed by BLAST and other bioinformatics software within Eucalyptus grandsis whole genome database.[Result] Six G6PDH genes,including one cytomic type and five plastids,were detected in the E.grandsis genome.All the G6PDHs have conserved motifs of motif 1,motif 2,motif 3,motif 7,motif 9 and motif 11.Furthermore,promoter sequences of all E.grandsis G6PDH contain TATA box,enhancer,light-responsive,hormone-responsive and stress-responsive regulatory elements.[Conclusion] This study provided reference for the further revealing molecular function of E.grandsis G6PDH gene family

文摘The solution for the forward displacement analysis(FDA) of the general 6-6 Stewart mechanism(i.e., the connection points of the moving and fixed platforms are not restricted to lying in a plane) has been extensively studied, but the efficiency of the solution remains to be effectively addressed. To this end, an algebraic elimination method is proposed for the FDA of the general 6-6 Stewart mechanism. The kinematic constraint equations are built using conformal geometric algebra(CGA). The kinematic constraint equations are transformed by a substitution of variables into seven equations with seven unknown variables. According to the characteristic of anti-symmetric matrices, the aforementioned seven equations can be further transformed into seven equations with four unknown variables by a substitution of variables using the Grobner basis. Its elimination weight is increased through changing the degree of one variable, and sixteen equations with four unknown variables can be obtained using the Grobner basis. A 40th-degree univariate polynomial equation is derived by constructing a relatively small-sized 9 × 9 Sylvester resultant matrix. Finally, two numerical examples are employed to verify the proposed method. The results indicate that the proposed method can effectively improve the efficiency of solution and reduce the computational burden because of the small-sized resultant matrix.
文摘[ Objective] To understand the functions of interteukin-2 (IL-2) and interleukin-6 (IL-6) in immune response. [ Method] Peripheral lymphocytas were isolated from a hybrid pig ( Duroc x Landrace x Shandong local pig) and stimulated with canavalin A in vitro. With total RNA as templates, the IL-2 and IL-6 cDNA were amplified by the RT-PCR and sequenced, respectively. [Result] The sequence of IL-2 gene had 100.0% nucleotide homology to the published IL-2 sequences. The sequence of IL-6 gene had 99.8% -100.0% nucleotide homology to the published IL-6 sequences, and the amino acid homology was 99.1% -100.0%. [ Conclusion] NO great variation of the IL-2 gene and IL-6 gene is observed in the hybdd pig, and these results provide a basis for research about functions of IL-2 and IL-6 protein.
文摘A new magnetic hydro-dynamics model for nozzle arc emphasizing the interaction of arc with PTFE (polytetrafluorethylene) vapour is established based on the conservation equations. The interruption of auto-expansion circuit breaker is simulated numerically by finite element method and the influence of PTFE vapour on the arc is analysed with this model. The results reveal that the flow field inside the arc chamber is determined by the arc current, the arcing time, the nozzle arc and the clogging of its thermal boundary. The establishment of quenching pressure relies on both SF6 gas and PTFE vapour that absorbed arc energy in the nozzle. The PTFE vapour leads to an increase in the pressure of nozzle arc obviously, and a decrease in the temperature of arc. But it enhances the temperature of arc at zero current and slows down the decreasing rate of arc temperature as the current decreases.

文摘Numerous studies currently compare the lipid metabolism in patients with cardiovascular disease(CVD)and healthy individuals to identify lipid markers for predicting CVD.In this study,multidimensional mass spectrometry-based shotgun lipidomics was used to examine the serum lipidomics of participants in a clinical randomized controlled feeding trial undergoing olive oil(OO),camellia seed oil(CSO),and soybean oil(SO)dietary interventions.189 lipid molecules are identified,including 14 species of phosphatidylinositol,45 species of ethanolamine glycerols(PE),47 species of choline glycerophospholipids(PC),39 species of triacylglycerols(TAG),18 species of lysophosphatidylcholine,and 26 species of sphingomyelin.After screening,10 lipid markers are found,among which 18:2 fatty acid(FA),16:1 FA,C54:4/C55:11,C54:3/C55:10,and C52:3/C53:10 in TAG pool,p18:0/20:0 and a18:0/18:1 in PC pool,and p18:1/20:4 in PE pool have differential regulation in the SO group compared to OO and CSO.The d16:0/18:1 in PC pool and C52:2/C53:9 in TAG pool are differentially regulated by OO and CSO.The C52:2/C53:9 in TAG pool has a significant negative correlation with aspartate aminotransferase(r=-0.363,P=0.048)and high-density lipoprotein cholesterol(r=-0.519,P<0.01).This study provides a reference for researching the effect of dietary fat on blood lipid metabolism.
文摘Lonicerae japonicaeFlos (LJF) is widely used in traditional Chinese medicines for the treatment of various diseases, which is now in great demand every year and has a broad development prospect. However, the flowering phase of common LJF varieties is so short, which seriously restricts the development of LJF industry. As a new cultivated kind of Lonicerae japonicae Flos, “Hua Jin 6” has characteristics in long flowering phase and conveniently picking, which makes it have a broad development prospect. The aim of this study is to provide scientific guidance for its suitable harvest period by measuring yield and quality of “Hua Jin 6” from different harvest time. Studies show that flower size had a slowly rising trend from the first day to the seventh day, and then slowly declined or kept stable. There were no significant differences of total phenolic acid contents in different samples from different days, but contents of total flavonoids were on the rise and up to maximum in the ninth day. The contents of total iridoids had an increasing tendency from the first day to the fifth day and then kept relatively stable in other days. We demonstrated that the quality of “Hua Jin 6” is relatively stable and suitable for harvesting in all flower buds white stage in term of HPLC fingerprints. Our findings can make it possible to select the suitable time for different harvest purpose.

文摘Rho-related GTPase from plants(ROP)proteins play an essential role in plant stress resistance.In this study,the full-length GlROP6 gene was cloned based on G.littoralis transcriptome sequencing data acquired in response to salt stress.The protein sequence,conserved domains,secondary structure,three-dimensional structure,phylogenetic relationships,and expression pattern of the GlROP6 gene were systematically analysed.Our results showed that the full-length GlROP6 gene had an open reading frame of 606 bp,which encoded 201 amino acid residues with a relative molecular weight of 22.23463 kDa and a theoretical isoelectric point of 9.06.Amino acid sequence analyses indicated that the structure of the GlROP6 protein was conserved,and included five G-box motifs(G1–G5),an effector binding region,a Rho insert region and a C-terminal hypervariable region.According to our phylogenetic analysis,the GlROP6 protein was closely related to the ROP protein of Daucus carota subsp.Sativus.Our quantitative real-time PCR results revealed that GlROP6 was highly expressed in flower,and GlROP6 expression was significantly upregulated in G.littoralis roots treated with NaCl.This study will facilitate investigations into the function of GlROP genes in response to salt stress in G.littoralis.
文摘Human papillomavirus 16 (HPV-16) is a major high-risk type causing cervical cancer. The E6 and E7 genes in HPV-16 are the major virulent genes in HPV, and we wanted to investigate the polymorphism of E6 and E7 genes of HPV-16 originated from Jingjiang, Jiangsu province. In research, HPV-16 sample was collected, and the E6 and E7 genes fragments were amplified by PCR assay. The sequences of E6 and E7 genes were used for phylogenetic analysis by Mega 5.0 software. By comparison, the major mutations of E6 gene were T178G (41.67%) and G658A (17%), as well as C58G, T61A and G188C (14%), T61C and C656A (11%). The mutation of E7 gene was mainly C491A and T935A (23%);G514C, G937C and G519C (11%). The 68 nucleotide site deficiency (69.4%) of E6 and 535 nucleotide site deficiency (23%) of E7 were dominant. In our study, we did not find the relevance between the E6 and E7 genes mutations and pathologic severity. But we think the E6 and E7 genes mutation of HPV-16 might affect vaccine prevention and treatment effect.
文摘This study focuses on testing and quality measurement and analysis of VoIPv6 performance. A client, server codes were developed using FreeBSD. This is a step before analyzing the Architectures of VoIPv6 in the current internet in order for it to cope with IPv6 traffic transmission requirements in general and specifically voice traffic, which is being attracting the efforts of research, bodes currently. These tests were conducted in the application level without looking into the network level of the network. VoIPv6 performance tests were conducted in the current tunneled and native IPv6 aiming for better end-to-end VoIPv6 performance. The results obtained in this study were shown in deferent codec's for different bit rates in Kilo bits per second, which act as an indicator for the better performance of G.711 compared with the rest of the tested codes.

文摘To confirm resistance and genetic rules of Xikemai 6 against physiological races of wheat stripe rust,physiological races CYR31,CYR32 and CYR33,Su11-4 and V26 were inoculated in Xikemai 6 and Mingxian 169 and their hybrid progenies F_1,F_2 and F_3 at adult plant stage on March 2015. The results showed that the resistance of Xikemai 6 against CYR31 was controlled by 2 pairs of dominant genes and a pair of recessive genes; the resistance against CYR32 was controlled by three pairs of dominant resistant genes( two pairs of genes performed cumulative effect); the resistance against CYR33 was controlled by a pair of dominant genes and a pair of recessive genes; the resistance against Su11-4 was controlled by a pair of dominant genes and a pair of recessive genes independently or collaboratively; the resistance against V26 was controlled by a pair of dominant genes independently. Due to good performance of Xikemai 6 in test and production,as well as years of resistance identification and genetic analysis,Xikemai 6 was proved to be an excellent cultivar with good resistance against stripe rust,and the inheritance of its resistance was stable,so Xikemai 6 could be used as a germplasm resource and resistance material with excellent comprehensive character. Molecular marker and localization could be further studied,to provide new resistance parents for disease-resistant breeding of wheat.

文摘[ Objective] This paper aimed to investigate the origin, characteristics and molecular evolution of duck derived H4N6 subtype avian influ- enza virus (DK/SH/Y20/06) and enrich the epidemiologic data of the waterfowl origin AIV. [Method] The entire genome of DK/SH/Y20/06 was amplified and subjected to genome sequencing. The molecular software was used for sequence analysis and phylogenetic tree construction of DK/ SH/Y20/06 with some other reference sequences in GenBank. [Result] The results indicated that the amino acid sequence adjacent to HA cleav- age site was PEKASR ↓ GLF, which was the typical characteristics of the LPAIV. The phylogenetic analysis indicated that the HA gene of the isolate was derived from the Eurasian lineage in the eastern hemisphere. The NA gene was at the same branch with A/rnallard/Yan chen/2005( H4N6), sharing 98.3% sequence identity. The PB2, PB1, NP and PA gene of this isolate had genetically close relationships with H6 subtype AIV which is epidemic in China at present. The M gene fell into the same branch with A/environment/Korea/CSM05/2004( H3N1 ). The NS segment had the highest similarity with A/wild duck/Korea/YS44/2004(H1N2). The eight genes were not at the same branch and shared a low similarity with other H4N6 subtype avian influenza viruses isolated in North America. [Condusion] These data showed that DK/SH/Y20/06(H4N6) was possibly a re- combinant virus derived from H4N6 subtype, H6N2, H6N5, H3N1 and H1 N2 subtype AIV by complex gene recombination in duck.
文摘N-aspartylchlorin e6 (talaporfin) concentrations in cancer cells, mouse liver tissues, and human plasma specimens were determined with fluorescence microtiter plate analysis. Talaporfin standard curves were obtained in each of the sample specimens of cells, mouse tissues, or human plasma including serial concentration of talaporfin. The correlation co-efficiencies (r2) of talaporfin standard curves were 0.99-1.00, and the CV less than 5%. Talaporfin incorporation into cells of human breast cancer cell line MCF-7 after incubating with 25 μg/mL talaporfin for up to 24 h revealed that the t-max of the drug incorporation was approximately 5 h, and the maximum drug concentration incorporated was 25 μg/107 cells. Talaporfin incorporation into MCF-7 cells was significantly decreased in the presence of 3 μg/mL cyc-losporine (p < 0.05). Balb/c nu/nu mice implanted human cholangiocarcinoma NOZ cells in liver were administered intravenously 5mg/mouse of talaporfin, and the tissues of normal liver and tumor, as well as plasma specimens, were analyzed for talaporfin concentrations. The mean (SD) of talaporfin concentration in plasma after 30 min of administration was 41.6 (2.3) μg/mL, while the level decreased to undetectable concentrations 2 h after administration. In contrast, the talaporfin concentrations in normal and tumor tissues after 30 min of administration were 1.1-7.8 μg/g tissue, and the level slightly increased or was almost maintained for up to 2-4 h after administration. The heparinized blood of the healthy subjects was incubated with 25 μg/mL talaporfin for up to 24 h. The plasma talaporfin concentration did not significantly change during the incubation, and thus talaporfin appears not to be incorporated into the blood cells. We established rapid and simple analysis procedures of talaporfin in biological specimens using a fluorescence microtiter plate assay. Using this assay procedure, the unique pattern of talaporfin disposition and pharmacokinetics were revealed in human cancer cells, liver tissues of tumor bearing mice, and human blood.
文摘The College English Test Band Six(CET-6)is one of the most widely applied exam across China.As a national teaching test,CET-6 is sponsored by the Department of higher education of the Ministry of education of The People’s Republic of China and held twice a year.In this study,77 sophomores of different majors were tested using the listening test in June 2018.It is ensured that the test method is exactly same as that of CET-6.The purpose of this paper is to analyze the content and specification of CET-6 listening comprehension test,and to evaluate this part in detail according to the real data,including score report and item analysis,trying to provide some enlightenment for the revision of the examination.
