Maternal one-carbon metabolism plays an important role in early life programming.There is a well-established connection between the fetal environment and the health status of the offspring.Howeve r,there is a knowledg...Maternal one-carbon metabolism plays an important role in early life programming.There is a well-established connection between the fetal environment and the health status of the offspring.Howeve r,there is a knowledge gap on how maternal nutrition impacts stro ke outcomes in offspring.The aim of our study was to investigate the role of maternal dietary deficiencies in folic acid or choline on stroke outcomes in 3-month-old offspring.Adult female mice were fed a folic acid-deficient diet,choline-deficient diet,or control diet 4 weeks before pregnancy.They we re continued on diets during pregnancy and la ctation.Male and female offspring were weaned onto a control diet and at 2 months of age were subjected to ischemic stroke within the sensorimotor cortex via photothrombotic damage.Mothers maintained on either a folic acid-deficient diet or choline-deficient diet had reduced levels of S-adenosylm ethionine in the liver and S-adenosylhomocysteine in the plasma.After ischemic stro ke,motor function was impaired in 3-month-old offspring from mothers receiving either a folic acid-deficient diet or choline-deficient diet compared to the animals receiving a control diet.In brain tissue,there was no difference in ischemic damage volume.When protein levels were assessed in ischemic brain tissue,there were lower levels of active caspase-3 and hypoxia-inducible factor 1α in males compared to females and betaine levels were reduced in offspring from the mothers receiving a choline-deficient diet.Our results demonstrate that a deficient maternal diet at critical time points in neurodevelopment results in worse stro ke outcomes.This study emphasizes the importance of maternal diet and the impact it can have on offspring health.展开更多

This study explores the enhancing effect of deep eutectic solvents(DESs) on the aqueous solubility of curcumin(CUR) via experiment and theoretical calculation. Choline chloride-based DESs with polyols 1,2-propanediol(1,2-PDO), 1,3-propanediol, ethylene glycol, and glycerol as hydrogen bond donors were prepared and used as co-solvents. The CUR aqueous solubility increased with increasing the DESs content at temperature of 303.15-318.15 K, especially in aqueous ChCl/1,2-PDO(mole ratio 1:4) solutions. The positive apparent molar volume values and reduced density gradient analysis confirmed the existence of strong interactions between CUR and solvent. The van der Waals interactions and hydrogen bonding coexisted in DESs monomer retained the stability of DESs structure after introducing CUR. Moreover,the lower interaction energy of DESs…CUR system than that of the counterpart DESs further proved the strong interaction between CUR and DESs. The lowest interaction energy of ChCl/1,2-PDO…CUR system indicated that this system was the most stable and ChCl/1,2-PDO was promising for CUR dissolution.This work provides efficient solvents for utilizing curcumin, contributing to a deep insight into the interactions between DES and CUR at the molecular level, and the role of DESs on enhancing drugs solubility.展开更多
Previous studies have shown that Vibrio splendidus infection causes mitochondrial damage in Apostichopus japonicus coelomocytes,leading to the production of excessive reactive oxygen species(ROS)and irreversible apopt...Previous studies have shown that Vibrio splendidus infection causes mitochondrial damage in Apostichopus japonicus coelomocytes,leading to the production of excessive reactive oxygen species(ROS)and irreversible apoptotic cell death.Emerging evidence suggests that mitochondrial autophagy(mitophagy)is the most effective method for eliminating damaged mitochondria and ROS,with choline dehydrogenase(CHDH)identified as a novel mitophagy receptor that can recognize non-ubiquitin damage signals and microtubule-associated protein 1 light chain 3(LC3)in vertebrates.However,the functional role of CHDH in invertebrates is largely unknown.In this study,we observed a significant increase in the mRNA and protein expression levels of A.japonicus CHDH(AjCHDH)in response to V.splendidus infection and lipopolysaccharide(LPS)challenge,consistent with changes in mitophagy under the same conditions.Notably,AjCHDH was localized to the mitochondria rather than the cytosol following V.splendidus infection.Moreover,AjCHDH knockdown using si RNA transfection significantly reduced mitophagy levels,as observed through transmission electron microscopy and confocal microscopy.Further investigation into the molecular mechanisms underlying CHDH-regulated mitophagy showed that AjCHDH lacked an LC3-interacting region(LIR)for direct binding to LC3 but possessed a FB1 structural domain that binds to SQSTM1.The interaction between AjCHDH and SQSTM1 was further confirmed by immunoprecipitation analysis.Furthermore,laser confocal microscopy indicated that SQSTM1 and LC3 were recruited by AjCHDH in coelomocytes and HEK293T cells.In contrast,AjCHDH interference hindered SQSTM1 and LC3 recruitment to the mitochondria,a critical step in damaged mitochondrial degradation.Thus,AjCHDH interference led to a significant increase in both mitochondrial and intracellular ROS,followed by increased apoptosis and decreased coelomocyte survival.Collectively,these findings indicate that AjCHDH-mediated mitophagy plays a crucial role in coelomocyte survival in A.japonicus following V.splendidus infection.展开更多
There are currently no prescribed requirements for B vitamins or choline for dairy cows during the transition period, but many recent studies have shown a variety of benefits from supplying these nutrients to peripart...There are currently no prescribed requirements for B vitamins or choline for dairy cows during the transition period, but many recent studies have shown a variety of benefits from supplying these nutrients to periparturient cows. The purpose of this research effort was to determine the potential benefit of including a rumen protected blend composed of B vitamins (riboflavin, folic acid, vitamin B12) and choline (RPBlend, Jefo Nutrition Inc., St. Hyacinthe, QC, Canada) for dairy cows during the transition period, based on results from 28 on-farm feeding studies (USA-12, Mexico-7, Canada-4, Chile-3, Australia-1, Brazil-1) conducted between 2011 and 2018. All farms participated in monthly herd management record keeping systems and were selected to participate in studies due to their excellent management. Meta-analyses in which risk differences were determined were used to assess the effects of the supplemental RPBlend on health parameters and reproduction. The effect size was used as the determinant of the possible contribution of RPblend on the yields of milk, energy corrected milk (ECM), fat and protein yields during the first four weeks of lactation. Results showed that the inclusion of RPblend reduced (P < 0.05) involuntary culling and mastitis by cows during the first 30 days after calving. There was a tendency (P < 0.10) for reduced retained placenta and metritis. The meta-analyses revealed that the risk of the displaced abomasum and milk fever did not diminish (P > 0.10) with the inclusion of the BPBlend. The incidence rate of subclinical ketosis, determined as blood beta hydroxy butyric acid greater than 1.2 mM was lower (P (than those not supplemented. The proportion of cows confirmed pregnant by 100 days in milk was greater for cows given the BPBlend (P < 0.05). Milk yield and ECM were greater for cows receiving the RPBlend (1.13 and 0.93 kg/cow respectively, P < 0.05). There was no change in fat yield (P > 0.10) while the yield of milk protein was greater (P < 0.05) when the cows received the blend. These results suggest that the inclusion of rumen protected B vitamins and choline can assist cow health, reproduction and production at the start of lactation.展开更多

Here, we demonstrated that the production of tabtoxin in a P. syringae strain increased at least 150%, when choline, betaine or dimethylglycine were used as nitrogen source, or when choline was added as osmoprotectant in hyperosmolar culture media. Besides, we investigated the induction of phosphorylcholine phosphatase (PchP) activity when choline or its metabolites were used as nitrogen sources. PchP is an enzyme involved in Pseudomonas aeruginosa pathogenesis through its contribution to the breakdown of choline-containing compounds of the host cells. Considering these results and that the success of a pathogenic microorganism depends on its ability to survive and proliferate in its target tissue, we propose that choline is one of the plant signals that contribute to establishment of the infection by tabtoxin-producing strains of P. syringae.展开更多
AIM: To investigate the normal hepatic magnetic resonance spectroscopy findings choline/lipid2 (Cho/Lip2) associated with age and body mass index (BMI).METHODS: A total of 58 single-voxel proton spectra of the liver w...AIM: To investigate the normal hepatic magnetic resonance spectroscopy findings choline/lipid2 (Cho/Lip2) associated with age and body mass index (BMI).METHODS: A total of 58 single-voxel proton spectra of the liver were acquired at 3.0 T using the eightchannel phased array abdominal coil as the receiver coil. Consecutive stacks of breath-hold spectra were acquired using the point resolved spectroscopy technique at a short echo time of 30 ms and a repetition time of 1500 ms. The spectra were processed with the SAGE software package. Areas and heights for metabolite resonance were obtained. Student's t test for unpaired data was used for comparisons of shimming, Cho/Lip2, and lipid content. RESULTS: There were significant negative correlations between the Cho/Lip2 peak height ratios and BMI (r=-0.615) and age (r=-0.398) (all P<0.01). Compared with the high-BMI group, the low-BMI group was younger (39.1±13.0 years vs 47.6±8.5 years, t=-2.954,P=0.005); had better water suppression (93.4%±1.4% vs 85.6%±11.6%, t=2.741, P=0.014); had higher Cho/Lip2 peak heights ratio (0.2±0.14 vs 0.05±0.04,t=6.033,P<0.000); and had lower lipid content (0.03±0.08 vs 0.29±0.31, t=-3.309, P=0.004). Compared with the older group, the younger group had better shimming effects (17.1±3.6 Hz vs 22.0±6.8 Hz, t=-2.919, P=0.008); higher Cho/Lip2 peak heights ratios (0.03±0.05vs 0.09±0.12,t=2.4, P=0.020); and lower lipid content (0.05±0.11 vs 0.23±0.32,t=-2.337,P=0.031). Compared with the lowcholine peak group, the high-choline peak group had lower lipid content (0.005±0.002 vs 0.13±0.23, t=-3.796,P<0.000); lower BMI (19.6±2.4vs 23.9±3.0, t=-4.410, P<0.000); and younger age (34.7±10.0 years vs 43.2±12.5 years, t=-2.088, P=0.041). CONCLUSION: Lipid accumulation could result from the increased fat in the body depending on age and BMI. Lipid can mask the resonance signal of choline.展开更多

The electrochemical corruption property of bipolar plates in simulated PEMFC environment was investigated. It was showed that the plating film was distributed on the surface of 316 stainless steel like isolated islands with height less than 50 nm. The XPS, XRD results showed that a smooth and strong chemical inert film of Nb O and Nb2O5 was formed on the surface of 316 stainless steel. In simulated cathodic condition, the corrosion potential of Nb coated stainless steel was improved by 244 m V, whilst in an anodic condition, it was improved by 105 m V. The current densities for the coated 316 stainless steel were decreased to 2.479 4 μA·cm-2 from 14.810 μA·cm-2 at-0.1 V and to 0.576 μA·cm-2 from 13.417 μA/·cm-2 at 0.6 V, respectively. It was implied that the niobium coating effectively decreased the corrosion rate. The results of the electrochemical tests indicated that the corrosion resistance of stainless steel was greatly improved after coated with niobium.展开更多

In traditional separation approaches, strong acid and strong base have to be used to remove these compounds, which may cause serious environmental problems. In this work, 19 kinds of cholinium ionic liquids have been developed to separate phenol from model oil by liquid–liquid extraction. Structural effect of anions of the ionic liquids in the separation is systematically investigated. It is found that depending on the chemical structure of ionic liquids, phenol can be removed from toluene with single-step removal efficiency from 86 to 99% under optimal conditions. The type of substituent groups and the-CH_2 number between two carboxylates have obvious effect on the removal efficiency, and more hydrophilic ionic liquids have a stronger extraction performance for phenol. Furthermore, thermodynamic,^(13) C NMR,~1 H NMR and density functional theory calculations have been performed to characterize the extraction process and to understand the extraction mechanism. It is shown that the extraction of phenol from oil to ionic liquid is a favorable process, and this process is mainly driven by enthalpy change. The formation of the hydrogen bond between anion of the ionic liquid and-OH of phenol is the main driving force for the extraction of phenol from oil to the ionic liquids.展开更多

The results show that the solubilities of gases increase with increasing pressure and decreasing temperature. The solubility of CO_2 is higher than that of CH_4, H_2, CO and N_2, which indicates that ChCl/Urea may be used as a potential solvent for CO_2 capture from the gas mixture. Solubility of CO_2 in ChCl/Urea was fitted by Non-Random Two-Liquid and Redlich-Kwong(NRTL-RK) model, and solubility of CH_4, H_2, CO or N_2 in ChCl/Urea was fitted by Henry's Law. The standard enthalpy, standard Gibbs energy and standard entropy of gases were calculated. Additionally, the CO_2/CH_4 selectivities in water, dry ChCl/Urea and aqueous ChCl/Urea were further discussed.展开更多

OBJECTIVE: To observe the effects of GSL combining with choline on learning and memory of AD model rats.DESIGN: Randomized grouping design and controlled animal study.SETTING: Department of Pharmacology, Taishan Medical College. MATERIALS: The experiment was carried out in the Pharmacological Department of Medical College of Jilin University from October 1996 to January 1997. Forty healthy male Wistar rats of clean grade were randomly divided into 5 groups, including sham-injury group, model group, GSL group, choline group and combination group, with 8 rats in each group. Main medications: GSL with the volume more than 92.8% was provided by Department of Chemistry, Norman Bethune Medical College of Jilin University. Panaxatriol, the main component, was detected with thin layer scanning technique and regarded as the index of GSL quality [(55±1)%, CV = 2%, n = 5]. Choline was provided by the Third Shanghai Laboratory Factory. METHODS: 150 nmol quinolinic acid was used to damage bilateral Meynert basal nuclei of adult rats so as to establish AD models. Rats in GSL, choline and combination groups were intragastric administrated with 400 mg/kg GSL, 200 mg/kg choline (20 mL/kg), and both respectively last for 17 days starting from two days before operation. Rats in sham-injury group and model group were perfused with the same volume of distilled water once in each morning for the same days. ① Passive avoidance step-down test: Five minutes later, rats jumped up safe platform when they were shocked with 36 V alternating current. If rats jumped down from the platform and the feet touched railings, the response was wrong. Numbers of wrong response were recorded within 3 minutes, and then the test was redone after 24 hours. ② Morris water-maze spatial localization task: Swimming from jumping-off to platform directly was regarded as right response. Additionally, 4 successively right responses were regarded as the standard. Each rat was trained 10 times a day with 120 s per time for 3 successive days. The interval was 30 s. Three days later, numbers of right response were recorded. The training times were increased to 30 for unlearned rats. ③ Measurement of activity of choilne acetylase in cerebral cortex: Rats were sacrificed at 17 days after operation to obtain cerebral cortex to measure activity of choilne acetylase with radiochemistry technique. ④ Synergistic effect: It was expressed as Q value: Q value = factual incorporative effect/anticipant incorporative effect; Q ≥ 1 was regarded as synergistic effect. Anticipant incorporative effect = (EA+EB-EA·EB), EA and EB were single timing effect, respectively in GSL group and choline group. E (step-down test and Morris water maze test) = (x in model group - factual value in medicine groups)/x in model group; E (activity of choilne acetylase) = (factual value in medicine groups -x in model group)/x in model group. MAIN OUTCOME MEASURES: ① Passive avoidance step-down test and Morris water-maze spatial localization task in the study of learning and memory; ② activity of choilne acetylase. RESULTS: All 40 rats were involved in the final analysis. ① Passive avoidance response: At learning phase on first day and retesting phase on the next day, numbers of wrong responses within 3 minutes were more in model group than sham operation group, and there was significant difference [(5.88±1.46), (2.25±0.87) times; (2.63±1.06), (0.50±0.53) times; P < 0.01]; numbers of wrong responses within 3 minutes were less in combination group than model group, and there was significant difference [learning phase: (1.12±0.83), (5.88±1.46) times; retesting phase: (0.38±0.74), (2.63±1.06) times, P < 0.01]; moreover, effect was stronger than that in GSL group and choline group. The Q value was 1.07 and 1.59, respectively and it showed synergistic effect. ② Spatial localization task: Training times were more in model group than sham operation group, and there was significant difference [(2.9±2.5), (12.6±3.5) times; P < 0.01]. Training times were less in combination group than model group, and there was significant difference [(11.8±2.4), (27.9±2.5) times, P < 0.01]; moreover, effect was stronger than that in GSL group and choline group. The Q value was 1.07 and it showed synergistic effect. ③ Activity of choilne acetylase: Activity was lower in model group than sham operation group, and there was significant difference [(30.56±8.33), (61.11±8.33) nkat/g; P < 0.01]. Activity was higher in combination group than model group and there was significant difference [(50.00±8.33), (30.56±8.33) nkat/g, P < 0.01]; moreover, effect was stronger than that in GSL group and choline group. The Q value was 1.5 and it showed synergistic effect. CONCLUSION: GSL in combination with choline can synergically improve the disorder of learning and memory of AD model rats. Its mechanism may be involved in enhancing the function of central cholinergic system.展开更多

The CMO cDNA fragment(1.3 kb) was generated by Reverse Transcription-Polymerase Chain Reaction (RT-PCR) with primers based on the report sequence of CMO in gene bank. A chimerical gene composed of the cauliflower mosaic virus (CaMV) 35S promoter fused to CMO cDNA and β-glucuronidase (GUS-marker gene) was transferred into Ti-derived disarmed binary vector pBI121. The new vector, p35SCMOp, was transferred into Agrobacterium tumefaciens C58/pMP90 by freeze-thaw method. Somatic embryogenesis (SE) initiation of Pinus. Strobus L. and Pinus.Koraiensis Sieb. et Zucc. depended on the manipulation of plant growth regulator (PGR) concentrations in the GLH culture medium. Transgenic embryos and regenerated plants of two Pine species were produced after co-culture of embryogenic tissue with the disarmed strain of A. tumefaciens C58/pMP90/ p35SCMOp and selected on medium containing 25mg/L kanamycin. We report a carbon/carbon capacitor based on micro/mesoporous carbon electrodes with cost-effective and eco-friendly aqueous choline bis(trifluoromethylsulfonyl)imide(Ch TFSI)electrolyte with a cosolvent enabling low-...We report a carbon/carbon capacitor based on micro/mesoporous carbon electrodes with cost-effective and eco-friendly aqueous choline bis(trifluoromethylsulfonyl)imide(Ch TFSI)electrolyte with a cosolvent enabling low-temperature operation down to-30℃.For this purpose,a Mg O-templated hierarchical carbon(MP98B)with an average mesopore diameter of 3.5 nm was prepared by pyrolysis of magnesium citrate hydrate at 900℃.To reach lower temperatures,the melting point and viscosity of the aqueous electrolyte were reduced by mixing water(W)with an organic solvent(methanol,M,or isopropanol,I)of high dielectric constant and low viscosity.5 mol kg^(-1)(5 m)Ch TFSI in an optimized volume fraction of cosolvent,M_(0.75)W_(0.25),and I_(0.75)W_(0.25),showed the highest conductivity;the higher conductivity in M_(0.75)W_(0.25)(22.8 and 3.1 m S cm^(-1) at 20 and-30℃,respectively)than in I_(0.75)W_(0.25)(8.5 and0.5 m S cm^(-1)at 20 and-30℃,respectively)is attributed to the lower viscosity of the M_(0.75)W_(0.25)solution.The electrochemical stability window(ESW)of 5 m Ch TFSI in M_(0.75)W_(0.25)and I_(0.75)W_(0.25)(1.6 V)on an MP98B electrode was determined by applying the S-method.Meanwhile,by adjusting the mass ratio of the two electrodes,a MP98B/MP98B capacitor using the 5 m electrolyte in M_(0.75)W_(0.25)could operate with a good life span up to 1.6 V while exhibiting a better charge propagation,greater specific capacitance,and higher specific energy than in I_(0.75)W_(0.25).展开更多

The formation of the choline salt of febuxostat wa...The objective of the present study was to exhibit the enhanced water-solubility and in vivo oral absorption when febuxostat(FXT) became the salt formation of choline. The formation of the choline salt of febuxostat was confirmed by X-ray powder diffraction, infrared spectroscopy analysis and differential scanning calorimetry. The direct filling method was used to develop a capsule formulation. Cellactose 80 was used as the filler due to its good fluidity, while cross-linked polyvinylpyrrolidone(PVPP) and magnesium stearate(MS) were used as the disintegrant and lubricant, respectively. Then the in vitro release of the formulation was carried out in five different dissolution media including HCl solution(pH 1.2),acetate buffer(pH 4.5), phosphate buffer(pH 6.8 and pH 7.2) and water. Evident improvement of release for choline febuxostat(CXT) was presented in water while the dissolution degree was decreased for CXT in the medium of phosphate buffer(pH 6.8) in comparison with FXT. Furthermore, the pharmacokinetics of CXT was studied in rats using UPLC-MS/MS compared with FXT. The data acquired illustrated that AUC0-24 h of CXT and FXT were22,245.96 ± 7342.92 μg·h/l and 12,249.70 ± 2024.04 μg·h/l, respectively. The relative bioavailability of CXT to FXT was about 181.6% and the P value of AUC0-24 h was less than 0.05. It showed significant difference between the two drugs after oral administration. In conclusion, the water-solubility and oral bioavailability were both improved remarkably for the choline salt of febuxostat and choline salinization was proved an effective way to increase the in vivo absorption for FXT.展开更多
AIM To investigate the repeatability of proton magnetic resonance spectroscopy in the in vivo measurement of human cerebral levels of choline-containing compounds(Cho).METHODS Two consecutive scans were carried out in...AIM To investigate the repeatability of proton magnetic resonance spectroscopy in the in vivo measurement of human cerebral levels of choline-containing compounds(Cho).METHODS Two consecutive scans were carried out in six healthy resting subjects at a magnetic field strength of 1.5 T. On each occasion, neurospectroscopy data were collected from 64 voxels using the same 2 D chemical shift imaging(CSI) sequence. The data were analyzed in the same way, using the same software, to obtain the values for each voxel of the ratio of Cho to creatine. The Wilcoxon related-samples signed-rank test, coefficient of variation(CV), repeatability coefficient(RC), and intraclass correlation coefficient(ICC) were used to assess the repeatability. RESULTS The CV ranged from 2.75% to 33.99%, while the minimum RC was 5.68%. There was excellent reproducibility, as judged by significant ICC values, in 26 voxels. Just three voxels showed significant differences according to the Wilcoxon related-samples signed-rank test. CONCLUSION It is therefore concluded that when CSI multivoxel proton neurospectroscopy is used to measure cerebral choline-containing compounds at 1.5 T, the reproducibility is highly acceptable.展开更多

The object of this study was to investigate the effects of chromium propionate replacing 25%rumen-protected choline(RPC)on production performance and blood indicators of perinatal dairy cows.According to the principle...The object of this study was to investigate the effects of chromium propionate replacing 25%rumen-protected choline(RPC)on production performance and blood indicators of perinatal dairy cows.According to the principle of ensuring that chromium propionate and RPC were fed 14 days prepartum,27 healthy Holstein cows(age,parity,lactation volume,body condition and expectancy were similar)were randomly divided into three groups(GroupsⅠ,ⅡandⅢ),with nine cows in each group.Cows in GroupⅠwere fed basal diets;cows in GroupⅡwere fed the same basal diets with 10 g•d-1 RPC per cow;and cows in GroupⅢwere fed the same basal diets,but 7.5 g RPC and 2.5 g chromium propionate(4 mg chromium)per cow.The results showed that dry matter intake(DMI)of prepartum in GroupsⅡandⅢwas significantly increased(P<0.05),compared with GroupⅠ;however,there was no significant difference between GroupsⅡandⅢ(P>0.05).The milk fat content in GroupⅢwas significantly higher than that in GroupⅠon the 7th day of postpartum(P<0.05).The urea nitrogen content in GroupsⅡandⅢsignificantly lower than that in GroupⅠon the 21st day of postpartum(P<0.05),but there were no significant difference between GroupsⅡandⅢ(P>0.05).The somatic cell counts in GroupⅢwere significantly lower than those in GroupsⅠandⅡon the 14th and 21st days of postpartum(P<0.05).The contents of the total cholesterol in GroupⅢdecreased significantly compared to GroupⅡon the 7th day of prepartum,on the day of calving and the 7th day of postpartum(P<0.05).The concentration of high-density lipoprotein(HDL)in GroupⅢwas significantly higher than that in GroupⅡon the 7th day of prepartum,on the day of calving and the 7th day of postpartum(P<0.05).On the 7th day of postpartum the concentration of insulin in GroupⅢwas obviously higher than that in GroupⅡ(P<0.05).In summary,replacing 25%of rumen-protected choline with chromium propionate reduced production costs and improved dairy farming benefits.展开更多

When deprived of their source of NGF by axotom...Many medial septal neurons of the basal forebrain are dependent on nerve growth factor (NGF) from the hippocampus for survival and maintenance of a cholinergic phenotype. When deprived of their source of NGF by axotomy, medial septal neuronal cell bodies atrophy and lose their cholinergic markers. This is similar to what is observed in the basal forebrain during Alzheimer’s disease (AD). In the present study, medial septal neurons were axotomized in female rats by way of a fimbria/fornix lesion. For fourteen days following axotomy, varying NGF doses (1 - 250 μg/ml) were administered to the lateral cerebral ventricle with either mini-osmotic infusion or daily injection. The responsiveness of medial septal neurons was evaluated with choline acetyltransferase immunohistochemistry. Within the mini-osmotic pumps, NGF activity diminished greatly during the first five days of implantation, but increased dramatically in the CSF after five days of infusion. The responsiveness of medial septal neurons to NGF was dose dependent and the ED50 for NGF injection was determined to be 14.08 μg/ml compared to 27.60 μg/ml for NGF infusion. Intermittent injections at varying intervals were evaluated with 30 μg/ml NGF over a fourteen-day time period (2, 3, 6, or 12 injections). No differences occurred in the number of choline acetyltransferase neurons from rats that received weekly injections to those that received daily injections of NGF. NGF administration has been suggested as a therapy for AD. The results of these studies continue to highlight the need for NGF stability within the delivery system and AD patient CSF, the choice of delivery system, frequency of administration, and the NGF dose for maintaining basal forebrain cholinergic neurons during AD.展开更多

With early emergence and rapid growth, weeds effectlively compete for water, nutrients, and light resources. The recent evolution of herbicide resistant weeds has made it increasingly difficult for growers to effectively control weeds in soybean. Glyphosate and ALS-resistant Palmer amaranth biotypes have spread rapidly throughout South Carolina, especially in areas where resistance management is lacking. Soybean varieties have been recently developed with tolerance to 2,4-D. Field experiments were conducted at the Clemson University Edisto Research and Education Center located near Blackville, SC in 2012 and 2013 to evaluate selected 2,4-D choline based herbicide programs for weed management in 2,4-D tolerant soybean. Overall, all herbicide treatments were effective in controlling weeds at the POST2 timing. Palmer amaranth control was excellent;however, pitted morningglory was the most difficult. The 2,4-D plus glyphosate pre-mixture provided excellent control for all three weed species with >95% control at POST2 timing. In these treatments, the rate of 2,4-D choline plus glyphosate (1.09 kg ae ha-1 or 1.64 kg ae ha-1) did not have a significant effect on weed control (P = 0.3772). There was a decrease in pitted morningglory control 3 WAP in 2012 vs 2013 in plots treated with S-metolachlor + fomesafen because of a lack of activating soil moisture in 2012. Results from this study showed that all treatments evaluated provided good to excellent control of all 3 weed species. Based on the herbicide programs evaluated in the study, herbicide resistant weeds, such as Palmer amaranth, can be effectively controlled when treated at the correct growth stage.展开更多
文摘Maternal one-carbon metabolism plays an important role in early life programming.There is a well-established connection between the fetal environment and the health status of the offspring.Howeve r,there is a knowledge gap on how maternal nutrition impacts stro ke outcomes in offspring.The aim of our study was to investigate the role of maternal dietary deficiencies in folic acid or choline on stroke outcomes in 3-month-old offspring.Adult female mice were fed a folic acid-deficient diet,choline-deficient diet,or control diet 4 weeks before pregnancy.They we re continued on diets during pregnancy and la ctation.Male and female offspring were weaned onto a control diet and at 2 months of age were subjected to ischemic stroke within the sensorimotor cortex via photothrombotic damage.Mothers maintained on either a folic acid-deficient diet or choline-deficient diet had reduced levels of S-adenosylm ethionine in the liver and S-adenosylhomocysteine in the plasma.After ischemic stro ke,motor function was impaired in 3-month-old offspring from mothers receiving either a folic acid-deficient diet or choline-deficient diet compared to the animals receiving a control diet.In brain tissue,there was no difference in ischemic damage volume.When protein levels were assessed in ischemic brain tissue,there were lower levels of active caspase-3 and hypoxia-inducible factor 1α in males compared to females and betaine levels were reduced in offspring from the mothers receiving a choline-deficient diet.Our results demonstrate that a deficient maternal diet at critical time points in neurodevelopment results in worse stro ke outcomes.This study emphasizes the importance of maternal diet and the impact it can have on offspring health.

文摘The development of green solvents for enhancing aqueous solubility of drug curcumin remains a challenge. This study explores the enhancing effect of deep eutectic solvents(DESs) on the aqueous solubility of curcumin(CUR) via experiment and theoretical calculation. Choline chloride-based DESs with polyols 1,2-propanediol(1,2-PDO), 1,3-propanediol, ethylene glycol, and glycerol as hydrogen bond donors were prepared and used as co-solvents. The CUR aqueous solubility increased with increasing the DESs content at temperature of 303.15-318.15 K, especially in aqueous ChCl/1,2-PDO(mole ratio 1:4) solutions. The positive apparent molar volume values and reduced density gradient analysis confirmed the existence of strong interactions between CUR and solvent. The van der Waals interactions and hydrogen bonding coexisted in DESs monomer retained the stability of DESs structure after introducing CUR. Moreover,the lower interaction energy of DESs…CUR system than that of the counterpart DESs further proved the strong interaction between CUR and DESs. The lowest interaction energy of ChCl/1,2-PDO…CUR system indicated that this system was the most stable and ChCl/1,2-PDO was promising for CUR dissolution.This work provides efficient solvents for utilizing curcumin, contributing to a deep insight into the interactions between DES and CUR at the molecular level, and the role of DESs on enhancing drugs solubility.

文摘Previous studies have shown that Vibrio splendidus infection causes mitochondrial damage in Apostichopus japonicus coelomocytes,leading to the production of excessive reactive oxygen species(ROS)and irreversible apoptotic cell death.Emerging evidence suggests that mitochondrial autophagy(mitophagy)is the most effective method for eliminating damaged mitochondria and ROS,with choline dehydrogenase(CHDH)identified as a novel mitophagy receptor that can recognize non-ubiquitin damage signals and microtubule-associated protein 1 light chain 3(LC3)in vertebrates.However,the functional role of CHDH in invertebrates is largely unknown.In this study,we observed a significant increase in the mRNA and protein expression levels of A.japonicus CHDH(AjCHDH)in response to V.splendidus infection and lipopolysaccharide(LPS)challenge,consistent with changes in mitophagy under the same conditions.Notably,AjCHDH was localized to the mitochondria rather than the cytosol following V.splendidus infection.Moreover,AjCHDH knockdown using si RNA transfection significantly reduced mitophagy levels,as observed through transmission electron microscopy and confocal microscopy.Further investigation into the molecular mechanisms underlying CHDH-regulated mitophagy showed that AjCHDH lacked an LC3-interacting region(LIR)for direct binding to LC3 but possessed a FB1 structural domain that binds to SQSTM1.The interaction between AjCHDH and SQSTM1 was further confirmed by immunoprecipitation analysis.Furthermore,laser confocal microscopy indicated that SQSTM1 and LC3 were recruited by AjCHDH in coelomocytes and HEK293T cells.In contrast,AjCHDH interference hindered SQSTM1 and LC3 recruitment to the mitochondria,a critical step in damaged mitochondrial degradation.Thus,AjCHDH interference led to a significant increase in both mitochondrial and intracellular ROS,followed by increased apoptosis and decreased coelomocyte survival.Collectively,these findings indicate that AjCHDH-mediated mitophagy plays a crucial role in coelomocyte survival in A.japonicus following V.splendidus infection.
文摘There are currently no prescribed requirements for B vitamins or choline for dairy cows during the transition period, but many recent studies have shown a variety of benefits from supplying these nutrients to periparturient cows. The purpose of this research effort was to determine the potential benefit of including a rumen protected blend composed of B vitamins (riboflavin, folic acid, vitamin B12) and choline (RPBlend, Jefo Nutrition Inc., St. Hyacinthe, QC, Canada) for dairy cows during the transition period, based on results from 28 on-farm feeding studies (USA-12, Mexico-7, Canada-4, Chile-3, Australia-1, Brazil-1) conducted between 2011 and 2018. All farms participated in monthly herd management record keeping systems and were selected to participate in studies due to their excellent management. Meta-analyses in which risk differences were determined were used to assess the effects of the supplemental RPBlend on health parameters and reproduction. The effect size was used as the determinant of the possible contribution of RPblend on the yields of milk, energy corrected milk (ECM), fat and protein yields during the first four weeks of lactation. Results showed that the inclusion of RPblend reduced (P < 0.05) involuntary culling and mastitis by cows during the first 30 days after calving. There was a tendency (P < 0.10) for reduced retained placenta and metritis. The meta-analyses revealed that the risk of the displaced abomasum and milk fever did not diminish (P > 0.10) with the inclusion of the BPBlend. The incidence rate of subclinical ketosis, determined as blood beta hydroxy butyric acid greater than 1.2 mM was lower (P (than those not supplemented. The proportion of cows confirmed pregnant by 100 days in milk was greater for cows given the BPBlend (P < 0.05). Milk yield and ECM were greater for cows receiving the RPBlend (1.13 and 0.93 kg/cow respectively, P < 0.05). There was no change in fat yield (P > 0.10) while the yield of milk protein was greater (P < 0.05) when the cows received the blend. These results suggest that the inclusion of rumen protected B vitamins and choline can assist cow health, reproduction and production at the start of lactation.
文摘Some Pseudomonas syringae pathovars secrete tabtoxin, a monocyclic β-lactam antibiotic, responsible for chlorosis, the principal halo blight symptom in susceptible plants as oats, rye, barley, wheat and sorghum, among other. Here, we demonstrated that the production of tabtoxin in a P. syringae strain increased at least 150%, when choline, betaine or dimethylglycine were used as nitrogen source, or when choline was added as osmoprotectant in hyperosmolar culture media. Besides, we investigated the induction of phosphorylcholine phosphatase (PchP) activity when choline or its metabolites were used as nitrogen sources. PchP is an enzyme involved in Pseudomonas aeruginosa pathogenesis through its contribution to the breakdown of choline-containing compounds of the host cells. Considering these results and that the success of a pathogenic microorganism depends on its ability to survive and proliferate in its target tissue, we propose that choline is one of the plant signals that contribute to establishment of the infection by tabtoxin-producing strains of P. syringae.

文摘AIM: To investigate the normal hepatic magnetic resonance spectroscopy findings choline/lipid2 (Cho/Lip2) associated with age and body mass index (BMI).METHODS: A total of 58 single-voxel proton spectra of the liver were acquired at 3.0 T using the eightchannel phased array abdominal coil as the receiver coil. Consecutive stacks of breath-hold spectra were acquired using the point resolved spectroscopy technique at a short echo time of 30 ms and a repetition time of 1500 ms. The spectra were processed with the SAGE software package. Areas and heights for metabolite resonance were obtained. Student's t test for unpaired data was used for comparisons of shimming, Cho/Lip2, and lipid content. RESULTS: There were significant negative correlations between the Cho/Lip2 peak height ratios and BMI (r=-0.615) and age (r=-0.398) (all P<0.01). Compared with the high-BMI group, the low-BMI group was younger (39.1±13.0 years vs 47.6±8.5 years, t=-2.954,P=0.005); had better water suppression (93.4%±1.4% vs 85.6%±11.6%, t=2.741, P=0.014); had higher Cho/Lip2 peak heights ratio (0.2±0.14 vs 0.05±0.04,t=6.033,P<0.000); and had lower lipid content (0.03±0.08 vs 0.29±0.31, t=-3.309, P=0.004). Compared with the older group, the younger group had better shimming effects (17.1±3.6 Hz vs 22.0±6.8 Hz, t=-2.919, P=0.008); higher Cho/Lip2 peak heights ratios (0.03±0.05vs 0.09±0.12,t=2.4, P=0.020); and lower lipid content (0.05±0.11 vs 0.23±0.32,t=-2.337,P=0.031). Compared with the lowcholine peak group, the high-choline peak group had lower lipid content (0.005±0.002 vs 0.13±0.23, t=-3.796,P<0.000); lower BMI (19.6±2.4vs 23.9±3.0, t=-4.410, P<0.000); and younger age (34.7±10.0 years vs 43.2±12.5 years, t=-2.088, P=0.041). CONCLUSION: Lipid accumulation could result from the increased fat in the body depending on age and BMI. Lipid can mask the resonance signal of choline.

文摘Niobium was electrodeposited on 316 stainless steel bipolar plates of a fuel cell in water and air-stable choline chloride based ionic liquids. The electrochemical corruption property of bipolar plates in simulated PEMFC environment was investigated. It was showed that the plating film was distributed on the surface of 316 stainless steel like isolated islands with height less than 50 nm. The XPS, XRD results showed that a smooth and strong chemical inert film of Nb O and Nb2O5 was formed on the surface of 316 stainless steel. In simulated cathodic condition, the corrosion potential of Nb coated stainless steel was improved by 244 m V, whilst in an anodic condition, it was improved by 105 m V. The current densities for the coated 316 stainless steel were decreased to 2.479 4 μA·cm-2 from 14.810 μA·cm-2 at-0.1 V and to 0.576 μA·cm-2 from 13.417 μA/·cm-2 at 0.6 V, respectively. It was implied that the niobium coating effectively decreased the corrosion rate. The results of the electrochemical tests indicated that the corrosion resistance of stainless steel was greatly improved after coated with niobium.

文摘Phenolic compounds exist in crude oil as pollutants, and their removal is vital important for the refining and further application of oils. In traditional separation approaches, strong acid and strong base have to be used to remove these compounds, which may cause serious environmental problems. In this work, 19 kinds of cholinium ionic liquids have been developed to separate phenol from model oil by liquid–liquid extraction. Structural effect of anions of the ionic liquids in the separation is systematically investigated. It is found that depending on the chemical structure of ionic liquids, phenol can be removed from toluene with single-step removal efficiency from 86 to 99% under optimal conditions. The type of substituent groups and the-CH_2 number between two carboxylates have obvious effect on the removal efficiency, and more hydrophilic ionic liquids have a stronger extraction performance for phenol. Furthermore, thermodynamic,^(13) C NMR,~1 H NMR and density functional theory calculations have been performed to characterize the extraction process and to understand the extraction mechanism. It is shown that the extraction of phenol from oil to ionic liquid is a favorable process, and this process is mainly driven by enthalpy change. The formation of the hydrogen bond between anion of the ionic liquid and-OH of phenol is the main driving force for the extraction of phenol from oil to the ionic liquids.

文摘Solubilities of CO_2, CH_4, H_2, CO and N_2 in choline chloride/urea(ChCl/Urea) were investigated at temperatures ranging from 308.2 to328.2 K and pressures ranging from 0.6 to 4.6 MPa. The results show that the solubilities of gases increase with increasing pressure and decreasing temperature. The solubility of CO_2 is higher than that of CH_4, H_2, CO and N_2, which indicates that ChCl/Urea may be used as a potential solvent for CO_2 capture from the gas mixture. Solubility of CO_2 in ChCl/Urea was fitted by Non-Random Two-Liquid and Redlich-Kwong(NRTL-RK) model, and solubility of CH_4, H_2, CO or N_2 in ChCl/Urea was fitted by Henry's Law. The standard enthalpy, standard Gibbs energy and standard entropy of gases were calculated. Additionally, the CO_2/CH_4 selectivities in water, dry ChCl/Urea and aqueous ChCl/Urea were further discussed.

OBJECTIVE: To observe the effects of GSL combining with choline on learning and memory of AD model rats.DESIGN: Randomized grouping design and controlled animal study.SETTING: Department of Pharmacology, Taishan Medical College. MATERIALS: The experiment was carried out in the Pharmacological Department of Medical College of Jilin University from October 1996 to January 1997. Forty healthy male Wistar rats of clean grade were randomly divided into 5 groups, including sham-injury group, model group, GSL group, choline group and combination group, with 8 rats in each group. Main medications: GSL with the volume more than 92.8% was provided by Department of Chemistry, Norman Bethune Medical College of Jilin University. Panaxatriol, the main component, was detected with thin layer scanning technique and regarded as the index of GSL quality [(55±1)%, CV = 2%, n = 5]. Choline was provided by the Third Shanghai Laboratory Factory. METHODS: 150 nmol quinolinic acid was used to damage bilateral Meynert basal nuclei of adult rats so as to establish AD models. Rats in GSL, choline and combination groups were intragastric administrated with 400 mg/kg GSL, 200 mg/kg choline (20 mL/kg), and both respectively last for 17 days starting from two days before operation. Rats in sham-injury group and model group were perfused with the same volume of distilled water once in each morning for the same days. ① Passive avoidance step-down test: Five minutes later, rats jumped up safe platform when they were shocked with 36 V alternating current. If rats jumped down from the platform and the feet touched railings, the response was wrong. Numbers of wrong response were recorded within 3 minutes, and then the test was redone after 24 hours. ② Morris water-maze spatial localization task: Swimming from jumping-off to platform directly was regarded as right response. Additionally, 4 successively right responses were regarded as the standard. Each rat was trained 10 times a day with 120 s per time for 3 successive days. The interval was 30 s. Three days later, numbers of right response were recorded. The training times were increased to 30 for unlearned rats. ③ Measurement of activity of choilne acetylase in cerebral cortex: Rats were sacrificed at 17 days after operation to obtain cerebral cortex to measure activity of choilne acetylase with radiochemistry technique. ④ Synergistic effect: It was expressed as Q value: Q value = factual incorporative effect/anticipant incorporative effect; Q ≥ 1 was regarded as synergistic effect. Anticipant incorporative effect = (EA+EB-EA·EB), EA and EB were single timing effect, respectively in GSL group and choline group. E (step-down test and Morris water maze test) = (x in model group - factual value in medicine groups)/x in model group; E (activity of choilne acetylase) = (factual value in medicine groups -x in model group)/x in model group. MAIN OUTCOME MEASURES: ① Passive avoidance step-down test and Morris water-maze spatial localization task in the study of learning and memory; ② activity of choilne acetylase. RESULTS: All 40 rats were involved in the final analysis. ① Passive avoidance response: At learning phase on first day and retesting phase on the next day, numbers of wrong responses within 3 minutes were more in model group than sham operation group, and there was significant difference [(5.88±1.46), (2.25±0.87) times; (2.63±1.06), (0.50±0.53) times; P < 0.01]; numbers of wrong responses within 3 minutes were less in combination group than model group, and there was significant difference [learning phase: (1.12±0.83), (5.88±1.46) times; retesting phase: (0.38±0.74), (2.63±1.06) times, P < 0.01]; moreover, effect was stronger than that in GSL group and choline group. The Q value was 1.07 and 1.59, respectively and it showed synergistic effect. ② Spatial localization task: Training times were more in model group than sham operation group, and there was significant difference [(2.9±2.5), (12.6±3.5) times; P < 0.01]. Training times were less in combination group than model group, and there was significant difference [(11.8±2.4), (27.9±2.5) times, P < 0.01]; moreover, effect was stronger than that in GSL group and choline group. The Q value was 1.07 and it showed synergistic effect. ③ Activity of choilne acetylase: Activity was lower in model group than sham operation group, and there was significant difference [(30.56±8.33), (61.11±8.33) nkat/g; P < 0.01]. Activity was higher in combination group than model group and there was significant difference [(50.00±8.33), (30.56±8.33) nkat/g, P < 0.01]; moreover, effect was stronger than that in GSL group and choline group. The Q value was 1.5 and it showed synergistic effect. CONCLUSION: GSL in combination with choline can synergically improve the disorder of learning and memory of AD model rats. Its mechanism may be involved in enhancing the function of central cholinergic system.

文摘A transformation procedure of choline monooxygenase(CMO) gene, involved in stress tolerance, was established in white pine embryogenic tissue by using A. tumefaciens C58/pMP90. The CMO cDNA fragment(1.3 kb) was generated by Reverse Transcription-Polymerase Chain Reaction (RT-PCR) with primers based on the report sequence of CMO in gene bank. A chimerical gene composed of the cauliflower mosaic virus (CaMV) 35S promoter fused to CMO cDNA and β-glucuronidase (GUS-marker gene) was transferred into Ti-derived disarmed binary vector pBI121. The new vector, p35SCMOp, was transferred into Agrobacterium tumefaciens C58/pMP90 by freeze-thaw method. Somatic embryogenesis (SE) initiation of Pinus. Strobus L. and Pinus.Koraiensis Sieb. et Zucc. depended on the manipulation of plant growth regulator (PGR) concentrations in the GLH culture medium. Transgenic embryos and regenerated plants of two Pine species were produced after co-culture of embryogenic tissue with the disarmed strain of A. tumefaciens C58/pMP90/ p35SCMOp and selected on medium containing 25mg/L kanamycin. The transformed embryogenic tissue was initially confirmed by histochemical GUS assay followed by PCR. One copy of T-DNA was detected by transgenic lines analysis in Pinus. Strobus L. and transgenic plants were regenerated for two species using modified protocols for maturation and germination of somatic embryos.

文摘We report a carbon/carbon capacitor based on micro/mesoporous carbon electrodes with cost-effective and eco-friendly aqueous choline bis(trifluoromethylsulfonyl)imide(Ch TFSI)electrolyte with a cosolvent enabling low-temperature operation down to-30℃.For this purpose,a Mg O-templated hierarchical carbon(MP98B)with an average mesopore diameter of 3.5 nm was prepared by pyrolysis of magnesium citrate hydrate at 900℃.To reach lower temperatures,the melting point and viscosity of the aqueous electrolyte were reduced by mixing water(W)with an organic solvent(methanol,M,or isopropanol,I)of high dielectric constant and low viscosity.5 mol kg^(-1)(5 m)Ch TFSI in an optimized volume fraction of cosolvent,M_(0.75)W_(0.25),and I_(0.75)W_(0.25),showed the highest conductivity;the higher conductivity in M_(0.75)W_(0.25)(22.8 and 3.1 m S cm^(-1) at 20 and-30℃,respectively)than in I_(0.75)W_(0.25)(8.5 and0.5 m S cm^(-1)at 20 and-30℃,respectively)is attributed to the lower viscosity of the M_(0.75)W_(0.25)solution.The electrochemical stability window(ESW)of 5 m Ch TFSI in M_(0.75)W_(0.25)and I_(0.75)W_(0.25)(1.6 V)on an MP98B electrode was determined by applying the S-method.Meanwhile,by adjusting the mass ratio of the two electrodes,a MP98B/MP98B capacitor using the 5 m electrolyte in M_(0.75)W_(0.25)could operate with a good life span up to 1.6 V while exhibiting a better charge propagation,greater specific capacitance,and higher specific energy than in I_(0.75)W_(0.25).
文摘The objective of the present study was to exhibit the enhanced water-solubility and in vivo oral absorption when febuxostat(FXT) became the salt formation of choline. The formation of the choline salt of febuxostat was confirmed by X-ray powder diffraction, infrared spectroscopy analysis and differential scanning calorimetry. The direct filling method was used to develop a capsule formulation. Cellactose 80 was used as the filler due to its good fluidity, while cross-linked polyvinylpyrrolidone(PVPP) and magnesium stearate(MS) were used as the disintegrant and lubricant, respectively. Then the in vitro release of the formulation was carried out in five different dissolution media including HCl solution(pH 1.2),acetate buffer(pH 4.5), phosphate buffer(pH 6.8 and pH 7.2) and water. Evident improvement of release for choline febuxostat(CXT) was presented in water while the dissolution degree was decreased for CXT in the medium of phosphate buffer(pH 6.8) in comparison with FXT. Furthermore, the pharmacokinetics of CXT was studied in rats using UPLC-MS/MS compared with FXT. The data acquired illustrated that AUC0-24 h of CXT and FXT were22,245.96 ± 7342.92 μg·h/l and 12,249.70 ± 2024.04 μg·h/l, respectively. The relative bioavailability of CXT to FXT was about 181.6% and the P value of AUC0-24 h was less than 0.05. It showed significant difference between the two drugs after oral administration. In conclusion, the water-solubility and oral bioavailability were both improved remarkably for the choline salt of febuxostat and choline salinization was proved an effective way to increase the in vivo absorption for FXT.
文摘AIM To investigate the repeatability of proton magnetic resonance spectroscopy in the in vivo measurement of human cerebral levels of choline-containing compounds(Cho).METHODS Two consecutive scans were carried out in six healthy resting subjects at a magnetic field strength of 1.5 T. On each occasion, neurospectroscopy data were collected from 64 voxels using the same 2 D chemical shift imaging(CSI) sequence. The data were analyzed in the same way, using the same software, to obtain the values for each voxel of the ratio of Cho to creatine. The Wilcoxon related-samples signed-rank test, coefficient of variation(CV), repeatability coefficient(RC), and intraclass correlation coefficient(ICC) were used to assess the repeatability. RESULTS The CV ranged from 2.75% to 33.99%, while the minimum RC was 5.68%. There was excellent reproducibility, as judged by significant ICC values, in 26 voxels. Just three voxels showed significant differences according to the Wilcoxon related-samples signed-rank test. CONCLUSION It is therefore concluded that when CSI multivoxel proton neurospectroscopy is used to measure cerebral choline-containing compounds at 1.5 T, the reproducibility is highly acceptable.
文摘Choline chloride can increase the activities of Hill reaction and ATPase in isolatedchloroplast from wheat leaves. It can also increase the activities of ribulose-1, 5-bisphosphate car-boxylase (RuBPCase EC 4. 1. 1. 39) and glyceraldehyde-3-phosphate dehydrogenase(G-3-PDHEC 1. 2. 1. 13) with the method of vacuum infiltration. But higher concentrations of choline chlo-ride tends to inhibite the activities of these enzymes. As a result of choline chloride treatment theamounts of both protein and chlorophyll can be increased, and the photosynthetic rate can beraised,too. The above data suggest that the choline chloride may act on the enzymes involved inlight reaction and dark reaction in the photosynthesis.

文摘Green solvents for cellulose dissolution is a key topic for green chemistry,especially natural cellulose with high molecular weight,and there are scarce solvents reported.Deep eutectic solvent(DES)is a typical kind of green solvent that has been attracted much attention recently.Here,high molecular weight natural cellulose(DP>3000)was first isolated from wheat straw and then be directly dissolved in the choline/L-lysine(Ch/Lys)DES.The solution owns excellent stability,and the solubility reaches^5%.Rheological studies revealed that the natural cellulose can be well dispersed in the DES solution and showed gelation at high concentrations.The dissolved cellulose can be regenerated when the dilute acid aqueous solution was added into the solution.It provides an energy conversation and an environmentally friendly route to prepare a cellulose solution,which makes it possible to convert cellulose to valuable chemicals and materials in its homogeneous solution.

文摘The object of this study was to investigate the effects of chromium propionate replacing 25%rumen-protected choline(RPC)on production performance and blood indicators of perinatal dairy cows.According to the principle of ensuring that chromium propionate and RPC were fed 14 days prepartum,27 healthy Holstein cows(age,parity,lactation volume,body condition and expectancy were similar)were randomly divided into three groups(GroupsⅠ,ⅡandⅢ),with nine cows in each group.Cows in GroupⅠwere fed basal diets;cows in GroupⅡwere fed the same basal diets with 10 g•d-1 RPC per cow;and cows in GroupⅢwere fed the same basal diets,but 7.5 g RPC and 2.5 g chromium propionate(4 mg chromium)per cow.The results showed that dry matter intake(DMI)of prepartum in GroupsⅡandⅢwas significantly increased(P<0.05),compared with GroupⅠ;however,there was no significant difference between GroupsⅡandⅢ(P>0.05).The milk fat content in GroupⅢwas significantly higher than that in GroupⅠon the 7th day of postpartum(P<0.05).The urea nitrogen content in GroupsⅡandⅢsignificantly lower than that in GroupⅠon the 21st day of postpartum(P<0.05),but there were no significant difference between GroupsⅡandⅢ(P>0.05).The somatic cell counts in GroupⅢwere significantly lower than those in GroupsⅠandⅡon the 14th and 21st days of postpartum(P<0.05).The contents of the total cholesterol in GroupⅢdecreased significantly compared to GroupⅡon the 7th day of prepartum,on the day of calving and the 7th day of postpartum(P<0.05).The concentration of high-density lipoprotein(HDL)in GroupⅢwas significantly higher than that in GroupⅡon the 7th day of prepartum,on the day of calving and the 7th day of postpartum(P<0.05).On the 7th day of postpartum the concentration of insulin in GroupⅢwas obviously higher than that in GroupⅡ(P<0.05).In summary,replacing 25%of rumen-protected choline with chromium propionate reduced production costs and improved dairy farming benefits.
文摘Many medial septal neurons of the basal forebrain are dependent on nerve growth factor (NGF) from the hippocampus for survival and maintenance of a cholinergic phenotype. When deprived of their source of NGF by axotomy, medial septal neuronal cell bodies atrophy and lose their cholinergic markers. This is similar to what is observed in the basal forebrain during Alzheimer’s disease (AD). In the present study, medial septal neurons were axotomized in female rats by way of a fimbria/fornix lesion. For fourteen days following axotomy, varying NGF doses (1 - 250 μg/ml) were administered to the lateral cerebral ventricle with either mini-osmotic infusion or daily injection. The responsiveness of medial septal neurons was evaluated with choline acetyltransferase immunohistochemistry. Within the mini-osmotic pumps, NGF activity diminished greatly during the first five days of implantation, but increased dramatically in the CSF after five days of infusion. The responsiveness of medial septal neurons to NGF was dose dependent and the ED50 for NGF injection was determined to be 14.08 μg/ml compared to 27.60 μg/ml for NGF infusion. Intermittent injections at varying intervals were evaluated with 30 μg/ml NGF over a fourteen-day time period (2, 3, 6, or 12 injections). No differences occurred in the number of choline acetyltransferase neurons from rats that received weekly injections to those that received daily injections of NGF. NGF administration has been suggested as a therapy for AD. The results of these studies continue to highlight the need for NGF stability within the delivery system and AD patient CSF, the choice of delivery system, frequency of administration, and the NGF dose for maintaining basal forebrain cholinergic neurons during AD.
文摘Weeds are the most limiting factor in soybean production in South Carolina. With early emergence and rapid growth, weeds effectlively compete for water, nutrients, and light resources. The recent evolution of herbicide resistant weeds has made it increasingly difficult for growers to effectively control weeds in soybean. Glyphosate and ALS-resistant Palmer amaranth biotypes have spread rapidly throughout South Carolina, especially in areas where resistance management is lacking. Soybean varieties have been recently developed with tolerance to 2,4-D. Field experiments were conducted at the Clemson University Edisto Research and Education Center located near Blackville, SC in 2012 and 2013 to evaluate selected 2,4-D choline based herbicide programs for weed management in 2,4-D tolerant soybean. Overall, all herbicide treatments were effective in controlling weeds at the POST2 timing. Palmer amaranth control was excellent;however, pitted morningglory was the most difficult. The 2,4-D plus glyphosate pre-mixture provided excellent control for all three weed species with >95% control at POST2 timing. In these treatments, the rate of 2,4-D choline plus glyphosate (1.09 kg ae ha-1 or 1.64 kg ae ha-1) did not have a significant effect on weed control (P = 0.3772). There was a decrease in pitted morningglory control 3 WAP in 2012 vs 2013 in plots treated with S-metolachlor + fomesafen because of a lack of activating soil moisture in 2012. Results from this study showed that all treatments evaluated provided good to excellent control of all 3 weed species. Based on the herbicide programs evaluated in the study, herbicide resistant weeds, such as Palmer amaranth, can be effectively controlled when treated at the correct growth stage.
