AIM: Genetic polymorphism in enzymes of carcinogen metabolism has been found to have the influence on the susceptibility to cancer. Cytochrome P450 2E1 (CYP2E1) is considered to play an important role in the metabolic activation of procarcinogens such as N-nitrosoamines and low molecular weight organic compounds. The purpose of this study is to determine whether CYP450 2E1 polymorphisms are associated with risks of gastric cancer. METHODS: We conducted a population based case-control study in Changle county, Fujian Province, a high-risk region of gastric cancer in China. Ninety-one incident gastric cancer patients and ninety-four healthy controls were included in our study. Datas including demographic characteristics, diet intake, and alcohol and tobacco consumption of individuals in our study were completed by a standardized questionnaire.PCR-RFLP revealed three genotypes:heterozygote (C1/C2) and two homozygotes (C1/C1 and C2/C2) in CYP2E1. RESULTS: The frequency of variant genotypes (C1/C2 and C2/C2) in gastric cancer cases and controls was 36.3% and 24.5%, respectively. The rare homozygous C2/C2 genotype was found in 6 individuals in gastric cancer group(6.6%), whereas there was only one in the control group (1.1%). However, there was no statistically significant difference between the two groups (two-tailed Fisher's exact test P=0.066). Individuals in gastric cancer group were more likely to carry genotype C1/C2 (odds ratio, OR=1.50) and C2/C2 (OR=7.34) than individuals in control group (chi(2) =4.597, for trend P=0.032). The frequencies of genotypes with the C2 allele (C1/C2 and C2/C2 genotypes) were compared with those of genotypes without C2 allele (C1/C1 genotype) among individuals in gastric cancer group and control group according to the pattern of gastric cancer risk factors. The results show that individuals who exposed to these gastric cancer risk factors and carry the C2 allele seemed to have a higher risk of developing gastric cancer. CONCLUSION: Polymorphism of CYP2E1 gene may have some effect in the development of gastric cancer in Changle county, Fujian Province. 展开更多

AIM:To test the hypothesis that,in the Southeastern Brazilian population,the GSTT1,GSTM1 and CYP2E1 polymorphisms and putative risk factors are associated with an increased risk for gastric cancer. METHODS:We conducted a study on 100 cases of gastric cancer (GC),100 cases of chronic gastritis (CG),and 150 controls (C).Deletion of the GSTT1 and GSTM1 genes was assessed by multiplex PCR.CYP2E1/Pst1 genotyping was performed using a PCR-RFLP assay. RESULTS:No relationship between GSTT1/GSTM1 deletion and the c1/c2 genotype of CYP2E1 was observed among the three groups.However,a significant difference between CG and C was observed,due to a greater number of GSTT1/GSTM1 positive genotypes in the CG group.The GSTT1 null genotype occurred more frequently in Negroid subjects,and the GSTM1 null genotype in Caucasians,while the GSTM1 positive genotype was observed mainly in individuals with chronic gastritis infected with H pylori. CONCLUSION:Our findings indicate that there is no obvious relationship between the GSTT1,GSTM1 and CYP2E1 polymorphisms and gastric cancer. 展开更多

AIM To investigate the contribution of polymorphisms in the CYP1A1, CYP2E1 and EPHX1 genes on sporadic colorectal cancer(SCRC) risk. METHODS Six hundred forty-one individuals(227 patients with SCRC and 400 controls) were enrolled in the study. The variables analyzed were age, gender, tobacco and alcohol consumption, and clinical and histopathological tumor parameters. The CYP1A1 *2A, CYP1A1 *2C CYP2E1 *5B and CYP2E1 *6 polymorphisms were analyzed by polymerase chain reaction-restriction fragment length polymorphism(PCR-RFLP). The EPHX1 Tyr113 His, EPHX1 His139 Arg and CYP1A1 *2C polymorphisms were detected by real-time PCR. Chisquared test and binary logistic regression were used in the statistical analysis. Haplotype analysis was conducted using the Haploview program, version 2.05.RESULTS Age over 6 2 years was a risk factor for SCRC development(OR = 7.54, 95%CI: 4.94-11.50, P < 0.01). Male individuals were less susceptible to SCRC(OR = 0.55, 95%CI: 0.35-0.85, P < 0.01). The CYP2E1*5B polymorphism was associated with SCRC in the codominant(heterozygous genotype: OR = 2.66, 95%CI: 1.64-4.32, P < 0.01), dominant(OR = 2.82, 95%CI: 1.74-4.55, P < 0.01), overdominant(OR = 2.58, 95%CI: 1.59-4.19, P < 0.01), and log-additive models(OR = 2.84, 95%CI: 1.78-4.52, P < 0.01). The CYP2E1*6 polymorphism was associated with an increased SCRC risk in codominant(heterozygous genotype: OR = 2.81, 95%CI: 1.84-4.28, P < 0.01; homozygous polymorphic : OR = 7. 3 2, 9 5 % C I : 1.85-28.96, P < 0.01), dominant(OR = 2.97, 95%CI: 1.97-4.50, P < 0.01), recessive(OR = 5.26, 95%CI: 1.35-20.50, P = 0.016), overdominant(OR = 2.64, 95%CI: 1.74-4.01, P < 0.01), and log-additive models(OR = 2.78, 95%CI: 1.91-4.06, P < 0.01). The haplotype formed by the minor alleles of the CYP2E1*5B(C) and CYP2E1*6(A) polymorphisms was associated with SCRC(P = 0.002). However, the CYP1A1 *2A, CYP1A1 *2C, EPHX1 Tyr113 His and EPHX1 His139 Arg polymorphisms were not associated with SCRC.CONCLUSION In conclusion, the results demonstrated that CYP2E1*5B and CYP2E1*6 minor alleles play a role in the development of SCRC. 展开更多

BACKGROUND Cholangiocarcinoma(CCA)is an intractable cancer,and its incidence in north eastern Thailand is the highest worldwide.Infection with the liver fluke Opisthorchis viverrini(OV)has been associated with CCA risk.However,animal experiments have suggested that OV alone does not induce CCA,but its combination with a chemical carcinogen like nitrosamine can cause experimentally induced CCA in hamsters.Therefore,in humans,other environmental and genetic factors may also be involved.AIM To examine relations between risk for CCA and genetic polymorphisms in carcinogenmetabolizing and inflammation-related genes.METHODS This hospital-based case-control study enrolled 95 case-control pairs matched by age(±5 years)and sex.We examined relations between risk for CCA and genetic polymorphisms in carcinogenmetabolizing and inflammation-related genes,serum anti-OV,alcohol consumption,and smoking.Polymorphisms of CYP2E1,IL-6(-174 and-634),IL-10(-819),and NF-κB(-94)and their cooccurrence with polymorphisms in the drug-metabolizing enzyme gene GSTT1 or GSTM1 were also analyzed.RESULTS Although CCA risk was not significantly associated with any single polymorphism,persons with the GSTT1 wild-type and CYP2E1 c1/c2+c2/c2 genotype had an increased risk(OR=3.33,95%CI:1.23-9.00)as compared with persons having the GSTT1 wild-type and CYP2E1 c1/c1 wild genotype.The presence of anti-OV in serum was associated with a 7-to 11-fold increased risk,and smoking level was related to an OR of 1.5-1.8 in multivariable analyses adjusted for each of the seven genetic polymorphisms.CONCLUSION In addition to infection with OV,gene-gene interactions may be considered as one of the risk factors for CCA development. 展开更多

目的观察银杏叶制剂(CGB)对酒精性肝损伤的防护作用。方法白酒-玉米油混悬液ig给予大鼠造肝损伤模型,连续10周,造模同时分别ig给予银杏叶提取物(GBE)0.8 g·kg-1,CGB 2.4,0.8和0.4 g·kg-1或联苯双酯0.15 g·kg-1。取血测... 目的观察银杏叶制剂(CGB)对酒精性肝损伤的防护作用。方法白酒-玉米油混悬液ig给予大鼠造肝损伤模型,连续10周,造模同时分别ig给予银杏叶提取物(GBE)0.8 g·kg-1,CGB 2.4,0.8和0.4 g·kg-1或联苯双酯0.15 g·kg-1。取血测定各组大鼠血清丙氨酸氨基转移酶(ALT)、天门冬氨酸氨基转移酶(AST)及肝匀浆丙二醛(MDA)、超氧化物歧化酶(SOD)和谷胱甘肽(GSH)。并观察肝组织病理及肝细胞线粒体超微结构变化。采用"cocktail"探针药物法测定大鼠ig给予CGB前后血浆中相应的CYP2E1和CYP3A4酶探针氯唑沙宗和氨苯砜的血药浓度,并计算药代动力学参数。通过药代动力学参数的变化,评价CGB对CYP2E1和CYP3A4酶活性的影响。结果连续ig给予乙醇后,大鼠血清转氨酶显著高于正常对照组(P<0.01),与模型组比较,CGB 0.8和2.4 g·kg-1组ALT和AST显著降低(P<0.05);CGB 0.8和2.4 g·kg-1组肝匀浆MDA显著降低、GSH和SOD明显升高(P<0.05)。ig给予CGB前,模型组大鼠氯唑沙宗和氨苯砜的AUC0-24,cmax均较正常对照组显著降低(P<0.05);CGB可使正常大鼠血浆氯唑沙宗和氨苯砜AUC0-24下降(P<0.05),使模型组大鼠血浆氯唑沙宗和氨苯砜的AUC0-24和cmax显著升高(P<0.05)。结论 CGB对酒精性肝损伤有防护作用,其机制与氧化应激干预相关。 展开更多

CYP4502E1基因多态型对青海藏族男性饮酒行为的影响

目的分析细胞色素P450 2E1(CYP450 2E1)不同基因型及等位基因频率,探讨其与藏族男性饮酒行为之间的关系。方法采用自评式问卷,对325名藏族男性进行调查分析,其中饮酒者193人,不饮酒者132人,另选取266名汉族男性作为对照,采用聚合酶链式... 目的分析细胞色素P450 2E1(CYP450 2E1)不同基因型及等位基因频率,探讨其与藏族男性饮酒行为之间的关系。方法采用自评式问卷,对325名藏族男性进行调查分析,其中饮酒者193人,不饮酒者132人,另选取266名汉族男性作为对照,采用聚合酶链式反应-限制性内切酶片段长度多态性(PCR-RFLP)对基因的多态型进行分析,探讨其与饮酒行为之间的相关性。结果不同职业﹑文化程度﹑经济收入﹑婚姻情况﹑吸烟情况人群组中的饮酒率差异具有显著性(P<0.05)。藏汉族男性基因型及等位基因频率分布差异无显著性(P>0.05)。藏族男性饮酒组和不饮酒组比较,CYP450 2E1三种基因型c1/c1、c1/c2和c2/c2的分布差异具有显著性(χ2=10.101,P<0.05),c1/c2基因型频率在饮酒组比在不饮酒组中的高;c1和c2两个等位基因分布差异也具有显著性(χ2=9.223,P<0.05),c2等位基因频率在饮酒组也比在不饮酒组中的高,与安全饮酒相比,危险饮酒组中c2等位基因频率更高。c2等位基因与是否饮酒以及饮酒量的关联程度分别为0.17和0.20。多因素分析显示:影响饮酒行为的因素依次为CYP450 2E1基因、吸烟和经济收入。结论CYP450 2E1基因是影响藏族男性饮酒行为的主要因素。 展开更多

目的通过Meta分析的方法系统评价细胞色素P4502E1(CYP2E1)基因RsaⅠ/PstⅠ位点多态性与中国人食管癌发病风险的相关性。方法检索PubMed、Embase、CBM、CNKI、VIP和WANFANG数据库中1990年1月1日至2011年11月30日有关CYP2E1基因多态性与... 目的通过Meta分析的方法系统评价细胞色素P4502E1(CYP2E1)基因RsaⅠ/PstⅠ位点多态性与中国人食管癌发病风险的相关性。方法检索PubMed、Embase、CBM、CNKI、VIP和WANFANG数据库中1990年1月1日至2011年11月30日有关CYP2E1基因多态性与食管癌发病风险关系的文献,进行文献筛选、质量评价和数据提取后,采用RevMan5.1软件进行Meta分析。结果最终纳入18篇病例对照研究共19个试验,其中食管癌患者1663例,对照2603例。Meta分析结果显示CYP2E1基因多态性与食管癌的关联有统计学意义[c2vs.c1:OR=0.64,95%CI=0.50~0.81,P=0.0003;c2/c2vs.c1/c1:OR=0.70,95%CI=0.56~0.89,P=0.003;c1/c2vs.c1/c1:OR=0.54,95%CI=0.38~0.75,P=0.0003;c2/c2vs.(c1/c1+c1/c2):OR=0.73,95%CI=0.58~0.92,P=0.008;(c1/c2+c2/c2)vs.c1/c1:OR=0.48,95%CI=0.34~0.70,P=0.0001];汉族的亚组分析结果也显示CYP2E1基因多态性与食管癌的关联有统计学意义[c2vs.c1:OR=0.71,95%CI=0.57~0.89,P=0.002;c2/c2vs.c1/c1:OR=0.75,95%CI=0.59~0.95,P=0.02;c1/c2vs.c1/c1:OR=0.62,95%CI=0.46~0.84,P=0.002;c2/c2vs.(c1/c1+c1/c2):OR=0.78,95%CI=0.61~0.99,P=0.04;(c1/c2+c2/c2)vs.c1/c1:OR=0.56,95%CI=0.40~0.79,P=0.001]。结论对目前相关研究结果的Meta分析显示CYP2E1基因多态性与中国人群食管癌发生风险具有相关性,其RsaⅠ/PstⅠ位点的c2等位基因可能是食管癌的保护或抑制因素。 展开更多

目的研究1,2-二氯乙烷(1,2-dichloroethane,1,2-DCE)染毒过程中不同脑区细胞色素P450 2E1(cytochrome P-450 2E1,CYP2E1)表达的变化,探讨CYP2E1对1,2-DCE中毒引起的脑水肿形成的影响和作用。方法于2018年12月,选择雌性SPF级昆明种小鼠4... 目的研究1,2-二氯乙烷(1,2-dichloroethane,1,2-DCE)染毒过程中不同脑区细胞色素P450 2E1(cytochrome P-450 2E1,CYP2E1)表达的变化,探讨CYP2E1对1,2-DCE中毒引起的脑水肿形成的影响和作用。方法于2018年12月,选择雌性SPF级昆明种小鼠40只随机分为对照组、染毒1、2和3 d组共4组,每组10只。将染毒组小鼠置于100 L静式染毒柜中(5只/柜),以1.2 mg/L 1,2-DCE每天吸入染毒3.5 h,分别染毒1、2和3 d,对照组不进行染毒,其他处理方式与染毒组相同。分别于染毒结束后次日处死,取脑组织并分区,采用HE染色法对不同脑区进行组织病理学观察,检测不同脑区丙二醛(malondialdehyde,MDA)、还原型谷胱甘肽(glutathione,GSH)含量和过氧化氢酶(catalase,CAT)活力;同时以蛋白免疫印迹(Western blotting)法检测不同脑区中CYP2E1、occludin和claudin5蛋白含量,再以实时荧光定量PCR法检测CYP2E1、occludin和claudin5 mRNA表达水平。实验数据采用SPSS 20.0进行统计分析,多组组间比较采用单因素方差分析(one-way ANOVA),组间两两比较采用SNK(q检验)法分析。结果与对照组比较,染毒2 d和3 d组小鼠小脑组织出现明显的脑水肿改变;与对照组比较,染毒3 d组小鼠小脑组织中MDA含量、CYP2E1蛋白和mRNA表达水平明显增高,差异均有统计学意义(P<0.05);与对照组比较,各个染毒组小鼠小脑组织中GSH含量、CAT活力及occludin和claudin5蛋白表达水平明显降低,差异均有统计学意义(P<0.05)。各组小鼠额叶皮质组织中上述指标差异均无统计学意义(P>0.05)。结论 1,2-DCE染毒可以诱导小鼠小脑组织中CYP2E1的表达增强,引起脑组织氧化损伤和脑水肿,对小鼠额叶皮质组织中CYP2E1的表达无明显影响。 展开更多

Cytochrome P450 CYP2E1 is mainly present in hepatocytes in the livers of mammals,where it plays an important role in the metabolism of xenobiotic organic substances. Previous studies showed that transgenic petunia(Petunia hybrid) plants harboring a mammalian cyp2e1 gene(designated cyp2e1-transgenic petunia) exhibited increased resistance to formaldehyde stress. In this study,we used cyp2e1-transgenic petunia plants to analyze physiological indexes related to formaldehyde stress responses. The results indicated that under formaldehyde stress,the malondialdehyde content in cyp2e1-transgenic petunia plants was lower than in β-glucuronidase gene(gus)-transgenic and wild-type petunia plants. The activities of both superoxide dismutase and peroxidase in the cyp2e1-transgenic plants were higher than in gus-transgenic and wild-type plants. The alcohol dehydrogenase activity was slightly increased and more glutathione was consumed. Additionally,under formaldehyde stress,the levels of plant hormones including indole-3-acetic acid,zeatin and abscisic acid in cyp2e1-transgenic petunia plants displayed decreasing trends,whereas the level of gibberellic acid displayed an increasing trend. In contrast,the indole-3-acetic acid,zeatin and abscisic acid levels in gus-transgenic and wild-type petunia plants displayed increasing trends,whereas the gibberellic acid level displayed a decreasing trend. At 72 h after incubation of 0.5 g of cyp2e1-transgenic petunia plants in 40 mL of treatment solution containing formaldehyde at 50 mg·L^(-1),the formaldehyde content remaining in the treatment solution was close to zero while approximately half of original formaldehyde remained in the treatment solutions containing gus-transgenic and wild-type petunia plants. 展开更多
