Understanding the molecular mechanisms underlying Pt effector-mediated wheat susceptibility remains largely unexplored. In this study, an effector Pt_21 was identified to interact with the apoplast-localized wheat thaumatin-like protein TaTLP1 using a yeast two-hybrid assay and the Pt_21-TaTLP1 interaction was characterized. The interaction between Pt_21 and TaTLP1 was validated by in vivo co-immunoprecipitation assay. A TaTLP1 variant,TaTLP1C71A, that was identified by the site-directed mutagenesis failed to interact with Pt_21. Pt_21was able to suppress Bax-mediated cell death in leaves of Nicotiana benthamiana and inhibit TaTLP1-mediated antifungal activity. Furthermore, infiltration of recombinant protein Pt_21 into leaves of transgenic wheat line overexpressing TaTLP1 enhanced the disease development of leaf rust compared to that in wild-type leaves. These findings demonstrate that Pt_21 suppresses host defense response by directly targeting wheat TaTLP1 and inhibiting its antifungal activity, which broadens our understanding of the molecular mechanisms underlying Pt effector-mediated susceptibility in wheat.展开更多
Delays in sowing have significant effects on the grain yield,yield components,and grain protein concentrations of winter wheat.However,little is known about how delayed sowing affects these characteristics at differen...Delays in sowing have significant effects on the grain yield,yield components,and grain protein concentrations of winter wheat.However,little is known about how delayed sowing affects these characteristics at different positions in the wheat spikes.In this study,the effects of sowing date were investigated in a winter wheat cultivar,Shannong 30,which was sown in 2019 and 2020 on October 8(normal sowing)and October 22(late sowing)under field conditions.Delayed sowing increased the partitioning of ^(13)C-assimilates to spikes,particularly to florets at the apical section of a spike and those occupying distal positions on the same spikelet.Consequently,the increase in grain number was the greatest for the apical sections,followed by the basal and central sections.No significant differences were observed between sowing dates in the superior grain number in the basal and central sections,while the number in apical sections was significantly different.The number of inferior grains in each section also increased substantially in response to delayed sowing.The average grain weights in all sections remained unchanged under delayed sowing because there were parallel increases in grain number and ^(13)C-assimilate partitioning to grains at specific positions in the spikes.Increases in grain number m^(–2) resulted in reduced grain protein concentrations as the limited nitrogen supply was diluted into more grains.Delayed sowing caused the greatest reduction in grain protein concentration in the basal sections,followed by the central and apical sections.No significant differences in the reduction of the grain protein concentration were observed between the inferior and superior grains under delayed sowing.In conclusion,a 2-week delay in sowing improved grain yield through increased grain number per spike,which originated principally from an increased grain number in the apical sections of spikes and in distal positions on the same spikelet.However,grain protein concentrations declined in each section because of the increased grain number and reduced N uptake.展开更多

[Objective] The aim of this study was to determine the variation of phytic acid content (PAC) and its frequency distribution, as well as the relationship between PAC and protein content, kernel characteristics in 16...[Objective] The aim of this study was to determine the variation of phytic acid content (PAC) and its frequency distribution, as well as the relationship between PAC and protein content, kernel characteristics in 161 Chinese winter wheat cultivars from four regions. [Method] One hundred and sixty-one winter wheat varieties from China Wheat Zones I, II, III and IV (Table 1) were grown in a randomized block de- sign, in the 2009-2011 cropping season; and then the indexes for describing the grain morphological characteristics such as the thousand kernel weight (TKW), kernel length (KL), kernel width (KW) and kernel thickness (KT) were measured; the phytic acid content (PAC), protein content and sedimentation value were also determined; finally, the relationship between PAC and protein content, kernel characteristics were analyzed. [Result] The PAC in the cultivars tested ranged from 0.92% to 1.95% with a mean value of 1.41%. Protein content ranged from 12.60% to 19.20%, with a mean of 15.24%. Most (53.4%) of the wheat genotypes had a PAC value in the range of 1.25% to 1.55%. No significant correlation was found between PAC and protein content, sedimentation value, while protein content and SDS sedimentation value was significant correlated, which suggested the possibility of breeding wheat cultivars that have a low PAC but a high protein content and good gluten quality. There was a high correlation between TKW and KW (,.=0.79), KL (r=0.50) and KT (r=0.64). PAC was found having no significant correlation with TKW, KW, KL and KT. [Conclusion] The result suggests the possibility of breeding wheat cultivars that have a low PAC but high kernel weight.展开更多
Photosynthesis and chlorophyll a fluorescence parameters, photochemical efficiency of PS II (Fv/Fm), photochemical quenching of PS II (qP), nonphotochemical quenching of PS II (NPQ), maximum activity of PS II (...Photosynthesis and chlorophyll a fluorescence parameters, photochemical efficiency of PS II (Fv/Fm), photochemical quenching of PS II (qP), nonphotochemical quenching of PS II (NPQ), maximum activity of PS II (Fv/Fo) as well as electron transport rate (ETR), and quantum yield of PS II (ФPS II) were measured on flag leaves of the winter wheat treated by methanol at different concentrations. The results revealed that photosynthesis was greatly improved by methanol, as indicated by higher photosynthetic rates and stomatal conductance. The enhancement effect of methanol on photosynthesis was maintained for 3-4 days. Different methanol concentration treatments also increased intercellular CO2 concentration and transpiration rates. No significant decline was found in Fv/Fm, Fv/Fo, and ФPS II, which revealed no photoinhibition during methanol application in different methanol concentrations. Methanol showing no apparent inhibitory effects indicated higher potential photosynthetic capacity of flag leaves of winter wheat. However, the increase in photosynthesis was not followed by an increase in the photosynthetic activity (Fv/Fm), and fluorescence parameters did not indicate an improvement in intercellular CO2 concentration and PS II photochemical efficiency compared with the control, thereby encouraging us to propose that lower leaf temperatures caused by applied methanol would reduce both dark respiration and photorespiration (most importantly), thus, increasing net CO2 uptake and photosynthetic rates.展开更多

The results showed an obvious increase in soluble protein content. Compared with control, Chl a, Chl b, total Chl contents and the ratio of Chl a to Chl b all decreased to some extent. ATP content increased in some varieties and decreased in others, but the stronger the drought resistance of the variety , the higher the ATP content. The correlation coefficient(r)of the soluble protein content, ratio of Chl a to Chl b and ATP content as a percentage of the drought-resistant sweet potato control variety are 0. 8968, - 0. 8509 and 0. 8200, respectively, P<0. 01. So these indices can be used to evaluate the drought resistance of different sweet potato varieties.展开更多

The intermediates of chlorophyll biosynthesis were analyzed in winter wheat cultivars in soil culture, miniblock culture, and solution culture to study the effects of Mo on chlorophyll biosynthesis without Mo addition (CK, soil available Mo 0.112 mg kg^-1) and Mo addition (+ Mo, 0.13 mg kg^-1 Mo was added). Laevulinic acid (LA), the competitive analog of δ-aminolaevulinic acid (ALA) was also introduced in the experiment. The ratio of Chl a/Chl b was constant between CK and + Mo treatment, whereas it increased at low temperature, which indicated that Mo deficiency did not inhibit the transformation of Chl a to Chl b at low temperature. Under Mo deficiency, the contents of protochlorophyll (Pchl), Mg-protoporphyrin Ⅸ (Mg-Proto Ⅸ), protoporphyrin Ⅸ (proto IX), and uroporphyrinogen Ⅲ (Uro Ⅲ) decreased [Uro Ⅲ decreased significantly (P 〈0.01)], whereas ALA and glutamate increased significantly (P 〈 0.01) compared with that of Mo addition, which suggested that the transformation from ALA to Uro Ⅲ might be inhibited. The content of ALA reversed after addition of LA, it was significantly higher (P 〈 0.01) in Mo addition than in CK. The results indicated that the transformation from ALA to Uro Ⅲ was blocked under Mo deficiency, which resulted in the inhibition of the biosynthesis of chlorophyll and led to the decrease of chlorophyll in winter wheat cultivars.展开更多

Results showed that the actual photochemical efficiency of PSⅡ reaction center (F PSⅡ)decreased significantly as leaf water stress progressed, however, the F PS was increased by nitrogen fertilization. The F PSⅡ of 0, 90 and180 kg ha-1 nitrogen treatments at noon were 0.197, 0.279 and 0.283, respectively, which decreased by 57.7, 56.4 and 40.2% as comparedto those in the morning. In the afternoon, the F PSⅡ partialy or completely recovered to the levels in the morning. The values of F PS Ⅱin 0 and 90kgha-1 treatments recovered to 87.3 and 81.5% of those in the morning. In 180kgha-1 treatment, the F PSⅡ in the afternoonwas even higher than that in the morning. Application of nitrogen fertilizer significantly increased maximum photochemical efficiency(Fv/Fm), photochemical quenching coefficient (qP) and non-photochemical quenching coefficient (qNP). These results indicated thatapplication of nitrogen fertilizer could increase the light energy conversion efficiency, the potential activity of photosynthetic reactioncenter, and the non-photochemical dissipation of excess light energy, which can prevent leaf photosynthetic apparatus from damage ofenvironmental stress. However, there was no significant difference in the values of F PSⅡbetween 90 and 180kgha-1 nitrogentreatments, indicating that the excess nitrogen was unfavorable to photosynthesis.展开更多

We characterized the Lhca and Lhcb gene families of Z. marina from the expressed sequence tags (EST) database. In total, 13 unigenes were annotated as ZmLhc, 6 in Lhca family and 7 in ZmLhcb family. ZmLHCA and ZmLHCB contained the conservative LHC motifs and amino acid residues binding chlorophyll. The average similarity among mature ZmLHCA and ZmLHCB was 48.91% and 48.66%, respectively, which indicated a high degree of diver- gence within ZmLHChc gene family. The reconstructed phylogenetic tree showed that the tree topology and phylogenetic relation- ship were similar to those reported in other high plants, suggesting that the Lhc genes were highly conservative and the classification of ZmLhc genes was consistent with the evolutionary position of Z. marina. Real-time reverse transcription (RT) PCR analysis showed that different members of ZmLhca and ZmLhcb responded to a stress in different expression patterns. Salinity, temperature, light intensity and light quality may affect the expression of most ZmLhca and ZmLhcb genes. Inorganic carbon concentration and acidity had no obvious effect on ZmLhca and ZmLhcb gene expression, except for ZmLhca6.展开更多

T1 plants were further tested by PCR and Southern hybridization. Results demonstrated that the alien resistance gene had been integrated into the wheat genome. The segregation ratio of CP1+ to CP1- in T1 generation was 1.0 to 1. 3, and didn't agree with Mendelian rule. RT-PCR result from T2 plants showed that the alien gene CWMV-CP1 had stable expression in wheat genetic background.展开更多

Here we...The pathogenesis-related proteins 1 (PR-1) gene family play important roles in the plant metabolism in response to biotic and abiotic stresses. The wheat TdPR1.2 has been previously isolated and characterized. Here we showed by bio-informatic analysis that TdPR1.2 contains six cysteine residues that are conserved between all PR-1 proteins tested. Using ScanProsite tool, we found that TdPR1.2 structure has a CRISP family signature 1 and 2 located at the C-terminal part of the protein. Those two domains are conserved in many identified PR1.2 proteins in plants. Moreover, SignalIP-5.0 analysis revealed that TdPR1.2 contains a putative signal peptide formed by 25 amino acids at the N-terminal extremity. The presence of this signal peptide suggested that the mature proteins will be secreted after the cleavage of the signal sequence. Further, we investigate the role of the TdPR1.2 proteins in the growth of <i>Escherichia coli</i> transformants cells under different abiotic stresses. Our results showed that the full-length form of TdPR1.2 enhanced tolerance of <i>E. coli</i> against salt and osmotic stress but not to KCl. Moreover, TdPR1.2 protein confers bacterial tolerance to heavy metals in solid and liquid mediums. Based on these results, we suggest that the TdPR1.2 protein could play an important role in response to abiotic stress conditions.展开更多

OTC is toxic to wheat. The wheat growth, especially wheat root was significantly decreased. Further OTC also significantly decreased root activity, chlorophyll contents, and photosynthetic parameters except for intercellular CO2 concentrations. The different responses of indicators such as root number, root activity and so on to OTC were also observed. The IC50 values for the tested indicators to OTC ranged from 7.1 to 113.4 mmol L-1 OTC. The order of indicator sensitivity to OTC was root number stomatal conductance chlorophyll a total chlorophyll photosynthetic rates total surface area transpiration rate chlorophyll b fresh weight of root dry weight of root total length dry weight of shoot = fresh weight of shoot total volume. The root number was more sensitive than other indicators with the IC50 value of 7.1 mmol L-1 OTC, and could be taken as the sensitive indicator to predict the hazards of OTC to wheat.展开更多

Year, cultivar, and the interaction between the year and...The effects of protein characteristics and the proportion of gluten on end-use quality in 13 Korean wheat cultivars for three years were verified in this study. Year, cultivar, and the interaction between the year and the cultivar influenced protein characteristics, the proportion of gluten except for γ-and ω-gliadin using RP-HPLC(reversed-phase high-performance liquid chromatography), and end-use quality. Protein characteristics and the proportion of gluten in Korean wheat cultivars were between those of Australian standard white(ASW) and hard wheat(AH). Korean wheat cultivars exhibited a higher average α+β gliadin proportion than imported wheat, a γ-gliadin proportion similar to that of dark northern spring wheat, and the same ω-gliadin proportion as AH. They showed a bread-loaf volume intermediate between those of ASW and AH and a texture of cooked noodles similar to that of soft white wheat, but less springiness than imported wheat. The cookie diameter of Korean wheat cultivars was similar to that of hard red winter wheat. There was a correlation between bread-loaf volume and protein characteristics, except for the protein content in Korean wheat cultivars. Springiness and cohesiveness of cooked noodles were not correlated with protein characteristics, while hardness was correlated with the protein content and water absorption of a mixograph. Cookie diameter was negatively correlated with the sodium dodecyl sulfate(SDS) sedimentation volume and water absorption of a mixograph. The end-use quality was not correlated with any proportion of gluten composition. Principal component analysis(PCA) showed that the proportion of gluten was not related to the quality of the bread(both PCs, 81.3%), noodle(77.7%), and cookie(82.4%). PCA explained that Keumkang is suitable for superior bread, while Uri is good for cooked noodles and cookies.展开更多

Among various genes involved in the plant-pathogen related (PR) protein genes determine different defense responses tritici) with wheat plants activate a w^ae range OT nost nteractions, the expressions of particular pathogenesis-Different types of resistance have been recognized and utilized for developing wheat cultivars for resistance to stripe rust. All-stage resistance can be detected in seedling stage and remains at high levels throughout the plant growth stages. This type of resistance is race-specific and not durable. In contrast, plants with only high-temperature adult-plant (HTAP) resistance are susceptible in seedling stage, but become resistant when plants grow older and the weather becomes warmer. HTAP resistance controlled by a single gene is partial, but usually non-race specific and durable. The objective of this study was to analyze the expression of PR protein genes involved in different types of wheat resistance to stripe rust. The expression levels of 8 PR protein genes (PR1, PRI.2, PR2, PR3, PR4, PR5, PR9 and PRIO) were quantitatively evaluated at 0, 1, 2, 7 and 14 days after inoculation in single resistance gene lines of wheat with all-stage resistance genes YrTrl, Yr76, YrSP and YrExp2 and lines carrying HTAP resistance genes Yr52, Yr59, Yr62 and Yr7B. Races PSTv-4 and PSTv-37 for compatible and incompatible interactions were used in evaluation of PR protein gene expression in wheat lines carrying all-stage resistance genes in the seedling- stage experiment while PSTv-37 was used in the HTAP experiment. Analysis of quantitative real-time polymerase chain reaction (qRT-PCR) revealed that all of the PR protein genes were involved in the different types of resistance controlled by different Yr genes. However, these genes were upregulated at different time points and at different levels during the infection process among the wheat lines with different Yr genes for either all-stage resistance or HTAP resistance. Some of the genes were also induced in compatible interactions, but the levels were almost always higher in the incompatible interaction than in the compatible interaction at the same time point for each Yr gene. These results indicate that both salicylic acid and jasmonate signaling pathways are involved in both race-specific all-stage resistance and non-race specific HTAP resistance. Although expressing at different stages of infection and at different levels, these PR protein genes work in concert for contribution to different types of resistance controlled by different Yr genes.展开更多
A field experiment using a split-plot randomized complete block design with three replications was carried out to determine relationships between spectral indices and wheat grain yield (GY), to compare the performan...A field experiment using a split-plot randomized complete block design with three replications was carried out to determine relationships between spectral indices and wheat grain yield (GY), to compare the performance of four vegetation indices (VIs) for GY prediction, and to study the feasibility of VI to estimate grain protein content (GPC) in winter wheat. Two typical winter wheat (Triticum aestivum L.) cultivars 'Xuzhou 26' (high protein content) and 'Huaimai 18' (low protein content) were used as the main plot treatments and four N rates, i.e., 0, 120, 210, and 300 kg N ha^-1, as the sub-plot treatments. Increasing soil N supply significantly increased GY and GPC (P ≤ 0.05). For the two cultivars combined, significant and positive correlations were found between four VIs and GY, with the strongest relationship observed when using the green ratio vegetation index (GRVI) at mid-filling. Cumulative VI estimates improved yield predictions substantially, with the best interval being heading to maturity stage. Similar results were found between VI and grain protein yield. However, when using cumulative VI, GPC showed no significant improvement. The strong relationship between leaf N status and GPC (R2 =0.9144 for 'Xuzhou 26' and R2 = 0.8285 for 'Huaimai 18') indicated that canopy spectra could be used to predict GPC. The strong fit between estimated and observed GPC (R2 = 0.7939) indicated that remote sensing techniques were potentially useful predictors of grain protein content and quality in wheat.展开更多

Kohdasht) and two winter (cvs. Sardari and Norstar) cul...The relationship between vernalization requirement and quantitative and qualitative changes in total leaf soluble proteins were determined in one spring (cv. Kohdasht) and two winter (cvs. Sardari and Norstar) cultivars of wheat (Triticum aestivum L.) exposed to 4℃. Plants were sampled on days 2, 14, 21 and 35 of exposure to 4℃. The final leaf number (FLN) was determined throughout the vernalization periods (0, 7, 14, 24, and 35 d) at 4℃. The final leaf number decreased until days 24 and 35 in Sardari and Norstar eultivars, respectively, indicating the vernalization saturation at these times. No clear changes were detected in the final leaf number of Kohdash cultivar, verifying no vernalization requirement for this spring wheat cultivar. Comparing with control, clear cold-induced 2-fold increases in proteins quantity occurred after 48 h following the 4℃-treatment in the leaves of the both winter wheat cultivars but, such response was not detected in the spring cultivar. However, the electrophoretic protein patterns showed between-cultivar and between-temperature treatment differences. With increasing exposure time to 4℃, the winter cultivars tended to produce more HMW polypeptides than the spring cultivar. Similar proteins were induced in both Sardari and Norstar winter wheat cultivars, however, the long vernalization requirement in Norstar resulted in high level and longer duration of expression of cold-induced proteins compared to Sardari with a short vernalization requirement. Optimization of tissue culture conditions and attempts of adding signa...The regeneration rate of wheat immature embryo varies among genotypes, howbeit many elite agriculture wheat varieties have low regeneration rates. Optimization of tissue culture conditions and attempts of adding signal molecules are effective ways to increase plant regeneration rate. Inter-culture is one of ways that have not been investigated in plant tissue culture. Moreover, the use of arabinogalactan proteins (AGPs) and hydrogen peroxide (H202) have been reported to increase regeneration rate in a few plant species other than wheat. The current research pioneeringly uses inter-culture of immature embryos of different wheat genotypes, and also investigates impacts of AGP and H2O2 on the induction of embryogenic calli and plant regeneration. As a result, high-frequency regeneration wheat cultivars Kenong 199 (KN 199) and Xinchun 9 (XC9), together with low-frequency regeneration wheat line Chinese Spring (CS), presented striking increase in the induction of embryogenic calli and plant regeneration rate of CS through inter-culture strategy, up to 52.19 and 67.98%, respectively. Adding 50 to 200 mg L-1 AGP or 0.005 to 0.01‰ H2O2 to the callus induction medium, enhanced growth of embryogenic calli and plant regeneration rate in quite a few wheat genotypes. At 50 mg L-1 AGP application level in callus induction medium plant regeneration rates of 8.49,409.06 and 283.16% were achieved for Jimai 22 (JM22), Jingdong 18 (JD18) and Yangmai 18 (YM18), respectively; whereas at 100 mg L-1 AGP level, CS (105.44%), Chuannong 16 (CN16) (80.60%) and Ningchun 4 (NC4) (62.87%) acted the best. Moreover CS (79.05%), JM22 (7.55%), CN16 (101.87%), YM18 (365.56%), Yangmai 20 (YM20) (10.48%), and CB301 (187.40%) were more responsive to 0.005 %o of H2O2, and NC4 (35.37%) obtained the highest shoot regeneration rates at 0.01%o of H2O2. Overall, these two methods, inter-culture and AGP (or H2O2) application, can be further applied to wheat transgenic research.展开更多
Waterlogging is one of the most factors limiting wheat production in the middle and lower reaches of the Yangtze River Plain of China,especially in the middle and late stages of wheat.Wheat varieties‘Jingmai102’(JM1...Waterlogging is one of the most factors limiting wheat production in the middle and lower reaches of the Yangtze River Plain of China,especially in the middle and late stages of wheat.Wheat varieties‘Jingmai102’(JM102)and‘Yangmai158’(YM158)were planted to study the dynamic changes of photosynthetic characteristics in flag leaf and the influence of waterlogging at anthesis on the yield and components and dry matter accumulation and remobilization of winter wheat in above ground.The results showed that the SPAD values slightly increased at 1 day after anthesis(d),and then kept decreasing with the increase of waterlogging time.The decrease in SPAD value was more remarkably in YM158 than that in JM102.As for the chlorophyll fluorescence parameters,the photochemical efficiency(Fv/Fm),potential activity(Fv/Fo)of photosystem II,and electronic transmission(Fm/Fo)on photosystem II increased first and then decreased with the increase of waterlogging days after anthesis.The quantum ratio of heat dissipation(Fo/Fm)had a tendency opposite to that of Fv/Fm,and the change range of JM102 was lower than that of YM158.For the grain yield and components,waterlogging at anthesis decreased the dry weight of single stem,grain yield,1 000-kernel weight,spikelet per panicle,and harvest index,and the reduction of JM102 was smaller than that of YM158.As for the accumulation and remobilization of dry matter,the accumulation of dry matter after anthesis decreased significantly under waterlogging condition(WL),and the reduction of JM102 was smaller than that of YM158.In conclusion,waterlogging at anthesis significantly affected the photosynthetic characteristics,yield and components in both varieties,but different varieties exhibited different tolerances to waterlogging stress and YM158 was more sensitive to water stress than JM102.展开更多

文摘The grain protein content(GPC)is the key parameter for wheat grain nutritional quality.This study conducted a resampling GWAS analysis using 406 wheat accessions across eight environments,and identified four previously reported GPC QTLs.An analysis of 87 landraces and 259 modern cultivars revealed the loss of superior GPC haplotypes,especially in Chinese cultivars.These haplotypes were preferentially adopted in different agroecological zones and had broad effects on wheat yield and agronomic traits.Most GPC QTLs did not significantly reduce yield,suggesting that high GPC can be achieved without a yield penalty.The results of this study provide a reference for future GPC breeding in wheat using the four identified QTLs.

文摘Puccinia triticina(Pt), as the causal agent of wheat leaf rust, employs a plethora of effector proteins to modulate wheat immunity for successful colonization. Understanding the molecular mechanisms underlying Pt effector-mediated wheat susceptibility remains largely unexplored. In this study, an effector Pt_21 was identified to interact with the apoplast-localized wheat thaumatin-like protein TaTLP1 using a yeast two-hybrid assay and the Pt_21-TaTLP1 interaction was characterized. The interaction between Pt_21 and TaTLP1 was validated by in vivo co-immunoprecipitation assay. A TaTLP1 variant,TaTLP1C71A, that was identified by the site-directed mutagenesis failed to interact with Pt_21. Pt_21was able to suppress Bax-mediated cell death in leaves of Nicotiana benthamiana and inhibit TaTLP1-mediated antifungal activity. Furthermore, infiltration of recombinant protein Pt_21 into leaves of transgenic wheat line overexpressing TaTLP1 enhanced the disease development of leaf rust compared to that in wild-type leaves. These findings demonstrate that Pt_21 suppresses host defense response by directly targeting wheat TaTLP1 and inhibiting its antifungal activity, which broadens our understanding of the molecular mechanisms underlying Pt effector-mediated susceptibility in wheat.

文摘Delays in sowing have significant effects on the grain yield,yield components,and grain protein concentrations of winter wheat.However,little is known about how delayed sowing affects these characteristics at different positions in the wheat spikes.In this study,the effects of sowing date were investigated in a winter wheat cultivar,Shannong 30,which was sown in 2019 and 2020 on October 8(normal sowing)and October 22(late sowing)under field conditions.Delayed sowing increased the partitioning of ^(13)C-assimilates to spikes,particularly to florets at the apical section of a spike and those occupying distal positions on the same spikelet.Consequently,the increase in grain number was the greatest for the apical sections,followed by the basal and central sections.No significant differences were observed between sowing dates in the superior grain number in the basal and central sections,while the number in apical sections was significantly different.The number of inferior grains in each section also increased substantially in response to delayed sowing.The average grain weights in all sections remained unchanged under delayed sowing because there were parallel increases in grain number and ^(13)C-assimilate partitioning to grains at specific positions in the spikes.Increases in grain number m^(–2) resulted in reduced grain protein concentrations as the limited nitrogen supply was diluted into more grains.Delayed sowing caused the greatest reduction in grain protein concentration in the basal sections,followed by the central and apical sections.No significant differences in the reduction of the grain protein concentration were observed between the inferior and superior grains under delayed sowing.In conclusion,a 2-week delay in sowing improved grain yield through increased grain number per spike,which originated principally from an increased grain number in the apical sections of spikes and in distal positions on the same spikelet.However,grain protein concentrations declined in each section because of the increased grain number and reduced N uptake.

文摘[ Objective] The experiment aimed to provide a theoretical base of optimal cultivation management for the high yield and good quality and high efficiency of winter wheat. [ Method] The effects of two sulfur fertilizer application methods on dynamic changes of grain protein content and glutenin content of Yumai 49 and Yumai 66 during wheat grain filling stage were studied under the field conditions. [Result] Both the grain protein and glutenin content of two cultivars were increased by sulfur fertilizer, particularly, the effects on Yumai 49 were more significant.[ Conclusion] The grain content and glutenin content of different wheat cultivars could be increased by taking different sulfur fertilizer application methods.

文摘[Objective] The aim of this study was to determine the variation of phytic acid content (PAC) and its frequency distribution, as well as the relationship between PAC and protein content, kernel characteristics in 161 Chinese winter wheat cultivars from four regions. [Method] One hundred and sixty-one winter wheat varieties from China Wheat Zones I, II, III and IV (Table 1) were grown in a randomized block de- sign, in the 2009-2011 cropping season; and then the indexes for describing the grain morphological characteristics such as the thousand kernel weight (TKW), kernel length (KL), kernel width (KW) and kernel thickness (KT) were measured; the phytic acid content (PAC), protein content and sedimentation value were also determined; finally, the relationship between PAC and protein content, kernel characteristics were analyzed. [Result] The PAC in the cultivars tested ranged from 0.92% to 1.95% with a mean value of 1.41%. Protein content ranged from 12.60% to 19.20%, with a mean of 15.24%. Most (53.4%) of the wheat genotypes had a PAC value in the range of 1.25% to 1.55%. No significant correlation was found between PAC and protein content, sedimentation value, while protein content and SDS sedimentation value was significant correlated, which suggested the possibility of breeding wheat cultivars that have a low PAC but a high protein content and good gluten quality. There was a high correlation between TKW and KW (,.=0.79), KL (r=0.50) and KT (r=0.64). PAC was found having no significant correlation with TKW, KW, KL and KT. [Conclusion] The result suggests the possibility of breeding wheat cultivars that have a low PAC but high kernel weight.
文摘Photosynthesis and chlorophyll a fluorescence parameters, photochemical efficiency of PS II (Fv/Fm), photochemical quenching of PS II (qP), nonphotochemical quenching of PS II (NPQ), maximum activity of PS II (Fv/Fo) as well as electron transport rate (ETR), and quantum yield of PS II (ФPS II) were measured on flag leaves of the winter wheat treated by methanol at different concentrations. The results revealed that photosynthesis was greatly improved by methanol, as indicated by higher photosynthetic rates and stomatal conductance. The enhancement effect of methanol on photosynthesis was maintained for 3-4 days. Different methanol concentration treatments also increased intercellular CO2 concentration and transpiration rates. No significant decline was found in Fv/Fm, Fv/Fo, and ФPS II, which revealed no photoinhibition during methanol application in different methanol concentrations. Methanol showing no apparent inhibitory effects indicated higher potential photosynthetic capacity of flag leaves of winter wheat. However, the increase in photosynthesis was not followed by an increase in the photosynthetic activity (Fv/Fm), and fluorescence parameters did not indicate an improvement in intercellular CO2 concentration and PS II photochemical efficiency compared with the control, thereby encouraging us to propose that lower leaf temperatures caused by applied methanol would reduce both dark respiration and photorespiration (most importantly), thus, increasing net CO2 uptake and photosynthetic rates.
文摘Some indices concerning the metabolism of substance and energy in sweet potato leaves under water stress were studied. The results showed an obvious increase in soluble protein content. Compared with control, Chl a, Chl b, total Chl contents and the ratio of Chl a to Chl b all decreased to some extent. ATP content increased in some varieties and decreased in others, but the stronger the drought resistance of the variety , the higher the ATP content. The correlation coefficient(r)of the soluble protein content, ratio of Chl a to Chl b and ATP content as a percentage of the drought-resistant sweet potato control variety are 0. 8968, - 0. 8509 and 0. 8200, respectively, P<0. 01. So these indices can be used to evaluate the drought resistance of different sweet potato varieties.

In different upgrowth phases,or different heat-resistant crop varieties,the heat shock protein exist differences. Therefore,study on differences of heat shock protein has significant theoretical and practical meaning for researching heat resistance of wheat. By using SDS-PAGE method,the inducing conditions and manifestation of heat shock protein were studied,which provided theoretical basis for yield increasing of wheat.

文摘The objective was to probe the site where the biosynthesis of chlorophyll was blocked under Mo deficiency at low temperature, which led to the decrease of chlorophyll in winter wheat cultivars. The intermediates of chlorophyll biosynthesis were analyzed in winter wheat cultivars in soil culture, miniblock culture, and solution culture to study the effects of Mo on chlorophyll biosynthesis without Mo addition (CK, soil available Mo 0.112 mg kg^-1) and Mo addition (+ Mo, 0.13 mg kg^-1 Mo was added). Laevulinic acid (LA), the competitive analog of δ-aminolaevulinic acid (ALA) was also introduced in the experiment. The ratio of Chl a/Chl b was constant between CK and + Mo treatment, whereas it increased at low temperature, which indicated that Mo deficiency did not inhibit the transformation of Chl a to Chl b at low temperature. Under Mo deficiency, the contents of protochlorophyll (Pchl), Mg-protoporphyrin Ⅸ (Mg-Proto Ⅸ), protoporphyrin Ⅸ (proto IX), and uroporphyrinogen Ⅲ (Uro Ⅲ) decreased [Uro Ⅲ decreased significantly (P 〈0.01)], whereas ALA and glutamate increased significantly (P 〈 0.01) compared with that of Mo addition, which suggested that the transformation from ALA to Uro Ⅲ might be inhibited. The content of ALA reversed after addition of LA, it was significantly higher (P 〈 0.01) in Mo addition than in CK. The results indicated that the transformation from ALA to Uro Ⅲ was blocked under Mo deficiency, which resulted in the inhibition of the biosynthesis of chlorophyll and led to the decrease of chlorophyll in winter wheat cultivars.

文摘The effect of nitrogen fertilization on leaf chlorophyll fluorescence was studied in field-grown winter wheat during grain filling underrainfed conditions in Loess Plateau. Results showed that the actual photochemical efficiency of PSⅡ reaction center (F PSⅡ)decreased significantly as leaf water stress progressed, however, the F PS was increased by nitrogen fertilization. The F PSⅡ of 0, 90 and180 kg ha-1 nitrogen treatments at noon were 0.197, 0.279 and 0.283, respectively, which decreased by 57.7, 56.4 and 40.2% as comparedto those in the morning. In the afternoon, the F PSⅡ partialy or completely recovered to the levels in the morning. The values of F PS Ⅱin 0 and 90kgha-1 treatments recovered to 87.3 and 81.5% of those in the morning. In 180kgha-1 treatment, the F PSⅡ in the afternoonwas even higher than that in the morning. Application of nitrogen fertilizer significantly increased maximum photochemical efficiency(Fv/Fm), photochemical quenching coefficient (qP) and non-photochemical quenching coefficient (qNP). These results indicated thatapplication of nitrogen fertilizer could increase the light energy conversion efficiency, the potential activity of photosynthetic reactioncenter, and the non-photochemical dissipation of excess light energy, which can prevent leaf photosynthetic apparatus from damage ofenvironmental stress. However, there was no significant difference in the values of F PSⅡbetween 90 and 180kgha-1 nitrogentreatments, indicating that the excess nitrogen was unfavorable to photosynthesis.

文摘Photosynthesis includes the collection of light and a/b-binding (LHC) proteins. In high plants, the LHC gene family constituting the light-harvesting complex ofphotosystems I and II. the transfer of solar energy using light-harvesting chlorophyll includes LHCA and LHCB sub-families, which encode proteins Zostera marina L. is a monocotyledonous angiosperm and inhab- its submerged marine environments rather than land environments. We characterized the Lhca and Lhcb gene families of Z. marina from the expressed sequence tags (EST) database. In total, 13 unigenes were annotated as ZmLhc, 6 in Lhca family and 7 in ZmLhcb family. ZmLHCA and ZmLHCB contained the conservative LHC motifs and amino acid residues binding chlorophyll. The average similarity among mature ZmLHCA and ZmLHCB was 48.91% and 48.66%, respectively, which indicated a high degree of diver- gence within ZmLHChc gene family. The reconstructed phylogenetic tree showed that the tree topology and phylogenetic relation- ship were similar to those reported in other high plants, suggesting that the Lhc genes were highly conservative and the classification of ZmLhc genes was consistent with the evolutionary position of Z. marina. Real-time reverse transcription (RT) PCR analysis showed that different members of ZmLhca and ZmLhcb responded to a stress in different expression patterns. Salinity, temperature, light intensity and light quality may affect the expression of most ZmLhca and ZmLhcb genes. Inorganic carbon concentration and acidity had no obvious effect on ZmLhca and ZmLhcb gene expression, except for ZmLhca6.

文摘CWMV-CP1 target gene and bar selection gene were co-transferred into commercial wheat variety of Yangmai158 by particle bombardment. In total, 145 resistant plants to 3 -5 mg L-1 Bialaphos were obtained, 21 plants were identified to be positive in T0 generation by PCR-Southern test, and the transformation frequency had 0.99%. T1 plants were further tested by PCR and Southern hybridization. Results demonstrated that the alien resistance gene had been integrated into the wheat genome. The segregation ratio of CP1+ to CP1- in T1 generation was 1.0 to 1. 3, and didn't agree with Mendelian rule. RT-PCR result from T2 plants showed that the alien gene CWMV-CP1 had stable expression in wheat genetic background.
文摘The pathogenesis-related proteins 1 (PR-1) gene family play important roles in the plant metabolism in response to biotic and abiotic stresses. The wheat TdPR1.2 has been previously isolated and characterized. Here we showed by bio-informatic analysis that TdPR1.2 contains six cysteine residues that are conserved between all PR-1 proteins tested. Using ScanProsite tool, we found that TdPR1.2 structure has a CRISP family signature 1 and 2 located at the C-terminal part of the protein. Those two domains are conserved in many identified PR1.2 proteins in plants. Moreover, SignalIP-5.0 analysis revealed that TdPR1.2 contains a putative signal peptide formed by 25 amino acids at the N-terminal extremity. The presence of this signal peptide suggested that the mature proteins will be secreted after the cleavage of the signal sequence. Further, we investigate the role of the TdPR1.2 proteins in the growth of <i>Escherichia coli</i> transformants cells under different abiotic stresses. Our results showed that the full-length form of TdPR1.2 enhanced tolerance of <i>E. coli</i> against salt and osmotic stress but not to KCl. Moreover, TdPR1.2 protein confers bacterial tolerance to heavy metals in solid and liquid mediums. Based on these results, we suggest that the TdPR1.2 protein could play an important role in response to abiotic stress conditions.

文摘A solution culture experiment was performed to investigate the effects of oxytetracycline (OTC) on wheat (Triticum aestivum L.) growth, chlorophyll contents, and photosynthesis at five levels of 0, 10, 20, 40, and 80 mmol L-1 OTC. OTC is toxic to wheat. The wheat growth, especially wheat root was significantly decreased. Further OTC also significantly decreased root activity, chlorophyll contents, and photosynthetic parameters except for intercellular CO2 concentrations. The different responses of indicators such as root number, root activity and so on to OTC were also observed. The IC50 values for the tested indicators to OTC ranged from 7.1 to 113.4 mmol L-1 OTC. The order of indicator sensitivity to OTC was root number stomatal conductance chlorophyll a total chlorophyll photosynthetic rates total surface area transpiration rate chlorophyll b fresh weight of root dry weight of root total length dry weight of shoot = fresh weight of shoot total volume. The root number was more sensitive than other indicators with the IC50 value of 7.1 mmol L-1 OTC, and could be taken as the sensitive indicator to predict the hazards of OTC to wheat.

文摘The effects of protein characteristics and the proportion of gluten on end-use quality in 13 Korean wheat cultivars for three years were verified in this study. Year, cultivar, and the interaction between the year and the cultivar influenced protein characteristics, the proportion of gluten except for γ-and ω-gliadin using RP-HPLC(reversed-phase high-performance liquid chromatography), and end-use quality. Protein characteristics and the proportion of gluten in Korean wheat cultivars were between those of Australian standard white(ASW) and hard wheat(AH). Korean wheat cultivars exhibited a higher average α+β gliadin proportion than imported wheat, a γ-gliadin proportion similar to that of dark northern spring wheat, and the same ω-gliadin proportion as AH. They showed a bread-loaf volume intermediate between those of ASW and AH and a texture of cooked noodles similar to that of soft white wheat, but less springiness than imported wheat. The cookie diameter of Korean wheat cultivars was similar to that of hard red winter wheat. There was a correlation between bread-loaf volume and protein characteristics, except for the protein content in Korean wheat cultivars. Springiness and cohesiveness of cooked noodles were not correlated with protein characteristics, while hardness was correlated with the protein content and water absorption of a mixograph. Cookie diameter was negatively correlated with the sodium dodecyl sulfate(SDS) sedimentation volume and water absorption of a mixograph. The end-use quality was not correlated with any proportion of gluten composition. Principal component analysis(PCA) showed that the proportion of gluten was not related to the quality of the bread(both PCs, 81.3%), noodle(77.7%), and cookie(82.4%). PCA explained that Keumkang is suitable for superior bread, while Uri is good for cooked noodles and cookies.

文摘Interactions of the stripe rust pathogen (Puccinia striiformis f. sp responses. Among various genes involved in the plant-pathogen related (PR) protein genes determine different defense responses tritici) with wheat plants activate a w^ae range OT nost nteractions, the expressions of particular pathogenesis-Different types of resistance have been recognized and utilized for developing wheat cultivars for resistance to stripe rust. All-stage resistance can be detected in seedling stage and remains at high levels throughout the plant growth stages. This type of resistance is race-specific and not durable. In contrast, plants with only high-temperature adult-plant (HTAP) resistance are susceptible in seedling stage, but become resistant when plants grow older and the weather becomes warmer. HTAP resistance controlled by a single gene is partial, but usually non-race specific and durable. The objective of this study was to analyze the expression of PR protein genes involved in different types of wheat resistance to stripe rust. The expression levels of 8 PR protein genes (PR1, PRI.2, PR2, PR3, PR4, PR5, PR9 and PRIO) were quantitatively evaluated at 0, 1, 2, 7 and 14 days after inoculation in single resistance gene lines of wheat with all-stage resistance genes YrTrl, Yr76, YrSP and YrExp2 and lines carrying HTAP resistance genes Yr52, Yr59, Yr62 and Yr7B. Races PSTv-4 and PSTv-37 for compatible and incompatible interactions were used in evaluation of PR protein gene expression in wheat lines carrying all-stage resistance genes in the seedling- stage experiment while PSTv-37 was used in the HTAP experiment. Analysis of quantitative real-time polymerase chain reaction (qRT-PCR) revealed that all of the PR protein genes were involved in the different types of resistance controlled by different Yr genes. However, these genes were upregulated at different time points and at different levels during the infection process among the wheat lines with different Yr genes for either all-stage resistance or HTAP resistance. Some of the genes were also induced in compatible interactions, but the levels were almost always higher in the incompatible interaction than in the compatible interaction at the same time point for each Yr gene. These results indicate that both salicylic acid and jasmonate signaling pathways are involved in both race-specific all-stage resistance and non-race specific HTAP resistance. Although expressing at different stages of infection and at different levels, these PR protein genes work in concert for contribution to different types of resistance controlled by different Yr genes.

文摘A field experiment using a split-plot randomized complete block design with three replications was carried out to determine relationships between spectral indices and wheat grain yield (GY), to compare the performance of four vegetation indices (VIs) for GY prediction, and to study the feasibility of VI to estimate grain protein content (GPC) in winter wheat. Two typical winter wheat (Triticum aestivum L.) cultivars 'Xuzhou 26' (high protein content) and 'Huaimai 18' (low protein content) were used as the main plot treatments and four N rates, i.e., 0, 120, 210, and 300 kg N ha^-1, as the sub-plot treatments. Increasing soil N supply significantly increased GY and GPC (P ≤ 0.05). For the two cultivars combined, significant and positive correlations were found between four VIs and GY, with the strongest relationship observed when using the green ratio vegetation index (GRVI) at mid-filling. Cumulative VI estimates improved yield predictions substantially, with the best interval being heading to maturity stage. Similar results were found between VI and grain protein yield. However, when using cumulative VI, GPC showed no significant improvement. The strong relationship between leaf N status and GPC (R2 =0.9144 for 'Xuzhou 26' and R2 = 0.8285 for 'Huaimai 18') indicated that canopy spectra could be used to predict GPC. The strong fit between estimated and observed GPC (R2 = 0.7939) indicated that remote sensing techniques were potentially useful predictors of grain protein content and quality in wheat.

文摘The relationship between vernalization requirement and quantitative and qualitative changes in total leaf soluble proteins were determined in one spring (cv. Kohdasht) and two winter (cvs. Sardari and Norstar) cultivars of wheat (Triticum aestivum L.) exposed to 4℃. Plants were sampled on days 2, 14, 21 and 35 of exposure to 4℃. The final leaf number (FLN) was determined throughout the vernalization periods (0, 7, 14, 24, and 35 d) at 4℃. The final leaf number decreased until days 24 and 35 in Sardari and Norstar eultivars, respectively, indicating the vernalization saturation at these times. No clear changes were detected in the final leaf number of Kohdash cultivar, verifying no vernalization requirement for this spring wheat cultivar. Comparing with control, clear cold-induced 2-fold increases in proteins quantity occurred after 48 h following the 4℃-treatment in the leaves of the both winter wheat cultivars but, such response was not detected in the spring cultivar. However, the electrophoretic protein patterns showed between-cultivar and between-temperature treatment differences. With increasing exposure time to 4℃, the winter cultivars tended to produce more HMW polypeptides than the spring cultivar. Similar proteins were induced in both Sardari and Norstar winter wheat cultivars, however, the long vernalization requirement in Norstar resulted in high level and longer duration of expression of cold-induced proteins compared to Sardari with a short vernalization requirement. These observations indicate that vernalization response regulates the expression of low temperature (LT) tolerance proteins and determines the duration of expression of LT- induced proteins.

文摘The regeneration rate of wheat immature embryo varies among genotypes, howbeit many elite agriculture wheat varieties have low regeneration rates. Optimization of tissue culture conditions and attempts of adding signal molecules are effective ways to increase plant regeneration rate. Inter-culture is one of ways that have not been investigated in plant tissue culture. Moreover, the use of arabinogalactan proteins (AGPs) and hydrogen peroxide (H202) have been reported to increase regeneration rate in a few plant species other than wheat. The current research pioneeringly uses inter-culture of immature embryos of different wheat genotypes, and also investigates impacts of AGP and H2O2 on the induction of embryogenic calli and plant regeneration. As a result, high-frequency regeneration wheat cultivars Kenong 199 (KN 199) and Xinchun 9 (XC9), together with low-frequency regeneration wheat line Chinese Spring (CS), presented striking increase in the induction of embryogenic calli and plant regeneration rate of CS through inter-culture strategy, up to 52.19 and 67.98%, respectively. Adding 50 to 200 mg L-1 AGP or 0.005 to 0.01‰ H2O2 to the callus induction medium, enhanced growth of embryogenic calli and plant regeneration rate in quite a few wheat genotypes. At 50 mg L-1 AGP application level in callus induction medium plant regeneration rates of 8.49,409.06 and 283.16% were achieved for Jimai 22 (JM22), Jingdong 18 (JD18) and Yangmai 18 (YM18), respectively; whereas at 100 mg L-1 AGP level, CS (105.44%), Chuannong 16 (CN16) (80.60%) and Ningchun 4 (NC4) (62.87%) acted the best. Moreover CS (79.05%), JM22 (7.55%), CN16 (101.87%), YM18 (365.56%), Yangmai 20 (YM20) (10.48%), and CB301 (187.40%) were more responsive to 0.005 %o of H2O2, and NC4 (35.37%) obtained the highest shoot regeneration rates at 0.01%o of H2O2. Overall, these two methods, inter-culture and AGP (or H2O2) application, can be further applied to wheat transgenic research.

文摘Waterlogging is one of the most factors limiting wheat production in the middle and lower reaches of the Yangtze River Plain of China,especially in the middle and late stages of wheat.Wheat varieties‘Jingmai102’(JM102)and‘Yangmai158’(YM158)were planted to study the dynamic changes of photosynthetic characteristics in flag leaf and the influence of waterlogging at anthesis on the yield and components and dry matter accumulation and remobilization of winter wheat in above ground.The results showed that the SPAD values slightly increased at 1 day after anthesis(d),and then kept decreasing with the increase of waterlogging time.The decrease in SPAD value was more remarkably in YM158 than that in JM102.As for the chlorophyll fluorescence parameters,the photochemical efficiency(Fv/Fm),potential activity(Fv/Fo)of photosystem II,and electronic transmission(Fm/Fo)on photosystem II increased first and then decreased with the increase of waterlogging days after anthesis.The quantum ratio of heat dissipation(Fo/Fm)had a tendency opposite to that of Fv/Fm,and the change range of JM102 was lower than that of YM158.For the grain yield and components,waterlogging at anthesis decreased the dry weight of single stem,grain yield,1 000-kernel weight,spikelet per panicle,and harvest index,and the reduction of JM102 was smaller than that of YM158.As for the accumulation and remobilization of dry matter,the accumulation of dry matter after anthesis decreased significantly under waterlogging condition(WL),and the reduction of JM102 was smaller than that of YM158.In conclusion,waterlogging at anthesis significantly affected the photosynthetic characteristics,yield and components in both varieties,but different varieties exhibited different tolerances to waterlogging stress and YM158 was more sensitive to water stress than JM102.
